This dissertation is comprised of several studies over three unrelated areas of focus. Chapters two and three investigate the role of the hormone ethylene on growth of creeping bentgrass through (1) exposure of turf to exogenous ethylene and (2) testing an ethylene inhibiting compound for use to alleviate stress related injury. Previous research demonstrated a correlation of stress injury and ethylene and lessening of injury through ethylene inhibition, but little work has been conducted evaluating the direct effects of ethylene. The performativity of gender through cross-dressing has been a staple in Japanese media throughout the centuries. This thesis engages with the pervasiveness of cross-dressing in popular Japanese media, from the modern shōjo gender-bender genre of manga and anime to the traditional Japanese theatre. Two experiments were conducted to evaluate effects of white striping and woody breast hardness on meat quality traits in broiler breast fillets. The goal of this dissertation is to present my play, Poor Relations, to my thesis committee and discuss both the process of bringing it to the stage and the results of that process.

The dissertation opens with a narrative essay detailing the process of writing Poor Relations and bringing it to the stage in a full production. Pectin is a soluble fiber with demonstrated health benefits such as the ability to lower blood glucose and cholesterol, increase satiety leading to lower caloric intake, improve insulin resistance and lower inflammation. Certain pathogenic Escherichia coli known as Shiga toxin (Stx)-producing Escherichia coli (STEC) are commensals in cattle, and typically cause bloody diarrhea in humans once the Stx toxin is secreted in invaded intestinal epithelial cells. Infections with STEC cells can lead to hemolytic uremic syndrome, which is commonly associated with kidney failure. Several STEC serogroups have been declared adulterants in raw, non-intact ground meat, and future regulations could potentially lead to a higher number of STEC serogroup detection strategies for these pathogenic microorganisms. Using Peptoids To Build Robust, Efficient Microarray Systems, Dhaval Sunil Shah

Recent studies have shown microarrays to be indispensable for various biological applications, allowing for high-throughput processing and screening of biological samples such as RNA, DNA, proteins and peptides using a small sample volume (< 1 µL). Peptoids (poly-N-substituted glycine oligomers) can be used as a substitute for antibodies as capture molecules, as well as coatings for slides in antibody microarrays. The ease of synthesis of peptoids, high customizability with desired bioactivity, and speed of synthesis allows us to build a diagnostic system with a large dynamic range that can detect biomolecules from a minimal sample size. In this study, peptoid-based antibody mimics are designed to have both structural and functional features similar to those of antibodies, including a stable constant region (scaffolding) and a variable region for protein recognition. Peptoids previously screened via combinatorial library synthesis to be specific to bind Mdm-2 (mouse double minute 2 homolog) and GST (gluthathione S-transferase), have been synthesized. The protein recognition peptoids have been conjugated to PEG (polyethylene glycol) molecules with modified end groups; an amine group on one end that allows for immobilization and orientation on the slide, and an azide group on the other end that will allow for attachment to the peptoid through “click chemistry”. The number of capture molecules printed on the slides can be increased by making the available surface area of the slide larger via coating with microspheres. We have determined that partially water soluble peptoids that are also helical, can self-assemble into microspheres. Sequences have been developed that can consistently produce uniform microsphere coatings on slides that increase the overall surface area. A high surface area corresponds to a higher number of binding sites, and therefore a more sensitive system. The work done has shown that slides may be successfully coated in order to potentially improve the detection system.

Investigations into the distribution and ecosystem functions of fruiting amoebae revealed that local-scale environmental conditions can largely explain broad biogeographical patterns in species assemblage, the way in which amoeboid predators shape bacterial communities and how this top-down influence may affect global biogeochemical processes in a changing climate. The distribution and assemblage of protosteloid amoebae on the islands of New Zealand and Hawaii did not yield any expected patterns of island biogeography, and conformed to other global regions studied. Islet amyloid polypeptide (IAPP) also called amylin is an amyloid-forming protein; IAPP is a proteinaceous hormone that comprises 37 amino acid residues. It is secreted along with insulin from the pancreatic β-cells to help it regulate the uptake and removal of glucose in the bloodstream. This dissertation analyzes Pakistani and Indian plays that illustrate the nexus of power relations that operate in Pakistan and India to disempower women and the way women resist by creating dialogic spaces or fissures in the exploitative system. This thesis reads the novels of Cormac McCarthy as posthuman southern literature to explain why fiction from the South after World War II could no longer convey a sense of place during postmodernity: that is, because the region's culture and economy were transitioning from predominantly humanistic thinking (i.e., believing that humans [and especially southern humans] are supreme beings) to predominantly posthumanistic thinking (i.e., believing that humans are not as supreme as they think they are). This study investigated the impact of different flour-batter types on protein rich food in regard to people’s overall liking and sensory preferences (saltiness, flavor, texture, moisture, and color). In addition, all battered fried chicken, fish, shrimp, pork, and beef were analyzed for fat, calories, and protein content in the Central Analytical Laboratory at the University of Arkansas. The accumulation of amyloid-beta (Aβ) oligomers is believed to be the driving force behind Alzheimer’s disease (AD) pathogenesis. Due to the metastable nature of Aβ oligomers, the knowledge of Aβ aggregation and accumulation is not well understood. Here, we use capillary electrophoresis (CE) and photo-induced crosslinking of unmodified proteins (PICUP) to learn about the aggregation of Aβ.

The purpose of this study was to prioritize subwatersheds using water quality data and watershed characteristics. Water quality data was provided through studies by the Arkansas Natural Resources Commission, Beaver Water District, Arkansas Water Resources Center, and publicly available USGS gage data. A total of 114 sites across five HUC-8 watersheds were analyzed, including 12 USGS gages. Watershed characteristic data was retrieved from USGS and Arkansas GeoStore geodatabase repositories. A significant linear relationship between baseflow and stormflow nutrient concentrations was established allowing for the use of baseflow concentrations in the prioritization methodology. Weedy rice (Oryza sativa L.) competes aggressively with rice, reducing yields and grain quality. ClearfieldTM rice, is nontransgenic herbicide-resistant (HR) rice introduced in 2002 to control weedy rice, has resulted in ALS-resistant weedy rice due to gene flow. Volunteers of ClearfieldTM rice (F2) accelerate the HR weedy rice evolution by acting as agents of gene flow. Weedy rice (89) collected from 11 counties in Arkansas were screened for resistance in a field experiment in Stuttgart (2011). Seventy-nine percent of accessions were resistant to imazethapyr and harbored S653N mutation. These HR weedy accessions were outcrosses of ClearfieldTM rice and weedy rice. …

Various studies have suggested that chir pine (Pinus roxburghii) is replacing banj oak (Quercus leucotrichophora) in the Central Himalaya. Five sites with three different types of forests (banj oak, chir pine and mixed oak-pine) were sampled to compare the diversity of their vegetation and to assess the impact of this ongoing conversion on biodiversity. Soil samples collected from oak and pine forests were analyzed and compared. Six experiments were conducted to determine effects of in ovo feeding of dextrin (Dext, 18% maltodextrin, 10% potato starch dextrin) and iodinated casein (IC) on hatch and growth performance in broilers. In each experiment, approximately 15,000 eggs (2500 eggs per group) were treated at 18.5 to 19 d of embryonic development. This work applies molecular genetic tools to distinguish the identity and understand the biology of termites, particularly Reticulitermes Holmgren and Coptotermes formosanus Shiraki (Isoptera: Rhinotermitidae) in the southeastern U.S. Termites are important economic pests costing billions of dollars annually to Americans but also are important wood decomposers in natural settings. Molecular tools are essential for species identification because of the morphological ambiguities among species. The cryptic nest structure of subterranean termites which prevents adequate sampling makes molecular genetic tools essential to examine an entire colony.
