Tlr4 Signaling Is Involved In Brain Vascular Toxicity Of Pcb153 Bound To Nanoparticles, Bei Zhang, Jeong June Choi, Sung Yong Eum, Sylvia Daunert, Michal Toborek

Graduate Center for Nutritional Sciences Faculty Publications

PCBs bind to environmental particles; however, potential toxicity exhibited by such complexes is not well understood. The aim of the present study is to study the hypothesis that assembling onto nanoparticles can influence the PCB153-induced brain endothelial toxicity via interaction with the toll-like receptor 4 (TLR4). To address this hypothesis, TLR4-deficient and wild type control mice (males, 10 week old) were exposed to PCB153 (5 ng/g body weight) bound to chemically inert silica nanoparticles (PCB153-NPs), PCB153 alone, silica nanoparticles (NPs; diameter, 20 nm), or vehicle. Selected animals were also subjected to 40 min ischemia, followed by a 24 h reperfusion. …

Lrp5 And Lrp6 Exert Overlapping Functions In Osteoblasts During Postnatal Bone Acquisition, Ryan C. Riddle, Cassandra R. Diegel, Julie M. Leslie, Kyle K. Van Koevering, Marie-Claude Faugere, Thomas L. Clemens, Bart O. Williams

Internal Medicine Faculty Publications

The canonical Wnt signaling pathway is critical for skeletal development and maintenance, but the precise roles of the individual Wnt co-receptors, Lrp5 and Lrp6, that enable Wnt signals to be transmitted in osteoblasts remain controversial. In these studies, we used Cre-loxP recombination, in which Cre-expression is driven by the human osteocalcin promoter, to determine the individual contributions of Lrp5 and Lrp6 in postnatal bone acquisition and osteoblast function. Mice selectively lacking either Lrp5 or Lrp6 in mature osteoblasts were born at the expected Mendelian frequency but demonstrated significant reductions in whole-body bone mineral density. Bone architecture measured by microCT revealed …

Entomology Faculty Publications

BACKGROUND: The sweetpotato whitefly, Bemisia tabaci (Hemiptera: Aleyrodidae), is one of the most widely distributed agricultural pests. Although it has developed resistance to many registered insecticides including the neonicotinoid insecticide thiamethoxam, the mechanisms that regulate the resistance are poorly understood. To understand the molecular basis of thiamethoxam resistance, "omics" analyses were carried out to examine differences between resistant and susceptible B. tabaci at both transcriptional and translational levels.

RESULTS: A total of 1,338 mRNAs and 52 proteins were differentially expressed between resistant and susceptible B. tabaci. Among them, 11 transcripts had concurrent transcription and translation profiles. KEGG analysis mapped 318 …

Microbiology, Immunology, and Molecular Genetics Faculty Publications

L. monocytogenes are facultative intracellular bacterial pathogens that cause food borne infections in humans. Very little is known about the gastrointestinal phase of listeriosis due to the lack of a small animal model that closely mimics human disease. This paper describes a novel mouse model for oral transmission of L. monocytogenes. Using this model, mice fed L. monocytogenes-contaminated bread have a discrete phase of gastrointestinal infection, followed by varying degrees of systemic spread in susceptible (BALB/c/By/J) or resistant (C57BL/6) mouse strains. During the later stages of the infection, dissemination to the gall bladder and brain is observed. The …

With Liberty And Access For Some: The Aca's Disconnect For Women's Health, Nicole Huberfeld

Law Faculty Scholarly Articles

This Article will scrutinize the separation of abortion from other aspects of women's health through the vehicle of the Patient Protection and Affordable Care Act (ACA). Part I will examine briefly why the fragmented nature of American healthcare has facilitated the separation of abortion from women's health, despite the fact that abortion is a medically necessary procedure for many women. To that end, this Part will explore the disjointed history of access to medicine juxtaposed against the strangely non-woman-centric nature of the fundamental rights at play in reproductive health. Part II will provide an overview of the ACA to explain …

Keeping Up With Ebooks: Automated Normalization And Access Checking With Normac, Kathryn Lybarger Apr 2013

Keeping Up With Ebooks: Automated Normalization And Access Checking With Normac, Kathryn Lybarger

Library Faculty and Staff Publications

Cataloging ebooks is difficult to do well, as they are often purchased in large collections, sometimes with only low-quality cataloging copy available. MARC records may be provided upfront in a large batch, or trickle in one at a time as they become available. Records may contain links that point nowhere, to the wrong book, or to an offer to sell you the book you already own. Loading records sight unseen may introduce inconsistency or overlay good print records with poor electronic ones, making the catalog much more difficult to search.

This article describes in more detail the major challenges in …

Pd123319 Augments Angiotensin Ii-Induced Abdominal Aortic Aneurysms Through An At2 Receptor-Independent Mechanism, Alan Daugherty, Debra L. Rateri, Deborah A. Howatt, Richard Charnigo, Lisa A. Cassis

Pharmacology and Nutritional Sciences Faculty Publications

BACKGROUND: AT2 receptors have an unclear function on development of abdominal aortic aneurysms (AAAs), although a pharmacological approach using the AT2 receptor antagonist PD123319 has implicated a role. The purpose of the present study was to determine the role of AT2 receptors in AngII-induced AAAs using a combination of genetic and pharmacological approaches. We also defined effects of AT2 receptors in AngII-induced atherosclerosis and thoracic aortic aneurysms.

METHODS AND RESULTS: Male AT2 receptor wild type (AT2 +/y) and deficient (AT2 -/y) mice in an LDL receptor -/- background were fed a saturated-fat enriched diet, and infused with either saline or …

Physics and Astronomy Faculty Publications

The Fe unresolved transition arrays (UTAs) produce prominent features in the ~15-17 Å wavelength range in the spectra of active galactic nuclei (AGNs). Here, we present new calculations of the energies and oscillator strengths of inner-shell lines from Fe XIV, Fe XV, and Fe XVI. These are crucial ions since they are dominant at inflection points in the gas thermal stability curve, and UTA excitation followed by autoionization is an important ionization mechanism for these species. We incorporate these, and data reported in previous papers, into the plasma simulation code Cloudy. This updated physics is subsequently employed to reconsider the …

Community & Leadership Development Faculty Publications

Scotland has one of the most uneven land ownership patterns in the world. In a country of 5.2 million people, about 969 people control 60% of the land. Over 20% of privately owned land in Scotland is held in some form of offshore or beneficial ownership (Committee on the Inquiry of Crofting, 2008). This land ownership pattern has a unique expression in the northern and western parts of the Scottish Highlands and Islands with a 300-year-old system of tenant farmers known as crofters. Unlike other tenant farmers across the world, crofters have gained legal rights to stay on the land …

Prevalence And Risk Factor Analysis Of Resistant Escherichia Coli Urinary Tract Infections In The Emergency Department, Abby M. Bailey, Kyle A. Weant, Stephanie N. Baker

Emergency Medicine Faculty Publications

BACKGROUND: Escherichia coli (E. coli) is a frequent uropathogen in urinary tract infections (UTI). Widespread resistance to sulfamethoxazole-trimethoprim (SMX-TMP) and increasing resistance to fluoroquinolones amongst these isolates has been recognized. There are limited data demonstrating risk factors for resistance to both SMX-TMP and fluoroquinolones.

OBJECTIVE: This study was conducted to assess for the prevalence of community resistance amongst E. coli isolates to SMX-TMP and levofloxacin in ambulatory patients discharged from the emergency department (ED).

METHODS: Adults presenting for evaluation and discharged from the ED with a diagnosis of an E. coli UTI were retrospectively reviewed. Utilizing demographic and clinical data …

Center of Excellence in Rural Health Presentations

Kentucky continues to have the highest lung cancer rate in the nation and is in the top 10 states for heart disease. Despite progress in local smoke-free policies, rural populations remain disproportionately affected by secondhand smoke (SHS) exposure. Due to the longstanding cultural heritage of tobacco, many rural residents think of SHS as a nuisance rather than a serious health hazard.

The aims were to determine the biodistribution, translocation, and persistence of nanoceria in the brain and selected peripheral organs. Nanoceria is being studied as an anti-oxidant therapeutic. Five, 15, 30, or 55 nm ceria was iv infused into rats which were terminated 1, 20, or 720 h later. Cerium was determined in blood, brain, liver, and spleen. Liver and spleen contained a large percentage of the dose, from which there was no significant clearance over 720 h, associated with adverse changes. Very little nanoceria entered brain parenchyma. The results suggest brain delivery of nanoceria will be a challenge.

Osteoblast-Restricted Disruption Of The Growth Hormone Receptor In Mice Results In Sexually Dimorphic Skeletal Phenotypes, Vandana Singhal, Brian C. Goh, Mary L. Bouxsein, Marie-Claude Faugere, Douglas J. Digirolamo

Internal Medicine Faculty Publications

Growth hormone (GH) exerts profound anabolic actions during postnatal skeletal development, in part, through stimulating the production of insulin-like growth factor-1 (IGF-1) in liver and skeletal tissues. To examine the requirement for the GH receptor (GHR) in osteoblast function in bone, we used Cre-LoxP methods to disrupt the GHR from osteoblasts, both in vitro and in vivo. Disruption of GHR from primary calvarial osteoblasts in vitro abolished GH-induced signaling, as assessed by JAK2/STAT5 phosphorylation, and abrogated GH-induced proliferative and anti-apoptotic actions. Osteoblasts lacking GHR exhibited reduced IGF-1-induced Erk and Akt phosphorylation and attenuated IGF-1-induced proliferation and anti-apoptotic action. In …
