Microscopic Traffic Simulation As A Decision Support System For Road Diet And Tactical Urbanism Strategies, Bernice Liu, Amirarsalan Mehrara Molan, Anurag Pande, Jonathan Howard, Serena Alexander, Zhiliang Lou

Faculty and Student Publications

Urban street networks in the United States have been primarily designed for automobile traffic with negligible considerations to non-motorized transportation users. Due to environmental issues and quality of life concerns, communities are reclaiming street spaces for active modes and slowing the speeds in their downtown. Moreover, tactical urbanism, i.e., the use of street space for innovative purposes other than moving automobile traffic, is becoming attractive due to reduced automobile travel demand and the need for outdoor activities in the age of the COVID-19 pandemic. This study provides details of the modeling of an urban downtown network (in the City of …

Assessment Of Evolutionary Relationships For Prioritization Of Myxobacteria For Natural Product Discovery, Andrew Ahearne, Hanan Albataineh, Scot E. Dowd, D. Cole Stevens Jul 2021

Assessment Of Evolutionary Relationships For Prioritization Of Myxobacteria For Natural Product Discovery, Andrew Ahearne, Hanan Albataineh, Scot E. Dowd, D. Cole Stevens

Faculty and Student Publications

Discoveries of novel myxobacteria have started to unveil the potentially vast phylogenetic diversity within the family Myxococcaceae and have brought about an updated approach to myxobacterial classification. While traditional approaches focused on morphology, 16S gene sequences, and biochemistry, modern methods including comparative genomics have provided a more thorough assessment of myxobacterial taxonomy. Herein, we utilize long-read genome sequencing for two myxobacteria previously classified as Archangium primigenium and Chondrococcus macrosporus, as well as four environmental myxobacteria newly isolated for this study. Average nucleotide identity and digital DNA–DNA hybridization scores from comparative genomics suggest previously classified as A. primigenium to instead be …

Transcriptomic Changes And The Roles Of Cannabinoid Receptors And Pparγin Developmental Toxicities Following Exposure To Δ9-Tetrahydrocannabinol And Cannabidiol, Zacharias Pandelides, Neelakanteswar Aluru, Cammi Thornton, Haley E. Watts

Faculty and Student Publications

Human consumption of cannabinoid-containing products during early life or pregnancy is rising. However, information about the molecular mechanisms involved in early life stage Δ9-tetrahydrocannabinol (THC) and cannabidiol (CBD) toxicities is critically lacking. Here, larval zebrafish (Danio rerio) were used to measure THC- and CBD-mediated changes on transcriptome and the roles of cannabinoid receptors (Cnr) 1 and 2 and peroxisome proliferator activator receptor γ(PPARγ) in developmental toxicities. Transcriptomic profiling of 96-h postfertilization (hpf) cnr+/+ embryos exposed (6 - 96 hpf) to 4 μM THC or 0.5 μM CBD showed differential expression of 904 and 1095 genes for THC and CBD, respectively, …

Background: Episodic memory (EM) is particularly sensitive to pathological conditions and aging. In a neurocognitive context, the paired-associate learning (PAL) paradigm, which requires participants to learn and recall associations between stimuli, has been used to measure EM. The present study aimed to explore whether functional near-infrared spectroscopy (fNIRS) can be employed to determine cortical activity underlying encoding and retrieval. Moreover, we examined whether and how different aspects of task (i.e., novelty, difficulty) affects those cortical activities. Methods: Twenty-two male college students (age: M = 20.55, SD = 1.62) underwent a face-name PAL paradigm under 40-channel fNIRS covering fronto-parietal and middle …

Investigations at the Native American site complex of Stark Farms in Mississippi, USA, have yielded numerous examples of metal artifacts of European origin. Our study suggests that they derive from contact between the AD 1540-1541 winter encampment of the Spanish Hernando de Soto expedition and the local Indigenous polity. The artifacts display a wide range of modifications, uses, and depositional contexts congruent with hybrid practices. We argue that the early colonial setting of Stark Farms requires a different perspective on cultural mixing than is often applied in studies of European colonialism. This is highlighted by the strongly improvisational nature of …

On The Move: Sloths And Their Epibionts As Model Mobile Ecosystems, Maya Kaup, Sam Trull, Erik F. Y. Hom Jun 2021

On The Move: Sloths And Their Epibionts As Model Mobile Ecosystems, Maya Kaup, Sam Trull, Erik F. Y. Hom

Faculty and Student Publications

Sloths are unusual mobile ecosystems, containing a high diversity of epibionts living and growing in their fur as they climb slowly through the canopies of tropical forests. These epibionts include poorly studied algae, arthropods, fungi, and bacteria, making sloths likely reservoirs of unexplored biodiversity. This review aims to identify gaps and eliminate misconceptions in our knowledge of sloths and their epibionts, and to identify key questions to stimulate future research into the functions and roles of sloths within a broader ecological and evolutionary context. This review also seeks to position the sloth fur ecosystem as a model for addressing fundamental …

Rage Differentially Altered In Vitro Responses In Vascular Smooth Muscle Cells And Adventitial Fibroblasts In Diabetes-Induced Vascular Calcification, Amber M. Kennon, James A. Stewart Jun 2021

Rage Differentially Altered In Vitro Responses In Vascular Smooth Muscle Cells And Adventitial Fibroblasts In Diabetes-Induced Vascular Calcification, Amber M. Kennon, James A. Stewart

Faculty and Student Publications

The Advanced Glycation End-Products (AGE)/Receptor for AGEs (RAGE) signaling pathway exacerbates diabetes-mediated vascular calcification (VC) in vascular smooth muscle cells (VSMCs). Other cell types are involved in VC, such as adventitial fibroblasts (AFBs). We hope to elucidate some of the mechanisms responsible for differential signaling in diabetes-mediated VC with this work. This work utilizes RAGE knockout animals and in vitro calcification to measure calcification and protein responses. Our calcification data revealed that VSMCs calcification was AGE/RAGE dependent, yet AFBs calcification was not an AGE-mediated RAGE response. Protein expression data showed VSMCs lost their phenotype marker, α-smooth muscle actin, and had …

Agricultural Conservation Practices And Aquatic Ecological Responses, Richard E. Lizotte, Peter C. Smiley, Robert B. Gillespie, Scott S. Knight Jun 2021

Agricultural Conservation Practices And Aquatic Ecological Responses, Richard E. Lizotte, Peter C. Smiley, Robert B. Gillespie, Scott S. Knight

Faculty and Student Publications

Conservation agriculture practices (CAs) have been internationally promoted and used for decades to enhance soil health and mitigate soil loss. An additional benefit of CAs has been mitigation of agricultural runoff impacts on aquatic ecosystems. Countries across the globe have agricultural agencies that provide programs for farmers to implement a variety of CAs. Increasingly there is a need to demonstrate that CAs can provide ecological improvements in aquatic ecosystems. Growing global concerns of lost habitat, biodiversity, and ecosystem services, increased eutrophication and associated harmful algal blooms are expected to intensify with increasing global populations and changing climate. We conducted a …

Rap1a Regulates Cardiac Fibroblast Contraction Of 3d Diabetic Collagen Matrices By Increased Activation Of The Age/Rage Cascade, Stephanie D. Burr, James A. Stewart Jun 2021

Rap1a Regulates Cardiac Fibroblast Contraction Of 3d Diabetic Collagen Matrices By Increased Activation Of The Age/Rage Cascade, Stephanie D. Burr, James A. Stewart

Faculty and Student Publications

Cardiovascular disease is a common diabetic complication that can arise when cardiac fibroblasts transition into myofibroblasts. Myofibroblast transition can be induced by advanced glycated end products (AGEs) present in the extracellular matrix (ECM) activating RAGE (receptor for advanced glycated end products) to elicit intracellular signaling. The levels of AGEs are higher under diabetic conditions due to the hyperglycemic conditions present in diabetics. AGE/RAGE signaling has been shown to alter protein expression and ROS production in cardiac fibroblasts, resulting in changes in cellular function, such as migration and contraction. Recently, a small GTPase, Rap1a, has been identified to overlap the AGE/RAGE …

Since its emergence in the early 1980’s HIV has killed approximately 32 million people, and continues to kill over half a million people every year. Significant research into potential vaccines and cures for HIV has been ongoing for decades but has been largely unsuccessful. One of the more promising technologies that is being investigated to develop an HIV vaccine is recombinant vaccine technology. The study presented in this thesis aims to use simian varicella virus (SVV) and simian immunodeficiency virus (SIV) to act as a model for recombinant vaccine development. If a recombinant SVV-SIV vaccine can be proven safe and …

Greco-Roman Paganism And Women Leaders: The Foundation Of Early Christian Art, Rowan Murry May 2021

Greco-Roman Paganism And Women Leaders: The Foundation Of Early Christian Art, Rowan Murry

Honors Theses

In this thesis, I explore the impact of Greco-Roman pagan motifs as well as women leaders and officials on the development of Early Christian art by analyzing catacomb paintings, sarcophagi, and minor arts such as finger rings and carved gemstones. I also discuss surviving primary sources written by Tertullian, Eusebius, St. Jerome, and Clement of Alexandria, to gain a better understanding of anti-art views in the first few centuries of the Church’s rise to power. These anti-art sentiments were often rooted in attempts to disassociate themselves from pagan practices while Early Christian art was emerging amongst the lower classes who …

Purification Of Pcbp2 And Its Effect On The Formation Of Dna I-Motifs And Secondary Structures, Nathan Redden May 2021

Purification Of Pcbp2 And Its Effect On The Formation Of Dna I-Motifs And Secondary Structures, Nathan Redden

Honors Theses

Several secondary, non-B forms of DNA are associated with the promoter regions of many oncogenes, including the intensely studied c-MYC gene that codes for a transcription factor. Two such secondary structures, G-quadruplexes, and i-motifs, have become topics of interest due to their prevalence within oncogenic promoters, as well as their potential accessibility for drug targeting. B-form DNA can adopt alternative structures under certain conditions. These alternative structures are favored when there is an asymmetric distribution of guanosine and cytosine on complimentary DNA strands. The strand rich in guanosine is able to form a quadruple stranded DNA complex termed a G-quadruplex. …

Chronic Exercise And Memory Interference, Lisa Vogelgesang May 2021

Chronic Exercise And Memory Interference, Lisa Vogelgesang

Honors Theses

This online study examined whether chronic exercise is associated with attenuated memory interference. Sixty-three healthy, young adults completed an interference task (AB/AC-paradigm) and self-reported the number of days and minutes a day they engaged in moderate-to-vigorous physical activity. We found that proactive interference (PI), but not retroactive interference (RI), occurred but none of the exercise modalities significantly impacted PI. Future studies should evaluate whether different interference tasks display diverse sensitivities to exercise-induced changes in memory interference. Moreover, other potential modulating factors, such as the duration and intensity of the exercise should be controlled for.

Analysis Of Multigenerational Behavioral Effects Of Dietary Benzo[A]Pyrene Exposure In Adult Zebrafish, Mary Beth Gillespie May 2021

Analysis Of Multigenerational Behavioral Effects Of Dietary Benzo[A]Pyrene Exposure In Adult Zebrafish, Mary Beth Gillespie

Honors Theses

Benzo[a]pyrene (BaP) is a polycyclic aromatic hydrocarbon that is linked to negative reproductive and developmental effects in humans and animals. Because BaP is carcinogenic, and its continued presence in the environment allows it to be inhaled and ingested, better understanding of the effects of BaP is needed. To determine the behavioral effects of BaP exposure, zebrafish were used as a model. Wild-type zebrafish (5D) underwent two separate 21-day dietary exposures to 2.5 and 25 μg BaP/g fish to compare how BaP exposure affects locomotor activity. Following the dietary exposure, fish were mated to obtain and raise the F1 generation to …
