Microbial Metal Tolerance In Bermuda Carbonate Sediments, Mark E. Hines, Galen E. Jones

Jackson Estuarine Laboratory

The recovery of aerobic heterotrophic bacteria from Bermuda carbonate sediments on metal-supplemented media varied as much as 44-fold over a 15-cm depth. Distributional relationships with sulfate-reducing bacteria and sediment character indicated that metal tolerance was a function of metal bioavailability.

Photometric immersion refractometry was used to determine the average apparent refractive index (n) of five types of dormant Bacillus spores representing a 600-fold range in moist-heat resistance determined as a D100 value. The n of a spore type increased as the molecular size of various immersion solutes decreased. For comparison of the spore types, the n of the entire spore and of the isolated integument was determined by use of bovine serum albumin, which is excluded from permeating into them. Morpho-Physiological Factors Affecting Plant Productivity In Bush And Vine Forms Of Winter Squash (Cucurbita Maxima Duch), Cyril Emery Broderick

The productivity of a new bush winter squash cultivar ('Autumn Pride') was compared to that of 'Blue Hubbard', a high-yielding vine cultivar. Yields of 'Autumn Pride' increased with increasing plant density and, at high plant densities, were greater than yields of 'Blue Hubbard' at its near optimum plant density.

Several factors could have contributed to the higher yield potential of 'Autumn Pride'. CO(,2) compensation points were determined to be 38 ppm and 77 ppm CO(,2) for 'Autumn Pride' and 'Blue Hubbard', respectively, at 30(DEGREES)C. 'Autumn Pride' had two layers of palisade parenchyma cells, occupying 74 percent of the mesophyll. In …

A study was undertaken to determine if natural growth irradiance regimes influence the pigment content and/or photosynthesis-irradiance relationships of the tropical brown seaweed, Lobophora variegata. Pigment analyses were performed on samples collected from a series of depths between 9 and 49m. Chlorophyll a, chlorophyll c, and fucoxanthin were constant on a thallus weight basis, but showed small decreases when expressed in terms of thallus area. The beta-carotene content of L. variegata was much greater in shallow than in deep-water samples regardless of the pigment units employed. The degree to which seed crops in seed orchards reflect the genetic variability and superiority present among orchard parents (genetic efficiency) was examined in two loblolly pine seed orchards using seven allozyme loci as genetic markers.

The frequency of outcrossed progeny in the seed crop was estimated using unique markers, a single locus mixed mating model, and a multilocus model. These estimates were 0.982, 0.992, and 0.994, respectively. Thus, selfing did not contribute significantly to deviations from genetic efficiency in these orchards.

Pollen contamination, on the other hand, was found to have a significant impact on genetic efficiency. Unique markers …

Kinetic studies of the hydrogenation of carbon monoxide to methane and methanol were carried out over iron-titanium alloy catalysts, in a continuous flow fixed bed differential reactor. The catalysts studied included FeTi, Fe(,1.5)Ti and Fe(,2.5)Ti alloys with pure Fe used as a comparison. The catalysts used were all unsupported with an averaged initial particle size of 270 microns. The range of feed compositions was varied from 10 to 33% CO with the remainder being H(,2). Temperatures were varied from 520 to 590 K at a total pressure of 34 atm for all the catalysts studied. A Study Of The Thermodynamics Of Anion Binding To Human Serum Transferrin, Donald Allen Folajtar

Electron paramagnetic resonance difference spectroscopy of diferric human serum transferrin indicates that certain inorganic anions alter the electronic environment of the ion centers of the protein by binding to specific anion sites. The binding of these anions (thiocyanate, perchlorate, adenosinetriphosphate, pyrophosphate, and chloride) is shown to occur pairwise with high positive cooperativity, an observation unique to nonstereospecific anion-protein interactions. It is shown that human serum transferrin has binding sites for four nonsynergistic anions, two in each domain. The association constants are reported.

A Characterization Of The Bacteriophage 41c And Its Lytic Cycle In Bacillus Subtilis 168, Denise Maratea

The ultrastructure of the lytic cycle of phage 41c in Bacillus subtilis 168 was examined by transmission electron microscopy. Thin sections of phage 41c-infected cells revealed that, prior to the appearance of phage structures, a marked rearrangement of the tightly packed, vesicular mesosomes was usually seen within the host cells. These mesosomes opened up into loosely arranged, lamellar forms. As phage accumulated, they appeared to be preferentially associated with the mesosomes. Towards the later portion of the lytic cycle, the mesosomes took on distinctive, loosely organized, spiral configurations. These ultrastructural changes occurred even in the presence of chloramphenicol, indicating that …

In a continuous fermentor with 100% recycle, growth of Escherichia coli and Bacillus polymyxa occurs in three sharply distinct phases: an initial phase of exponential growth that terminates when the glucose level in the fermentor falls below transport saturation levels; a second phase of linear growth whose rate is dependent on the rate of glucose provision to the fermentor, but whose length if fixed and independent of growth rate; and a third phase of slower glucose dependent linear growth. The specific growth rate falls in phase 2 and 3 while the mass doubling time lengthen, eventually reaching hundreds of hours. …

This dissertation evaluates the potential of chemiluminescence analysis for two types of clinical applications: (1) rapid analysis with a flow injection system which consumes small reagent volumes (25 (mu)L) and (2) a new approach to chemiluminescence immunoassay.

The first application studied uses the firefly reaction with a flow system to analyze for adenosine triphosphate (ATP). Two flow configurations called the "valve within a valve" and "merging zones" configurations were evaluated with respect to effect of parameters such as flow and sample volume on precision, sensitivity and sample throughput. The purpose of the present study was to investigate the salient factors in teachers' judgements of hyperactivity and conduct problems. A set of thirty eight vignettes was presented to fifty elementary school teachers. Five variables, two levels of each, were systematically varied in the vignettes: gender (male or female), quality of schoolwork (adequate or poor), activity level (medium or high), aggression (non-aggressive or aggressive), and peer acceptance (liked or not-liked). For each vignette, subjects rated the extent they considered the child to be hyperactive (or a conduct problem), how likely they would be to refer the child for evaluation, and …

The MHC of the chicken (B complex) has been shown to exert a decisive influence on the fate of Rous sarcoma virus (RSV)-induced tumors. However, noninbred chickens with identical B genotypes and inbred lines indistinguishable at the MHC have demonstrated considerable variation in response to RSV challenge. The major objective of this research was to determine the cause of variation in anti-Rous sarcoma response observed among chickens with "identical" B genotypes.

Hypotrichs represent a highly diverse species ensemble displaying a wide range of form and function that reflects extensive radiation in a wide variety of habitats. Because of their constancy through time, particular habitats such as marine sands harbor forms that may have persisted with little change for hundreds of millions of years and, hence, may hold clues to help unravel mysteries of ciliate evolution. Using light optical microscopy and both scanning and transmission electron microscopy, I examine cortical morphogenesis through cell division and interphase ultrastructure of several species of marine ciliates.
