Comparative Effects Of Event Detection Methods On The Analysis And Interpretation Of Ca2+ Imaging Data, Austin Neugornet, Bernadette O’Donovan, Pavel I. Ortinski

Calcium imaging has gained substantial popularity as a tool to profile the activity of multiple simultaneously active cells at high spatiotemporal resolution. Among the diverse approaches to processing of Ca2+ imaging data is an often subjective decision of how to quantify baseline fluorescence or F0. We examine the effect of popular F0 determination methods on the interpretation of neuronal and astrocyte activity in a single dataset of rats trained to self-administer intravenous infusions of cocaine and compare them with an F0-independent wavelet ridgewalking event detection approach. We find that the choice of the processing …

Colon adenocarcinoma is a common cause of cancer-related deaths worldwide. Epithelial-mesenchymal transition is a major regulator of cancer metastasis, and increased understanding of this process is essential to improve patient outcomes. Long non-coding RNA (lncRNA) are important regulators of carcinogenesis. To identify lncRNAs associated with colon carcinogenesis, we performed an exploratory differential gene expression analysis comparing paired colon adenocarcinoma and normal colon epithelium using an RNA-sequencing data set. This analysis identified lncRNA ZFAS1 as significantly increased in colon cancer compared to normal colon epithelium. This finding was validated in an institutional cohort using laser capture microdissection. ZFAS1 was also found …

OBJECTIVE: To determine whether assessment tools for non-randomised studies (NRS) address critical elements that influence the validity of NRS findings for comparative safety and effectiveness of medications.

DESIGN: Systematic review and Delphi survey.

DATA SOURCES: We searched PubMed, Embase, Google, bibliographies of reviews and websites of influential organisations from inception to November 2019. In parallel, we conducted a Delphi survey among the International Society for Pharmacoepidemiology Comparative Effectiveness Research Special Interest Group to identify key methodological challenges for NRS of medications. We created a framework consisting of the reported methodological challenges to evaluate the selected NRS tools.

STUDY SELECTION: Checklists …

Astrocytes with intracellular accumulations of misfolded phosphorylated tau protein have been observed in advanced-stage chronic traumatic encephalopathy (CTE) and in other neurodegenerative conditions. There is a growing awareness that astrocytic tau inclusions are also relatively common in the brains of persons over 70 years of age-affecting approximately one-third of autopsied individuals. The pathologic hallmarks of aging-related tau astrogliopathy (ARTAG) include phosphorylated tau protein within thorn-shaped astrocytes (TSA) in subpial, subependymal, perivascular, and white matter regions, whereas granular-fuzzy astrocytes are often seen in gray matter. CTE and ARTAG share molecular and histopathologic characteristics, suggesting that trauma-related mechanism(s) may predispose to the …

Nutraceutical Intervention With Colostrum Replacer: Can We Reduce Disease Hazard, Ameliorate Disease Severity, And Improve Performance In Preweaned Dairy Calves?, Melissa C. Cantor, D. L. Renaud, Joao H. C. Costa Mar 2021

Nutraceutical Intervention With Colostrum Replacer: Can We Reduce Disease Hazard, Ameliorate Disease Severity, And Improve Performance In Preweaned Dairy Calves?, Melissa C. Cantor, D. L. Renaud, Joao H. C. Costa

The objective of this randomized clinical trial was to determine if an intervention with colostrum replacer (CR; Premolac Plus, Zinpro; 125 g/d fed for 3 d) or a placebo milk replacer (Cows Match, Land O'Lakes Inc.; 125 g/d for 3 d) following a triggered alarm could ameliorate disease bouts in dairy calves. The alarm was set to detect negative deviations of milk intake (20% reduction) or drinking speed (30% reduction) in relation to a calf's 12-d rolling average feeding behavior. Calves were enrolled on this study (n = 42 CR, n = 42 placebo) when they triggered an alarm from …

Educating and training a multisectoral food systems workforce is a critical part of developing sustainable, resilient, and healthy food and water systems. This paper shares perspectives from a working group of educators, learners, and food systems subject matter experts that collaborated over the course of a year to develop, pilot test, and evaluate two interactive webinar series with a multi-site cohort of dietetics interns and graduate students. The three-part webinar series format included a training webinar, a practice activity, and a synthesis webinar. In reflecting on the effectiveness of this format, we provide direct assessments of student learning from subject …
