A large amount of parasite biomass production necessitates a higher volume of culture media, and this further increases the overall cost. For this reason, novel and alternative culture supplementary materials have been continuously investigated. The aim of this study was therefore to evaluate the effect of cell-conditioned media, collected as waste material during routine cell culture, on the growth and development of Leishmania parasites in vitro. In this study, for the first time, we show that cell-conditioned medium obtained from MCF-7 culture increased the growth of Leishmania parasites in vitro. In this study, our aim was to develop a new simple technique for isolation of mesenchymal stem cells from adipose tissue. For this purpose, mesenchymal stem cells were isolated from rat adipose tissue by using the primary explant culture technique. When the cells became confluent, they were passaged 4 times by using the standard trypsinization method with trypsin/EDTA solution. Twenty-seven aqueous extracts obtained from 21 plants used in the treatment of respiratory tract infections as folk remedies in Turkey were investigated for their relative total phenolic contents and antioxidant, antibacterial, antimycobacterial, and antifungal activities. Antibacterial and antifungal activities against dermatophytes and yeast-like fungi were determined using the microdilution method. Antibacterial activity of all extracts was more pronounced against gram-positive bacteria than against gram-negative bacteria. These extracts also exhibited higher antifungal activity against Candida krusei than ketaconazole. Six extracts displayed antidermatophytic activity against Epidermophyton floccosum, while only 3 extracts were highly active against Trichophyton rubrum. The extracts showed strong activity …

The Expression Of Gst Isoenzymes In Acinar Adenocarcinoma, Intraepithelial Neoplasia, And Benign Prostate Tissue: Correlation Of Clinical Parameters With Gst Isoenzymes, Gülçi̇n Şi̇mşek, Serpi̇l Oğuztüzün, Servet Güreşçi̇, Murat Kiliç, Ömer Faruk Bozkurt, Ali̇ Ünsal

This study investigated the immunohistochemical staining characteristics of glutathione-S-transferase (GST) alpha, pi, mu, and theta in prostatic acinar adenocarcinoma (PCA), prostatic intraepithelial neoplasia (PIN), and benign prostatic tissues from 19 patients. Relationships between GST isoenzyme expression in benign, PIN, and PCA tissue were examined by the Wilcoxon signed-rank test and clinicopathological data were examined by the Spearman correlation rank test. When the benign, PIN, and PCA tissues from these cases were compared according to their staining intensity, GST alpha, pi, mu, and theta expressions in tumor cells were significantly lower than in benign epithelial cells (P < 0.05). The GST alpha class displayed the lowest level of expression in PIN and PCA. Expression of GST pi was lower in PCA tissue than in PIN and benign epithelial tissue (P < 0.05). We hypothesize that carcinogenesis in the prostate results from impaired cellular handling of mutagenic agents owing to reduction or loss of expression of multiple GST isoenzymes and other detoxifying and antimutagenesis agents. This study confirms the down-regulation of GST isoenzymes in PCA of the prostate and shows that the loss of GST isoenzyme expression is a phenotype associated with malignant transformation. There was no statistical relationship between GST isoenzyme expression and the clinicopathological data (age, Gleason score, and total serum prostate-specific antigen levels) (P > 0.05).

Purification And Properties Of An Endoglucanase From Aspergillus Niger Vtcc-F021, Thi Hoa Pham, Dinh Thi Quyen, Ngoc Minh Nghiem

An extracellular endoglucanase (EG) from Aspergillus niger VTCC-F021 was purified 2.09-fold to homogeneity with a yield of 18.4%. The enzyme had a molecular mass of 31 kDa and a specific activity of 14.122 U/mg protein. Optimum temperature was observed at 55 °C and optimum pH at 5. The enzyme was stable up to 50 °C and from pH 5 to 6 with residual activity >80% and 60%, respectively. The kinetic constants K_m and V_{max} determined for EG, with carboxyl methyl cellulose as a substrate, were 8.5815 mg CMC/mL and 20.121 U/mg protein, respectively. A field experiment including 3 sainfoin cultivars (Makedonka, EG Norm, and the Sokobanja population) was set up in a seed production trial with the aim of determining adequate row spacing for seed and forage yield. The chemical fruit composition, phenolic profile, and corresponding total antioxidant activity of bilberry (Vaccinium myrtillus) and 2 commercial blueberry cultivars (Vaccinium corymbosum) were analyzed. Results from this study showed that cultivar Berkeley yielded the highest glucose and fructose contents (70.8 and 88.8 mg/g FW, respectively), and the sweetness index expressed a similar trend, achieving the highest value for cultivar Berkeley (279.2). Citric acid was the major organic acid in the berries tested. V. myrtillus yielded a total organic acids value 2-fold higher (0.35 mg/g FW) as well as the highest vitamin C content (25.8 mg/100 g FW). Among the flavonols …

Salinity has adverse effects on plant development and ultimately reduces production. Very few reports on the genes induced by salt stress in Gossypium arboreum are available. In this report salt-stress-responsive genes were screened from cotton leaves using the differential display (DD) technique, and gene expression was compared under control and salt stressed conditions. By using a 107 primer combination, 25 gene fragments were found to be up-regulated in response to salt stress. Out of 25 induced fragments, 12 were rejected as false positives after reamplification and quality control assay. The remaining 13 fragments were selected for cloning and genetic transformation. …

In this study, the water saving and conservation potential of various furrow irrigation management techniques for irrigated cotton were compared. Conventional every-furrow irrigation with open-end furrows (EFO) and blocked-end furrows (EFB), and alternate every-other-furrow management with open-end furrows (AFO) and blocked-end furrows (AFB), were considered. Considerable seasonal water savings were obtained with AFO and AFB flows, on average from 717 mm to 906 mm, respectively, when compared to EFO. Alternate furrows showed the ability to reduce tailwater runoff considerably. When compared with EFO, water use was reduced by 9063 m^3 ha^{-1} (60%) using AFB and 7167 m^3 ha^{-1} (48%) using …

Italian ryegrass (Lolium italicum L. cv. Tetraflorum) seed production is an important industrial activity in Serbia. Cleavers (Galium aparine L.) interferes in Italian ryegrass seed production as well as in many other crops. The effect of stand density of Italian ryegrass on its competitive interaction with cleavers was investigated in western Serbia in the 2005-2006 and 2006-2007 cropping seasons. Safflower (Carthamus tinctorius L.) callus cultures grown in the presence of different concentrations (0-100 µM) of cadmium were assayed for growth, Cd accumulations, and antioxidative responses. Cadmium inhibited the growth of calli by 25% and 46% at 75 and 100 µM Cd concentrations, respectively. Cadmium concentration in the calli increased with an increased Cd supply in the growth medium. Calli accumulated 334 mg Cd kg^{-1} of their dry weight at 100 µM Cd. Cadmium induced oxidative stress, which was indicated by modulating antioxidant (glutathione and \alpha-tocopherol) levels and antioxidative enzymes. The impact of pretreatment with high temperature (45 °C for 45 min) on the UV-B tolerance of 4 barley cultivars (Hordeum vulgare L. 'Bülbül-89', 'Kalaycı-97', 'Tarm-92', and 'Tokak-157/37') was examined. The response of the plants to treatment was evaluated by measuring the pigment content, chlorophyll a fluorescence, oxygen evolution, fraction of oxygen-evolving complex, proline content, UV-B-absorbing compounds (A_{535} and A_{300}), and stress markers (malondialdehyde, H_2O_2, and UV-B marker). Regardless of high temperature pretreatment, UV-B irradiation decreased the photosynthetic pigment content, photosystem II activity, oxygen evolution, and the fraction of oxygen-evolving complex in almost all of the barley cultivars. For this study, 76 fig accessions were collected from Hatay, in the Eastern Mediterranean Region of Turkey, in 2008 and 2009; the morphological diversity of plants and fruits was evaluated. The effectiveness of different arbuscular mycorrhizal fungi (AMF) on different hybrid cucumber cultivars has not been well documented, even under normal seedling conditions. The present study therefore aimed to evaluate colonization, nutrient uptake, dependency, and other seedling traits of 4 cucumber hybrids (Ceren F_1, Beta F_1, Silyon F_1 and Maraton F_1) inoculated by 3 different AMF [Glomus intraradices (Gi), Glomus etunicatum (Ge) and Gigaspora margarita (Gm)]. The effects of inoculum concentrations and the aggressiveness of spot blotch (Bipolaris sorokiniana (Sacc.) Shoemaker) isolates were evaluated on the resistance of 123 winter wheat advanced breeding lines developed from European winter wheat as well as on 3 control cultivars, BR8, BH1146, and Zentos. The test was conducted under laboratory conditions using a detached leaf technique. A total of 3 inoculum concentrations were used: 0.5 × 10^3, 1 × 10^3, and 5 × 10^3 spores mL^{-1}. The prevalence of Potato spindle tuber pospiviroid (PSTVd) infection in randomly selected potato tubers was determined via nonisotopic dot blot hybridization and reverse transcriptase polymerase chain reaction (RT-PCR). In January and February of 2010, 168 seedling tubers of Solanum tuberosum, representing 27 cultivars, were received from different seedling potato producers in Turkey. Digoxigenin-labeled RNA probes of PSTVd were synthesized from the cloned PCR products and applied in dot blot hybridization to detect viroids in RNA extracts isolated from dormant potato tubers. The aim of this study was to estimate patterns of the mating system in 2 natural Cedrus libani A.Rich. populations in Elmalı, Antalya, with the aid of isozyme markers. Isozyme analysis was performed on both maternal and embryo tissues of the seeds collected from Avlankuzu and Çamkuyusu populations. Eleven loci encoding 8 enzyme systems were analyzed. The Avlankuzu and Çamkuyusu populations were a homogeneous and similar gene pool for the loci studied. Boron efficiency of 16 oilseed rape genotypes was tested using both hydroponic and pot-soil growing techniques. From the nutrient solution experiment (0.1 and 10 µM B), 4 representative contrasting genotypes were selected based on relative root and shoot growth. These were then grown in pots with low-B soil (0.25 mg kg^{-1}). From the nutrient solution experiment, 2 genotypes selected as B-inefficient did not show any growth disorders, and the concentration of B in the shoots was above critical. The results of studying the effects of sunflower seed priming with an aqueous solution of ascorbic acid (A), tocopherol (T), and glutathione (G) performed prior to accelerated ageing and a cold test are presented in this paper. Germination, the percentage of abnormal seedlings, and the lengths of both roots and shoots were monitored. The results showed that the cold test caused a drastic drop in germination, an adverse effect on the shoot length, an increase in the percentage of abnormal seedlings, and no effect on the root length. The effect of soil chemical properties on the diversity and colonization of arbuscular mycorrhiza (AM) varies among ecosystems. This study was conducted to assess and compare the abundance of AM in a rehabilitated forest and a logged-over forest soil using the most probable number and spore number methods. Glomus (71.7%-82.1%) and Acaulospora (17.4%-19.5%) were found to be abundant in both sites, while Gigaspora was found only in the logged-over forest. The objectives of this research were to inspect the drop production of a twin fluid system with the Floodjet TK SS 10 042 deflector nozzle and to identify the drift of the droplets produced by the nozzle in a wind tunnel when using the nozzle with the individual settings provided by the company that enlisted our institute to do the described research task.
