Molecular techniques are used today in many areas and have increased the confidence level of studies. The aim of the study was to compare the classical morphological characters of coccoid green algae under field and culture conditions by using modern molecular analysis. To achieve this aim, we first isolated the coccoid green algae from Liman and Cernek lakes. The morphological variations of green algae were observed under culture conditions and the phylogenetic relationships of these strains were defined according to the 18S rRNA gene sequences. Beauveria bassiana (Balsamo) Vuillemin and Metarhizium anisopliae var. anisopliae (Metschnikoff) Sorokin are the most common entomopathogenic fungi used in microbial control programs all over the world. Assessments of the genetic variability of these 2 important species are useful for the development of effective biocontrol strategies and for evaluating the impact of artificial epizootics. The present experiment was undertaken to determine the mutagenic potential of lead on the cytomorphology of Trigonella foenum-graecum L. and to determine the maximum concentration of lead nitrate that induces maximum genetic variability. The seeds were treated with 6 different concentrations (10, 25, 50, 100, 200, and 400 ppm) of lead nitrate. The higher concentrations of lead nitrate significantly reduced plant height, number of branches per plant, pollen fertility, and yield; higher concentrations also caused variation in the plants at the seedling and mature stages. Mucuna pruriens belongs to the family Papillonaceae. The seeds and leaves of these plants contain a large amount of L-DOPA. L-DOPA (L-3,4-dihydroxyphenylalanine) is one of the most widely used drugs in the treatment of Parkinson disease. Development of an efficient gene transfer method is an absolute requirement for genetically improving Mucuna pruriens and creating plants with more desirable traits. Determination Of Genetic Relationships Among Velezia L. Caryophyllaceae) Species Using Rapd Markers, İlham Eröz Poyraz, Emel Sözen, Ebru Ataşlar, İsmai̇l Poyraz

Random amplified polymorphic DNA (RAPD) markers were used to determine genetic relationships among Velezia L. species from Turkey. A total of 432 amplified bands were obtained using 14 RAPD primers. The polymorphism in RAPD markers was high (98.61%) and was sufficient to distinguish each species. The degree of band-sharing was used for evaluating the genetic similarity between species and for constructing a dendrogram by the unweighted pair group method with arithmetic mean (UPGMA). İzmir Çamaltı Saltern is the biggest seawater-based saltern in Turkey. To date, it has not been investigated extensively for the existence of poly-3-hydroxybutyrate (PHB)-producing species. In this study, 14 extremely halophilic archaea were isolated, purified, and screened for PHB production. One strain was then selected as the best PHB producer, further cultivated in different PHB media, and compared with a positive control, Haloferax mediterranei ATCC 33500. PHB was extracted from cells and measured with a spectrophotometer, and then the amount of PHB was measured through comparison with standard PHB. Cooling towers are heat rejection systems that are used in some industrial applications, and they have the potential to develop infectious concentrations of Legionella pneumophila. SANIPACKING® cooling tower fill material and standard polypropylene fill material were compared in terms of biofilm formation potential and anti-Legionella activity within a 4-month period using a laboratory-scale recirculating water system. The recirculating water system was experimentally infected with a L. pneumophila standard strain (ATCC 33152) suspension and operated continuously until all experiments had been completed. To investigate the genetic diversity and phylogenetic relationships of Salvia miltiorrhiza and related species, we analyzed the nuclear ribosomal DNA internal transcribed spacer (ITS) region for 7 accessions of Salvia miltiorrhiza and another 23 samples from other taxa within the genus Salvia by maximum parsimony and Bayesian inference analyses. There were 257 variation sites amounting to 40.8% of the total base pairs. All of the data revealed abundant genetic diversity in the genus Salvia. The aim of this study was to compare the salinity tolerances of 2 oriental tobacco varieties (İzmir Özbaş and Akhisar 97). Salinity stress experiments were performed under both in vitro and in vivo conditions. Seedlings of each variety were subjected to 0, 50, 100, 150, 200, 250, 300, and 350 mM NaCl. Photosynthetic pigment levels, lipid peroxidation rate, total protein content, antioxidant enzyme activities, and proline concentrations were determined for seedlings treated with salt for 14 days. This study explored the level of infection caused by different inoculum concentrations (10^2, 10^3, and 10^4 zoospores mL^-^1) of Phytophthora capsici in 3 pepper cultivars at days 2, 4, and 6. The effect that the infection induced on the peroxidase (POX), catalase (CAT), and phenolics of resistant and sensitive seedlings, as well as the defense mechanism against the pathogen, were also investigated. The resistance of PM-702 against the isolate used was high, whereas KM-Hot and DEM-8 displayed sensitive reactions. Antioxidant Activity And Luteolin Content Of Marchantia Polymorpha L., Alper Gökbulut, Basri̇ Satilmiş, Kadi̇r Batçioğlu, Barbaros Çeti̇n, Engi̇n Şarer

Turkish Journal of Biology

Antioxidant activity of the methanol and ethyl acetate extracts of Marchantia polymorpha L., which grows naturally in Turkey, was tested by DPPH and ABTS methods. According to the applied methods, both extracts exhibited moderate activity. Some phenolic compounds that could be responsible for the activity were investigated by RP-HPLC, qualitatively and quantitatively, in the methanolic extract of liverwort. While caffeic acid, chlorogenic acid, rutin, myricetin, quercetin, and kaempferol were not found in the extract, luteolin as the liable antioxidant agent was determined as 0.0052 ± 0.0002% among the investigated compounds.

An obligate alkaliphilic Bacillus strain was isolated from a soil sample and identified as Bacillus cohnii APT5 based upon phylogenetic and phenotypic analyses. The optimum growth pH of B. cohnii APT5 was 10.0. B. cohnii APT5 was also found capable of producing an extracellular alkaline protease that showed optimum activity (693,318 U/min) at 50 °C and pH 11.0 when grown in a medium containing casein. The enzyme was partially purified 3.22-fold with a yield of 78.74% after acetone precipitation and cation exchange column chromatography, respectively. Gypsy moth, Lymantria dispar L. (Lepidoptera: Lymantriidae), is one of the most serious pests affecting fruit, forest, industrial, and foliage plants worldwide. The aim of this study was to isolate and characterize pathogenic bacteria from this pest and determine their pathogenicity. L. dispar larvae were collected from various populations in the forest areas of the Black Sea region of Turkey in May and June of 2008 and 2009, and 9 culturable bacterial isolates were obtained from these larvae. Copper-tolerant bacteria were isolated from soil samples taken from a region where metal industries are located. After selecting 2 isolates with relatively higher bioremoval efficiency, the effects of increasing copper concentration, pH, and temperature on the bioremoval efficiency of the growing isolates were determined. Strain N1c and strain N5a showed maximal bioremoval efficiency of 82% and 75%, respectively, in 20 mg/L copper-containing medium at pH 6.8 and 30 °C. One of the natural reservoirs of potentially human-pathogenic bacteria is believed to be the rhizosphere. The aim of the present work was to test nontuberculous mycobacterium Mycobacterium phlei MbP18 for its ability to colonize the rhizosphere of wheat and to evaluate its effect on plant growth under saline conditions. In competitive wheat root tip colonization assays, M. phlei MbP18 showed poor competitive colonization of the wheat rhizosphere compared to the reference strain. Oxidative stress responses were evaluated in the digestive gland tissue of Perna viridis exposed to water-accommodated fractions (WAFs) of petrol and diesel at 0.5% and 5% concentrations over 5-, 10-, and 15-day periods. Increased lipid peroxidation, protein carbonyl, DNA integrity, and hydrogen peroxide in WAF-exposed specimens are indicative of oxidative damage due to dissolved petroleum extracts. A consistent increase in superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase, glutathione-S-transferase, reduced glutathione, and ascorbic acid in WAF-exposed specimens signify the importance of antioxidants in protecting the cell against the oxidative damage arising from petroleum contamination. Glutathione S-transferase (GST) is the one of most widely used affinity tags in biotechnology applications. The present study named a GST gene as the TtGSTz1 gene, characterized with the conserved glutathione binding motif (SSTSWRVRIAL) under a GST zeta subfamily from Tetrahymena thermophila. Phylogenetic analysis of the TtGstz1p protein sequence, with its orthologs from different GST classes, showed that it is a member of the unicellular GSTz monophyletic clade, which is positioned close to the bacterial GSTz clade. In this study, the phytase from Lactobacillus brevis strains isolated from fermented food products was produced and characterized. Phytase production of this strain was determined by production of clear zones around the colonies on the sodium phytate containing medium, after 48 h of incubation at 30 °C. The molecular weight of the phytase from Lactobacillus brevis was estimated as 73 kDa and 34 kDa on SDS-PAGE. The optimum activity of Lactobacillus brevis extracellular and intracellular enzymes emerged at 120 °C, pH 3.4 and at 110 °C, pH 3.4, respectively. The genus Fritillaria embraces about 165 taxa in the family Liliaceae. In this study, the molecular phylogenetic relationships between 10 Fritillaria taxa were presented. Fritillaria spp. were collected from their natural habitats, and specimens with different morphological features were obtained via selective breeding. Specimens with the desired characters were presented as promising candidate cultivars for ornamental use. Phylogenetic analysis was based on DNA sequences of the chloroplast trnL-trnF region. The phylogeny was constructed using the neighbor joining, maximum parsimony, maximum likelihood, and Bayesian inference methods. The persistence of endosulfan and intermediate metabolite endosulfan sulfate in the environment and their toxic effects on biota necessitate their removal. This study investigated the bioaugmentation of endosulfan-contaminated soil by fungal inoculant Aspergillus niger ARIFCC 1053. The influence of bioaugmentation by A. niger on endosulfan-contaminated soil was evaluated with the help of change in pH and released chloride, and by thin layer chromatography and gas chromatography analysis. Its effects on soil functionality were monitored by estimating dehydrogenase and arylsulfatase enzyme activities. The endosulfan degradation reached an undetectable level on day 15. The aim of this study was to evaluate the genotoxic effect of a bovine bone-derived hydroxyapatite (HA), Boneplus-xs (B-P-XS). The genotoxicity of B-P-XS was investigated by examining the potential of the xenograft (B-P-XS) to induce gene mutation (evaluated using the Ames test), in vitro chromosome aberration (CA), and sister chromatid exchange (SCE) with tests in cultured human peripheral lymphocytes. Alocasia macrorrhizos (L.) (Araceae), a coarse and erect plant that is widely cultivated to be eaten as a vegetable throughout Bangladesh, has been reported to possess a number of medicinal properties. The antihyperglycemic, antioxidant, and cytotoxic effects of the methanol extract of A. macrorrhizos rhizomes were investigated. The extract was given at a single dose (250 and 500 mg/kg) in alloxan-induced hyperglycemic mice. The extract at 500 mg/kg produced a significant (P < 0.05) decrease in the blood glucose level (55.49%) at 8 h of treatment when compared with the control and was comparable with the reference drug, metformin. The extract was also subjected to antioxidant potentiality and brine shrimp lethality bioassays. The IC_{50} value of the extract was 693.0 \mug/mL and the LC_{50} was 188.14 \mug/mL.

In vitro antioxidant, antimicrobial, and antiproliferative activities of methanol, acetone, and ethyl acetate extracts of Sedum acre L. were determined. Their total phenolic contents as well as the concentrations of flavonoids were also evaluated. The total phenolic content was determined with Folin-Ciocalteu reagent and it ranged between 18.25 to 181.75 mg gallic acid/g. The concentration of flavonoids in various extracts of S. acre was determined by using a spectrophotometric method with aluminum chloride and the obtained results varied from 8.42 to 173.42 mg rutin/g. Wheat landraces could be unique sources of favorable genes for agronomic traits like yield and insect, pest, and disease resistances, as well as quality characteristics that are important for breeding programs. Molecular marker systems offer great opportunities to characterize wheat accessions, which in particular are not morphologically identifiable. In this study, 20 bread wheat landraces collected from different regions of Turkey were characterized by using microsatellite markers (SSRs) and morphological characters. Hyperthyroidism increases the vulnerability of the heart to ischemia-reperfusion (I/R) injury. Regarding the fact that diazepam also affects cardiac I/R injury, the current study was conducted to investigate the effect of diazepam on ischemia-reperfused isolated hyperthyroid rat heart. The animals were divided into 6 groups: the control, hyperthyroid, control diazepam (1 mg/kg), hyperthyroid diazepam (1 mg/kg), control diazepam (5 mg/kg), and hyperthyroid diazepam (5 mg/kg) groups. Langendorff-perfused rat hearts underwent 40 min of global ischemia and 45 min of reperfusion.
