Enhancing Soil Conservation Practice Adoption With Targeted Educational Programs, Elbert C. Dickey, David P. Shelton, Paul J. Jasa
Enhancing Soil Conservation Practice Adoption With Targeted Educational Programs, Elbert C. Dickey, David P. Shelton, Paul J. Jasa
Department of Agricultural and Biological Systems Engineering: Faculty Publications
Two independent, but closely related, grant funded educational programs were developed and implemented to reduce soil erosion in selected areas of eastern Nebraska. Traditional extension programming methods as well as other more non-traditional approaches were used extensively to enhance soil conservation practice adoption. In one program, encompassing 220 000 ha (540,000 acre) of cropland, annual soil erosion was reduced by 2.3 million t (2.5 million ton) and annual fuel savings of 1.5 million L (390,000 gal) were achieved through a reduction in the number of tillage operations. In the second project, more than 93 000 m (305,000 ft) of terraces …

Department of Animal Science: Faculty Publications
Two experiments were conducted to test the following hypotheses: 1) exposure of beef heifers to sterile bulls increases the proportion of heifers attaining puberty by 14 mo of age and 2) rate of growth interacts with bull exposure to influence age at puberty in beef heifers. In Exp. I, heifers were assigned to one of two treatments: 1) heifers were exposed to bulls (BE approximately 70-d period of exposure) or 2) heifers were isolated from bulls (NE) and served as controls. In Exp. 11, heifers were assigned to either BE or NE treatments (175-d period of exposure to bulls) and …

Purification And Characterization Of Soybean Root Nodule Ferric Leghemoglobin Reductase, Lin Ji, Stephen Wood, Manuel Becana, Robert V. Klucas
Department of Biochemistry: Faculty Publications
A ferric leghemoglobin reductase from the cytosol of soybean (Glyclne max) root nodules was purified to homogeneity and partlafly characterized. The enzyme is a flavoprotein with flavin adenine dinuclotide as the prosthetic group and consists of two identical subunits, each having a molecular mass of 54 kilodaltons. The pure enzyme shows a high activity for ferric leghemoglobin reduction with NADH as the reductant in the absence of any exogenous mediators. The enzyme also exhibits NADH-dependent 2,6-dichloroindophenol reductase activity. A sequence of the first 50 N-terminal amino acids of the purified protein was obtained. Comparisons with known protein sequences …

Detection Of Mercuric Ions In Water By Elisa With A Mercury- Specific Antibody, Dwane E. Wylie, Larry D. Carlson, Randy Carlson, Fred W. Wagner, Sheldon M. Schuster
Department of Biochemistry: Faculty Publications
An immunoassay that detects mercuric ions in water at concentrations of 0.5 ppb and above is described. The assay utilizes a monoclonal antibody that binds specifically to mercuric ions immobilized in wells of microtiter plates. Within the range of 0.5-10 ppb mercury, the absorbance in the enzyme-linked immunosorbent assay (ELISA) is linear to the log of the mercuric ion concentration. The quantitation of mercury by ELISA correlates closely with results from cold-vapor atomic absorption. Other divalent metal cations do not interfere with the assay, although there is interference in the presence of 1 mM chloride ions. The optimum pH for …

Department of Plant Pathology: Faculty Publications
The proposals to reinstate Xanthomonas citri (ex Hasse 1915) Gabriel et al. 1989 and X. phaseoli (Smith 1897) Gabriel et al. 1989 are examined in terms of conventional criteria for describing new species. We suggest that the descriptions presented are insufficient in terms of modern practice for the purposes of formal classification in the genus Xanthomonas. To create guidelines for future reinstatements, the Judicial Commission of the International Committee on Systematic Bacteriology is requested to arrange for the preparation and promulgation of minimal standards for Xanthomonas. The pathovars proposed, X. campestris pv. aurantifolii Gabriel et al. 1989 …
Fertilization Of Crops With Feedlot Manure, Elbert C. Dickey, Gerald Bodman
Fertilization Of Crops With Feedlot Manure, Elbert C. Dickey, Gerald Bodman
Department of Agricultural and Biological Systems Engineering: Faculty Publications
The application of animal manure to farmland is an appropriate and environmentally sound management practice for most feedlot operators. Land application returns nutrients from manure to the soil and helps build and maintain soil fertility. In addition to containing nitrogen, phosphorus and potassium, manure contains trace "elements such as calcium, magnesium, iron and zinc. Manure has also been shown to improve soil tilth, increase water-holding capacity, lessen wind and water erosion and improve aeration. Land application of manure should be viewed as a means to utilize crop nutrients present in the manure for crop production rather than utilizing the land …

Mechanisms Of Hybrid Sunflower Resistance To The Sunflower Midge (Diptera: Cecidomyiidae), Marc D. Anderson, Gary J. Brewer
Department of Entomology: Faculty Publications
A study was conducted to identify mechanisms of resistance in sunflower to the sunflower midge, Contarinia schulzi Gagné. Infestation ratings, larval counts, and damage ratings were used to quantify midge resistance in seven sunflower hybrids grown at three locations. Six of the seven hybrids possessed midge resistance. Each hybrid was further categorized with respect to infestation resistance, antibiosis, and tolerance. Two bud characteristics, percent open and size, were examined for their contribution to infestation resistance. The percentage of time that buds of each hybrid were open was negatively correlated with infestation (indicating a preference for the open characteristic). Infestations were …

Malaria is the world’s most ravaging infectious disease. Rampant throughout much of the tropics and some of the temperate areas, its numbers beggar the imagination. It threatens a third of the world’s population, presently afflicts hundreds of millions of people, causes several million deaths annually and may generate as many as 92 million new clinical cases each year [ 1, 21]. Its socioeconomic drain is enormous.
The resurgence of this pestilence during the past two decades has stimulated the search for a vaccine but, despite a prodigious research effort and some cautious optimism, an effective vaccine is still far from …

Metals And Organic Compounds In Missouri River Fish In 1988 Boyd County, Nebraska To Kansas City, Missouri, George T. Allen, R. Mark Wilson
United States Fish and Wildlife Service: Publications
We conducted this project to assess the concentrations of natural and anthropogenic contaminants in fish of the Missouri River from the Nebraska/South Dakota border to Kansas City, Missouri. We collected 45 composite samples of eight fish species from nine locations on the river. We were unable to collect all species at all locations, but the collections allowed us to compare concentrations within trophic levels and at different locations. Comparisons in this project are based on concentrations in composites of fish that may have been of different ages. Also, the comparisons of different species should be treated with caution because' different …

The Pangaean Megamonsoon- Evidence From The Upper Triassic Chinle Formation, Colorado Plateau, Russell F. Dubiel, Judith Totman Parrish, J. Michael Parrish, Steven C. Good
United States Geological Survey: Staff Publications
The Upper Triassic Chinle Formation was deposited at an exceptional time in Earth's paleogeographic and paleoclimatic history. During the Triassic, the supercontinent Pangaea was at its greatest size, in terms of both aggregated continental crust and exposed land area. Moreover, the exposed land was divided symmetrically about the paleoequator between the northern and south- ern hemispheres. These conditions were ideal for maximizing monsoonal circulation, as predicted from paleoclimate models. The Chinle was deposited between about 5° to 15° N paleolatitude in the western equatorial region of Pangaea, a key area for documenting the effects of the monsoonal climate. This study …

Uniformed Services University of the Health Sciences
Thirty-two clinical isolates of Shiga-like toxin (SLT)-producing Escherichia coli associated with single cases or outbreaks of bloody diarrhea, hemorrhagic colitis, the hemolytic uremic syndrome, or edema disease of swine were examined for multiple copies of genes belonging to the slt-I or slt-II toxin families. Five of 19 strains that were known to produce SLT-II or to hybridize to slt-II-specific probes by colony blot were found by Southern hybridization to contain two copies of toxin genes related to slt-II. The genes for two toxins closely related to slt-II were cloned from one of the isolates, Escherichia …

Dietary And Parasitological Aanalysis Of Coprolites Recovered From Mummy 5, Ventana Cave, Arizona, Karl J. Reinhard, Richard H. Hevly
Department of Anthropology: Faculty Publications
Four coprolites were excavated with Burial 5 at Ventena Cave, a partially mummified five-year-old child. Two coprolites were granular and dark in color and two were fibrous and light in color. The coprolites are remains of the child's intestinal contents and were submitted for dietary and parasitological analysis. No parasites were found. The fibrous coprolites proved to be remains of highly masticated mesquite pods (Prosopis). The granular coprolites consist of seeds of saguaro cactus (Carnegiea gigantea). Pollen analysis reveals two dietary pollen types, both derived from cactus. No evidence of cultivated plants except for a trace …

Using a cotransformation system to identify chloroplast transformants in Chlamydomonas reinhardtii, we converted histidine-195 of the photosystem H reaction center D1 protein to a tyrosine residue. The mutants were characterized by a reduced quantum efficiency for photosynthetic oxygen evolution, which varied in a pH-dependent manner, a reduced capacity to oxidize artificial donors to photosystem II, and P680+) reduction kinetics (microsecond) that were essentially similar to wild type. In addition, a dark-stable radical was detected by ESR in mutant photosystem II particles but not in wild-type particles. This radical was similar in g value and lineshape to chlorophyll …

Effect Of Starch-Based Corn Rootworm (Coleoptera: Chrysomelidae) Baits On Selected Nontarget Insect Species: Influence Of Semiochemical Composition, Thomas J. Weissling, Lance J. Meinke, Kandy A. Lytle
Department of Entomology: Faculty Publications
Various starch-encapsulated semiochemical-insecticide formulations, developed for potential use in adult corn rootworm (Diabrotica spp.) management programs, were evaluated in the laboratory and field for effectiveness on corn rootworm beetles: a carabid, Harpalus pennsylvanicus DeGeer; and a coccinellid, Coleomegilla maculata lengi Timberlake. Carbaryl was formulated in pregelatinized starch matrices along with Diabroticaspecific semiochemicals. The specific combination of feeding-gustatory stimulants encapsulated within or coating the outside of starch granules significantly influenced effectiveness. Allstarch formulations containing feeding-gustatory stimulants effectively killed Diabrotica virgifera virgifera LeConte adults in laboratory and field bioassays. However, H. pennsylvanicus and C. m. lengi mortality was greatly reduced when …

Ammonia-Labile Bonds In High- And Low-Digestibility Strains Of Switchgrass, J. O. Fritz, K. J. Moore, Kenneth P. Vogel
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
Improvement in the forage quality of switchgrass (Panicum virgatum L.) through phenotypic selection for increased in vitro dry matter digestibility (IVDMD) has been demonstrated. This study tested the hypothesis that genetic improvement of fiber digestibility in switchgrass has been achieved by selection for a strain with a decreased quantity of ammonia-labile bonds. Tissue samples of a high-digestibility (high-IVDMD) and a low-digestibility strain (low-IVDMD) of switchgrass were ammoniated at rates of 0, 10, 20, and 40 g kg-1 dry matter. Fiber composition and in vitro rate and extent of neutral-detergent fiber (NDF) digestion were determined on control and ammoniated …
