Pretreatment is an essential step in biofuel production from lignocellulose. Disruption of the lignin structure gives enzymes and fermentation organisms access to long chains of cellulose and hemicellulose. For this project’s purposes, the pretreatment must work within the framework of an on-farm butanol bioprocessing facility. Alkaline hydrogen peroxide (AHP) is a delignification method that potentially provides several advantages. At the alkaline pH, powerful hydroxyl radicals are formed; which attack lignin. Coal-fired electricity production is the major emitter of CO2 and other greenhouse gases including NOx and SOx. Microalgae-based CO2 mitigation systems have been proposed to reduce the net CO2 emission from coal-fired power plants. This study focused on developing an optimum culture media and exploring the possibilities for recycling nutrients, which were added as commercial mineralized chemicals at the beginning of cultivation.

My dissertation consists of three essays focused on identifying the strategic responses of governments and individuals following changes in the tax and transfer system. Two essays contribute to the literature on fiscal competition, focusing on state level polices aimed at redistributing income. A third essay contributes to the literature estimating the responsiveness of individual’s incomes to changing marginal tax rates. A better understanding of these responses contributes to our ability to design an optimal tax and transfer system in a federalist nation.

In many chronic inflammatory diseases, patients suffer from skeletal muscle weakness, exacerbating their symptoms. Serum levels of tumor necrosis factor-alpha (TNF) and sphingomyelinase are increased, suggesting their possible role in the progression of this weakness. This dissertation focuses on the role that reactive oxygen species (ROS) and nitric oxide (NO) play in mediating TNF-induced skeletal muscle weakness and to what extent sphingolipid signaling mediates cellular response to TNF.

Breastfeeding is the gold standard of infant feeding and its benefits extend beyond the mother and child. Multiple organizations recommend exclusive breastfeeding for the first six months of an infant’s life. Exclusive breastfeeding rates nationally and in the state of Kentucky fall below the Healthy People 2020 goals. A mother’s intention to breastfeed has been shown to impact actual breastfeeding behavior. The current state of the measurement of intention was explored through a literature review. A majority of the measures were single item scales. The reliability and validity of the scales should be further tested in diverse populations.

Obesity and drug abuse share common neural circuitries including the mesocoticolimbic and striatal dopamine reward system. In the current study, a rat model of diet-induced obesity (DIO) was used to determine striatal dopamine function, impulsivity and motivation as neurobehavioral outcomes and predictors of obesity. For the outcome study, rats were randomly assigned a high-fat (HF) or a low-fat (LF) diet for 8 wk. Following the 8-wk HF-diet exposure, rats were segregated into obesity-prone and obesity-resistant groups based on maximum and minimum body weight gain, respectively, and neurobehavioral outcomes were evaluated. Microneedle (MN) enhanced drug delivery is a safe, effective and efficient enhancement method for delivery of drug molecules across the skin. The “poke (press) and patch” approach employs solid stainless steel MN to permeablize the skin prior to application of a regular drug patch over the treated area. It has been previously shown that MN can be used to deliver naltrexone (NTX) at a rate that provides plasma concentrations in the lower end of the therapeutic range in humans. Chronic ethanol exposure results in neuroadaptations that drive the progression of an alcohol use disorder (AUD). One such driving force is alcohol-induced neurodegeneration. Neuroinflammation has been proposed as a mechanism underlying this damage. Although neuroinflammation is a physiological response to damage, overactivation of its pathways can lead to neurodegeneration. A hallmark indicator of neuroinflammation is microglial activation, but microglial activation is a heterogeneous continuum of phenotypes that can promote or inhibit neuroinflammation. Effects of pharmacotherapies for Attention Deficit/Hyperactivity Disorder (ADHD) on cocaine abuse liability in ADHD are not understood. Spontaneously Hypertensive Rats (SHR), an ADHD model, exhibited greater cocaine self-administration than control Wistar-Kyoto and Wistar rats. Methylphenidate, but not atomoxetine during adolescence enhanced cocaine self-administration in adult SHRs compared to controls. The mesocortical dopaminergic system, including medial prefrontal (mPFC) and orbitofrontal (OFC) cortices, is important for ADHD and cocaine addiction. Dopamine and norepinephrine transporter (DAT and NET) are molecular targets for methylphenidate, atomoxetine and cocaine action.

Maize anthracnose, caused by the fungus Colletotrichum graminicola, is an economically important species contributing to major yield losses. C. graminicola is a hemibiotroph; initially it invades its host while it is alive, and then it switches to destructive necrotrophic growth and the host is killed. Establishment of compatible interactions by biotrophic pathogens is usually associated with suppression of host defenses and cell death, while necrotrophic pathogens typically secrete phytotoxic compounds and induce cell death. Lawsonia intracellularis and the disease it causes in horses, equine proliferative enteropathy (EPE), is an emerging pathogen of increasing importance to the horse industry from both an economic and welfare standpoint. Long recognized as an economically important disease of swine, the hallmark of EPE is a protein-losing enteropathy, where affected horses suffer weight loss and some ultimately succumb to the disease despite aggressive treatment. Poison hemlock, Conium maculatum (Apiaceae), is an invasive plant in North America with a unique toxic chemistry. Previous research on this plant has focused on identifying herbivores as potential biological control agents or describing the toxic plant alkaloids. The goal of this study was to evaluate several sustainable turf maintenance techniques for their potential to increase beneficial insect populations, which could then provide ecosystem services including pest suppression and pollination. The three techniques in question were 1) raising mowing height in commercial and residential lawns, 2) establishing naturalized roughs on golf courses, and 3) creating pollinator refuges on golf courses through the program Operation Pollinator. We found that raising mowing heights did increase populations of some predators such as spiders and staphylinids, but did not increase predation, which was ubiquitously high because ant populations were unaffected by mowing …

Spiders are host to a plethora of heritable endosymbiotic bacteria. Broad-taxa screening studies indicate that endosymbionts are particularly common among spiders, however, little is known about how these bacteria affect their spider hosts. In insects these bacteria ensure vertical transmission by either conveying a benefit to the host or manipulating host reproduction to eliminate males that serve as evolutionary dead-ends for maternally-inherited bacteria. Common modes of reproductive manipulation include parthenogenesis, male killing, feminization, and cytoplasmic incompatibility. Screening an assemblage of Mermessus genus spiders, I detected a high frequency and diversity of endosymbiont infection. Within a single species, M. fradeorum …

Several socio-structural theoretical approaches attempt to explain the gendered division of household labor, but the dyadic process of dividing labor has gone largely unexplored. Therefore, a grounded theory approach was taken with 20 dual-earner married couples to uncover the process of dividing household labor between spouses. The theory that emerged indicated that couples seek to maximize benefits in their distribution of labor, and do so by dividing tasks according to personal preferences and proficiencies. A balanced diet rich in fruits and vegetables has been strongly associated with weight management, improved overall health, and decreased incidence of chronic disease. Availability of locally grown produce is associated with a higher intake of fruits and vegetables; there were 7,864 farmers markets operating in the U.S. in 2012, a 9.6% increase from 2011. The purpose of this study was to evaluate the demographics, nutrition knowledge, and dietary habits of farmers market patrons at ten diverse markets in Kentucky. The media has highlighted a proposed link between a gluten free diet (GFD) and weight loss. However, research related to weight gain and a GFD for persons with celiac disease (CD) has shown the opposite effect. A GFD is the only known treatment for persons with CD. If a patient with CD consumes a diet high in GF processed foods, weight management may be difficult to achieve.

Athletes appear healthy according to weight for height and body fat standards. Though athletes appear to be healthy, little is known about their diets. Research states there is a connection between athletic performance and nutrition. The objective of this study was to determine the diet quality of 138 collegiate athletes. Data were collected in a laboratory. Demographics were collected via survey. Anthropometrics were assessed using a free standing stadiometer and the BodPod® for body fat assessment. This study analyzed the perceptions of Kentucky Homebased Processors and Microprocessors of the benefits of and barriers to developing and selling value-added products. The final sample consisted of 141 participants, 60.5% (n=72) of which were from Central KY, 26.9% (n=32) were from Western KY, and 12.6% (n=15) were from Eastern KY. Overall, participants seemed to feel that their value-added products were successful in many different benefit categories. The primary barriers to developing value-added products were lack of time, lack of funding, and lack of legal knowledge. Eating disorders and disordered eating habits and behaviors were measured using the Eating Disorders Inventory-3 Referral Form (EDI-3 RF). The sample consisted of 159 elite, collegiate student athletes, males and females, aged 18 to 22. Five sports were represented, including basketball, gymnastics, soccer, swimming & diving, and volleyball. Overall, findings support the position that athletes, as a distinct population, and specifically female athletes and those that participate in lean sports, are at an increased risk for disordered eating behaviors and diagnosable eating disorders. The contribution phage display made to our fundamental understanding of how various protective molecules serve to safeguard plants and seeds from herbivores and microbes is discussed. The utility of phage display for directed evolution of enzymes with enhanced capacities to degrade the complex polymers of the cell wall into molecules useful for biofuel production is surveyed. Self-care can be seen as not only critical for individual professionals, but also for the growth of the helping professions and the quality of care which clients receive. The purpose of this study was to investigate use of career-sustaining behaviors and the levels of professional quality of life in music therapy professionals. This study investigated research questions regarding use of career sustaining behaviors and levels of professional quality of life, the relationship between these variables, the differences in the use of career sustaining behavior by demographics, and the use of music as a self-care strategy.

The chemical diversity and biological activities of terpene and terpenoids have served in the development of new flavors, fragrances, medicines and pesticides. While terpenes are made predominantly by plants and microbes in small amounts and as components of complex mixtures, chemical synthesis of terpenes remains technically challenging, costly and inefficient. In this dissertation, methods to create new yeast lines possessing a dispensable mevalonate biosynthetic pathway wherein carbon flux can be diverted to build any chemical class of terpene product are described.
