In vitro neurovirological studies of viral infectivity or viral gene expression may be confounded by the mulHple neural cell types and/or fibrob last contamination present in early passage cultures prepared from dissociated human central nervous system (eNS) tissue. We have developed highly enriched astrocyte cultures for neurovirological study by culturing in a serum-free defined medium, 816, supplemented with basic fibroblast growth factor (FGF-2). Subculture in this medium selects against fibroblast proliferation and favors sustained proliferation of a highly enriched glial fibrillary acidic protein (GFAP)-positive cell population. Changes in pulsatile secretion of LH, FSH, oestradiol and progesterone were related to the growth phase, early plateau phase and regression phase of the first ovarian dominant follicle of the oestrous cycle in Bos indicus heifers. Relationships between these hormones during the three phases were also investigated. Accurate measurements of episodic ovarian steroid secretion were obtained by catheterizing the caudal vena cava via the lateral saphenous vein; the tip of the catheter was positioned just cranial to the ovarian vein using transrectal ultrasonography. Six natural history institutions contributed video and other images to produce a single multimedia exhibit about famous paleontology sites throughout the United States. In Mesozoic Monsters. Mammals and Magnolias users can view videos of the original excavation of each of the sites and also play computer games relating to each location. This project provides a model for how collaboration among museums can reduce the cost of multimedia exhibits while improving quality and making them available to wider audiences.

Our working hypothesis was that doses of melengestrol acetate (MGA) greater than those typically administered in estrous synchrony regimens would regulate secretion of LH and 17β-estradiol (E2) as endogenous progesterone (P4) does during the midluteal phase of the estrous cycle. We also hypothesized that endogenous P4 from the CL would interact with MGA to further decrease the frequency of LH pulses and E2. This Extension Circular reviews what is known about the effects of rotations, tillage practices, and planting and harvest dates on crop insect management, focusing on major crops in Nebraska.

Before the development of synthetic organic insecticides (pre-DDT era), rotations, tillage practices, planting and harvest dates, and other nonchemical cultural controls were commonly recommended for insect management. Research focused on crop rotation and other cultural practices for insect management. With the development of DDT and later insecticides however, research on nonchemical controls decreased markedly.

The hydrologic cycle controls water available for use. Rainfall, evaporation, runoff, percolation, and transpiration combine to influence the water available at a specific location at a particular point in time. The speed at which water moves among stages in the hydrologic cycle and the amount of time it spends in storage at any stage affects water availability to users. The blue-winged teal is a small dabbling duck that is common in North America and northern South America. The species is highly mobile and has an opportunistic life history strategy. Breeding populations respond to variable wetland conditions in the drought-prone prairie regions of the north-central United States and southern Canada. Extensive habitat loss and degradation has occurred on the prairies and on neotropical wintering areas in recent decades. Renewed interest in the ecology and management of blue-winged teal has resulted from these environmental pressures. The red imported fire ant, Solenopsis invicta Buren, is considered an oil-loving feeder, however, carbohydrates are essential ingredients in the diet of the fire ant also. Comprehensive screening of mono-, di-, and tri-saccharides demonstrated that glucose, fructose, fucose, sucrose, maltose, turanose and raffinose were significant phagostimulants for fire ant workers. It was also found that while D-glucose and L-fucose, the naturally occurring isomers, were active, the opposite diastereomers were not. Any structural modification of the glucose molecule resulted in loss of activity. None of the sugar alcohols evaluated were active. The fire ant is an agricultural pest and the many …

This NebGuide describes how to use the calculation method to estimate the percent of the soil surface that will be covered with crop residue after using residue-disturbing implements and operations. Leaving crop residue on the soil surface is the easiest and most cost-effective method of reducing soil erosion. Research in Nebraska and other midwestern states found that leaving as little as 20 percent of the soil surface covered with crop residue can reduce soil erosion caused by rainfall and flowing water by one-half compared to residue-free conditions. Greater amounts of residue cover will further reduce erosion.

To investigate the effects of orientation of beef ribs in an electromagnetic field and storage time on the prediction of rib composition, 64 ribs (IMPS 103) were scanned (2.5 MHz) in three different orientations: posterior end first (POS), dorsal end first (DOR), and fat side first, blade end down (BLD). Scans were obtained after 1 and 5 d of storage. Scan peak, rib weight, 3/4 fat depth, length, and temperature were used to predict percentage and weight of dissectable and fat-free lean. The hypothesis tested was that 17β-estradiol (E2) would increase amounts of mRNA for α, LHβ, and FSHβ subunits during the follicular phase of the estrous cycle prior to the preovulatory surge of gonadotropins in cows. On Day 16 (Day 0 = estrus) of the estrous cycle, all cows were treated with prostaglandin F2α (PGF2α). The objective of this study was to determine whether there were doses at which the synthetic progestin, norgestomet, could mimic midluteal phase concentrations of progesterone in regulating the secretion of LH and 17β-estradiol in bovine females. Selection for litter size had been practiced for 21 generations and relaxed selection for 13 generations in mice. Three replicates were used with four selection criteria: index of components (ovulation rate and ova success), uterine capacity, litter size, and an unselected control. Especially with selection for litter size and the index relative to the control, number of pups born had increased, and differences also occurred in mating weight. This paper reassesses the areal extent of Mammoth Steppe in Eastern Beringia and discusses some of the ways in which grazing and topography entered into the maintenance of the Mammoth Steppe. Large, generalist grazing mammals, by their grazing style, helped maintain the Mammoth Steppe by removing old-growth and by stimulating grass plants to reproduce vegetatively. These grazers also promoted uniformity in growth-form and uniformity in plant biomass (phytomass) above and below the ground surface. During the late Pleistocene these large, generalist grazers were eliminated from interior Alaska by Paleolndian hunters and their predatory animal companions. The loss of these large, …

Luteolysis in sheep is associated with uterine secretion of pulses of prostaglandin F2α(PGF2α) due to the action of luteal oxytocin on endometrial oxytocin receptors. For pregnancy to become established inhibition of oxytocin receptors is important as an antiluteolytic mechanism. The maternal recognition of pregnancy in cattle and sheep involves production, by the trophoblast, of a type 1 interferon (IFN-τ)that suppresses uterine development of oxytocin receptors and the generation of luteolytic episodes of PGF2α. We present a method to analyze serial time-mortality data from bioassay experiments where successive observations are made on the same group of organisms exposed to 1 concentration of a stimulus (for example, a pesticide). Standard probit techniques are not applicable because such data are correlated. Methods are presented for calculating statistics for the time-mortality line by regressing complementary log-log, logit, or probit transformations of proportion of responders on untransformed time or logarithmic transformations of time; and for calculating confidence limits on lethal time values corresponding to given mortality levels. An Automated Plot Harvest System For Use With A Commercial Forage Harvester, Jeffrey F. Pedersen, Kenneth J. Moore

This automated harvest system is based on minimal modification of currently available commercial forage harvesters. Using 6.8 m plots separated by 0.75 m alleys, a single operator can harvest, finely chop, and collect wet plot weights of forage sorghum [Sorghum bicolor (L.) Moench] or pearl millet [Pennisetum glaucum (L.) R. Hr.] in <1 min per plot. Harvested material is deposited in a pile in the center of each plot. Subsamples for moisture and quality analysis can easily and safely be obtained from the pile.

Concentrations of particulate and dissolved nutrients in Saginaw Bay, Lake Huron, were examined relative to zebra mussel colonization which occurred summer 1991. The magnitude and spatial pattern of changes indicate that mussels had a significant impact on nutrients in Saginaw Bay. Annual means for total suspended solids, particulate organic carbon, particulate phosphorus, and particulate silica in the inner bay were significantly lower in 1992 and 1993 (post-zebra mussel) than in 1991 (pre-zebra mussel).
