Role Of The Cystine/Glutamate Exchanger In Glioma Cell Biology, Toyin Adeyemi Ogunrinu

All ETDs from UAB

Changes in the glioma microenvironment including oxygen (O2) levels, supply of amino acid such as L-glutamate and L-cystine and glutathione (GSH) concentrations play a critical role in glioma biology. Previous data from our laboratory and others have implicated the L-cystine/L-glutamate exchanger, system xc- in the invasion and proliferation of cancers including glioma. The central aim of this dissertation was to characterize the contribution of L-cystine uptake, GSH synthesis and L-glutamate release to migration and proliferation of glioma cells. In my first study, I examined the role of system xc- mediated L-glutamate release on glioma migration. I show that activation of …

A Topological Approach To Shape Analysis And Alignment, Benjamin David O'Gwynn Jan 2011

A Topological Approach To Shape Analysis And Alignment, Benjamin David O'Gwynn

All ETDs from UAB

When analyzing multiple mesh models, an important first step is to find the rigid transform that best aligns them. Many mesh alignment techniques exist, but because they treat input meshes as a collection of surface samples, they tend to be sensitive to surface variations between those meshes. We instead wish to align shapes based upon an analysis of mesh topology, thus making it possible to align shapes with similar part structures but varying surface geometry. To this end, we present a novel algorithm that extracts a curve-skeleton from a mesh based on its connectivity, rather than its surface geometry. We …

The Effects Of Apolipoprotein Mimetic Peptides On Inflammation And Oxidative Stress, Toral Rohit Patel Jan 2011

The Effects Of Apolipoprotein Mimetic Peptides On Inflammation And Oxidative Stress, Toral Rohit Patel

All ETDs from UAB

Coronary Artery Disease (CAD) is the leading cause of deaths worldwide, including the United States. Atherosclerosis remains the primary cause of CAD, and is a leading target for drug treatments. Despite advances in treatments, many questions remain unanswered pertaining to the mechanisms of the treatments on inflammation and oxidative stress. We hypothesize that peptides 4F and Ac-hE18A-NH2 act as anti-inflammatory factors and reduce oxidative stress. We also hypothesize that these peptides require apolipoprotein A-I (apoA-I) to increase paraoxonase-1 (PON-1) activity. The effects of administering peptides 4F and Ac-hE18A-NH2 on oxidative stress were evaluated through the analysis of plasma from apoA-I-null …

Reactivation Of Estrogen Receptor-Α (Erα) By Bioactive Dietary Compounds Through Epigenetic Mechanisms In Erα-Negative Breast Cancer Cells, Shweta Naran Patel Jan 2011

Reactivation Of Estrogen Receptor-Α (Erα) By Bioactive Dietary Compounds Through Epigenetic Mechanisms In Erα-Negative Breast Cancer Cells, Shweta Naran Patel

All ETDs from UAB

In breast cancer treatment, the presence of the estrogen receptor-alpha (ERα) plays an important role with hormonal-responsive drugs such as tamoxifen. ERα-negative breast cancers are more aggressive not only because of the increased uncontrollable growth but also because of the lack of target-directed treatments. Bioactive dietary components such as green tea polyphenols (GTPs) and sulforaphane (SFN, found in cruciferous vegetables) have been shown to have anti-cancer properties. In this study, we found that the dietary combination of a DNA methyltransferases (DNMTs) inhibitor, GTPs, and a histone deacetylase (HDAC) inhibitor, SFN, dose-dependently inhibited the proliferation of ERα-negative MDA-MB-231 human breast cancer …

The Graphic Novel: A Frontier Literary Form Edges Towards The English Classroom, Malgorzata Pflueger-Pieczynska Jan 2011

The Graphic Novel: A Frontier Literary Form Edges Towards The English Classroom, Malgorzata Pflueger-Pieczynska

All ETDs from UAB

Graphic novels, as not yet fully researched and examined forms under academia, suffer from their association in the public mind with the advertisement industry and cheap subject matter. This negative affiliation is especially popular in teachers' circles that use the graphic novel mostly as an instrument to improve reading skills rather than to present it as an extraordinary example of a frontier literary form. The graphic novel is one of many products that took off dynamically with the technological revolution using the visual as a communication's medium. Visual communication is not only one of the leading forms of passing information, …

Robust Nonparametric Regression Approach For Competing Cause Censored Survival Mortality Data With Covariates, Milind Arvind Phadnis Jan 2011

Robust Nonparametric Regression Approach For Competing Cause Censored Survival Mortality Data With Covariates, Milind Arvind Phadnis

All ETDs from UAB

In the case of two or more competing risks when only the time to the event due to one risk is observable with times due to the other outcomes treated as censored observations, standard methods of survival analysis fall short of assessing the association of a set of predictors with the outcomes of interest. This problem is particularly severe when there is a differential association of a predictor with two competing risks (harmful for one risk and protective for the other risk) such as stroke and myocardial infarction and in situations where the predictor is associated with only one risk …

Regulation Of Neuronal Death By The Autophagy Lysosomal Pathway: Implications For Parkinson Disease, Violetta N. Pivtoraiko

All ETDs from UAB

Parkinson Disease (PD) is the second most common age-related neurodegenerative disorder and is characterized pathologically by the loss of dopaminergic (DA) neurons in the stubstantia nigra pars compacta (SNpc). Mitochondrial dysfunction, increased oxidative stress, and accumulation of aggregated α-synuclein (α-syn), an intracellular protein involved in synaptic function, are all pathological hallmarks of PD have been implicated in PD pathogenesis. However, it is debated whether α-syn aggregates themselves are responsible for neurodegeneration in PD, cellular pathways involved in degradation of α-syn aggregates are believed to promote neuron survival. The autophagy lysosomal pathway (ALP), a physiological mechanism for recycling of intracellular components, …

Examination Of Ideal Physical Parameter Choices For Efficacious Hyperthermic Treatment Of Cancer, Lauren Rast Jan 2011

Examination Of Ideal Physical Parameter Choices For Efficacious Hyperthermic Treatment Of Cancer, Lauren Rast

All ETDs from UAB

This dissertation presents results on the computational modeling of electromagnetic and thermal effects during the hyperthermic treatment of cancer using magnetic nanoparticles. Magnetic nanoparticle hyperthermia can be used for direct targeting and destruction of tumors through heat treatment or as a complement to chemotherapy. This method of treatment would be much less invasive than many current treatment options. Additionally, since these are nanoscale devices, the hyperthermic treatment of the cancer cells would be extremely localized. This would cause very minimal damage to surrounding tissue, making these systems superior to traditional hyperthermic treatment. The use of ferrofluids for cancer treatment requires …

Proper Orthogonal Decomposition For Parameter Estimation In Oscillating Biological Networks, Amanda Marie Rehm Jan 2011

Proper Orthogonal Decomposition For Parameter Estimation In Oscillating Biological Networks, Amanda Marie Rehm

All ETDs from UAB

Proper orthogonal decomposition (POD) is frequently applied to estimate parameters of partial differential equations. This study examines the application of the POD method in estimating the parameters of an Ordinary Differential Equation (ODE) model of stable oscillating biological networks. The model is related to the Lotka-Volterra equations. The findings reveal that POD generates accurate estimates of the parameters; furthermore, extrapolating biologically measured data points to a number of oscillations improves the curve fits, C1 approximations, and parameter estimations. KEYWORDS: Proper Orthogonal Decomposition, Ordinary Differential Equations, Biological Networks, Oscillations, Parameters.

Black Hollow, Candice Cottingham Reyes Jan 2011

Adolescents present a unique collection of characteristics and challenges for which middle school interdisciplinary teams were designed to address. Current research focuses on teacher-specific strategies which can be utilized in order to increase parental involvement. However, there is little literature addressing how an interdisciplinary team approach to involving parents alters the face of parent-teacher communication. The central research question for this study was, "What are the strategies utilized by interdisciplinary middle school teams to effectively involve the parents of their students in the educational process?" The researcher utilized a multiple-case study approach with three central Alabama middle school interdisciplinary teams: …

Design Space Decomposition Using Concept Maps, David E. Robbins Jan 2011

Design Space Decomposition Using Concept Maps, David E. Robbins

All ETDs from UAB

Decomposition of engineering design spaces has been performed primarily through statistical and system identification techniques. However, no approach has yet attempted to capture the semantics of a design through decomposition. This thesis presents graphical techniques for capturing the semantics of a design by developing an information architecture through design space decomposition using concept maps. In particular, concept maps are used to begin identifying the top layers of the information architecture of a problem, which has the dual effects of performing an initial decomposition of the design space and refining the parameters that will define more detailed design spaces for further …

Myosin Ii In Hippocampal Synapses: Regulation Of Synaptic Plasticity, Strength And Actin Dynamics By Two Distinct Isoforms, Maria Dolores Rubio Jan 2011

Myosin Ii In Hippocampal Synapses: Regulation Of Synaptic Plasticity, Strength And Actin Dynamics By Two Distinct Isoforms, Maria Dolores Rubio

All ETDs from UAB

Cytoskeletal actin filaments underlie dendritic spine plasticity, critical for several forms of learning and memory. Therefore, understanding the mechanisms that regulate actin dynamics is essential to elucidate memory formation pathways. Myosins, a superfamily of actin binding proteins, have emerged as candidates for regulation of actin dynamics in the brain. Several myosin class II isoforms have been identified in brain, but their individual contribution to synaptic activity is still unknown. Based on the finding that myosin IIB regulates actin polymerization in the growth cone of developing neurons and that it is necessary for maintenance of dendritic spine structure, I hypothesized that …

Identification Of Putative Gene Targets Of Pgc-1alpha In The Brain: Insights Into Inhibitory Neuron Maturation, Physiology, And Behavior, Elizabeth Lucas

All ETDs from UAB

The transcriptional coactivator peroxisome proliferator activated receptor γ coactivator 1α (PGC-1α) is a master regulator of energy homeostasis in peripheral tissues, and it has been assumed that PGC-1α plays a similar role in the brain. Previous work has shown that PGC-1α localizes to neurons that express high levels of the enzyme glutamic acid decarboxylase of 67 kDa (GAD67) during early postnatal development, leading us to hypothesize that PGC-1α regulates gene expression primarily in GABAergic neurons. We initially investigated the distribution of inhibitory neuron markers in PGC-1α -/- mice and observed a striking reduction in expression of the Ca2+-binding protein parvalbumin …

The Effects Of Simvastatin On Learning And Memory Mechanisms In Mice, Robert Mans Jan 2011

The Effects Of Simvastatin On Learning And Memory Mechanisms In Mice, Robert Mans

All ETDs from UAB

Statins, a widely prescribed class of cholesterol-lowering drug, inhibit HMG-CoA reductase, the rate-limiting enzyme in the cholesterol biosynthetic pathway. Due to the identification of cholesterol as a risk factor for developing Alzheimer's disease (AD), a number of studies have examined whether statins are neuroprotective against developing AD or dementia. While some epidemiological studies do indicate a lower prevalence of AD in people taking statins, other reports are contradictory. A consensus has therefore not been reached regarding the neuroprotective effects of statin treatment. Aside from the well-characterized reduction in circulating cholesterol resulting from statin therapy, numerous cholesterol-independent, pleiotropic effects have been …

EFAs were conducted for the entire NACC sample as well …

Gabaergic Signaling To Adult-Generated Neurons In Hippocampus, Sean Markwardt Jan 2011

Gabaergic Signaling To Adult-Generated Neurons In Hippocampus, Sean Markwardt

All ETDs from UAB

In the central nervous system of adult mammals, new neurons are produced throughout life in at least two regions, the subventricular zone and dentate gyrus of the hippocampus. New neurons in the SVZ migrate via the rostral migratory stream and eventually participate in adaptive olfactory processes. In the dentate gyrus, a variety of physiological functions for continued addition of neurons have been proposed, but its role in normal or pathological conditions remains largely unclear. Neurogenesis in the dentate gyrus consists of the stepwise process of generating fully functional granule cells (GCs) from adult neural stem cells. Regulation of neurogenesis can …
