Self-assembled cylindrical tubules of chiral phospholipids are interesting supramolecular structures. Understanding the molecular-tilt order is a key step in controlling the size and shape of the tubules and designing new functional materials. The current theories based on the chiral interactions, coupled with molecular tilt, have predicted that the tubules could have both uniform and modulated tilt states. Here, we image the molecular-tilt order in the self-assembled tubules of a chiral phospholipid by using liquid crystals as an optical amplification probe. We demonstrate that the organization of the molecular-tilt azimuth in the lipid tubules can induce an azimuthal orientation in the …

The currently available human vaccine for anthrax, derived from the culture supernatant of Bacillus anthracis, contains the protective antigen (PA) and traces of the lethal and edema factors, which may contribute to adverse side effects associated with this vaccine. Therefore, an effective expression system that can provide a clean, safe, and edicacious vaccine is required. In an effort to produce anthrax vaccine in large quantities and free of extraneous bacterial contaminants, PA was expressed in transgenic tobacco chloroplasts by inserting the pagA gene into the chloroplast genome. Chloroplast integration of the pagA gene was confirmed by PCR and Southern analysis. …

Before European settlement, 23% of Illinois (3.2 million of 14 million ha) was covered by wetlands. It is estimated that 90% of those wetlands Were lost during conversion of the landscape to agriculture and urban, use. Champaign County was one of the most extensively drained counties in Illinois, with 39-60% of original county area estimated to have been drained. Current and future efforts to conserve and restore wetlands would benefit from information on the number and distribution of former wetlands. Background The use of mega-vitamin intervention began in the 1950s with the treatment of schizophrenic patients. Pyroxidine (vitamin B6) was first used with children diagnosed with "autism syndrome" when speech and language improvement was observed in some children as a result of large doses of B6. A number of studies attempted to assess the effects of vitamin B6-Magnesium (Mg) was found to reduce undesirable side effects from B6) on characteristics such as verbal communication, non-verbal communication, interpersonal skills, and physiological function, in individuals with autism. Background: Rates of synonymous nucleotide substitutions are, in general, exceptionally low in plant mitochondrial genomes, several times lower than in chloroplast genomes, 10-20 times lower than in plant nuclear genomes, and 50-100 times lower than in many animal mitochondrial genomes. Several cases of moderate variation in mitochondrial substitution rates have been reported in plants, but these mostly involve correlated changes in chloroplast and/or nuclear substitution rates and are therefore thought to reflect whole-organism forces rather than ones impinging directly on the mitochondrial mutation rate. The phospholipase A(2) (PLA(2)) enzymes are activated by binding to phospholipid membranes. Although the N-terminal alpha-helix of group I/II PLA(2)s plays an important role in the productive mode membrane binding of the enzymes, its role in the structural aspects of membrane-induced activation of PLA(2)s is not well understood. In order to elucidate membrane-induced conformational changes in the N-terminal helix and in the rest of the PLA(2), we have created semisynthetic human group IB PLA(2) in which the N-terminal decapeptide is joined with the C-13-labeled fragment, as well as a chimeric protein containing the N-terminal decapeptide from human group IIA PLA(2) …

Optical imaging technique has strong potential for sensitive cancer diagnosis, particularly at the early stage of cancer development. This is a sensitive, non-invasive, non-ionizing (clinically safe) and relatively inexpensive technique. Cancer imaging with optical technique however greatly relies upon the use of sensitive and stable optical probes. Unlike the traditional organic fluorescent probes, fluorescent nanoparticle probes such as dye-doped nanoparticles and quantum dots (Qdots) are bright and photostable. Fluorescent nanoparticle probes are shown to be very effective for sensitive cancer imaging with greater success in the cellular level. Two-photon fluorescence microscopy is a powerful tool for the study of dynamic cellular processes and live-cell imaging. Many commercially available fluorescent probes have been used in multiphoton-based imaging studies despite exhibiting relatively low two-photon absorption cross-section values in the tunability range of ultrafast Ti:sapphire lasers commonly used in multiphoton microscopy imaging. Furthermore, available fluorophores may be plagued with low fluorescence quantum yield and/or photoinstability (i.e., photobleaching) on exposure to the high peak power and photon density provided by the ultrafast laser source. Discrete models are used in describing various microscopic phenomena in many branches of science, ranging from biology through chemistry to physics. Arrays of evanescently coupled, equally spaced, identical waveguides are prime examples of optical structures in which discrete dynamics can be easily observed and investigated. As a result of discretization, these structures exhibit unique diffraction properties with no analogy in continuous systems. Recently nonlinear discrete optics has attracted a growing interest, triggered by the observation of discrete solitons in AlGaAs waveguide arrays reported by Eisenberg et al. in 1998. Recent findings suggest that leptin may be regulated in response to abrupt changes in energy homeostasis. Therefore, it is conceivable that transient changes in energy balance induced by exercise may also regulate leptin synthesis and secretion. As such, we hypothesized that acute increases energy expenditure (i.e. exercise), may regulate leptin concentrations in obese individuals. Fifteen healthy obese males underwent either a single exercise session of moderate intensity (58.4 % ± 4.0 of VO2max) for 60 min (n=8), or served as controls (n=7). The exercise session elicited an energy expenditure of 567±80 Kcal. No significant changes in plasma leptin (pre 23.5± …

Enhanced Biological Phosphorous Removal (EBPR) performance was found to be adequate with reduced Return Activated Sludge (RAS) flows (50% of available RAS) to the anaerobic tank and smaller than typical anaerobic zone volume (1.08 hours hydraulic retention time or HRT). Three identical parallel biological nutrient removal (BNR) pilot plants were fed with strong, highly fermented (160 mg/L VFAs), domestic/industrial wastewater from a full scale wastewater treatment facility (WWTF). The pilot plants were operated at 100%, 50%, 40% and 25% RAS (percent of available RAS) flows to the anaerobic tank with the remaining RAS to the anoxic tank. In addition, varying …

Since their inception, the actual use of nuclear weapons in conflict is extremely limited. There have been only two documented occurrences which were committed exclusively by the United States. By contrast, however, state posturing with nuclear weapons occurs with regularity transcending historical situations, national wealth, military power, or even the actual possession of nuclear weapons. Rationalist arguments that depict nuclear posturing as a means of deterrence appear insufficient given its tendency to unbalance perceptions of equilibrium, and the public nature in which it occurs. The present investigation modeled the expectancy memory organization and likely memory activation patterns of marijuana expectancies of children across age and marijuana use. The first phase of the study surveyed 142 children to obtain their first associate to marijuana use. From their responses, the Marijuana Expectancy Inventory for Children and Adolescents (MEICA) was developed. The second phase of the study administered the MEICA to a second sample of 392 children to model marijuana expectancy organization and probable memory activation paths of marijuana users versus never-users. In NiTi, a reversible thermoelastic martensitic transformation can be induced by temperature or stress between a cubic (B2) austenite phase and a monoclinic (B19') martensite phase. Ni-rich binary compositions are cubic at room temperature (requiring stress or cooling to transform to the monoclinic phase), while Ti-rich binary compositions are monoclinic at room temperature (requiring heating to transform to the cubic phase). The stress induced transformation results in the superelastic effect, while the thermally induced transformation is associated with strain recovery that results in the shape memory effect. Total lead release in drinking water in the presence of free chlorine and chloramine residuals was investigated in field, laboratory and fundamental investigations for finished waters produced from ground (GW), surface (SW), saline (RO) and blended (B) sources. Field investigations found more total lead was released in the presence of chloramines than in the presence of free chlorine for RO and blended finished waters; however, there were no statistical differences in total lead release to finished GW and SW. The primary purpose of this study is to identify the 'universal' rate formulations with scaled-dependent parameters for the biological reduction of hematite. Three possible rate formulations were proposed to describe the bioreduction rate of hematite, and two kinds of simulation were conducted to validate the formulations and parameters with both batch and column experimental data: a reaction-based biogeochemical (batch) modeling with BIOGEOCHEM 1.0 and a reactive biogeochemical transport (column) modeling via HYDROGEOCHEM 4.0. This dissertation addresses the role of individual fouling mechanisms on productivity decline and solute mass transport in nanofiltration (NF) of surface waters. Fouling mechanisms as well as solute mass transport mechanisms and capabilities must be understood if NF of surface waters is to be successful. Nanofiltration of surface waters was evaluated at pilot-scale in conjunction with advanced pretreatment processes selected for minimization of nanofilter fouling, which constituted several integrated membrane systems (IMSs). Membrane fouling mechanisms of concern were precipitation, adsorption, particle plugging, and attached biological growth. In this dissertation, we describe the portable, open-source distributed simulation framework (WINDS) targeting simulations of wireless network infrastructures that we have developed. We present the simulation framework which uses modular architecture and apply the framework to studies of mobility pattern effects, routing and intrusion detection mechanisms in simulations of large-scale wireless ad hoc, infrastructure, and totally mobile networks. The distributed simulations within the framework execute seamlessly and transparently to the user on a symmetric multiprocessor cluster computer or a network of computers with no modifications to the code or user objects. In realizing a portable chemical analysis system, adequate partitioning of a reusable component and a disposable is required. For successful implementation of micromachined sensors in an instrument, reliable methods for interconnection and interface are in great demand between these two major parts. This thesis work investigates interconnection methods of micromachined chip devices, a hybrid fluidic interface system, and measurement circuitry for completing instrumentation. The interconnection method based on micromachining and injection molding techniques was developed and an interconnecting microfluidic package was designed, fabricated and tested. Proper stormwater management acknowledges both water quantity and water quality. Historically, stormwater quantity and quality have been separately considered; runoff was routed as quickly as possible into the nearest body of water. Although this alleviates potential flooding concerns, water quality is often compromised. Common stormwater quality problems include gross pollutants, sediment, nutrients, and heavy metals. The chronic elevated presence of these pollutants is detrimental to the environment. As a result, the government has passed legislature to protect waterways. This dissertation investigates the community structure of web-like networks (i.e., large, random, real-life networks such as the World Wide Web and the Internet). Recently, it has been shown that many such networks have a locally dense and globally sparse structure with certain small, dense subgraphs occurring much more frequently than they do in the classical Erdös-Rényi random graphs. This peculiarity--which is commonly referred to as community structure--has been observed in seemingly unrelated networks such as the Web, email networks, citation networks, biological networks, etc. The pervasiveness of this phenomenon has led many researchers to believe that such cohesive groups of …
