For the endangered green turtle, Chelonia mydas, a fundamental component of recovery and conservation is an understanding of its foraging ecology. Foraging optimality models suggest animals will select resources of high quality over those of low quality. For green turtles, this behavior is important, as sufficient quantities of nutritionally adequate forage items are necessary for growth and reproduction. One intrinsic element in the understanding of green turtle foraging ecology is to identify and document the availability and quality of forage resources preferred by green turtles. Creating engaging, interactive, and immersive synthetic characters is a difficult task and evaluating the success of a synthetic character is often even more difficult. The later problem is solved by extending Turing's Imitation Game thusly: computational construct should be evaluated based on the criteria of how well the character can mimic a human. In order to accomplish a successful evaluation of the proposed metric, synthetic characters must be consistently believable and capable of role-appropriate emotional expression. Between World Wars I and II, allied forces girded themselves to quash yet another enemy bent on world conquest: fascism. In England, however, the British fascists set about to save what they saw as a dying empire. In an effort to restore Britain's greatness, British fascism held to fascist principles and doctrine to stem the flow of immigration, which fascists saw as darkening the pure British culture. While many of the British fascists strongly admired Nazi Germany's version of fascism, they were unique in that they forged their solutions from social ills that were distinctly British. Currently, forensic scientists are only able to determine time since death (or postmortem interval) up to the first 60 hours. This is based largely on insect activity. Herein, it is proposed to use the degradation of the intervertebral disc (IVD) after death to determine a relationship between the mechanical properties of cadaveric tissue and time since death in order to extend the 60-hour window. To that end, 1 fresh human spine and 6 pig spines were each separated into sections (6 human and 48 pig), with each section having one intact disc. Calcium phosphate (CP) materials have been used extensively for bone replacement and augmentation due to their similarity to the mineral component of bone. In addition to being non-toxic, they are biocompatible, not recognized as foreign materials in vivo, and most importantly, exhibit osteoconductive behavior, being able to help in bone formation during healing. CPs form an intimate physicochemical bond with the host tissue, termed osseointegration. However the main limitation of calcium phosphates is their inherent brittle nature and poor mechanical performance under complex stress states. An After Action Review (AAR) is the Army training system's performance feedback mechanism. The purpose of the AAR is to improve team (unit) and individual performance in order to increase organizational readiness. While a large body of knowledge exists that discusses instructional strategies, feedback and training systems, neither the AAR process nor the AAR systems have been examined in terms of learning effectiveness and efficiency for embedded trainers as part of a holistic training system. This thesis aims at providing a new approach for detecting R-waves in the ECG signal and generating the corresponding R-wave impulses with the delay between the original R-waves and the R-wave impulses being lesser than 100 ms. The algorithm was implemented in Matlab and tested with good results against 90 different ECG recordings from the MIT-BIH database. The Discrete Wavelet Transform (DWT) forms the heart of the algorithm providing a multi-resolution analysis of the ECG signal. A change in surface condition of a watershed, which is usually caused by development, can have measured effects on the naturally occurring hydrologic cycle and nitrogen cycle. This could result in environmental problems, such as reduced springflow and eutrophication. In an effort to address these issues, a combination of best management practices (BMPs) can be adhered to. The post-translational farnesylation of proteins serves to anchor a subset of intracellular proteins to membranes in eukaryotic organisms and also promotes protein-protein interactions. Inhibition of protein farnesyltransferase (PFT) is lethal to the pathogenic protozoa Plasmodium falciparum. Parasites were isolated that were resistant to BMS-388891, a tetrahydroquinoline (THQ) PFT inhibitor. Resistance was associated with a 12-fold decrease in drug susceptibility. Genotypic analysis revealed a single point mutation in the beta subunit in resistant parasites. The resultant tyrosine 837 to cysteine alteration in the beta subunit corresponded to the binding site for the THQ and peptide substrate. Biochemical analysis of Y837C-PFT demonstrated …

The recent proposal of optical induction for producing nonlinear photonic lattices has revolutionized the study of nonlinear waves in waveguide arrays. In particular, it enabled the first observation of (2+1) dimensional lattice solitons, which were the first 2D solitons observed in any nonlinear periodic system in nature. Since then, progress has been rapid, with many fundamental discoveries made within the past two years. Here, we review our theoretical and experimental contributions to this effort.

Coverage control has many applications such as security patrolling, land mine detectors, and automatic vacuum cleaners. This Thesis presents an analytical approach for generation of control inputs for a non-holonomic mobile robot in coverage control. Neural Network approach is used for complete coverage of a given area in the presence of stationary and dynamic obstacles. A complete coverage algorithm is used to determine the sequence of points. A novel synthetic methodology has been developed for attaching fluorene derivatives, containing different types of electron donating and accepting groups at the 2 and 7 positions, to gold nanoparticles of different sizes by exploiting the affinity of the thiol functional group for gold. The distance between the dye and nanoparticles was varied by introducing two alkyl chains containing different number of carbon atoms at the 9 position on the fluorene ring system. Optical Coherence Tomography (OCT) is a novel optical imaging technique that has assumed significant importance in bio-medical imaging in the last two decades because it is non-invasive and provides accurate, high resolution images of three dimensional cross-sections of body tissue, exceeding the capabilities of the current predominant imaging technique ultrasound. In this thesis, high resolution OCT is investigated for in vivo detection of abnormal skin pathology for the early diagnosis of cancer. We have studied the effect of shape of an amphiphilic molecule on micellization properties by carrying out stochastic molecular dynamics simulation on a bead-spring model of amphiphiles for several sizes of hydrophilic head group with a fixed hydrophobic tail length. Our studies show that the effect of geometry of an amphiphile on shape and cluster distribution of micelles is significant. We find the critical micelle concentration increases with the increasing size of the hydrophilic head. We demonstrate that the onset of micellization is accompanied by (i) a peak in the specific heat as found earlier in the simulation studies of …

This study was conducted to determine: 1) If healthy subjects can be conditioned to tolerate clinically useful electrically induced muscle contraction; and 2) If there is a gender difference in response to such conditioning. Healthy volunteers ( 10 males, 11 females, mean age of 27.6 +/- 5.8 yrs) were tested during each of 6 testing sessions. Maximal voluntary isometric contractions (MVIC) of the right quadriceps femoris (RQF) recorded by a computerized dynamometer. Electrical stimulation delivered through two surface electrodes and stimulation amplitude increased until the subject indicated to stop. After a 1 min rest the amplitude increased again to the …

It is theoretically shown that multi-component discrete vector surface waves can exist in arrays of coupled waveguides. These mutually trapped surface states primarily reside in the first waveguide of a semi-infinite array. The existence and stability of such surface waves are systematically investigated.

Lymphocytes are the central mediators of the immune response, requiring cytokines for survival and proliferation. Survival signaling targets the Bcl-2 family of apoptotic mediators, however, the pathway for the cytokine-driven proliferation of lymphocytes is poorly understood. Here we show that cytokine-induced cell cycle progression is not solely dependent on the synthesis of cyclin-dependent kinases (Cdks) or cyclins. Rather, we observe that in lymphocyte cell lines dependent on interleukin-3 or interleukin-7, or primary lymphocytes dependent on interleukin 7, the phosphatase Cdc25A is the critical mediator of proliferation. Withdrawal of IL-7 or IL-3 from dependent lymphocytes activates the stress kinase, p38 MAPK, …

Telomere homeostasis is regulated by telomerase and a collection of associatedproteins. Telomerase is, in turn, regulated by post-translational modifications of the rate-limiting catalytic subunit hTERT. Here we show that disruption of Hsp90 by geldanamycin promotes efficient ubiquitination and proteasome-mediated degradation of hTERT. Furthermore, we have used the yeast two-hybrid method to identify a novel RING finger gene (MKRN1) encoding an E3 ligase that mediates ubiquitination of hTERT. Overexpression of MKRN1 in telomerase-positive cells promotes the degradation of hTERT and decreases telomerase activity and subsequently telomere length. Previous studies have established constitutive activation of Stat3 protein as one of the molecular changes required for tumorigenesis. To develop novel therapeutics for tumors harboring constitutively active Stat3, compounds from the NCI 2000 diversity set were evaluated for inhibition of Stat3 DNA-binding activity in vitro. Of these, a novel platinum (IV) compound, IS3 295, interacted with Stat3 and inhibited its binding to specific DNA-response elements. Along the east coast of central Florida in the Indian River Lagoon system, intense recreational boating activity occurs year-round, and intertidal reefs of the eastern oyster Crassostrea virginica (Gmelin) with dead margins (mounds of disarticulated shells) on their seaward edges are commonly found adjacent to major boating channels. These dead margins are caused, at least in part, by boat wakes and extend significantly higher above the high water line than reefs lacking dead margins (pristine reefs). One of the critical problems in the short oligo microarray technology is how to deal with cross-hybridization that produces spurious data. Little is known about the details of cross-hybridization effect at molecular level. Here, we report a free energy analysis of crosshybridization on short oligo microarrays using data from a spike-in study. Our analysis revealed that cross-hybridization on the arrays is mostly caused by oligo fragments with a run of 10 - 16 nt complementary to the probes. Mismatches were estimated to be energetically much more costly in crosshybridization than that in gene-specific hybridization, implying that the sources of cross-hybridization …

Some pathogenesis-related genes are expressed in fungi only when the pathogen is in the host, but the host signals that trigger these gene expressions have not been identified. Virulent Nectria haematococca infects pea plants and requires either pelA, which is induced by pectin, or pelD, which is induced only in planta. However, the host signal(s) that trigger pelD expression was unknown. Here we report the isolation of the host signals and identify homoserine and asparagine, two free amino acids found in uniquely high levels in pea seedlings, as the pelD-inducing signals.
