The effects of different carbon sources (ethanol, methanol and acetic acid) and C/N ratios (0,5; 1; 1,5; 2; 2,5) on the microbial denitrification in batch culture were investigated. For this purpose Paracoccus denitrificans and Pseudomonas stutzeri were used as denitrifiers. For all experiments, the initial NO 3 concentrations were constant at 100 mg/l. During the incubation period of 66 hours, nitrate, nitrite and microorganism concentrations were determined spectrophotometrically at certain intervals. The aim of the present study, which was carried out on 720 individuals between October 1995 and September 1996 in the mid Black Sea region was to estimate some parameters of the population of horse mackerel (Trachurus trachurus L. 1758) including age, size, weight, sex compositions, growth and mortality rates. Ages between 0 and 7 were established in horse mackerel stock. Cytogenetic Effects Of Copper Chloride On The Root Tip Cells Of Viciahirsuta (L.) S. F. Gray, Hüseyi̇n İnceer, Osman Beyazoğlu

The cytogenetic effects of copper chloride on root tip cells of Vicia hirsuta were investigated. Seeds were treated with 10, 25, 50 and 100 mg/L of the copper chloride for 4, 12, 24 and 48 hours. It was found that copper chloride has a marked mitodepressive action on mitosis. Mitotic abnormalities were increased and the mitotic index was decreased depending on the concentration and duration time of the copper chloride applied.

The Effect Of Soy Flour On Alcohol And Cell Concentration In Alcoholfermentation Of Molasses, Tuncay Ari, Sedat Dönmez

The effects of soy flour on alcohol fermentation of molasses by different yeast strains were investigated. Six strains of Saccharomyces cerevisiae and one Saccharomyces bayanus were used. It was found that soy flour affected fermentation time, alcohol concentration and total cell count in all strains. But the degree of this effect was different among strains. The maximum ethanol concentration were 63.9 g/l in Saccharomyces cerevisiae N3 strains. The maximum theoritical yield, however was 91.6% in Saccharomyces cerevisiae W4 strains. The effects of cadmium nitrate on the fecundity, larval survival and sex ratio of D. melanogaster were tested. According to our findings, the effects of cadmium nitrate can be summarized as follows: it appeared that when used as a feeding substance during the developmental period, cadmium nitrate did not have any effect on fecundity. On the other hand, if cadmium nitrate was given throughout adult life it caused a proportional decrease in the daily mean with increasing concentration. Furthermore, the larval survival ratio decreased due to the concentration of cadmium nitrate. Also cadmium nitrate application appeared to have no effect …

The effect of cadmium nitrate on the longevity of Drosophila melanogaster was tested. When cadmium nitrate was given throughout the adult life it caused a decrease in the mean life-span of flies in all treated groups. Furthermore, the effect of cadmium nitrate on young populations was also compared with aged populations. The young populations survived longer than aged populations with the food medium containing cadmium nitrate.

The Isolation And Characterization Of Glucoamylase Enzyme Of Anaspergillus Niger Natural Isolate, Gökhan Coral, Ömer Çolak

An extracellular glucoamylase enzyme was isolated from Aspergillus niger Z10 natural isolate in a liquid Czapek Dox medium which contained soluble starch (2%) as the sole carbon source. The molecular weight of the enzyme was found to be about 100.000 Daltons through Starch-SDS Polyacrylamide Gel Electrophoresis. The optimal pH range of the enzyme was found to be 4.5-5.5 and it was determined that the thermal stability of this enzyme is quite high.

Olive oil mill effluent (OOME) is one of the major pollutants produced by the agricultural industry in Mediterranean countries. The olive oil mills in these countries generate about 2-3 million tonnes of effluent annually. OOME is a black liquor containing a water-soluble fraction of ripe olives and water that is used in the process of olive oil extraction. This waste has a high mineral content and a chemical oxygen demand of 25.000-100.000 mg/l. depending on the olive oil extraction process used. The seasonal variation in total lipid and total fatty acid contents of the gonads of Cyprinion macrostomus(HECKEL, 1843) living in the Topardiç Brook (Kangal-Sivas) were investigated. It was observed that the variations in the gonad weights and the amount of total lipid and total fatty acid in the gonad of the male and female fish were more noticeable especially at the time of gonad development and the spawning period. The total lipid and total fatty acid amount which increased during the spawning period showed a decrease at the end of this period.

The effects of various concentrations (0.01, 1, 2mM) of spermine (Spm) on germination and some early growth parameters of sunflower ( Helianthus annuus L. cv. Santafe) seeds were investigated in media containing different concentrations of NaCl (50, 100, 200mM). Regardless of its concentrations, application of Spm under non-saline conditions remained ineffective on germination percentage as well as on the early growth parameters studied (length of radicle, fresh and dry weights). Karyotype analysis was performed in Chalcalburnus mossulensis (Heckel, 1843) belonging to the carp family by investigating the number and structures of their chromosomes. The fish used in this study were caught with fishing nets from Kizilirmak river and taken to the laboratory. They received an abdominal injection of 0.6 mg colchicine (Colchicum Dispert) dissolved in 1 ml of distilled water per 1 g body weight. As a result of the C-metaphase investigation, it was determined that C. mossulensis had 2n=48 chromosomes. Samples of water, bottom clay, plankton and fish were collected over a period of one year. The boron contents of the samples taken were determined in the laboratory. The differences and relationships in the obtained data were identified using various statistical methods. Boron gradually accumulates in the plankton, bottom clay and water of Çatören and Kunduzar. The effect of heavy metals, namely mercury (Hg^{+2}), zinc (Zn^{+2}), nickel (Ni^{+2}) and cobalt (Co^{+2}), which were applied in the form of chloride salt, and that of lead (Pb^{+2}) and cadmium (Cd^{+2}) applied in the form of nitrate salt, on the pollen germination and tube growth were investigated on apples ( Malus sylvestris cv. Apoptosis induced by hydrogen peroxide (H_2O_2 ) was investigated in Chinese hamster ovarian (CHO) cells, and compared to apoptosis induced by cisplatin (25 µM), a well-established apoptosis-inducing agent. In preliminary experiments, CHO cells were treated with low concentrations of H_2O_2 (250-500 µM) continuously or for 3 hours, followed by washing and further incubation in complete growth medium for 1-10 days. Fluorescence microscopy of acridine orange-stained cells indicated morphological changes of apoptosis. Similar changes were observed in cisplatin-treated cells. We report here the development of hybrid cells producing monoclonal antibodies specific for two different steroid hormones with mixed immunization using hybridoma technology. BALB/c mice were immunized with a mixture of three steroid antigens: progesterone, estradiole and testosterone linked to bovine serum albumine. These mice were used for fusion. In the two fusion experiments, ELISA tests showed that among 645 wells only 2 hybrids reacted with progesterone (MAM 3C2, MAM 3E3) and one of them reacted specifically with estradiol (MAM 7H7). This study was performed to determine the capability of embryonic stem (ES) cells at late passage number to form functional germ cells and to orientate the sexual development of chimeric mice produced from ES cell injection into embryos. When an R1 ES cell line was routinely cultured on mitotically inactivated feeder cells in culture medium supplemented with leukemia inhibitory factor (LIF), the colonies maintained an undifferentiated ideal ES cell morphology. Germin, a molecular marker of wheat embryo germination, is a remarkably protease-resistant, apoplastic, homopentameric glycoprotein. Different forms of germin found to be related to various aspects of plant development have been discovered. Germins were determined in cereals, dicotyledonous angiosperms, gymnosperms and in a prosit, all these proteins are together called germin-like proteins due to their DNA sequence homology. Cereal germins have oxalate oxidase activity, an activity which generates hydrogen peroxide by degrading oxalic acid. Thermomonospora fuscaBD25 produces relatively high levels of activity of endoxylanases. Only oat spelt xylan, RBB-xylan and birchwood xylan reacted with endoxylanases with the maximum specific activity of 47.65 U mg -1 protein on birchwood xylan. The endoxylanase remained stable up to 70 °C. At 60 °C endoxylanase activity showed stability (92 %) for 16 h, however it showed a half-life of 8 h at 70 °C and 30 min at 80 °C in the absence of substrate at pH 8.0. Thermomonospora fuscaBD25 produces relatively high levels of activity of \beta-xylosidase and \alpha-L-arabinofuranosidase. The aim of the work described in this study was to characterize some properties of \beta-xylosidase and \alpha-L-arabinofuranosidase produced by T. fusca BD25 when growing on oat spelt xylan as the main carbon and energy sources. The substrates p-NPX and p-NPA reacted with \beta-xylosidase and \alpha-L-arabinofuranosidase with specific activities of 4.01 U mg -1 protein and 0.35 U mg -1 protein, respectively. The fungus Phanerochaete chrysosporiumME446 was entrapped on polyurethane foam to synthesize auxin and abscisic acid (ABA). The maximum amounts of auxin produced on the 18 th day in both free and immobilized cells were 55.91mg/mL and 76.07mg/mL, respectively. Maximum ABA concentrations on the 12 th day in both free and immobilized cells were 6.87 mg/mL and 10.69 mg/mL, respectively. An immobilized system of auxin and ABA production resulted in a comparatively higher yield than free fungal mycelium.

In the current study, the biosynthesis of germin was determined in the organs of wheat embryos during germination. The temporal and spatial determination of germin protein was carried out by employing an anti-germin sera in gel-blot analysis. Germin was detected in the coleoptile, roots and vascular transition region of and 6-day-old germinated wheat seedlings. It was shown that the leaf tissue acquires germin accumulation only at the late stage of germination. The pellet fraction of protein homogenate obtained from embryos at different ages revealed higher germin accumulation than the supernatant fraction from the same sources.

The cultural characteristics of Morchella esculenta mycelium were investigated in potato dextrose agar, malt extract agar and complete medium yeast plates prepared with casein (cas), Casamino Acids (caa), peptone (p) and sodium nitrate (NaNO 3 ). Aerial and vegetative mycelium types were determined in the agar media with cas, caa and p. The third colonial characteristic was observed in the agar media with NaNO 3 . In this media, a colonization increase in circular forms spreading from the centre to the edges was determined. Mycelium colonization periods were 20-24 days in agar media with NaNO 3 .

Eighty-six Pseudomonas syringae pv. phaseolicola isolates collected from different bean growing areas in Eskişehir were typed for the production of bacteriocin.All the isolates tested produced bacteriocin and 24 bacteriosin groups were determined. No correlation was found between the bacteriocin groups and geographical origin. Authentic isolates of the bacterium representing 3 different races were also tested for bacteriocin production and bacteriocin types did not correlate with the races.

Native-polyacrylamide gel electrophoresis (N-PAGE) was used to characterize Candida albicansstrains isolated from vaginal specimens of patients suffering from vaginitis in Ankara hospitals, Turkey. Three different protein band profiles were observed among these 38 C. albicansisolates when whole cell protein extracts of these strains were used in N-PAGE. 34 of the 38 isolates were shown to belong to a single group or clone with a dissemination percentage of 89, while three isolates belonged to another group with a percentage of 8. Furthermore only one C. albicans isolate was detected with a dissemination percentage of 3 and assigned to a third group. …

The present study was undertaken to determine the relationship between Methicillin-Resistant Staphylococcus aureus (MRSA) strains isolated from various clinical specimens in Gülhane Military Medical Academy, in Ankara, Turkey. In 1997, at this hospital, the percentage of S. aureus among all nosocomial isolates was 20% and the percentage of MRSA among these S. aureus isolates was 51%. There are no reports in the literature dealing with gram-positive cocci typing by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of extracellular proteins. In this study, Staphylococcus aureus, Staphylococcus warneri, Staphylococcus chromogenes, Micrococcus lysodeikticus and Micrococcus ssp. of the Micrococcaceae family, and Pediococcus halophilus, Streptococcus ssp. and Enterococcus faecalis of the Streptococcaceae family were characterized by SDS-PAGE of whole cell proteins (WCPs) and total extracellular proteins (TEPs). Comparison of SDS-PAGE of WCP profiles with banding patterns produced by that of TEPs provided additional criteria and characteristic banding patterns for the study of the characterization of gram-positive cocci. The effect of crude extracts, obtained from Aconitum nasutum, Daphne glomerata, Hypericum androsaemum, Laurus nobilis, Nerium oleander, Olea europaea, Prunus laurocerasus, Punica granatum, Rhododendron caucasicum, and Urtica dioica on the replication of Autographa californica nuclear polyhedrosis virus (AcNPV) grown in Spodoptera frugiperda cell culture were investigated by observing changes in cytophatic effects, progeny virus concentration and viral protein (polyhedrin) synthesis.
