Analysis Of Nphp Complex Genetic Interactions Associated With Human Cilia Disorders, Svetlana Viktorovna Masyukova
Analysis Of Nphp Complex Genetic Interactions Associated With Human Cilia Disorders, Svetlana Viktorovna Masyukova
All ETDs from UAB
Primary cilia are antenna-like organelles that extend from the surface of almost all mammalian cell types. They regulate many signaling pathways and sense physical and chemical changes in the extracellular environment. Defects in primary cilia cause several human disorders of different severity collectively called ciliopathies, including nephronophthisis (NPHP), Joubert syndrome (JBTS), and Meckel-Gruber syndrome (MKS). Numerous MKS, JBTS and NPHP genes have been identified but in most cases of these ciliopathies the genetic defect is unknown. Despite the fact that NPHP, JBTS and MKS patients present with distinct clinical features, they have mutations in identical genes. This can be explained …
Hemoglobin Switching, Thalassemia And Sickle Cell Disease In Humanized Knockin Mice, Sean Mcconnell
Hemoglobin Switching, Thalassemia And Sickle Cell Disease In Humanized Knockin Mice, Sean Mcconnell
All ETDs from UAB
In this dissertation we describe knockin mouse models for the study of human hemoglobin disorders. These knockin human globin genes, in contrast to transgenes, replace the adult mouse globin genes, remain under control of endogenous mouse globin enhancer sequences and are inherited in a manner identical to globin alleles in man. Starting with various knockin alleles composed of unique human alpha, beta and gamma globin gene sequences, we evaluate several allele combinations towards our goal of generating improved models of thalassemia and sickle cells disease. We show that humanized knockin mice complete a fetal to adult hemoglobin switch during postnatal …

Individual, Socio-Cultural And Structural Factors Associated With Cervical Cancer Screening And Treatment In Tanzania, Renicha Tomeka Mccree-Hale
All ETDs from UAB
Most African countries including Tanzania have limited cervical cancer screening and treatment programs. Only about 5% of women have ever been screened for cervical cancer. A constellation of individual and structural factors contributes to the challenge of cervical cancer screening and treatment in countries that bear significant burdens of disease. Yet, little is known about country specific factors that may constrain or facilitate scale up of health services. This dissertation is an attempt to identify and understand the country specific individual and structural barriers along with facilitators to cervical cancer screening and treatment in Tanzania. Individual, institutional and structural level …
An Investigation Of The Effects Of Ocean Acidification On Larval Development, Juvenile Settlement And Mortality, Growth, Onset Of Egg Production, Shell Integrity And Feeding Activity In The Common Barnacle Amphibalanus Amphitrite, Michelle R. Mcdonald
All ETDs from UAB
ABSTRACT Ocean acidification is of great concern among the scientific community and is caused by the anthropogenic release of CO2 from the burning of fossil fuels. Higher levels of atmospheric CO2 are anticipated to cause a decrease in oceanic pH. Recent studies have shown that this can have adverse effects on a variety of marine organisms; calcified organisms are predicted to be the most heavily affected and of these types of organisms, barnacles have received little attention. The barnacle Amphibalanus amphitrite was utilized to examine the effects of Ocean acidification across various life history stages. No adverse effects of reduced …

OPTIMAL BRACING TYPE AND LAYOUTS TO MINIMIZE LATERAL DRIFT IN HIGH-RISE STEEL FRAMED BUILDINGS SUBJECTED TO HIGH WINDS USING SAP2000 NICHOLAS F. MCEWEN Civil, Const. & Environmental Engineering (ABSTRACT) As cost controls almost all projects and wind forces typically control the design of high-rise buildings, it is crucial to understand the most optimal use of steel when concerning protection against lateral forces. One of the largest tasks of the designer in highrise building design simply reduces to limiting the lateral drift that is associated with wind forces. Thus, the primary objective of this research was to use the SAP2000 software …
Role Of Calcium-Activated Potassium Channels In Glioblastoma Volume Regulation, Michael Bryan Mcferrin
Role Of Calcium-Activated Potassium Channels In Glioblastoma Volume Regulation, Michael Bryan Mcferrin
All ETDs from UAB
The most common and most malignant gliomas are the Grade IV Glioblastoma Multiforme (GBM), characterized by a highly proliferative tumor mass and extremely invasive phenotype that allows for profuse dispersal of tumor cells throughout the brain. GBM cells must specifically regulate their cell volume to thrive within the edematous tumor mass and infiltrate throughout the tortuous extracellular spaces of the brain. These rapid and directed volume changes are governed by the controlled flux of potassium and chloride ions across the cell membrane, which move osmotically obliged water. The goal of this dissertation was to investigate the role of calcium-activated potassium …

Alcoholic Women In Recovery: A Phenomenological Inquiry Of Spirituality And Recidivism Prevention, Jennifer Irene Mcleod
All ETDs from UAB
In the United States, 7-12% of women compared to 20% of men, abuse alcohol, yet the social and medical consequences impact women much harder and faster than men. Women aggrieved by alcoholism have unique health and social consequences which are not well understood because women have been marginalized in studies of alcoholism. Little is known about the nature of the lived experience of the recovery process in women who have achieved a stable recovery and prevented recidivism. The aim of this study was to describe the lived experiences of alcoholic women in a stable recovery, defined as sustained abstinence for …
Contracts, Consent, And Fate: Framing The Chinese Civil Service Examinations As Mechanisms Of Social Conditioning And Political Domination, Forrest Cale Mcsweeney
Contracts, Consent, And Fate: Framing The Chinese Civil Service Examinations As Mechanisms Of Social Conditioning And Political Domination, Forrest Cale Mcsweeney
All ETDs from UAB
Though the form of civil service examinations in pre-modern Imperial China was dynamic, their function was comparatively static. In every era, they were principally an institution designed to either assist or outright effect the creation of an economy of social hierarchy designed to ensure social an economic domination. This function changed only with respect to what kind of hierarchy was to be created: either aristocratic, gentry, or just as often ethnic. The exams' evolution culminated in an era of understood autocracy after the Song characterized by the state's structure as one ultimately tailored to enable the despotic rule of one …

Killer Immunoglobulin-Like Receptors And Human Leukocyte Polymorphisms In Hiv-1 Pathogenesis, Aimee Marie Merino
All ETDs from UAB
Human leukocyte antigen (HLA) molecules influence the adaptive and innate immune responses through interactions with cytotoxic T lymphocytes and natural killer cells (NK). HLA influence the activation state of NK through direct binding to killer immunoglobulin-like receptors (KIR) and presentation of HLA-derived peptides to NKG2 receptors. Activated NK can kill HIV-infected cells through direct cytolysis and release cytokines that influence other aspects of the immune system. Variations in KIR gene content and allelic variants have been associated with autoimmunity, transplantation success, and infectious diseases such as hepatitis C and HIV/AIDS. We investigated the impact of KIR genes and potential KIR-HLA …

All ETDs from UAB
Antarctic ecosystems are extreme environments to study the biodiversity, survival and adaptation of bacteria. The current study describes the rich and diverse heterotrophic bacterial communities from seasonally ice-free open lakes in East Antarctic Schirmacher Oasis and a perennially ice-covered Lake Joyce in West An-tarctic McMurdo Dry Valleys using culture-dependent and culture-independent approaches. Heterotrophic bacteria from three different Phyla: Proteobacteria, Bacteroidetes and Actinobacteria were cultured from lakes of Schirmacher Oasis using 16S rRNA gene-based taxonomic analysis. However, similar approach applied on the community DNA of the Black Hole sediments in Lake Joyce exhibited previously undescribed heterotrophs from seven different phyla: Proteobacteria, …
Supporting Autonomy In Early Childhood: A Case Study Of Educators Offering Choice, Jann Montgomery
Supporting Autonomy In Early Childhood: A Case Study Of Educators Offering Choice, Jann Montgomery
All ETDs from UAB
The purpose of this study was to explore the behaviors of early childhood educators that support their students' autonomy. Researchers have shown that the most prevalent style of teaching is an autonomy-suppressive style that has been determined to undermine student functioning and achievement (Assor, Kaplan, & Roth, 2002; Reeve, 2009). Teachers are often unaware of the atmosphere they create in their classroom (DeVries & Zan, 1994) and this can result in learning environments that are coercive which has been found to impede children's autonomous thinking (Reeve, 2009). This study sought to explore autonomy-supportive teacher behaviors in the early childhood classroom. …

Competition between northern sympatric mosquitoes, Culex restuans and Culex pipiens pipiens, was evaluated in Illinois using different ratios of the two species. Similarly, competition between southern sympatric species, Culex restuans and Culex pipiens quinquefasciatus, was evaluated in Alabama. Species ratios were repeated at biologically relevant temperatures and densities. Results for the northern study suggested that Cx. restuans survival was greater than Cx. pipiens across all treatment variables, whereas survival did not differ between the southern strains. Survival of both northern and southern strains of Cx. restuans increased with increasing competition, but southern strains of this species had poor survival in …

All ETDs from UAB
Traditional construction materials have numerous shortcomings such as poor durability, low penetration resistance against wind-borne debris and lower strength compared to composites. Accordingly, a new type of Composite Structural Insulated Panels (CSIPs) was developed and evaluated in this study to replace the conventional structural materials. CSIPs are prefabricated components that can be shipped to the site and assembled into the finished structure. These hybrid panels have the potential to be used as structural wall, floor, and roof elements. Analytical and finite element (FE) models were developed in this study to design and simulate the behavior of CSIPs. These models were …

LEGAL CHALLENGES TO PUBLIC EDUCATION FINANCE: EQUITY V. ADEQUACY J. MICHAEL MUNGER DEPARTMENT OF GOVERNMENT -- PUBLIC ADMINISTRATION ABSTRACT This research examines the nature of public education financing in the United States. When education became a cornerstone of public policy in America, federalism dictated that the responsibility of implementation be delegated to the states, and from the states to the constituent school districts. Oftentimes these districts came to be divided--accidentally or on purpose--along lines of race/ethnicity or socioeconomic status; this led to wide variations in education outcomes. Such after effects have come under greater scrutiny over the past few decades. …

Development And Characterization Of Thermal Diffusion Doped Fe2+:Znse/Zns Polycrystalline Gain Media For Room Temperature Mid-Ir Lasing, Nosoung Myoung
All ETDs from UAB
The dissertation is focused on thermo-diffusion doping of Fe2+:ZnSe, Fe2+:ZnS, Fe2+:Co2+:ZnSe, and Fe2+:Co2+:ZnS polycrystals and their laser-spectroscopic characterization. Most materials absorb infrared (IR) light at specific patterns of frequencies directly related to their structure. IR spectra can therefore be used to identify and quantify the constituents of an unknown material, making detection of chemicals by their absorption of IR light an important analytical tool. For instance, Mid-infrared lasers (ë = 3 to 5µm) are widely used for an environmental monitoring, remote sensing of atmosphere and defense-related applications. Tunable middle infrared lasers are essential for highly sensitive detection of organic molecules …

Cancer is an abnormal and uncontrolled growth of cells and is one of the most common causes of millions of deaths worldwide. Currently used chemotherapeutic drugs have severe side effects. Hence, there is a need to develop new cancer therapeutic drugs that can offer higher survival rates and fewer limitations. About 60 % of the known anticancer drugs are either natural products or are derived from natural products. Development of marine natural products into drugs has received increasing attention as a part of humanity's quest for newer sources of cancer drugs. Marine natural products are isolated only in small quantities …

Altered Dna Methylation Of Repetitive Elements May Explain The Modified Risk Of Cervical Intraepithelial Neoplasia (Cin) Associated With Genetic Polymorphisms Of The Folate Metabolic Pathway In The Us Post Folic Acid Fortification Era, Suguna Badiga
All ETDs from UAB
Despite the controversy regarding the adverse effects of exposure to higher folate concentrations on health outcomes particularly cancer, there has been paucity of research to understand the biological mechanisms affected by such exposures in relation to the risk of developing pre-cancer or cancer. We have recently addressed this concern by demonstrating that higher plasma folate concentrations were not associated with greater risk of being diagnosed with higher grades of cervical intraepithelial neoplasia (CIN 2+) especially in women with sufficient plasma vitamin B12 concentrations. Higher plasma folate was also associated with a pre-cancer protective level of peripheral blood mononuclear cells (PBMC) …

Adsorption Characteristics Of Activated Carbon Fibers (Acfs) For Toluene, Jo Anne Goot Balanay
All ETDs from UAB
Granular activated carbon (GAC) is currently the standard adsorbent in respirators against several gases and vapors because of its efficiency, low cost and available technology. However, a drawback of GAC due to its granular form is its need for containment, adding weight and bulkiness to respirators. This makes respirators uncomfortable to wear, resulting to poor compliance in its use. Activated carbon fiber (ACF) is considered a viable alternative adsorbent material for developing thinner, light-weight and efficient respirators because of their larger surface area, lighter weight and fabric form. This study aims to determine the critical bed depth and adsorption capacity …

Gas Phase Laser Synthesis And Processing Of Calcium Phosphate Nanoparticles For Biomedical Applications, Parimal Vilas Bapat
All ETDs from UAB
Biochemical processes make pervasive use of calcium and phosphate ions. Calcium phosphate salts that are naturally nontoxic and bioactive have been used for several medical applications in form of coatings and micropowders. Nanoparticle-based calcium phosphates have been shown to be internalized by living cells and be effective in DNA transfection, drug delivery, and transport of fluorophores for imaging of intracellular processes. They are also expected to interact strongly with cell adhesive proteins and are therefore promising elements in approaches to mimic the complex environment of the extra cellular matrix of bone. Harnessing this biomedical potential requires the ability to control …

An Examination Of Statistical Methods For Longitudinal Mediation Modeling, John Paul Bentley
All ETDs from UAB
The use of mediation modeling is quite prevalent in a number of disciplines to answer questions about how or why one variable exerts its influence on another variable. Although mediation can be assessed in the context of several types of study designs, the use of cross-sectional data and a single-mediator model tend to be the most commonly reported features in empirical tests of mediation. There are several limitations associated with assessing mediation with cross-sectional data, perhaps the most significant is that mediated effect estimates are biased in the case of true longitudinal mediation. For this and several other reasons, there …

Three Topics In Eye Injury Research, Justin Blackburn
All ETDs from UAB
Eye injuries are a leading cause of monocular blindness, with approximately 2 million Americans sustaining an eye injury annually. Occupational eye injuries resulting in at least one missed day of work total nearly 28,000 each year. Eye injuries have many different etiologies, although most are "minor" injuries such as corneal abrasions and foreign bodies. Chemical-related eye injuries have not been described epidemiologically. The most widely advocated prevention method for eye injuries is protective eyewear, but evidence to support this is surprisingly absent. Furthermore, the use of eye protection among workers who have previously sustained an eye injury remains a question …
