Lead-Induced Cytotoxicity And Mutagenicity In Grass Pea, Girjesh Kumar, Ritambhara Tripathi

Chromosomal variation is an important tool in genetic analysis. The objectives of the present study were to determine the mutagenic potential of lead (Pb) on the meiotic cells of grass pea (Lathyrus sativus) and to determine the maximum concentration of lead nitrate that induces maximum genetic variability in grass pea. The different concentrations of lead nitrate used were 25, 50, 100, 200, and 300 ppm. The percentage of abnormalities increased with the lead nitrate concentration. Different types of chromosomal abnormalities were observed, such as condensed bivalents, secondary association, laggards, bridges, cytomixis, stickiness, etc.

The effect of salinity on growth, photosynthetic pigment content, and alkaloid secondary metabolite accumulation were studied in an economically important medicinal plant, Catharanthus roseus (L.) G. Don., under pot culture conditions. Plants were treated with different concentrations of NaCl, (e.g. 50 and 100 mM) 30, 45, 60, and 75 days after sowing (DAS). The plants were uprooted randomly 90 DAS to analyse growth, and chlorophyll and alkaloid content. Salinity affected all the morphological parameters and decreased growth performance. At low salinity regimes, a slight decrease was noted in chlorophyll a and b, and total chlorophyll content, but under high salinity …

Soil salinization is a major problem in irrigated agriculture. A field study was conducted in the Sariosiyo district in the Surkhandarya region of southeast Uzbekistan to evaluate soil nitrogenase activity and nitrogen-fixing bacteria populations in saline serozem soils under wheat, maize, and alfalfa, as well as from adjacent fallow land. Composite soil samples were randomly collected from depths of 0-10, 10-20, and 20-30 cm in autumn, winter, spring, and summer, which were then 2-mm sieved and subjected to selected microbial population analysis and enzymatic studies. Protein quality in soybean seeds is strongly influenced by nutritional conditions. Glycinin (11S globulin) and \beta-conglycinin (7S globulin) are the 2 main proteins in soybean seeds. Protein quality of glycinin (11S) is higher than that of \beta-conglycinin (7S), due to the presence of higher amounts of s-containing amino acids (methionine and cysteine) in glycinin than \beta-conglycinin. Of the amino acids in glycinin, 3%-4.5% are methionine and cysteine but these important amino acids are not found in \beta-conglycinin (especially \beta-subunit). Evaluation Of Antioxidant And Antimicrobial Activity Of Seaweed (Sargassum Sp.) Extract: A Study On Inhibition Of Glutathione-S-Transferase Activity, Jayanta Kumar Patra, Sakti Kanta Rath, Karmabeer Jena, Vijaya Kumar Rathod, Hrudayanath Thatoi

Turkish Journal of Biology

In the present study, the free radical scavenging potentials (DPPH radical and hydroxyl radical), inhibition of lipid peroxidation, and glutathione-S-transferase and antimicrobial properties of Sargassum sp. extract were investigated. The tested extract exhibited a dose-dependent free radical scavenging action against DPPH radical and hydroxyl radical and antimicrobial activity. In addition, inhibition of lipid peroxidation and glutathione-S-transferase activities were also observed. The overall results have established that this seaweed could be used against several diseases and, in the food processing industry, to preserve foods.

In vitro culture systems were used to assess the growth inhibition of kinetoplastids by a variety of aminoglycosides. The parasites were allowed to grow to the stationary phase in the presence of varying concentrations of the drugs. The IC-50 for every drug was calculated by comparison with the control. The sensitivity of various leishmanial strains to these drugs was in the order of: G418 > hygromycin > paromomycin > neomycin; however, under our assay conditions gentamycin and kanamycin were non-leishmanicidal. Kluyveromyces lactis and Kluyveromyces marxianus are industrial yeasts widely used in the production of the \beta-galactosidase enzyme. Biosynthesis of \beta-galactosidase is controlled by glucose repression. In this study it was demonstrated that the derepression of \beta-galactosidase biosynthesis in these yeast strains is inhibited by high osmotic stress. It was found that the \beta-galactosidase activity of K. lactis and K. marxianus remained approximately at the repressed level when these yeast cells were subjected to NaCl-, KCl-, or sucrose-induced high osmotic stress. Derepression of \beta-galactosidase biosynthesis seems to be more sensitive to high osmotic stress in K. marxianus than in K. lactis. …

Comparative Antioxidant Activity Of Some Edible Plants, Zaha A. El-Agbar, Ashok K. Shakya, Nooman A. Khalaf, Makbula Al-Haroon

Turkish Journal of Biology

Terminalia chebula, Salvia officinalis L., Calamintha incana, Thymus vulgaris L., Matricaria chamomilla, and Nigella sativa are commonly used spices. In the present study the methanolic extracts of these spices were evaluated for their antioxidant activity by the 1,1-diphenyl-2-picryl-hydrazyl (DPPH) free radical scavenging method. All the methanolic extracts showed significant antioxidant activity. The IC_{50} of the methanolic extracts ranged between 3.8 ± 0.1 and 168.8 ± 2.1 \mug/ml, and that of ascorbic acid was 8.6 ± 0.1 \mug/ml. The study revealed that the consumption of these spices could exert several beneficial effects by virtue of their antioxidant activity.

The antioxidant properties of the methanol extracts of 10 medicinal plants were evaluated with the DPPH (1,1-diphenyl-2-picryl hydrazyl) test, hydroxyl scavenging test, and lipid peroxidation assay. The methanol extract of Emblica officinalis was the most active according to the DPPH test, with an IC_{50} value of 7.92 ± 0.1 \mug/ml, followed by Asparagus racemosus, with an IC_{50} value of 9.28 ± 0.2 \mug/ml. The minimum free radical scavenging activity was shown by Zingiber officinale, with an IC_{50} value of 44.48 ± 1.8 \mug/ml. Amygdalus lycioides var. horrida (Spach) Browicz (Rosaceae), also known as Prunus lycioides (Spach.) Schneid., is an endemic Iranian species of the genus Amygdalus. In Iranian traditional medicine, the aerial parts and roots of A. lycioides are used in the treatment of diabetes. Six flavonoids, i.e. quercetin 3-O-rhamnoside (1), luteolin 7-O-rhamnoside (2), isorhamnetin 3-O-rutinoside (3), kaempferol 3-O-rhamnoside (4), apigenin (5), and naringenin (6), have been isolated from the aerial parts of this plant. Using demand-feeders, time-place learning association of food was investigated in rainbow trout (Oncorhynchus mykiss) in 2 small raceways (TR1 and TR2). In order for the fish to be visually identified, they were tagged individually and their behaviour and distribution were monitored by cameras. Equipping with a demand-feeder, TR1 was assigned to test 2 times/1 place (2T/1P) while 2 demand-feeders were used in TR2 assigned to test 2 times/2 places (2T/2P). Variously substituted N-(4-phenyl-3-aroylthiazol-2(3H)-ylidene)benzamides (2a-t) were prepared by base-catalyzed cyclization of corresponding 1-aroyl-3-aryl thioureas with acetophenone in the presence of bromine. Some of the synthesized compounds were assayed in vitro for their antifungal activity and were found to exhibit low to moderate activity.

A series of novel di-[3(thiophen-2-yl-methyl)-4,5-dihydro-1H- [1,2,4]triazole-5-one-4yl]n-alkanes (2a-h) were obtained by the reaction of N'-1-ethoxy-2-thiophen-2-yl-ethylydene hydrazino carboxylic acid ethyl ester (1) and diamines. Compound 3 was reacted with aldehydes and 4-(arylidene-amino)-3-thiophen-2-yl-methyl-4,5-dihydro-1H-[1,2,4] triazole-5-ones (4, 5, and 8) with Schiff base character were synthesized. (4-(arylidene-amino)-5-oxo-3-thiophen-2-yl-methyl-4,5- dihydro-1H-[1,2,4]triazole-1-yl)-acetic acid ethyl esters (6, 7, and 9) were obtained by the reaction of 4-(arylidene-amino)-3-thiophen-2-yl- methyl-4,5-dihydro-1H-[1,2,4]triazole-5-ones (4, 5, and 8) and ethyl bromoacetate. The structures of the new compounds were inferred through IR, ^1H/^{13}C NMR, elemental analyses, and mass spectral data. An HPLC-ESI/MS method that enables fast detection and identification of iridoid glycosides is described. Eleven iridoid glycosides known to occur in the genus Lamium-lamalbide, sesamoside, 6-\beta-OH ipolamiide, shanzhiside methyl ester, dehydropenstemoside, barlerin (= 8-O-acetylshanzhiside methyl ester), 6-O-syringyl-8-O- acetylshanzhiside methyl ester, lamerioside, lamiide, eriobioside, and ipolamiide, were identified by means of their retention time and ESI/MS data. In this study, the antioxidant activity of ethanol extract of Salvia halophila Hedge (Lamiaceae) was evaluated by 2 antioxidant assays, including phosphomolybdenum reduction and free radical scavenging activity. The ethanol extract showed free radical scavenging activity with IC_{50} of 67.73 \mug/mL in 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. The antioxidant activity of the extract was found to be 84.87 ± 0.7 mg ascorbic acid equivalent (AAE)/g extract in phosphomolybdenum assay. The interaction effects of kinetin and flooding on growth indexes, chlorophyll content, and the activity of 5-aminolevulinic acid dehydratase (ALA-D; EC 4.2.1.24) in seedlings of corn (Zea mays L. cv. 704) were investigated. Samples were taken 48, 96, 144, and 192 h after the start of treatment. Growth criteria (root length, shoot height, and dry weight) were inhibited more in flooded plants than in control plants. The pattern of growth changes was similar to chlorophyll a and b, as well as ALA-D activity. Production of alkaline protease from B. licheniformis N-2 was studied in shake flask experiments in growth medium comprised of (g/l): glucose, 10; soybean meal, 10; K_2HPO_4, 5; MgSO_4.7H_2O, 0.5; NaCl, 0.5; CaCl_2.2H_2O, 0.5. Different process parameters, such as fermentation period, initial pH, incubation temperature, agitation speed, and size and age of inoculum, were optimized for the maximum yield of the alkaline protease. Maximum enzyme production (991.04 PU/ml) was obtained after 24 h of incubation at 37 °C in growth medium of pH 10. Swollen chitin, flake chitin, powder chitin, and mushroom paste were used as substrates for chitinase production by Serratia marcescens GG5 in submerged fermentation. Enzyme production was 0.3 U/ml when the organism was grown in M9 medium supplemented with 0.5% swollen chitin and 0.5% soluble starch. Scanning electron microscopy revealed that Serratia marcescens GG5 digested the chitin flakes by producing chitinase.

The aerial part of Equisetum telmateia Ehrh. is used against various conditions including stomachache and peptic ulcer in Turkish folk medicine. In order to prove the claimed anti-ulcerogenic potential of the plant, aqueous and methanol extracts were prepared from aerial parts of E. telmateia and investigated for their anti-ulcerogenic effect on ethanol-induced gastric ulcer model in rats. Pharmacological experiments clearly demonstrated that the oral administration of both methanol and aqueous extracts of Equisetum telmateia Ehrh. plant showed significant stomach protection (77.9% and 100% ulcer inhibition, respectively) against the applied model of ulcerogenesis (P < 0.001). Since both extracts were found significantly active, 80% ethanol was preferred for the extraction of the plant material and this crude methanolic extract was further fractionated by successive solvent extractions with chloroform and H_2O saturated n-BuOH to obtain the chloroform and n-BuOH extracts as well as the remaining aqueous extract. The 80% ethanol extract and the solvent extracts were then submitted to pharmacological assay using the same in vivo experimental ulcer model. All the extracts statistically showed potent anti-ulcerogenic effect on this model (P < 0.001). However, the remaining aqueous extract was found prominent (98.2% ulcer inhibition). Consequently, folkloric utilization of E. telmateia on peptic ulcer has been confirmed in the present study by using ethanol-induced experimental ulcer model in rats.

The aim of this study was to evaluate the antimutagenic activity of the methanol extract of Origanum vulgare L. subsp. vulgare. Antimutagenic activity was estimated by employing the plate incorporation AMES/Salmonella histidine reversion assay. The base pair substitution tester strain Salmonella typhimurium TA1535 and the frame shift mutagen strain S. typhimurium TA1538 were used against direct acting mutagens - sodium azide (NaN_3), 4-nitro-1-quinoline oxide (4NQO) and the S9-dependent mutagen 2-aminofluorene (2AF). The results suggested that in the absence of S9 metabolic activation, all 3 doses (5, 0.5, and 0.05 \mug/plate) of the plant extract tested caused statistically significant (P < 0.05) antimutagenic activity on TA1535 strain, but not on TA1538 strain. However, in the presence of S9 microsomal fraction, the plant extract exerted moderate antimutagenic activity against the 2AF mutagen and reduced mutant colonies in TA1535 and TA1538 strains. As a result, the methanol extract of Origanum vulgare subsp. vulgare showed antimutagenic effects at 5, 0.5, and 0.05 \mug/plate concentrations. These effects may be explained with the antioxidant activity mechanism, changes in membrane lipids, and permeability of ion channels.

Phytoplankton Species Composition And Seasonal Variation At Wadi Al-Arab Dam Lake, Jordan, Ismail M. Saadoun, Erwah Bataineh, Adil Alhandal

The composition of phytoplankton species thriving in Wadi Al-Arab Dam Reservoir, Jordan and their seasonal variation in relation to water ecology were studied between February 2001 and February 2002. A total of 75 species of phytoplankton were identified: Bacillariophyceae (diatoms) contributed with the highest number of taxa (39) followed by Chlorophyceae (23), Cyanophyceae (9), and Dinophyceae (4). Diatoms constituted 52% of the total phytoplankton flora of the reservoir and the most common species were Synedra ulna, Cymbella affinis, and Cyclotella meneghiniana with pennate and centric diatoms being the dominant species in May and October-November, respectively.
