Diagnostic Electrophysiology Of The Eye, Brooke Nicole Conner-Tsang Jan 2022

Diagnostic Electrophysiology Of The Eye, Brooke Nicole Conner-Tsang

All ETDs from UAB

Aging and age-related changes in the brain are known to parallel functional and anatomical changes in the eye, especially in certain degenerative diseases. This study explores the potential of combining ERG, specifically the Pattern Electroretinogram (PERG), which has a black and white checkerboard pattern, and OCT tests to monitor retinal ganglion cell health. We compared ERG and OCT findings from ten normal females, ages ranging from 18 to 30 years old to ascertain if ganglion cell layer volume is a reliable indicator of ganglion cell function as measured by ERG. Our results from these healthy subjects will serve as prelude …

Use Of Force By Correctional Officers: A Test Of General Strain And Scial Bond Theories, Benjamin Hixon Jan 2022

Use Of Force By Correctional Officers: A Test Of General Strain And Scial Bond Theories, Benjamin Hixon

All ETDs from UAB

Use of force in corrections is significantly less studied when compared to other fields, such as policing. This is a problem as unnecessary and unjustified use of force in corrections needlessly raises the violence level in correctional facilities, which endangers persons in custody and correctional officers alike. The current study examines use of force in corrections using a theoretical approach. First, Robert Agnew’s general strain theory is applied to understand use of force encounters between persons in custody and correctional officers. Second, Travis Hirschi’s social bond theory is used to investigate whether certain social bonds may relate to reductions in …

Correlates Of Early Adversity In Children And Adolescents: Examining The Interrelatedness Of Aggression, Post-Traumatic Stress, And Suicidal Behaviors, Elizabeth Mcrae Jan 2022

Correlates Of Early Adversity In Children And Adolescents: Examining The Interrelatedness Of Aggression, Post-Traumatic Stress, And Suicidal Behaviors, Elizabeth Mcrae

All ETDs from UAB

The negative outcomes associated with adverse childhood experiences, especially child maltreatment ACEs (CM-ACEs), often emerge concomitantly with these experiences and have cascading negative effects across the lifespan. For example, childhood aggression, post-traumatic stress, and suicidal behaviors have all been identified as negative sequelae of CM-ACEs. They have been associated with each other, as well, and share several common underlying mechanisms. Thus, it might be that these outcomes collectively exacerbate the risk of experiencing negative effects of CM-ACEs, yet most existing research has examined these risk factors independently. Moreover, these outcomes often emerge early in development, yet most related research has been …

Nox2-Derived Superoxide And Mda5-Dependant Responses Contribute To Autoimmune Diabetes, Samuel Isaac Blum Jan 2022

Nox2-Derived Superoxide And Mda5-Dependant Responses Contribute To Autoimmune Diabetes, Samuel Isaac Blum

All ETDs from UAB

NADPH oxidase 2 (NOX2)-derived superoxide, a precursor for many reactive oxygen species (ROS), regulates cell signaling, cellular trafficking, melanoma differentiation-associated protein 5 (MDA5) responses, and Type 1 diabetes (T1D). Previously, we demonstrated that NOX2-derived superoxide promotes chemokine synthesis, spontaneous and virus-accelerated T1D, and MDA5 responses to coxsackievirus B3 (CVB3), an associated environmental trigger for T1D. Yet, it remains unknown if ROS is necessary for diabetogenic T cell trafficking into islets or how MDA5 responses affect T1D. First, using non-obese diabetic (NOD) mice that contain an in-frame deletion within the helicase 1 domain of MDA5 (ΔHel1) or lack MDA5 expression (KO), …

Mice With Ghrh Deficiency Are Resistant To The Detrimental Health Effects Of High Fat Diet, Joseph Jablonsky

All ETDs from UAB

According to our previous study, mice with a CRISPR/Cas9 mediated deletion of the GHRH gene have decreased bodyweight, respiratory exchange ratio (RER), energy expenditure compared to wild type animals, and increased insulin sensitivity and enhanced longevity. Calorie restriction enhances lifespan and delays multiple aging related phenotypes; however it is not known how these animals respond to nutritional stressors such as high fat diet (HFD) during the aging process. We subjected 18 month old GHRH-/- male and female mice to a high fat diet to determine differences in response to HFD in aged animals. After HFD exposure, GHRH-/- mice have increased …

Spike-And-Slab Additive Models And Scalable Algorithms For High Dimensional Data Analysis, Boyi Guo Jan 2022

Spike-And-Slab Additive Models And Scalable Algorithms For High Dimensional Data Analysis, Boyi Guo

All ETDs from UAB

There are proposals that extend classical additive models (AMs) to accommodate high-dimensional data (p >> n) using group sparse regularization. However, the sparse regularization may induce excess shrinkage when estimating smooth functions, damaging predictive performance. Moreover, most of these AMs consider an “all-in-allout” approach for functional selection, rendering them difficult to answer if nonlinear effects are necessary. While some Bayesian models can address these shortcomings, model fitting can create a new challenge, scalability. In this dissertation, we propose Bayesian hierarchical additive models to address the previous shortcomings: we consider the smoothing penalty for proper shrinkage of curve interpolation via reparameterization. A …

Analysing The Structural Dynamics Of Retinoid X Receptor Using Hydrogen Deuterium Exchange Mass Spectrometry, Nathalia Melo

All ETDs from UAB

Nuclear receptors (NR) are ligand-activated transcription factors that are targets for drug discovery due to their ability to directly modulate gene expression. There are 48 NRs and approximately half of them require heterodimerization with retinoid X receptor (RXR) to function. Targretin (bexarotene) is an FDA-approved RXR specific (rexinoid) currently used to treat Cutaneous T-cell Lymphoma (CTCL); however, it causes hyperlipidemia toxicities. A functionally similar rexinoid developed at UAB, 9-cis-UAB30 (UAB30), is non-toxic and weak agonist in the liver. While UAB30 shows promise as a low toxicity chemopreventative drug, the mechanism of UAB30 action at the molecular level is poorly understood. …

Elucidating The Role Of Stat4 In Cd4 T Cell Mediated Pathogenicity, Ashlyn E. Anderson

All ETDs from UAB

While the exact cause of many autoimmune diseases is currently unknown, many genetic factors are associated with increased disease susceptibility such as single nucleotide polymorphisms in the STAT4 gene locus. Signal Transducer and Activator of Transcription 4 (STAT4) is critical for the development and function of Th1 cells; however, many Th1 associated molecules are dispensable for CD4 mediated inflammation such as IFNγ, T-bet, and IL-12Rβ2, suggesting that STAT4 may function outside of the Th1 cell lineage. Importantly, Stat4 has recently been correlated with an inflammatory Th17 cell phenotype. Using a murine model of neuroinflammation, we illustrate that Th17 intrinsic STAT4 …
