Characterization Of Secretory Sphingomyelinase Activity, Lipoprotein Sphingolipid Content And Ldl Aggregation In Ldlr-/- Mice Fed On A High-Fat Diet, Gergana M. Deevska, Manjula Sunkara, Andrew J. Morris, Mariana N. Nikolova‑Karakashian Oct 2012

Characterization Of Secretory Sphingomyelinase Activity, Lipoprotein Sphingolipid Content And Ldl Aggregation In Ldlr-/- Mice Fed On A High-Fat Diet, Gergana M. Deevska, Manjula Sunkara, Andrew J. Morris, Mariana N. Nikolova‑Karakashian

Physiology Faculty Publications

The propensity of LDLs (low-density lipoproteins) for aggregation and/or oxidation has been linked to their sphingolipid content, specifically the levels of SM (sphingomyelin) and ceramide. To investigate this association in vivo, ldlr (LDL receptor)-null mice (ldlr-/-) were fed on a modified (atherogenic) diet containing saturated fats and cholesterol. The diet led to significantly elevated SM content in all serum lipoproteins. In contrast, ceramide increased only in the LDL particles. MS-based analyses of the lipid acyl chain composition revealed a marked elevation in C16:0 fatty acid in SM and ceramide, consistent with the prevalence of palmitic acid in the modified diet. …

Doxycycline Does Not Influence Established Abdominal Aortic Aneurysms In Angiotensin Ii-Infused Mice, Xiaojie Xie, Hong Lu, Jessica J. Moorleghen, Deborah A. Howatt, Debra L. Rateri, Lisa A. Cassis, Alan Daugherty

Saha Cardiovascular Research Center Faculty Publications

Background: There is no proven medical approach to attenuating expansion and rupture of abdominal aortic aneurysms (AAAs). One approach that is currently being investigated is the use of doxycycline. Despite being primarily used as an antimicrobial drug, doxycycline has been proposed to function in reducing AAA expansion. Doxycycline is effective in reducing the formation in the most commonly used mouse models of AAAs when administered prior to the initiation of the disease. The purpose of the current study was to determine the effects of doxycycline on established AAAs when it was administered at a dose that produces therapeutic serum …

Was There A ‘Race To The Bottom’ After Welfare Reform?, Sarah K. Burns Sep 2012

Was There A ‘Race To The Bottom’ After Welfare Reform?, Sarah K. Burns

University of Kentucky Center for Poverty Research Discussion Paper Series

Leading up to the passage of the 1996 welfare reform, there was much speculation and debate over the possibility that states would "race to the bottom" in setting welfare generosity if given more control over their individual programs. In the fifteen years after welfare reform, did such a race to the bottom ensue? Using a spatial dynamic econometric approach I investigate welfare competition across multiple policy instruments and across three distinct welfare periods - the AFDC regime, the experimental waiver period leading up to the reform, and the TANF era. Results suggest strategic policy setting occurs over multiple dimensions of …

Sex Determination In Beetles: Production Of All Male Progeny By Parental Rnai Knockdown Of Transformer, Jayendra Nath Shukla, Subba Reddy Palli Aug 2012

Sex Determination In Beetles: Production Of All Male Progeny By Parental Rnai Knockdown Of Transformer, Jayendra Nath Shukla, Subba Reddy Palli

Entomology Faculty Publications

Sex in insects is determined by a cascade of regulators ultimately controlling sex-specific splicing of a transcription factor, Doublesex (Dsx). We recently identified homolog of dsx in the red flour beetle, Tribolium castaneum (Tcdsx). Here, we report on the identification and characterization of a regulator of Tcdsx splicing in T. castaneum. Two male-specific and one female-specific isoforms of T. castaneum transformer (Tctra) were identified. RNA interference-aided knockdown of Tctra in pupa or adults caused a change in sex from females to males by diverting the splicing of Tcdsx pre-mRNA to male-specific isoform. All the pupa and adults developed from Tctra …

The Child And Adult Care Food Program And Food Insecurity, Colleen Heflin, Irma Arteaga, Sara Gable

University of Kentucky Center for Poverty Research Discussion Paper Series

High rates of food insecurity are a significant problem in the United States. It is currently estimated that more than 48.8 million people live in food insecure households, meaning that at some time during the previous year, they were unable to acquire or were uncertain of having enough food to meet basic needs due to inadequate household resources (Coleman-Jensen, Nord, Andrews, & Carlson, 2011). Rates of food insecurity are substantially higher among those in households with incomes below the poverty line (40.2%) and in households with children headed by a single woman (35.1%). Levels of food insecurity increased across U.S. …

Decomposing Trends In Income Volatility: The ‘Wild Ride’ At The Top And Bottom, Bradley Hardy, James P. Ziliak Aug 2012

Decomposing Trends In Income Volatility: The ‘Wild Ride’ At The Top And Bottom, Bradley Hardy, James P. Ziliak

University of Kentucky Center for Poverty Research Discussion Paper Series

We provide a detailed accounting of the trend increase in family income volatility in recent decades by quantifying the contributions of household head earnings, spouse earnings, non-transfer non-labor income, transfer income, and tax payments (inclusive of the refundable Earned Income Tax Credit), along with covariances among the income components. Using twoyear matched panels in the Current Population Survey from 1980 to 2009, we find that the volatility of family income, as measured by the variance of the arc percent change, doubled over the past three decades. The increase in volatility was most pronounced among the top 1% of the income …

Keeneland Conference Plenary Sessions: Thomas R. Frieden, Thomas R. Frieden

Frontiers in Public Health Services and Systems Research

One of the leading challenges in public health today is moving forward as one whether we are part of academia, a clinical health setting, or a public health department, in order to improve health outcomes. Right now in the United States, the leading causes of death are caused from diseases that are preventable. We also face a steady rise in health inequalities among those citizens with a lower socioeconomic status. Director of the CDC, Thomas Friedan addressed the Keeneland conference audience on the impact that public health initiatives have had on the health of our nation. Public health interventions have …

Frontiers in Public Health Services and Systems Research

Historically individual and community health problems fed off one another, nonetheless the connection eventually became clear and individual health and population health solutions started to emerge and built off one another. Somewhere along the line, population health and patient health seemed to become divided by a brighter line into 2 separate realms, few of us crossing from one to the otherToday we are looking out for the good of our respective communities. As hospitals move from volume-based payments to value-based payments, they are more concerned about the connection between population health and their own efforts to improve outcomes, care coordination …

Developing Service Delivery Measures For Studies Of Practice Variation: The Mprove Study, Glen P. Mays Jun 2012

Developing Service Delivery Measures For Studies Of Practice Variation: The Mprove Study, Glen P. Mays

Health Management and Policy Presentations

OBJECTIVES: A growing body of evidence indicates that the delivery of public health activities varies widely across states and communities, creating missed opportunities for prevention as well as inequities in health protection. Consequently, HHS recently used a consensus-based process to develop a framework for improving quality in public health and to identify a set of nine priority areas for improvement. As next steps in operationalizing this framework, measures of quality in public health are needed to guide public health improvement initiatives and to support research on the comparative effectiveness of alternative public health strategies. This presentation profiles the methodology developed …

Entomology Faculty Publications

BACKGROUND: Bemisia tabaci (Gennadius) is a phloem-feeding insect poised to become one of the major insect pests in open field and greenhouse production systems throughout the world. The high level of resistance to insecticides is a main factor that hinders continued use of insecticides for suppression of B. tabaci. Despite its prevalence, little is known about B. tabaci at the genome level. To fill this gap, an invasive B. tabaci B biotype was subjected to pyrosequencing-based transcriptome analysis to identify genes and gene networks putatively involved in various physiological and toxicological processes.

METHODOLOGY AND PRINCIPAL FINDINGS: Using Roche 454 pyrosequencing, …

The Use Of High-Solids Loadings In Biomass Pretreatment – A Review, Alicia A. Modenbach, Sue E. Nokes Apr 2012

The Use Of High-Solids Loadings In Biomass Pretreatment – A Review, Alicia A. Modenbach, Sue E. Nokes

Biosystems and Agricultural Engineering Faculty Publications

The use of high‐solids loadings (≥ 15% solids, w/w) in the unit operations of lignocellulose conversion processes potentially offers many advantages over lower‐solids loadings, including increased sugar and ethanol concentrations and decreased production and capital costs. Since the term lignocellulosic materials refers to a wide range of feedstocks (agricultural and forestry residues, distillery by‐products, and dedicated energy crops like grasses), the term “solids loading” here is defined by the amount of dry material that enters the process divided by the total mass of material and water added to the material. The goal of this study is to provide a consolidated …

Local Health Department Provision Of Wic Services Relative To Local “Need”— Examining 3 States And 5 Years, Betty Bekemeier, Matthew Bryan, Matthew D. Dunbar, Chris Fowler Apr 2012

Local Health Department Provision Of Wic Services Relative To Local “Need”— Examining 3 States And 5 Years, Betty Bekemeier, Matthew Bryan, Matthew D. Dunbar, Chris Fowler

Frontiers in Public Health Services and Systems Research

Great variation exists in the nature of LHD service delivery and it varies, in part, relative to jurisdiction population size. Larger LHD jurisdictions may achieve an economy of scale in WIC service delivery that is not matched in smaller areas. Overall, we found that WIC service provision appears relatively consistent across study states and in the presence of increasing need, with greater responsiveness to need in urban areas. As demand for some preventive services increases LHDs in rural areas may need greater support than LHDs in large jurisdictions for meeting local demand. Unlike WIC, LHD-provided services that have less consistently …
