Pathology Research and Scholarship

The mode of acquisition and causes for the variable clinical spectrum of coronavirus disease 2019 (COVID-19) remain unknown. We utilized a reverse genetics system to generate a GFP reporter virus to explore severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pathogenesis and a luciferase reporter virus to demonstrate sera collected from SARS and COVID-19 patients exhibited limited cross-CoV neutralization. High-sensitivity RNA in situ mapping revealed the highest angiotensin-converting enzyme 2 (ACE2) expression in the nose with decreasing expression throughout the lower respiratory tract, paralleled by a striking gradient of SARS-CoV-2 infection in proximal (high) versus distal (low) pulmonary epithelial cultures. This dissertation is an ethnographic study which examined the ritual performances of an interconnected Mexican American and Mexican immigrant danza Azteca and curanderismo ceremonial community located in central and northern New Mexico, and central México. This project also explored if and how these rituals recognize the practitioners’ indigeneity. As a Mexican American and Native scholar and ceremonial participant of this community, I provided an “insider’s” understanding of the epistemologies and ontologies that inform these ceremonies. My positionality and methodology acted as a lens to critically examine danzantes’ and promotoras tradicionales’ claims of indigeneity. Importantly, this work provides a fluid conceptualization …

Wonder and awe are often considered to be synonyms. But though these emotions are similar, a more nuanced look at their musical representations reveals that they are not entirely the same. This thesis examines the difference between musical wonder and awe in music from The Lion, The Witch and the Wardrobe, a 2005 film about siblings who find a magical world in a wardrobe. After a review of the literature on musical wonder, the music from two scenes is analyzed. The wardrobe scene, in which the youngest child discovers the land of Narnia, depicts wonder visually. Microalgae are of interest for the creation of sustainable and cost competitive alternatives to petroleum-based fuels and chemicals. However, cultivation, extraction and processing of algal biomass requires improved yields to achieve economic feasibility. The advancement of microalgal biotechnology and various genetic engineering techniques allow the improvement of microalgae biomass for this purpose. Here, the characterization of the overexpression of the native vacuolar H+ pumping pyrophosphate (AVP1) in Picochlorum soloecismus was examined. With the emerging antibiotic resistance, Staphylococcus aureus, primary causative agent of skin and soft tissue infections (SSTIs) as well as other life-threatening conditions has become a global health concern. While factors contributing to host defense against S. aureus SSTIs has been well studied, impact of sex as a variable has not been reported. In Chapter 2, we uncover the sex-specific differences in host response to S. aureus SSTIs and the contribution of female sex hormone, estrogen (E2) in providing protection in females. “Pre-Packaged Sovereignty”: The Fallacy Of Indian Self-Determination In The Bureau Of Indian Affairs (Bia) Tribal Social Services Programs, April Kateri Chavez

This thesis examines the Bureau of Indian Affairs (BIA) tribal social services programs in New Mexico Native reservation communities. I rely on interviews with current/former BIA social workers and administrators to contextualize my analysis, while revealing the limits of existing social work scholarship and offering recommendations for future scholarship and community work. Using critical Indigenous studies and feminisms along with critical social work, I advance two primary arguments. The first is, despite the so-called self-determination era, the Bureau of Indian Affairs (BIA) functions as a settler colonial administration that diminishes tribal sovereignty and perpetuates racist and gendered violence. Staphylococcus aureus is a pathogenic bacterium that inflicts a large amount of human suffering using an arsenal of virulence factors to cause minor to life-threatening infections. Both the accessory gene regulator (agr) operon, a virulence regulatory pathway that controls expression of over 200 virulence factors, and alpha-hemolysin (Hla), a key pore-forming toxin, play a significant role in infection severity. Given the clinical trial failures of S. aureus vaccine candidates and the prevalence of antibiotic-resistant strains, novel and effective treatment options are urgently needed. The Next Generation Radar (NEXRAD) was implemented as one of the first network-wide operational radar- estimated precipitation products in the United States in 1988. The algorithms within this system bring drastic advancements to operational flood forecasting when compared to prior radars and even rain gage systems. The objectives were; (1) to determine how radar plays a role in estimating precipitation by comparing rainfall accuracy to other sources such as calibrated radar data and gage data within Albuquerque, New Mexico and (2) to determine if distance from the radar affects estimation of rainfall. This dissertation explores the deployment of race and gender in comic books and graphic novels, paying close attention to how Black womanhood and girlhood operates in the speculative future. This project suggests that the framing of black womanhood and girlhood in post-apocalyptic/dystopian spaces provide a counter to the normative notions of both while simultaneously using normative tropes of Black womanhood and girlhood to produce new ways of understanding Black femininity in the future. This thesis analyzes Boubacar Boris Diop’s Murambi, le livre des ossements and Véronique Tadjo’s L’Ombre d’Imana: voyages jusqu’au bout du Rwanda. I argue that both authors write trauma by employing both a dominant realist style and the trauma aesthetic with attention to the embodied experiences of genocide victims and survivors in both styles. In doing so, each author contributes to impeding indifference surrounding the 1994 Rwandan genocide. The purpose of this study was to determine whether there is a difference in hemodynamic responses to head-up tilt (HUT) in subjects who do, and do not, experience acute mountain sickness (AMS) during exposure to hypobaric hypoxia. Secondarily, we aimed to determine if those hemodynamic variables altered during HUT correlated with AMS severity. Fifteen participants completed three testing sessions: 1) VO2max test to determine workload at 50% VO2max for hypoxia exposure; 2) HUT test consisting of supine rest for 20 min followed by 70° upright tilting for ≤ 40 min; and 3) six hours of hypobaric …

This dissertation reviews the results from an online survey created to identify and compare how second language (L2) learners and heritage language (HL) learners of Spanish, enrolled in beginning-level coursework at the college level, acquired and built vocabulary. A total of 451 participants completed the survey. The purpose of the online survey was to serve as a baseline for pedagogical purposes, since it provided information about participants’ language profiles and the way they build vocabulary based on sematic relatedness. The Magic Of Love: Love Magic In Medieval Romance, Dalicia Raymond

This project examines authorial representations of the morality of three functions of love magic: to induce, to disrupt, and to facilitate love in twelfth- through fifteenth-century Middle High German, Old French, and Middle English romances. Using a cultural studies approach with close textual analysis and informed by gender studies, it investigates medieval romance authors’ discomfort with love inducing magic and asserts that this discomfort is a response to the magic’s violation of free will, a central tenet of medieval theology. The current study examined the interactive effects of race/ethnicity and sex/gender on cognitive trajectories of older adults. Participants were a total of 5258 non-Hispanic White (NHW), Black, and Hispanic men and women in the Washington/Hamilton Heights/Inwood Columbia Aging Project. Multiple-group latent growth curve modeling was used to examine trajectories across sex/gender by racial/ethnic groups and to determine the relationship between specific socio-cultural/health indicators and cognitive decline, including childhood socioeconomic position, years of education, adult income and occupation, cardiovascular risk factors, and marital status. Despite widespread knowledge that bone marrow-resident breast cancer cells (BMRCs) affect tumor progression, signaling mechanisms of BMRCs implicated in maintaining long-term dormancy have not been characterized. To overcome these hurdles, we developed a new experimental model of clinical dormancy employing patient-isolated Circulating Tumor Cells (de novo CTCs) and their injection in xenografts with subsequent tumor monitoring and CTC characterization (ex vivo CTCs). We hypothesized that significant distinctions exist between signaling pathways of bone marrow-homing vs metastasis-competent CTCs upon transplantation in xenografts. Pathology Research and Scholarship

Maize is a cultigen of global economic importance, but when it first became a staple grain in the Americas, was unknown and contested. Here, we report direct isotopic dietary evidence from 52 radiocarbon-dated human skeletons from two remarkably well-preserved rock-shelter contexts in the Maya Mountains of Belize spanning the past 10,000 years. Individuals dating before ~4700 calendar years before present (cal B.P.) show no clear evidence for the consumption of maize. Evidence for substantial maize consumption (~30% of total diet) appears in some individuals between 4700 and 4000 cal B.P. Isotopic evidence after 4000 cal B.P. indicates that maize became …
