Animal models of retinal degeneration are critical for understanding disease and testing potential therapies. Inducing degeneration commonly involves the administration of chemicals that kill photoreceptors by disrupting metabolic pathways, signaling pathways, or protein synthesis. While chemically induced degeneration has been demonstrated in a variety of animals (mice, rats, rabbits, felines, 13-lined ground squirrels (13-LGS), pigs, chicks), few studies have used noninvasive high-resolution retinal imaging to monitor the in vivo cellular effects. Here, we used longitudinal scanning light ophthalmoscopy (SLO), optical coherence tomography, and adaptive optics SLO imaging in the euthermic, cone-dominant 13-LGS (46 animals, 52 eyes) to examine retinal structure …

BACKGROUND: Pain and dysfunction persist for most patients following hip-related pain treatment. Additionally, individuals with hip-related pain are typically less physically active than individuals without hip pain, despite evidence that regular physical activity reduces chronic musculoskeletal pain. Poor psychological health is common in patients with hip-related pain and further reinforces low physical activity. Mind-body interventions can improve psychological health and activity levels but have yet to be integrated to provide comprehensive, psychologically informed care for patients with hip-related pain. Thus, we are using the NCCIH intervention development framework to develop Helping Improve PSychological Health (HIPS), a novel, multimodal mind-body intervention …

Ultra‐Fast Finite Element Analysis Of Coreless Axial Flux Permanent Magnet Synchronous Machines, Yaser Chulaee, Dan Ionel May 2024

Ultra‐Fast Finite Element Analysis Of Coreless Axial Flux Permanent Magnet Synchronous Machines, Yaser Chulaee, Dan Ionel

Power and Energy Institute of Kentucky Faculty Publications

Large‐scale design optimisation techniques enable the design of high‐performance electric machines. Electromagnetic 3D finite element analysis (FEA) is typically employed in optimisation studies for accurate analysis of axial flux permanent magnet(AFPM) machines, which require extensive computational resources. To reduce the computational burden, a FEA‐based mathematical method relying on the geometric and magnetic symmetry of coreless AFPM machines is proposed to estimate the machine performance indicators using the least number of FEA solutions, thereby significantly lowering the running time. This method is generally applicable to AFPM machines with low saturation effects and cogging torque as exemplified for a printed circuit board …

Cytokines produced by peripheral T-helper 1/17 cells disproportionately contribute to the inflammation (i.e., metaflammation) that fuels type 2 diabetes (T2D) pathogenesis. Shifts in the nutrient milieu could influence inflammation through changes in T-cell metabolism. We aimed to determine whether changes in glucose utilization alter cytokine profiles in T2D. Peripheral blood mononuclear cells (PBMCs), CD4+ T-cells, and CD4+CD25- T-effector (Teff) cells were isolated from age-matched humans classified by glycemic control and BMI. Cytokines secreted by CD3/CD28-stimulated PBMCs and Teff were measured in supernatants with multiplex cytokine assays and a FLEXMAP-3D. Metabolic activity of stimulated CD4+ T-cells was measured by a Seahorse …

The oxidatively modified proteins showed a strong correlation with the reported changes in brain structure, carbohydrate …

Predicting The Pathway Involvement Of Metabolites Based On Combined Metabolite And Pathway Features, Erik D. Huckvale, Hunter N. B. Moseley May 2024

Predicting The Pathway Involvement Of Metabolites Based On Combined Metabolite And Pathway Features, Erik D. Huckvale, Hunter N. B. Moseley

Markey Cancer Center Faculty Publications

A major limitation of most metabolomics datasets is the sparsity of pathway annotations for detected metabolites. It is common for less than half of the identified metabolites in these datasets to have a known metabolic pathway involvement. Trying to address this limitation, machine learning models have been developed to predict the association of a metabolite with a “pathway category”, as defined by a metabolic knowledge base like KEGG. Past models were implemented as a single binary classifier specific to a single pathway category, requiring a set of binary classifiers for generating the predictions for multiple pathway categories. This past approach …

This paper presents a large-scale multi-objective design optimization for a direct-drive wind turbine generator concept that is based upon an experimentally validated computational model for a small-scale prototype motor of the same type. By integrating an outer reluctance-type rotor and a segmented stator with toroidally wound single-coil modules containing spoke-type PMs, the design optimization aims to minimize losses, active mass, and torque ripple while adhering to a power factor constraint. The AC windings and PMs are positioned in the stator and this concept enhances flux concentration, enabling the use of more affordable high energy non-rare-earth (special type) magnets. The exterior …

Optimization, Characterization, And Comparison Of Two Luciferase-Expressing Mouse Glioblastoma Models, Louis Rodgers, Julia A. Schulz Pauly, Bryan J. Maloney, Anika M. S. Hartz, Bjoern Bauer

UK CARES Faculty Publications

Glioblastoma (GBM) is the most aggressive brain cancer. To model GBM in research, orthotopic brain tumor models, including syngeneic models like GL261 and genetically engineered mouse models like TRP, are used. In longitudinal studies, tumor growth and the treatment response are typically tracked with in vivo imaging, including bioluminescence imaging (BLI), which is quick, cost-effective, and easily quantifiable. However, BLI requires luciferase-tagged cells, and recent studies indicate that the luciferase gene can elicit an immune response, leading to tumor rejection and experimental variation. We sought to optimize the engraftment of two luciferase-expressing GBM models, GL261 Red-FLuc and TRP-mCherry-FLuc, showing differences …

Influence Of Four Veterinary Antibiotics On Constructed Treatment Wetland Nitrogen Transformation, Matthew Russell, Tiffany L. Messer, Deborah A. Report, Richard L. Smith, Shannon L. Bartelt-Hunt, Daniel D. Snow, Ariel P. Reed

UK CARES Faculty Publications

The use of wetlands as a treatment approach for nitrogen in runoff is a common practice in agroecosystems. However, nitrate is not the sole constituent present in agricultural runoff and other biologically active contaminants have the potential to affect nitrate removal efficiency. In this study, the impacts of the combined effects of four common veterinary antibiotics (chlortetracycline, sulfamethazine, lincomycin, monensin) on nitrate-N treatment efficiency in saturated sediments and wetlands were evaluated in a coupled microcosm/mesocosm scale experiment. Veterinary antibiotics were hypothesized to significantly impact nitrogen speciation (e.g., nitrate and ammonium) and nitrogen uptake and transformation processes (e.g., plant uptake and …

Free And Forced Vibrations Of Elastically Restrained Cantilever With Lumped Oscillator, Alireza Babaei, Johné Parker, Paria Moshaver

Earth and Environmental Sciences Faculty Publications

The efficiency assessment of cantilever-based energy harvesters relies on vibrational analysis, which necessitates modi- fications aimed at enhancing efficiency. These modifications involve manipulating the fundamental frequency to lower values and encompassing a wider range of resonances within a specified bandwidth. Consequently, this paper introduces an original analytical-numerical exploration into the vibratory response of a cantilever with a novel boundary condi- tion involving an elastically restrained oscillator-spring arrangement. At the microbeam’s tip, an oscillator is elastically confined by a linear spring, resulting in a novel set of coupled governing equations and a distinct shearing boundary condition. Microbeam equations is derived from …
