Modulation Of High Affinity Hormone Binding To Lh/Cg Receptor, Meena Sundaramoorthy Jan 2002

Modulation Of High Affinity Hormone Binding To Lh/Cg Receptor, Meena Sundaramoorthy

University of Kentucky Master's Theses

Precise control of physiological phenomena is performed by various kinds of receptormediated signaling. The vast majority of receptors belong to the superfamily of G proteincoupled receptors (GPCRs), which form one of the largest protein families. In theclassical model of GPCR signaling, stimulation of seven transmembrane spanning GPCRleads to the activation of heterotrimeric G proteins, which dissociate into a and bgsubunits. The subunits activate effector molecules, which include second messengergenerating systems, giving rise to various kinds of cellular responses. The LH/CGreceptor is a member of the glycoprotein hormone receptor family along with the FSHand TSH receptors, which belongs to the GPCR …

The Jak-Stat Pathway Is Required For Multiple Early Events In Drosophila Oogenesis, Jennifer Renee Matlock Jan 2002

The Jak-Stat Pathway Is Required For Multiple Early Events In Drosophila Oogenesis, Jennifer Renee Matlock

University of Kentucky Master's Theses

The Janus kinase (JAK) pathway is an integral part of signaling through a variety of ligands and receptors in mammals. The extensive reutilization and pleiotropy of this pathway in vertebrate development is conserved in other animals as well. In Drosophila melanogaster, JAK signaling is involved in embryonic pattern formation, sex determination, larval blood cell development, wing venation, planar polarity in the eye, and formation of other adult structures. Here we describe several roles for JAK signaling in Drosophila oogenesis. The gene for a JAK pathway ligand, unpaired, is expressed specifically in the polar follicle cells, two pairs of somatic cells …

The Efficiacy Of Various Salmonella Intervention Methods Applied To Pork Carcasses During Slaughter, Nathan C. Clayton Jan 2002

The Efficiacy Of Various Salmonella Intervention Methods Applied To Pork Carcasses During Slaughter, Nathan C. Clayton

University of Kentucky Master's Theses

Twenty-one market-age swine slaughtered in the University of Kentuckyabattoir were inoculated with fecal slurry containing two strains of nalidixic acidresistant Salmonella typhimurium on the ham, belly, and jowl regions on eachside of the carcass. Trial 1 revealed that a 10 s hot water spray was just aseffective as the 20 s spray in removing S. typhimurium, as there was nodistinguishable difference in population. The shorter flame singe (10 s) was aseffective as the 20 s application and the two chlorine solutions (100, 200 ppm)had similar results. The 2% lactic acid spray reduced S. typhimurium populationssignificantly more than the 1% treatment. …

Compositional Perspectives On The Exchange Of Muna Slate Wares In The Late And Terminal Classic Northern Maya Lowlands, Christopher M. Gunn Jan 2002

Compositional Perspectives On The Exchange Of Muna Slate Wares In The Late And Terminal Classic Northern Maya Lowlands, Christopher M. Gunn

University of Kentucky Master's Theses

This thesis presents the results of petrographic point counting analyses of Muna Slate ware, thepredominant slipped ceramic ware in the Northern Maya Lowlands during the Late and TerminalClassic Periods (600-1000 A.D.) of Maya prehistory. Recently, it was hypothesized that MunaSlate wares were centrally produced and distributed from the Puuc Hills site of Sayil (Smyth andDore 1994; Smyth et al. 1995). Given that Muna Slate wares may be considered utilitariansubsistence items (sensu Brumfiel and Earle 1987), this suggestion runs counter to severalarguments that ancient Maya utilitarian ceramics production is associated with outlyingcommunities and that their distribution is localized. In the research …

Cognitive And Somatic Item Response Pattern Of Orofacial Pain Patients Compared To Fibromyalgia Patients And A Non-Pain Control Group, Morten Hadsel

University of Kentucky Master's Theses

Previous work has suggested that chronic pain patients report psychological distress through higher endorsement of somatic rather than cognitive signs of anxiety and depression. The present study compared female Non-Pain (11=52), Orofacial Pain (n=3 17) and Fibromyalgia (n=50) groups. on SCL-90-R Somatization, Anxiety and Depression raw scores and cognitive-somatic symptom patterning of the Anxiety and Depression scales. Comparisons were also made amongst orofacial pain diagnostic subgroups and subgroups based on Multidimensional Pain Inventory (MPI) classification groups. The Somatization. Anxiety and Depression scores were higher in the OroFacial Pain and Fibromyalgia than Non-Pain group and higher in the Dysfunctional than Adaptive …

The Place Of Dietary Practices In The Lives Of Older Women, Lisa Marie Curch Jan 2002

The Place Of Dietary Practices In The Lives Of Older Women, Lisa Marie Curch

University of Kentucky Doctoral Dissertations

Studies have supported the benefits of positive dietary behaviors in preventing or reducing morbidity and extending longevity, as well as the psychosocial function of dietary practices for quality of life. Research is needed though on the dietary behaviors of elders in general and older women in particular, for whom gender affects lifelong dietary patterns. Health behavior theory has typically focused on psychological factors, to the neglect of sociocultural processes. This investigation utilized a life course perspective, enhanced by social interactionist elements, to address aspects of development and change in behavior neglected by health behavior theories, such as temporal dimensions and …

Structure And Function Analysis Of Par-4, Nadia M. El-Guendy Jan 2002

Structure And Function Analysis Of Par-4, Nadia M. El-Guendy

University of Kentucky Doctoral Dissertations

Par-4 is a leucine zipper domain protein that induces apoptosis on its own in certain cancer cells and in Ras-transformed cells, but not in normal or immortalized cells. Par-4 induces apoptosis by activation of the Fas death receptor pathway and co-parallel inhibition of NF-B transcription activity. Cells that are resistant to apoptosis by Par-4 alone, however, are greatly sensitized by Par-4 to the action of other pro-apoptotic insults such as growth factor withdrawal, TNF, ionizing radiation, intracellular calcium elevation, or those involved in neuronal degeneration such as Alzheimer's, Parkinson's, Huntington's and Stroke. Previous studies have suggested that the apoptosis-sensitization potential …

Creation Of A Bacterial Mutagenicity Assay Highly Sensitive To Dialkylnitrosamines, Matthew Troy Cooper Jan 2002

Creation Of A Bacterial Mutagenicity Assay Highly Sensitive To Dialkylnitrosamines, Matthew Troy Cooper

University of Kentucky Doctoral Dissertations

Although dialkylnitrosamines are environmentally significant carcinogens, the use of short-term bioassays to assess the mutagenic potential of these compounds remains problematic. The Ames test, a mutagenicity assay based on the reversion of Salmonella typhimurium histidine auxotrophs, is the most widely used bioassay in genetic toxicology, but the traditional Ames tester strains are largely insensitive to dialkylnitrosamine mutagenicity. I have constructed several mutagenicity tester strains that co-express combinations of full-length human cytochrome P450 2E1, rat cytochrome P450 reductase, and human cytochrome b5 in S. typhimurium lacking ogt and ada methyltransferases (YG7104ER, ogt-; and YG7108ER, ogt-, ada-). These new strains are susceptible …

Roles Of The Jak Pathway In Follicular Patterning In Drosophila, Rongwen Xi Jan 2002

Roles Of The Jak Pathway In Follicular Patterning In Drosophila, Rongwen Xi

University of Kentucky Doctoral Dissertations

The JAK-STAT pathway is an intracellular signaling pathway that is found to have crucial roles in hematopoiesis, immune response and the development of many other tissues in mammals. The pathway is conserved in Drosophila melanogaster, and is much simpler: there is only one Drosophila JAK (Hopscotch, Hop) and STAT (STAT92E) respectively, while there are at least 4 JAKs and 7 STATs in mammals. The pathway has been intensively studied in Drosophila, and has been implicated in many tissue development and cellular processes. In this work, I present several roles of JAK signaling in oogenesis.First, JAK signaling is required for cell …

Identification And Characterization Of Contact Sites Between Human Chorionic Gonadotropin And The Amino Terminal Region Of The Luteinizing Hormone/Choriogonadotropin Receptor, Rebecca Mccaffrey Jan 2002

Identification And Characterization Of Contact Sites Between Human Chorionic Gonadotropin And The Amino Terminal Region Of The Luteinizing Hormone/Choriogonadotropin Receptor, Rebecca Mccaffrey

University of Kentucky Master's Theses

The luteinizing hormone / choriogonadotropin receptor (LH/CG-R) is a member of theG protein-coupled receptor family. The LH/CG-R has seven transmembrane helices, threeexoloops, three cytoloops, a C-terminal tail, and an extensive N-terminal exodomain. Theexodomain is capable of binding hormone with high affinity without hormone action. Previousstudies have shown that the amino-terminal region of the LH/CG receptor contacts both subunitsof human chorionic gonadotropin (hCG). In particular, three residues (Leu20, Cys22, and Gly24)were found to be crucial for hormone binding. In this thesis work, benzoylphenylalanine (Bpa),a photoactivatable reagent, was used to continue investigating the interactions of the N-terminalregion of the LH/CG-R with hCG. …

Interacting Color And Behavior Responses To Multiple Selection Pressures In The Sister Salamander Species Ambystoma Barbouri And Ambystoma Texanum., Tiffany Sacra Garcia Jan 2002

Interacting Color And Behavior Responses To Multiple Selection Pressures In The Sister Salamander Species Ambystoma Barbouri And Ambystoma Texanum., Tiffany Sacra Garcia

University of Kentucky Doctoral Dissertations

My research explores the complex strategies animals adapt to cope with multiple selection pressures. I studied the behavioral and color response of two salamander sister species, Ambystoma barbouri and A. texanum, to temperature, predation risk and ultraviolet radiation (UVR, 280-320 nm). Ambystoma barbouri undergo development in streams, while A. texanum larvae inhabit ponds. Thus, A. barbouri are exposed to increased habitat ephemerality, enhanced predation risk, and UVR exposure. I show how A. barbouri have evolved alternate coping mechanisms in response to these environmental factors, relative to A. texanum. In this comparison study, I've quantified the affects of these selection pressures …

Graduate Center for Nutritional Sciences Faculty Patents

The isolation and characterization of cDNAs encoding poly(ADP-ribose) glycohydrolase (PARG) enzymes and the amino acid sequences of PARGs from several species are described. PARG is involved in the cellular response to DNA damage and its proper function is associated with the body's response to neoplastic disorder inducing agents and oxidative stress. Expression vectors containing the cDNAs and cells transformed with the vectors are described. Probes and primers that hybridize with the cDNAs are described. Expression of the cDNA in E. coli results in an enzymatically active protein of about 111 kDa and an active fragment of about 59 kDa. Methods …

Farnesyl Pyrophosphate Analogs, Hans Peter Spielmann, Douglas A. Andres, Kareem A.H. Chehade

Molecular and Cellular Biochemistry Faculty Patents

The post-translational addition of a farnesyl moiety to the Ras oncoprotein is essential for its membrane localization and is required for both its biological activity and ability to induce malignant transformation. The present invention describes design and synthesis of a farnesylpyrophosphate (FPP) analog, 8-anilinogeranyl pyrophosphate (AGPP) that is transferred to Ras by farnesyltransferase (FTase), in which the ω-terminal isoprene unit of the farnesyl group has been replaced with an aniline functionality. AGPP potently inhibited FTase activity in vitro (IC50=0.6 μM) and is highly selective showing little inhibitory activity against either geranylgeranyl-protein transferase type I (GGTase I) (IC50 …

Textualism’S Limits On The Administrative State: Of Isolated Waters, Barking Dogs, And Chevron, Michael P. Healy

Law Faculty Scholarly Articles

In Solid Waste Agency of Northern Cook County (SWANCC) v. U.S. Army Corps of Engineers, the U.S. Supreme Court recently held that the U.S. Army Corps of Engineers (the Corps) does not have authority under the Clean Water Act (the Act or the CWA) to regulate the filling of “other waters.” This decision demonstrates a major shift in the Court's approach to statutory interpretation, particularly in the context of reviewing an agency’s understanding of a statute. The significance of the case is best gauged by contrasting it with United States v. Riverside Bayview Homes, Inc. There, the Court, acting …
