A Song Without Music Or Music Without Song: Noise Music And Lexington’S Modern Avant-Garde, Matthew H. Gibson

Kaleidoscope

While Lexington KY appears to be a fairly unassuming college town, it is actually a regional hub for noise, experimental, and improvisational music. This work explores the directions, motivations, and origins of Lexington’s experimental music scene through interviews with its artists, promoters, and participants. What I uncovered was a loose collective of fiercely independent artists who write their own music, create their own visual artwork, and organize their own performances in some of the most unlikely locations. These musicians and artists represent a peculiar contemporary folk art that emerges more from the psyche than the classroom and challenges all artistic …

The implementation of in vitro results to in vivo applications has limitations due to conventional two-dimensional (2D) in vitro conditions lacking the ability to create a physiologically representative model. This study investigated a three-dimensional (3D) cell culture technique to model lung tumors in vitro. A 3D lung cancer model was created by applying collagen (a semi-non-adhesive material) to a transwell, which allowed for nutrient transfer through the collagen. Two lung cancer cells lines (H358, a bronchioalveolar carcinoma and A549, a lung adenocarcinoma) were seeded on top of the collagen. The non-adhesive collagen allowed the cells to preferentially attach to one …

Oswald Physical And Engineering Sciences First Place: An Improved In Vitro Model For The Study Of Endothelial Cells Using Micropatterned Surfaces, Lindsay Gray

Kaleidoscope

Sickle cell anemia, malaria, and cancer are a few of the deadly diseases that utilize blood vessels as a means of migration throughout the body. Adhesion of harmful cells to the endothelial lining of the circulatory system is an integral step in the metastasis of blood borne diseases. As a result of shear stress produced by blood flow through veins and arteries, the endothelium undergoes a distinct morphological change resulting in a more elongated and unidirectional morphology. It has recently been suggested that such changes in cell morphology can affect surface expression profiles, which in turn affects cell-cell binding and …

Planning under uncertainty is a central problem in developing intelligent autonomous systems. The traditional representation for these problems is a Markov Decision Process (MDP). The MDP model can be extended to a Multi-criteria MDP (MMDP) for planning under uncertainty while trying to optimize multiple criteria. However, due to the trade-offs involved in multi criteria problems there may be infinitely many optimal solutions. The focus of this project has been to find a method that efficiently computes a subset of solutions that represents the entire set of optimal solutions for bi-objective MDPs.

Doxorubicin (DOX), one of the most effective anticancer drugs, is known to generate progressive cardiac damage, which is due, in part, to DOX-induced reactive oxygen species (ROS). The elevated ROS often induce oxidative protein modifications that result in alteration of protein functions. This study demonstrates that the level of proteins adducted by 4-hydroxy-2-nonenal (HNE), a lipid peroxidation product, is significantly increased in mouse heart mitochondria after DOX treatment. A redox proteomics method involving two-dimensional electrophoresis followed by mass spectrometry and investigation of protein databases identified several HNE-modified mitochondrial proteins, which were verified by HNE-specific immunoprecipitation in cardiac mitochondria from the …

Understanding Soilless Media Test Results And Their Implications On Nursery And Greenhouse Crop Management, Dewayne L. Ingram Jul 2014

Understanding Soilless Media Test Results And Their Implications On Nursery And Greenhouse Crop Management, Dewayne L. Ingram

Agriculture and Natural Resources Publications

Soilless media, sometimes called growth media or substrates, are one of the most important components of crop production in containers or raised beds with restricted volume. The word media is the plural of medium. The physical and chemical properties of media greatly influence optimal management protocols. Primary components of these media may be peat, pine bark, perlite, vermiculite, compost and/or perhaps coarse sand. Understanding the chemical and physical properties of optimal media, or the chosen medium for a production system, allows more effective system management. Although choosing or formulating media with optimum physical properties (such as pore air space and …

Skeletal muscle is a unique tissue because of its structure and function, which requires specific protocols for tissue collection to obtain optimal results from functional, cellular, molecular, and pathological evaluations. Due to the subtlety of some pathological abnormalities seen in congenital muscle disorders and the potential for fixation to interfere with the recognition of these features, pathological evaluation of frozen muscle is preferable to fixed muscle when evaluating skeletal muscle for congenital muscle disease. Additionally, the potential to produce severe freezing artifacts in muscle requires specific precautions when freezing skeletal muscle for histological examination that are not commonly used when …

Spinal Cord and Brain Injury Research Center Faculty Publications

Alternative activation of microglia/macrophages (M2a) by interleukin (IL)-4 is purported to support intrinsic growth and repair processes after CNS injury. Nonetheless, alternative activation of microglia is poorly understood in vivo, particularly in the context of inflammation, injury, and aging. Here, we show that aged mice (18-19 months) had reduced functional recovery after spinal cord injury (SCI) associated with impaired induction of IL-4 receptor α (IL-4Rα) on microglia. The failure to successfully promote an IL-4/IL-4Rα response in aged mice resulted in attenuated arginase (M2a associated), IL-1β, and chemokine ligand 2 (CCL2) expression, and diminished recruitment of IL-4Rα+ macrophages to …

Activation of the Wnt/β-catenin pathway has been observed in at least 1/3 of hepatocellular carcinomas (HCC), and a significant number of these have mutations in the β-catenin gene. Therefore, effective inhibition of this pathway could provide a novel method to treat HCC. The purposed of this study was to determine whether FH535, which was previously shown to block the β-catenin pathway, could inhibit β-catenin activation of target genes and inhibit proliferation of Liver Cancer Stem Cells (LCSC) and HCC cell lines. Using β-catenin responsive reporter genes, our data indicates that FH535 can inhibit target gene activation by endogenous and exogenously …

Identification Of A Dominant Gene In Medicago Truncatula That Restricts Nodulation By Sinorhizobium Meliloti Strain Rm41, Jinge Liu, Shengming Yang, Qiaolin Zheng, Hongyan Zhu

Plant and Soil Sciences Faculty Publications

BACKGROUND: Leguminous plants are able to form a root nodule symbiosis with nitrogen-fixing soil bacteria called rhizobia. This symbiotic association shows a high level of specificity. Beyond the specificity for the legume family, individual legume species/genotypes can only interact with certain restricted group of bacterial species or strains. Specificity in this system is regulated by complex signal exchange between the two symbiotic partners and thus multiple genetic mechanisms could be involved in the recognition process. Knowledge of the molecular mechanisms controlling symbiotic specificity could enable genetic improvement of legume nitrogen fixation, and may also reveal the possible mechanisms that restrict …

In the pSiM24 vector, the size of the backbone of the early binary vector pKYLXM24 (GenBank Accession No. HM036220; a derivative of pKYLX71) was reduced from 12.8 kb to 7.1 kb. The binary vector pSiM24 is composed of the following genetic elements: left and right T-DNA borders, a modified full-length transcript promoter (M24) of Mirabilis mosaic virus with duplicated enhancer domains, three multiple cloning sites, a 3'rbcsE9 terminator, replication functions for Escherichia coli (ColE1) and Agrobacterium tumefaciens (pRK2-OriV) and the replicase trfA …

Numerical Investigation Of Pyrolysis Gas Blowing Pattern And Thermal Response Using Orthotropic Charring Ablative Material, Haoyue Weng, Alexandre Martin Jun 2014

Numerical Investigation Of Pyrolysis Gas Blowing Pattern And Thermal Response Using Orthotropic Charring Ablative Material, Haoyue Weng, Alexandre Martin

Mechanical Engineering Faculty Publications

An orthotropic material model is implemented in a three-dimensional material response code, and numerically studied for charring ablative material. Model comparison is performed using an iso-Q sample geometry. The comparison is presented using pyrolysis gas streamlines and time series of temperature at selected virtual thermocouples. Results show that orthotropic permeability affects both pyrolysis gas flow and thermal response, but orthotropic thermal conductivity essentially changes the thermal performance of the material. The effect of orthotropic properties may have practical use such that the material performance can be manipulated by altering the angle of orthotropic orientation.

Arrhythmogenic Calmodulin Mutations Disrupt Intracellular Cardiomyocyte Ca2+ Regulation By Distinct Mechanisms, Guo Yin, Faisal Hassan, Ayman R. Haroun, Lisa L. Murphy, Lia Crotti, Peter J. Schwartz, Alfred L. George, Jonathan Satin

Physiology Faculty Publications

BACKGROUND: Calmodulin (CaM) mutations have been identified recently in subjects with congenital long QT syndrome (LQTS) or catecholaminergic polymorphic ventricular tachycardia (CPVT), but the mechanisms responsible for these divergent arrhythmia-susceptibility syndromes in this context are unknown. We tested the hypothesis that LQTS-associated CaM mutants disrupt Ca2+ homeostasis in developing cardiomyocytes possibly by affecting either late Na current or Ca2+-dependent inactivation of L-type Ca2+ current.

METHODS AND RESULTS: We coexpressed CaM mutants with the human cardiac Na channel (NaV1.5) in tsA201 cells, and we used mammalian fetal ventricular cardiomyocytes to investigate LQTS- and CPVT-associated CaM …
