Adipose tissue lipoprotein lipase (LPL) activity is decreased in patients with poorly controlled diabetes, and this contributes to the dyslipidemia of diabetes. To study the mechanism of this decrease in LPL, we studied adipose tissue LPL expression in male rats with streptozotocin-induced diabetes. Heparin releasable and extractable LPL activity in the epididymal fat decreased by 75-80% in the diabetic group and treatment of the rats with insulin prior to sacrifice reversed this effect. Northern blot analysis indicated no corresponding change in LPL mRNA levels. However, LPL synthetic rate, measured using [35S]methionine pulse labeling, was decreased by 75% in …

Wild and domesticated animals can harbor a pathogenic Escherichia coli strain designated as O157:H7. Potential health problems could occur if strain O157:H7 is a more robust survivor in defecated waste than commonly used indicator bacteria. A laboratory study was conducted to assess E. coli O157:H7 survival relative to a nonpathogenie E. coli strain in two soils with different physical and chemical characteristics. Bacteria in the inoculated soils were enumerated on a weekly basis for 8 wk using a most probable number (MPN) technique. The Soil and Water Assessment Tool (SWAT) was evaluated and parameter sensitivities were determined while modeling daily streamflows in a small central Kentucky watershed over a two-year period. Streamflow data from 1996 were used to calibrate the model and streamflow data from 1995 were used for evaluation. The model adequately predicted the trends in daily streamflow during this period although Nash-Sutcliffe R2 values were –0.04 and 0.19 for 1995 and 1996, respectively. The model poorly predicted the timing of some peak flow values and recession rates during the last half of 1995. Excluding daily peak flow values from August …

This report provides an overview of the water quality problems and associated state and federal programs in the 40 counties that make up the PRIDE region. The 2000 Kentucky 305(b) stream assessment has identified over 1000 miles of impaired stream within the region. The major cause of pollution in the region is related to problems with pathogens. Much of these problems are related to straight pipes and failing septic and wastewater systems. It has been estimated that there are over 35,000 straight pipes and failing septic systems in the PRIDE region. A second major environmental impact in the region is …

A general assessment of the associated environmental problems and programs in the region can be found in the companion report: PRIDE Water Quality Assessment Report I: Problems and Programs while the report PRIDE Water Quality Assessment Report II: Chemical, Biological and Habitat Assessments provides a 10 year baseline assessment of environmental conditions in the region as measured by indicators of pH, fecal coliforms, habitat assessment, and macro-invertebrate assessment.

Experimental trials were carried out to evaluate the technical feasibility of reconditioning overly dry corn and soybeans to optimal market and processing moisture contents. Data obtained from experimental trials were used to validate an aeration simulation model. This model was used to evaluate the feasibility of reconditioning soybeans and corn. Reconditioning of grain was feasible at low airflow rates (0.11 m3 min–1 t–1) over a six-month period when an automatic aeration controller was used. Using downflow aeration and monthly unloading of the bin allowed for the greatest net economic gain. Predicted reconditioning in Des Moines, Iowa, …

An amplification primer and probe which can be used in an in vitro diagnostic test for the presence of S. neurona in equine blood or cerebrospinal fluid. Sarcocystis neurona is responsible for the equine condition of protozoal myelitis. The amplification primer is seventeen nucleotides in length and complementary to a unique section of the small ribosomal subunit of Sarcocystis neurona. The primer encompasses nucleotide positions 1470-1487 of the small ribosomal subunit of S. neurona. The primer has the sequence 5' CCATTCCGGACGCGGGT SEQ ID NO:1.

Compositions containing as the active ingredient a spin-trapping reagent, preferably α-phenyl butyl nitrone (PBN) or spin-trapping derivatives thereof, in a suitable pharmaceutical carrier for administration to a patient are disclosed for treating or preventing symptoms associated with aging or other conditions associated with oxidative tissue damage. Other spin-trapping agents can also be used, such as 5,5-dimethyl pyrroline N-oxide (DMPO) or α-(4-pyridyl 1-oxide)-N-tert-butylnitrone (POBN), and other spin-trapping derivatives thereof. Qualitative transcriptional regulatory sequences functional in plants, plant tissue and in plant cells for inducible gene expression and quantitative transcriptional regulatory sequences for increasing the transcriptional expression of downstream genetic information in plants, plant tissue and plant cells are disclosed. Also disclosed are methods and recombinant DNA molecules for improving the disease resistance of transgenic plants, especially wherein an inducible promoter controls the expression of a protein capable of evoking the hypersensitive response in a plant.

Reconciling Arguments About The Self-Preserving Soul With Evolutionary Theory, James M. Donovan

Advocates of a self-preserving soul must somehow integrate that soul with an organic body which is the product of an evolutionary history. Efforts to resolve this version of the sorites paradox (at what point in the gradual evolution of Homo sapiens was the step taken from the soul-less to the ensouled?) can result in conflict between the two theories. The position is taken here that, given the standard of Popperian verisimilitude, any such conflict signals a weakness in the soul-theory, and not in evolutionism.

Chronic arsenic exposure has been associated with a range of neurologic, vascular, dermatologic, and carcinogenic effects. However, limited research has been directed at the association of arsenic exposure and human reproductive health outcomes. The principal aim of this study was to investigate the trends in infant mortality between two geographic locations in Chile: Antofagasta, which has a well-documented history of arsenic exposure from naturally contaminated water, and Valparaíso, a comparable low-exposure city. The arsenic concentration in Antofagasta's public drinking water supply rose substantially in 1958 with the introduction of a new water source, and remained elevated until 1970. We used …

Disclosed is a chimeric isoprenoid synthase polypeptide including a first domain from a first isoprenoid synthase joined to a second domain from a second, heterologous isoprenoid synthase, whereby the chimeric isoprenoid synthase is capable of catalyzing the production of isoprenoid reaction products that are not produced in the absence of the second domain of the second, heterologous isoprenoid synthase. Increased plasma concentrations of angiotension II (Ang II) have been implicated in atherogenesis. To examine this relationship directly, we infused Ang II or vehicle for 1 month via osmotic minipumps into mature apoE–/– mice. These doses of Ang II did not alter arterial blood pressure, body weight, serum cholesterol concentrations, or distribution of lipoprotein cholesterol. However, Ang II infusions promoted an increased severity of aortic atherosclerotic lesions. These Ang II–induced lesions were predominantly lipid-laden macrophages and lymphocytes; moreover, Ang II promoted a marked increase in the number of macrophages present in the adventitial tissue underlying lesions. Disclosed is a method for preparing a composition that is capable of activating the expression of a gene involved in the synthesis of an isoprenoid, the method involving: (a) contacting a plant cell with an elicitor under conditions that allow an elicitor-induced release of a compound that activates the synthesis of an isoprenoid; and (b) recovering a composition including the compound, wherein the compound is diffusible and has a molecular weight less than or equal to 10,000 daltons. This study presents evidence that phosphoinositide (PI) 3-kinase is involved in T cell Ca(2+) signaling via a phosphatidylinositol 3,4, 5-trisphosphate PI(3,4,5)P(3)-sensitive Ca(2+) entry pathway. First, exogenous PI(3,4,5)P(3) at concentrations close to its physiological levels induces Ca(2+) influx in T cells, whereas PI(3,4)P(2), PI(4, 5)P(2), and PI(3)P have no effect on [Ca(2+)](i). This Ca(2+) entry mechanism is cell type-specific as B cells and a number of cell lines examined do not respond to PI(3,4,5)P(3) stimulation. Second, inhibition of PI 3-kinase by wortmannin and by overexpression of the dominant negative inhibitor Deltap85 suppresses anti-CD3-induced Ca(2+) response, which could be reversed by subsequent …

Combination drying, based on computer simulation, was evaluated as an alternative drying technique to traditional high-temperature drying and dryeration. Simulation models of high-temperature crossflow drying and in-bin drying and conditioning were used to evaluate the performance of conventional crossflow drying and full-heat crossflow drying followed by dryeration or natural-air drying for Indianapolis, Indiana, and Des Moines, Iowa. Energy costs from propane, electricity, moisture shrink below the market moisture content, and dry matter loss were estimated to find the total average drying cost over 29 years. Dryeration and combination drying reduced the total drying cost by approximately 10% compared to conventional …

In some areas of the country, producers store a substantial portion of their forage for winter feeding as silage or haylage. However, hay remains the most popular storage method for forage. Hay stores well for long periods and is better suited to cash sale and transportation than silage. Mechanical conditioning, which gained acceptance during the 1950's is probably still the greatest single change in hay harvesting and storage technology during this century. However, a number of other noteworthy changes and innovations have occurred in recent years which have helped to reduce the extent of losses during hay harvesting and storage.

Intimate partner violence (IPV) is associated with a range of adverse physical health outcomes, including chronic and infectious diseases. An emerging literature suggests that partner violence and specifically sexual violence may be associated with an increased risk of cervical neoplasia. During the fall of 1997 and spring of 1998 about 40 individuals and more than 30 organizations and agencies provided input into the development of a monitoring strategy for the Kentucky River basin under the Kentucky Watershed Management Framework. In addition to the agencies that were able to commit resources to the monitoring effort, citizen input was sought in the design of the strategy. Citizen monitoring efforts are also providing valuable information in many portions of the state. The purpose of this study was to investigate the failures seen in the moisture barrier of fire fighting turnout gear. Moisture barriers taken from garments in field were evaluated to establish a baseline for comparison. Moisture barriers were exposed as part of a three-piece ensemble and as a single layer moisture barrier to instrumental light exposure in a Carbon Arc Fade-ometer and natural sunlight exposure according to AATCC and ASTM test methods. After exposure, moisture barriers were visually examined using stereo and compound microscopes. Heat shock transcription factors (HSFs) function to regulate the expression of heatshock proteins (hsps) or molecular chaperones in the cell. Mammalian cells have twowell-characterized HSFs, HSF1 and HSF2. HSF1 functions to regulate the stress-inducedexpression of hsps. The function of HSF2 appears to be in regulating hsp expressionduring development and differentiation.In this work, I describe two distinct HSF1 mRNA isoforms (HSF1-a and HSF1-b)that are generated by alternative splicing of the HSF1 pre-mRNA. The two HSF1 mRNAisoforms result from the inclusion (HSF1-a), or omission (HSF1-b), of a 66 nucleotideexon of the HSF1 gene, which encodes a 22 amino acid sequence. These results …
