Tuberous sclerosis complex (TSC) 1 and TSC2 are thought to be involved in protein translational regulation and cell growth, and loss of their function is a cause of TSC and lymphangioleiomyomatosis (LAM). However, TSC1 also activates Rho and regulates cell adhesion. We found that TSC2 modulates actin dynamics and cell adhesion and the TSC1-binding domain (TSC2-HBD) is essential for this function of TSC2. Expression of TSC2 or TSC2-HBD in TSC2-/- cells promoted Rac1 activation, inhibition of Rho, stress fiber disassembly, and focal adhesion remodeling. The down-regulation of TSC1 with TSC1 siRNA in TSC2-/- cells activated Rac1 and induced loss of …

On December 15, 2003, a metal fabrication shop owner died when a 4,000 pound steel base plate fell on him. While inside the shop building, the work crew of least three men, including the owner, fabricated a 10’x 20’ steel base plate to be used at a water treatment plant. The base plate was to house or serve as a bas anchor a large water pump. After welding connectors and channel iron beams to one side of the plate, the crew pushed the plate through a bay door with a forktruck to an outside area in back of the shop. …

Women entering the court system face a challenging experience, in part, because a courtroom can be an intimidating and difficult place for any person, and in part because women victimized by crimes in which the offender is known to them face distinctive difficulties when they seek the court’s remedies. The interface is also made more challenging for women as the literature offers disparate findings as to the efficacy of criminal justice responses and civil remedies. This article briefly explores the unique characteristics of intimate partner violence cases that influence the interface of these victims with the court system.Areviewis provided of …

ApoE is expressed in multiple mammalian cell types in which it supports cellular differentiated function. In this report we demonstrate that apoE expression in adipocytes is regulated by factors involved in modulating systemic insulin sensitivity. Systemic treatment with pioglitazone increased systemic insulin sensitivity and increased apoE mRNA levels in adipose tissue by 2-3-fold. Treatment of cultured 3T3-L1 adipocytes with ciglitazone increased apoE mRNA levels by 2-4-fold in a dose-dependent manner and increased apoE secretion from cells. Conversely, treatment of adipocytes with tumor necrosis factor (TNF) α reduced apoE mRNA levels and apoE secretion by 60%. Neither insulin nor a peroxisome …

Methods and apparatus for analyzing the chemical composition of vulnerable plaques with an intravascular catheter having a near-infrared light source, a fiber-optic probe, a mechanism for directing the light from the light source into a blood vessel, and detectors for detecting light reflected or scattered by the tissue. The light source may be a tunable laser, and may transmit an incident beam having a wavelength ranging from 1400 to 4100 nm. A computer may be included to receive and process the spectral data in the analysis of the vulnerable plaques. A catheter system may be configured to provide near-IR spectrometric …

On November 3, 2003, a 38-year-old male construction laborer died when the unprotected 8-foot high walls of the trench he was working in collapsed. The decedent was working for a subcontractor who was contracted by a construction company to remove old gas, storm and sanitary sewer lines and install similar new lines. The old abandoned 6-inch diameter gas line was a high pressure, 300 – 320 psi gas line in 10-inch steel casing and was 600 feet long. On September 19, 2003, a 23-year-old male laborer (decedent) died when he became caught in the activated arms of a skidsteer loader. A work crew consisting of a contractor and 3 laborers had spent the day installing fence on a dairy farm. Earlier in the day, the contractor had borrowed a skidsteer from the farm owner. The work crew used the skidsteer to set fence posts in the ground. At the end of the work day (approximately 4:00 PM), the contractor instructed the decedent to return the skidsteer to the barn area. This report examines the provision of a variety of government services within Kentucky. The provision of these public services, specifically the cost of providing these services is examined for the years 1992, 1997, and 2002. In addition, employment and salaries in government services are also examined. In addition to comparing costs within Kentucky during this period, the costs of providing public services are also compared to costs of the same government services by its neighboring states (Illinois, Indiana, Missouri, Ohio, Tennessee, Virginia, and West Virginia).

When trying to design a forage and grazing program, producers often try to consider so many factors that it becomes mind-boggling. If basic plant physiology, how and when a forage plant grows is considered, improving a forage program will usually become easier, especially in developing a controlled grazing system. The purpose of a controlled grazing system should be to improve the utilization of the high quality forage produced on the farm. In order to improve this utilization, it is helpful to understand plant growth and quality, and the factors that affect them.

Tall fescue has played a key role in forage production over the past century in the USA. It has other uses besides forage applications, such as turf, mine reclamation, and highway rights-of-way, but in this presentation we will focus on forage varieties. Several hundred turf-type tall fescue varieties have been developed over the past 30 years, mainly by private seed companies using material from Reed Funk at Rutgers University in New Jersey. Forage varieties aren’t as numerous. I will discuss some of the older and current forage tall fescue varieties, and finish by discussing the newest varieties, and those that …

It has been known for centuries that providing certain nutrients to plants is critical for high productivity. The nutrient source and manner in which it is applied may vary, but the desired outcome is the same: high crop yield and quality. Fertilization is the most practical means that producers have to ensure that crops receive the nutrients they need. While increasing fertilizer application and managing pH by liming can lead to increase forage production, your investment should be measured against the potential return. Also, over application of nutrients can potentially cause environmental problems.

On July 4, 2003, a 36-year-old male lead electrician died after being electrocuted with 480 volts of electricity. A crew of five licensed electricians were working at an automotive supply manufacturing facility running wires to connect service for two air conditioning units (3-phase; 480 volts; 30 amp and 35 amp) and service for a lighting panel (3-phase; 277/480 volts and 200 amps). Natural killer (NK) T cells are activated by synthetic or self-glycolipids and implicated in innate host resistance to a range of viral, bacterial, and protozoan pathogens. Despite the immunogenicity of microbial lipoglycans and their promiscuous binding to CD1d, no pathogen-derived glycolipid antigen presented by this pathway has been identified to date. In the current work, we show increased susceptibility of NK T cell–deficient CD1d−/− mice to Leishmania donovani infection and Leishmania-induced CD1d-dependent activation of NK T cells in wild-type animals. Kentucky Transportation Center Research Report
The Kentucky Transportation Cabinet (KYTC) conducted an experimental project to investigate the use of accelerated maintenance painting on several overpass and elevated steel structures (74,000 ft2) in Louisville. Accelerated maintenance painting procedures specified for this project entailed limited access for the painting contractor coupled with incentives/disincentives to complete each phase of the work involving traffic closures on the underlying roadways. The approach was coupled with the use of abrasive blasting for surface preparation and the application of a rapid-cure 2-coat paint system to facilitate the work.
The project was let for $402,800. The contractor proceeded to paint 4 spans designated …

America's population is growing older. According to the 2000 census, more than 35 million people in the United States (12% of the total population) are over 65 years old. Does Regional Variation In Multiple Measures Of Health Status Differ Across Income Levels?, Janet M. Bronstein, Shailender Swaminathan, Joshua Klapow

This study examines whether regional variations in health status measures are consistent across the income gradient, or whether they are more pronounced at the lowest income levels. We use data from the Community Tracking Survey, a large randomized telephone survey of residents in 60 U.S. communities. Controlling for individual risk factors and county level income inequality, lowest income individuals have poorer scores on counts of chronic diseases, global health ratings, and the physical and mental components of the SF-12. Residents of the South have poorer scores on chronic disease counts, global health and physical health than residents of the Northeast, …

In this paper I exploit the fact that the social and economic reforms over the past two decades differentially affected the opportunity costs of non-participation in work, welfare, and disability programs for single mothers across different birth-year and education cohorts. This cohort variation in after-tax wages and transfer benefits is used to identify own- and cross-price elasticities of demand for and substitution across wage, welfare, and disability income over 1979 to 2001 in the Current Population Survey. To estimate these key parameters I model household preferences with a conditional Almost Ideal Demand System that admits corner solutions, nonseparability, endogenous wages …

Our objective was to determine whether insulin sensitivity was related to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) in Vietnam veterans exposed to Agent Orange. Air Force veterans of Operation Ranch Hand, the unit responsible for spraying Agent Orange and other herbicides in Vietnam from 1962 to 1971, and comparison veterans who did not spray herbicides were included. We measured insulin sensitivity (SI) using a frequently sampled iv glucose tolerance test in a matched study of 29 matched pairs of veterans and a quantitative insulin sensitivity check index (QUICKI) based on fasting glucose and insulin in 71 matched pairs. This study was initiated to provide a review of safety and health issues related to Kentucky Transportation Cabinet (KYTC) construction activities including both KYTC and contractor personnel. Work included a literature search, interviews with KYTC resident engineers, and both a survey of and subsequent meeting with district construction safety coordinators. Recommendations are provided for implementing changes to improve safety and health regulatory compliance for both KYTC and contractor personnel.
