Coreless Axial Flux Halbach Array Permanent Magnet Generator Concept For Direct-Drive Wind Turbine, Matin Vatani, Ali Mohammadi, Donovin Lewis, John F. Eastham, Dan M. Ionel

Electrical and Computer Engineering Graduate Research

This paper proposes a concept for a coreless axial flux permanent magnet (CAFPM) generator with a double sided Halbach array rotor for direct-drive offshore wind turbine applications. The application of printed circuit board (PCB) technology is explored as applied to a concentrated winding stator due to its high reliability, repeatability, and fast mass production. A double-sided Halbach array rotor is employed to intensify the airgap flux density and to enable the elimination of the rotor back iron, decreasing the generator’s total active mass. The effect of the Halbach array rotor pole number and magnet-to-magnet (M2M) gap on the airgap flux …

Nucleus-accumbens-associated protein-1 (NAC1) is a cancer-related transcriptional factor encoded by the NACC1 gene, which is amplified and overexpressed in various human cancers and has been appreciated as one of the top potential cancer driver genes. NAC1 has therefore been explored as a potential therapeutic target for managing malignant tumors. Here, we show that NAC1 is a negative regulator of NF-κB signaling, and NAC1 depletion enhances the level of the nuclear NF-κB in human melanoma. Furthermore, the inhibition of NF-κB signaling significantly potentiates the antineoplastic activity of the NAC1 inhibition in both the cultured melanoma cells and xenograft tumors. This study …

Zika virus (ZIKV) is a flavivirus primarily transmitted by Aedes species mosquitoes, first discovered in Africa in 1947, that disseminated through Southeast Asia and the Pacific Islands in the 2000s. The first ZIKV infections in the Americas were identified in 2014, and infections exploded through populations in Brazil and other countries in 2015/16. ZIKV infection during pregnancy can cause severe brain and eye defects in offspring, and infection in adults has been associated with higher risks of Guillain-Barré syndrome. We initiated a study to describe the natural history of Zika (the disease) and the immune response to infection, for which …

Background: Regulatory T cell (Treg) therapy is considered an alternative approach to induce tolerance in transplantation. If successful, this therapy may have implications on immunosuppression minimization/withdrawal to reduce drug-induced toxicity in patients. The aim of this study was to assess the efficacy of the mTORC1/C2 inhibitor, AZD8055, in the manufacturing of clinically competent Treg cells and compare the effects with those induced by rapamycin (RAPA), another mTOR inhibitor commonly used in Treg expansion protocols.

Methods: Primary human Treg cells were isolated from leukapheresis product. Cell viability, expansion rates, suppressive function, autophagy, mitochondrial unfolded protein response (mitoUPR), and cell metabolic profile …

Phosphatase And Pseudo-Phosphatase Functions Of Phosphatase Of Regenerating Liver 3 (Prl-3) Are Insensitive To Divalent Metals In Vitro, Jeffery T. Jolly, Ty C. Cheatham, Jessica S. Blackburn

Markey Cancer Center Faculty Publications

Phosphatase of regenerating liver 3 (PRL-3) is associated with cancer metastasis and has been shown to interact with the cyclin and CBS domain divalent metal cation transport mediator (CNNM) family of proteins to regulate the intracellular concentration of magnesium and other divalent metals. Despite PRL-3’s importance in cancer, factors that regulate PRL-3’s phosphatase activity and its interactions with CNNM proteins remain unknown. Here, we show that divalent metal ions, including magnesium, calcium, and manganese, have no impact on PRL-3’s structure, stability, phosphatase activity, or CNNM binding capacity, indicating that PRL-3 does not act as a metal sensor, despite its interaction …

The Toxin Complex (Tc) superfamily consists of toxin translocases that contribute to the targeting, delivery, and cytotoxicity of certain pathogenic Gram-negative bacteria. Membrane receptor targeting is driven by the A-subunit (TcA), which comprises IgG-like receptor binding domains (RBDs) at the surface. To better understand XptA2, an insect specific TcA secreted by the symbiont X. nematophilus from the intestine of entomopathogenic nematodes, we determined structures by X-ray crystallography and cryo-EM. Contrary to a previous report, XptA2 is pentameric. RBD-B exhibits an indentation from crystal packing that indicates loose association with the shell and a hotspot for possible receptor binding or a …

Innovative Methods To Strengthen Internal Kytc Communications, P. Gayle Marks, Jeff Jasper Aug 2023

Innovative Methods To Strengthen Internal Kytc Communications, P. Gayle Marks, Jeff Jasper

Kentucky Transportation Center Research Report

Communication is the linchpin of every organization. Poor communication leads to inefficiencies, frustration among staff, and employee disaffection and disengagement. Although some state transportation agencies have adopted comprehensive strategic plans for internal communications, many have taken a more piecemeal approach, with some Offices and units (e.g., incident management) developing their own plans that apply only to their operations. The latter approach is less than ideal because establishing a comprehensive communications plan provides an organizing framework for all communications within an organization and is critical for uniting staff around a shared sense of purpose and equipping them with the tools they …

Facilitated Identification Of Kytc Performance Measures, Candice Wallace, Jon Wilcoxson Aug 2023

Facilitated Identification Of Kytc Performance Measures, Candice Wallace, Jon Wilcoxson

Kentucky Transportation Center Research Report

Federal and state government agencies are embracing performance management as a means to (1) justify the use of tax dollars and (2) monitor and report program accomplishments. The Kentucky Transportation Cabinet’s (KYTC) efforts to fold Transportation Performance Management (TPM) into its business processes and workplace culture required training that would educate personnel about TPM’s benefits, objectives, and activities. After reviewing training resources on performance management from FHWA, NHI, AASHTO, and state DOTs, Cabinet leadership concluded that their emphasis on federal requirements was not exactly what KYTC was looking for in a training program. Instead, KYTC wanted a training program that …

Evaluating The Use Of A Near-Miss Reporting Program To Enhance Employee Safety Performance, Gabriel Dadi, Farshid Taherpour, Seth Atkins, Ashtarout Ammar, Jon Wilcoxson

Kentucky Transportation Center Research Report

Using safety incidents and crashes to improve safety performance is an outdated and reactive practice in safety and health programs. One component of a modern approach to safety is a near-miss reporting program. This type of program tracks close calls or near misses that do not result in an incident or crash, but which often can be an early indicator that one may happen. While most construction companies have implemented some methods for reporting near-miss events, many agencies, such as the Kentucky Transportation Cabinet (KYTC), struggle getting employees to report near misses. To fill this gap, potential factors that result …

Friction And Texture Equipment Correlation Study, David Hacker, Kean H. Ashurst Jr., Clark Graves

Kentucky Transportation Center Research Report

The Kentucky Transportation Cabinet’s (KYTC) foremost goal is building and maintaining a safe road network. To accomplish this goal, KYTC needs reliable pavement friction and macrotexture data, both of which influence road performance and safety. KYTC currently measures fiction using a lock-wheel skid tester (AASHTO T 242), however, the agency has not been able to collect data at the network level due to equipment and personnel limitations. In 2020 the Cabinet commissioned a study to collect friction and macrotexture data. Investigators used a sideway-force coefficient routine investigation machine (SCRIM) (AASHTO TP 143) to measure friction. To understand the performance of …

Combined Behavioral And Engineering Approach To Preventing Highway Fatalities, Benjamin Blandford, Reginald Souleyrette, Caitlin N. Pope, Tony Fields, Erin Lammers-Staats

Kentucky Transportation Center Research Report

Traditional, single-discipline highway safety approaches can be augmented through multidisciplinary approaches that consider both engineering and behavioral interventions (e.g., education, enforcement, public outreach campaigns). Leveraging a systems-based conceptual framework of roadway safety, multiple forms of statistical and geospatial analysis, and SPF modeling and network screening, this report proposes and demonstrates methods for unpacking the influence of behavioral-related factors on crash occurrences and outcomes. The primary focus is on behaviors targeted in the Strategic Highway Safety Plan — aggressive driving, distracted driving, impaired driving, and driving without proper restraint (i.e., seatbelts). Based on application of these methods, the report highlights areas …

Organic mixed ionic–electronic conductors (OMIECs) have varied performance requirements across a diverse application space. Chemically doping the OMIEC can be a simple, low-cost approach for adapting performance metrics. However, complex challenges, such as identifying new dopant materials and elucidating design rules, inhibit its realization. Here, these challenges are approached by introducing a new n-dopant, tetrabutylammonium hydroxide (TBA-OH), and identifying a new design consideration underpinning its success. TBA-OH behaves as both a chemical n-dopant and morphology additive in donor acceptor co-polymer naphthodithiophene diimide-based polymer, which serves as an electron transporting material in organic electrochemical transistors (OECTs). The combined effects enhance OECT …

Trichloroethylene (TCE) is a prominent groundwater pollutant due to its stability, widespread contamination, and negative health effects upon human exposure; thus, an immense need exists for enhanced environmental remediation techniques. Temperature-responsive domains and catalyst incorporation in membrane domains bring significant advantages for toxic organic decontamination. In this study, hollow fiber membranes (HFMs) were functionalized with stimuli-responsive poly-N-isopropylacrylamide (PNIPAm), poly-methyl methacrylate (PMMA), and catalytic zero- valent iron/palladium (Fe/Pd) for heightened reductive degradation of such pollutants, utilizing methyl orange (MO) as a model compound. By utilizing PNIPAm’s transition from hydrophilic to hydrophobic expression above the LCST of 32 ◦C, increased pollutant diffusion …

The global water crisis growth has led to a tremendous increase in membrane technology research. Membranes are favored over many other technologies for water treatment because, in principle, they require no chemical additives and can be used isothermally at low temperatures. Membranes that can reject contaminants and salts, produce adequate permeate flux values, and require minimal cleaning are highly demanded. However, most synthesized membranes on the market have associated problems, such as membrane fouling; inverse relationships between flux and solute rejection; and the high cost of synthesis, operation, and maintenance. Therefore, there is a continuied need to produce membranes with …

Perovskite solar cells (PSCs) consisting of interfacial two- and three-dimensional heterostructures that incorporate ammonium ligand intercalation have enabled rapid progress toward the goal of uniting performance with stability. However, as the field continues to seek ever-higher durability, additional tools that avoid progressive ligand intercalation are needed to minimize degradation at high temperatures. We used ammonium ligands that are nonreactive with the bulk of perovskites and investigated a library that varies ligand molecular structure systematically. We found that fluorinated aniliniums offer interfacial passivation and simultaneously minimize reactivity with perovskites. Using this approach, we report a certified quasi–steady-state power-conversion efficiency of 24.09% …

Epithelial-to- mesenchymal transition (EMT) underlies immunosuppression, drug resistance, and metastasis in epithelial malignancies. However, the way in which EMT orchestrates disparate biological processes remains unclear. Here, we identify an EMT- activated vesicular trafficking network that coordinates promigratory focal adhesion dynamics with an immunosuppressive secretory program in lung adenocarcinoma (LUAD). The EMT-activating transcription factor ZEB1 drives exocytotic vesicular trafficking by relieving Rab6A, Rab8A, and guanine nucleotide exchange factors from miR-148a-dependent silencing, thereby facilitating MMP14-dependent focal adhesion turnover in LUAD cells and autotaxin-mediated CD8+ T cell exhaustion, indicating that cell- intrinsic and extrinsic processes are linked through a microRNA that coordinates vesicular …

To improve the charge-carrier transport capabilities of thin-film organic materials, the intermolecular electronic couplings in the material should be maximized. Decreasing intermolecular distance while maintaining proper orbital overlap in highly conjugated aromatic molecules has so far been a successful way to increase electronic coupling. We attempted to decrease the intermolecular distance in this study by synthesizing cocrystals of simple benzoic acid coformers and dipyridyl-2,2′- bithiophene molecules to understand how the coformer identity and pyridine N atom placement affected solid-state properties. We found that with the 5-(3-pyridyl)-5′-(4-pyridyl)-isomer, the 4- pyridyl ring interacted with electrophiles and protons more strongly. Synthesized cocrystal powders …

Factors Affecting Bag Silo Densities And Losses, R. E. Muck, B. J. Holmes

IGC Proceedings (1977-2023)

Bag silos (polyethylene tubes, 30 to 90 m length, 2.4 to 3.7 m diameter, 0.22 mm thick) are used on approximately one-third of the dairy farms in the U.S.A. for making silage, and the level of adoption is increasing rapidly. Unfortunately, almost no research data have been published on these types of silos. Our objective was to measure densities and losses in bag silos at three farms, looking for causes of variation in both.

All possible diastereomeric C9-hydroxymethyl-, hydroxyethyl-, and hydroxypropyl-substituted 5-phenylmorphans were synthesized to explore the three-dimensional space around the C9 substituent in our search for potent MOR partial agonists. These compounds were designed to lessen the lipophilicity observed with their C9-alkenyl substituted relatives. Many of the 12 diastereomers that were obtained were found to have nanomolar or subnanomolar potency in the forskolin-induced cAMP accumulation assay. Almost all these potent compounds were fully efficacious, and three of those chosen for in vivo evaluation, 15, 21, and 36, were all extremely G-protein biased; none of the three compounds recruited beta-arrestin2. Electrical and Computer Engineering Graduate Research

As the smart grid involves more new technologies such as electric vehicles (EVs) and distributed energy resources (DERs), more attention is needed in research to general energy storage (GES) based energy management systems (EMS) that account for all possible load shifting and control strategies, specifically with major appliances that are projected to continue electrification such as the electric water heater (EWH). In this work, a methodology for a modified single-node model of a resistive EWH is proposed with improved internal tank temperature for user comfort modeling and capabilities for conservation voltage reduction (CVR) simulations as well as Energy Star and …
