Significance Of Absorption Spectra For The Chemotaxonomic Characterization Of Pigmented Bacteria, Nuretti̇n Şahi̇n

Turkish Journal of Biology

A method for characterizing pigmented bacteria based on the numerical analysis of their carotenoid absorption spectra was examined. To achieve this, some non-phototrophic pigmented bacteria that were isolated from various environments and closely related types of species were used. Bacterial carotenoid pigments were extracted with an acetone:methanol (7:2) mixture and the absorption spectra were determined. Data were normalized, and their first derivatives were analyzed using Ward clustering algorithms. Based on the spectral data, isolates were separated into 7 groups. Isolation And Identification Of Trichoderma Species From Different Habitats And Their Use For Bioconversion Of Solid Waste, Ahsanur Rahman, Most. Ferdousi Begum, Matiur Rahman, M. A. Bari, G.N.M. Illias, M. Firoz Alam

Turkish Journal of Biology

The native population of Trichoderma species collected from different habitats was quantified and characterised. The identified strains are T. harzianum (IMI-392432, 392433, 392434); T. pseudokoningii (IMI-392431) and T. virens (IMI-392430). Out of five strains, T. harzianum was the most common in all of the habitats. Colony forming units (cfu's) of Trichoderma species varied significantly (P = 0.05) in different habitats and were positively correlated with the physico-chemical characteristics of the habitat. Hot spring sediment and soil samples from West Anatolia in Turkey were investigated for the occurrence of thermophilic Actinomycetes. Isolation was done using Actinomycetes isolation agar (AIA), starch casein agar and glycerol yeast extract agar at 55 °C. Extracellular protease activity of the isolates were screened by using AIA plus 1% casein according to the hydrolysis zones surrounding the colonies. Antimicrobial activities of the isolates were also screened by using streaked plate method against a panel of test bacteria. The present investigation was undertaken to study the frequency and spectrum of morphological mutants induced by gamma rays and EMS in chickpea (Cicer arietinum L.). The seeds of 2 varieties of chickpea, Pusa-212 and Pusa-372, were treated with gamma rays (150 Gy, 200 Gy, 300 Gy, and 400 Gy), EMS (0.1%, 0.2%, 0.3%, and 0.4%) and combination treatments (200Gy+0.2%EMS, 300Gy+0.2%EMS, 200Gy+0.3%EMS, and 300 Gy+0.3%EMS). The M2 population was carefully screened for various viable macromutations. The aerial parts and seeds of Sophora alopecuroides L. var. alopecuroides growing in Turkey were investigated for their alkaloid compositions and antimicrobial activities. The alkaloid extracts were analysed by capillary gas chromatography-mass spectrometry (GC-MS). The main components were identified as sophoridine (33.07%), matrine (24.03%), and sophocarpine (8.01%) in the aerial parts, and matrine (36.67%) and sophoridine (34.53%) in the seeds of the plant. The alkaloid extracts were also evaluated for their in vitro antibacterial and antifungal activities using microdilution techniques. Levels Of Phenolic Compounds And Their Effects On Antioxidant Capacity Of Wild Vaccinium Arctostaphylos L. (Qare-Qat) Collected From Different Regions Of Iran, Tahereh Hasanloo, Roshanak Sepehrifar, Homa Hajimehdipoor

Antioxidant activities in the leaves and fruits of wild Vaccinium arctostaphylos (locally named Qare-Qat) from different regions of Iran were evaluated using the stable 1, 1-diphenyl-2-picrylhydrazyl free radical and ferric reducing/antioxidant power methods. Variations in antioxidant activity, total phenolic, anthocyanin, and flavonoid content in fruit samples among genotypes was much greater than the variation observed between growing seasons, indicating that genetics plays a more important role than growing season in V. arctostaphylos fruits. RNA interference (RNAi) pathway is a gene silencing process during which small double-stranded RNA (dsRNA) molecules trigger the degradation of homologous RNA targets. Small interfering RNAs (siRNAs) are the mediators of the RNAi that can be induced in vitro and in vivo by direct application of chemically synthesized siRNAs. : The present study aimed to determine the genetic relationship between Poa angustifolia and Poa trivialis as well as intraspecies genetic diversity using inter simple sequence repeat (ISSR) markers. ISSR amplification was evaluated for its applicability to narrow leaved bluegrass and rough bluegrass accessions identification. A total of 401 bands, 363 of which were polymorphic (90.52%), were generated using selected 20 primers. Venom from the endoparasitoid Pimpla turionellae L. (Hymenoptera: Ichneumonidae) contains a mixture of biologically active components, which display potent paralytic, cytotoxic, and cytolytic effects towards hosts. Here, we further investigate if parasitism or envenomation by P. turionellae alters total protein of its host Galleria mellonella L. (Lepidoptera: Pyralidae). Various venom concentrations representing doses previously determined to yield host responses yet fall below the calculated LD99 were used for pupae and larvae. Parasitization was only assayed for host pupa since P. turionellae females normally parasitize host prepupae and pupae in nature. Forty-five fungal isolates were obtained from sclerotia of Rhizoctonia solani on potato tubers in Erzurum, Turkey. The interaction between fungal isolates and R. solani was studied in dual culture technique. Some fungal isolates affected R. solani by antibiosis and/or parasitism. Results of the antagonism tests showed that Acremonium sp., Gliocladium viride, Paecilomyces marquandii, Paecilomyces sulphurellus, Penicillium camemberti, Penicillium expansum, Penicillium frequentans (ME-50), Penicillium nigricans, Penicillium olsonii, Penicillium phialosporum, Sporothrix sp. (MCY-4), Sporothrix schenckii, and Verticillium dahliae isolates produced an inhibition zone in front of the R. solani colony to a varying degree. A series of studies were carried out to determine the succession and phytoplankton community of the Melen River in the Western Black Sea region of Turkey. The study was conducted at 6 stations at monthly intervals between May 2003 and April 2004. It was observed that the abundance and biovolume of phytoplankton were low in winter, though higher values were recorded in late spring and summer. The aim of the present study was to investigate the antibacterial activities of 25 propolis samples collected from various geographical regions of Turkey against 2 food-borne pathogens, Salmonella Enteritidis ATCC 13076 and Listeria monocytogenes ATCC 1462. The chemical compositions of ethyl alcohol extracts of the propolis (EEP) samples were determined by gas chromatography coupled to mass spectrometry. The main components of EEP samples were flavonoids, aromatic acid esters, aromatic alcohols, aromatic acids, aliphatic carboxylic acids, terpens, and aliphatic carboxylic acid esters. We analyzed the effects of sodium hypochlorite and Plant Preservative Mixture (PPM) on surface sterilization. We also examined the effects that the addition of an antibiotic-antimycotic solution to the culture medium had on callus induction. Explants were initially sterilized with different concentrations of sodium hypochlorite and cultured on MS media containing kinetin (0.29 \muM) and naphthalene acetic acid (NAA, 10 \muM). No calluses were produced, either because of contamination of the explants, or loss of explants as a result of the high levels of sodium hypochlorite. To evaluate possible interactions between the renin-angiotensin system (RAS) and the sympathetic nervous system (SNS) in the systemic circulation upon the intravenous administration of angiotensin II and a set of adrenergic agonists in normal Sprague-Dawley (SD) rats. Sonchus erzincanicus (Asteraceae) is a plant species that is endemic to Turkey and grows in Erzincan. Lipid peroxidation inhibition, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, and total phenolic content of the methanol and water extracts of the plant were determined. Antimicrobial activity of the extracts was also tested against 107 clinical isolates of human pathogenic microorganism strains belonging to Enterobacter aerogenes, Escherichia coli, Proteus mirabilis, Pseudomonas aeruginosa, Staphylococcus aureus, and Candida albicans using the disc-diffusion method, and the minimal inhibition concentration (MIC) value of each active extract was determined. The efficacy of boric acid (BA) was examined on liver marker enzymes in aluminum (Al)-treated rats. Also, a liver micronucleus assay was performed to evaluate the genotoxicity in hepatocytes. With these aims, Sprague-Dawley rats were randomly separated into 10 groups of 5 animals. Aluminum chloride (5 mg/kg body weight (b.w.) AlCl3) and BA (3.25, 13, 36 and 58.5 mg/kg b.w.) alone were administered with injections to the experimental animals. A total of 94 marine actinobacterial isolates were found to be associated with 4 sponges: Callyspongia diffusa, Mycale mytilorum, Tedania anhelans, and Dysidea fragilis. Among the cultures 7 strains showed antagonism against fish and shellfish pathogens. Analysis of morphological, physiological, biochemical, and protein profile characteristics suggested that the actinobacterial strains belonged to the genus Streptomyces. The initial screening of the isolates by the spot inoculation method exhibited antibacterial activity against Aeromonas hydrophila. Determining Mitotic Index In Peripheral Lymphocytes Of Welders Exposed To Metal Arc Welding Fumes, Ceylan Alakoç, Hali̇l Erhan Eroğlu

Gas metal arc welding is a widely used method in a lot of industrial areas as it is cheap and produces high quality results. In the present study, peripheral blood lymphocytes of 23 welders and 25 non-exposed subjects were monitored for cytotoxicity. Information regarding the subjects' ages, smoking habits, alcohol consumption, duration of exposure, and medicine usage was recorded. According to the results, mitotic index (2.278 ± 1.325) of the welders was higher than that of those non-exposed (0.956 ± 0.616) (P < 0.01). There was no significant difference between mitotic index rates of smoking and non-smoking subjects. In contrast, there were significant differences between non-exposed subjects and welders for both smoking and non-smoking (P < 0.05). It was found that as the time of exposure to welding fumes increased so did the mitotic index rates (P < 0.05). Both a positive correlation in welders and a negative correlation in non-exposed subjects were observed between mitotic index frequency and age. In the light of the results obtained, it is advised that welders who work with gas metal arc welding be instructed and necessary protective measures be taken because of the risk caused by metal arc welding fumes.

In this study genetic variation was determined with isozyme analysis in 8 natural house fly (Musca domestica L.) populations from the western and southern coasts of Turkey, a part of the southern Palearctic region. For this purpose, 7 isozyme systems (ACP, AOX, CAT, HBDH, HXK, MDH, and SOD) were resolved using the native polyacrylamide gel electrophoresis (native-PAGE) technique. A total of 9 loci and 20 alleles were detected. The proportion of polymorphic loci in populations ranged from 44.44% to 66.67%. The Analgesic Effect Of The Ethanolic Extract Of Matricaria Aurea, Esam Qnais

The analgesic effect of the ethanolic whole plant extract of Matricaria aurea L. (Asteraceae) was studied in rats using the cold water tail flick assay and in mice using the tail immersion, tail clip, acetic-acid-induced writhing, and formalin pain tests. The results showed dose-dependent and significant (P < 0.001) increases in pain threshold at 60 min post-treatment with doses of 100, 200, and 300 mg/kg of the extract in tail flick, tail immersion, and tail clip tests. The effects of the extract were significantly (P < 0.001) lower than those produced by morphine (10 mg/kg) in the same tests. The extract (100, 200, and 300 mg/kg) exhibited a dose-dependent inhibition of writhing and also showed a significant (P < 0.001) inhibition in both phases of the formalin pain test, but with a less intense effect in the first phase. The results indicate that the analgesic effect of M. aurea ethanolic extract is both centrally and peripherally mediated.

In the present study, the phenolic contents and antioxidant activities of the water and methanol (MeOH) extracts obtained from the root, herb, and fruits of 4 species of Prangos (Prangos ferulacea, P. heyniae, P. meliocarpoides var. meliocarpoides, and P. uechtritzii) collected in Konya province were compared. The sensitivity of sugar beet leaf spot disease caused by Cercospora beticola to fungicides used in Turkey was determined using a mycelial growth assay in petri dishes with 100 fungal isolates. The isolates were obtained from samples collected from 6 sugar beet production areas in 2005 and 2006. Flutriafol, mancozeb, and fentin acetate were added to Aspergillus complete medium (ACM) at the rates of 0.0, 0.05, 0.1, 0.5, 1.0, 5.0, and 10.0 µg mL^{-1}, and then mycelial growth was measured.
