One of the natural reservoirs of potentially human-pathogenic bacteria is believed to be the rhizosphere. The aim of the present work was to test nontuberculous mycobacterium Mycobacterium phlei MbP18 for its ability to colonize the rhizosphere of wheat and to evaluate its effect on plant growth under saline conditions. In competitive wheat root tip colonization assays, M. phlei MbP18 showed poor competitive colonization of the wheat rhizosphere compared to the reference strain. Oxidative stress responses were evaluated in the digestive gland tissue of Perna viridis exposed to water-accommodated fractions (WAFs) of petrol and diesel at 0.5% and 5% concentrations over 5-, 10-, and 15-day periods. Increased lipid peroxidation, protein carbonyl, DNA integrity, and hydrogen peroxide in WAF-exposed specimens are indicative of oxidative damage due to dissolved petroleum extracts. A consistent increase in superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase, glutathione-S-transferase, reduced glutathione, and ascorbic acid in WAF-exposed specimens signify the importance of antioxidants in protecting the cell against the oxidative damage arising from petroleum contamination. Glutathione S-transferase (GST) is the one of most widely used affinity tags in biotechnology applications. The present study named a GST gene as the TtGSTz1 gene, characterized with the conserved glutathione binding motif (SSTSWRVRIAL) under a GST zeta subfamily from Tetrahymena thermophila. Phylogenetic analysis of the TtGstz1p protein sequence, with its orthologs from different GST classes, showed that it is a member of the unicellular GSTz monophyletic clade, which is positioned close to the bacterial GSTz clade. In this study, the phytase from Lactobacillus brevis strains isolated from fermented food products was produced and characterized. Phytase production of this strain was determined by production of clear zones around the colonies on the sodium phytate containing medium, after 48 h of incubation at 30 °C. The molecular weight of the phytase from Lactobacillus brevis was estimated as 73 kDa and 34 kDa on SDS-PAGE. The optimum activity of Lactobacillus brevis extracellular and intracellular enzymes emerged at 120 °C, pH 3.4 and at 110 °C, pH 3.4, respectively. The genus Fritillaria embraces about 165 taxa in the family Liliaceae. In this study, the molecular phylogenetic relationships between 10 Fritillaria taxa were presented. Fritillaria spp. were collected from their natural habitats, and specimens with different morphological features were obtained via selective breeding. Specimens with the desired characters were presented as promising candidate cultivars for ornamental use. Phylogenetic analysis was based on DNA sequences of the chloroplast trnL-trnF region. The phylogeny was constructed using the neighbor joining, maximum parsimony, maximum likelihood, and Bayesian inference methods. The persistence of endosulfan and intermediate metabolite endosulfan sulfate in the environment and their toxic effects on biota necessitate their removal. This study investigated the bioaugmentation of endosulfan-contaminated soil by fungal inoculant Aspergillus niger ARIFCC 1053. The influence of bioaugmentation by A. niger on endosulfan-contaminated soil was evaluated with the help of change in pH and released chloride, and by thin layer chromatography and gas chromatography analysis. Its effects on soil functionality were monitored by estimating dehydrogenase and arylsulfatase enzyme activities. The endosulfan degradation reached an undetectable level on day 15. The aim of this study was to evaluate the genotoxic effect of a bovine bone-derived hydroxyapatite (HA), Boneplus-xs (B-P-XS). The genotoxicity of B-P-XS was investigated by examining the potential of the xenograft (B-P-XS) to induce gene mutation (evaluated using the Ames test), in vitro chromosome aberration (CA), and sister chromatid exchange (SCE) with tests in cultured human peripheral lymphocytes. Counts of reverse mutations in Salmonella typhimurium TA98 and TA100 strains were not found to be significantly increased after treatment with B-P-XS (P > 0.05). Alocasia macrorrhizos (L.) (Araceae), a coarse and erect plant that is widely cultivated to be eaten as a vegetable throughout Bangladesh, has been reported to possess a number of medicinal properties. The antihyperglycemic, antioxidant, and cytotoxic effects of the methanol extract of A. macrorrhizos rhizomes were investigated. The extract was given at a single dose (250 and 500 mg/kg) in alloxan-induced hyperglycemic mice. The extract at 500 mg/kg produced a significant (P < 0.05) decrease in the blood glucose level (55.49%) at 8 h of treatment when compared with the control and was comparable with the reference drug, metformin. The extract was also subjected to antioxidant potentiality and brine shrimp lethality bioassays. The IC_{50} value of the extract was 693.0 \mug/mL and the LC_{50} was 188.14 \mug/mL.

In vitro antioxidant, antimicrobial, and antiproliferative activities of methanol, acetone, and ethyl acetate extracts of Sedum acre L. were determined. Their total phenolic contents as well as the concentrations of flavonoids were also evaluated. The total phenolic content was determined with Folin-Ciocalteu reagent and it ranged between 18.25 to 181.75 mg gallic acid/g. The concentration of flavonoids in various extracts of S. acre was determined by using a spectrophotometric method with aluminum chloride and the obtained results varied from 8.42 to 173.42 mg rutin/g. Wheat landraces could be unique sources of favorable genes for agronomic traits like yield and insect, pest, and disease resistances, as well as quality characteristics that are important for breeding programs. Hyperthyroidism increases the vulnerability of the heart to ischemia-reperfusion (I/R) injury. Regarding the fact that diazepam also affects cardiac I/R injury, the current study was conducted to investigate the effect of diazepam on ischemia-reperfused isolated hyperthyroid rat heart. The animals were divided into 6 groups: the control, hyperthyroid, control diazepam (1 mg/kg), hyperthyroid diazepam (1 mg/kg), control diazepam (5 mg/kg), and hyperthyroid diazepam (5 mg/kg) groups. In this study, our aim was to develop a new simple technique for isolation of mesenchymal stem cells from adipose tissue. For this purpose, mesenchymal stem cells were isolated from rat adipose tissue by using the primary explant culture technique. Twenty-seven aqueous extracts obtained from 21 plants used in the treatment of respiratory tract infections as folk remedies in Turkey were investigated for their relative total phenolic contents and antioxidant, antibacterial, antimycobacterial, and antifungal activities. Antibacterial and antifungal activities against dermatophytes and yeast-like fungi were determined using the microdilution method. Antibacterial activity of all extracts was more pronounced against gram-positive bacteria than against gram-negative bacteria. These extracts also exhibited higher antifungal activity against Candida krusei than ketaconazole. Six extracts displayed antidermatophytic activity against Epidermophyton floccosum, while only 3 extracts were highly active against Trichophyton rubrum. The extracts showed strong activity …

The Expression Of Gst Isoenzymes In Acinar Adenocarcinoma, Intraepithelial Neoplasia, And Benign Prostate Tissue: Correlation Of Clinical Parameters With Gst Isoenzymes, Gülçi̇n Şi̇mşek, Serpi̇l Oğuztüzün, Servet Güreşçi̇, Murat Kiliç, Ömer Faruk Bozkurt, Ali̇ Ünsal

This study investigated the immunohistochemical staining characteristics of glutathione-S-transferase (GST) alpha, pi, mu, and theta in prostatic acinar adenocarcinoma (PCA), prostatic intraepithelial neoplasia (PIN), and benign prostatic tissues from 19 patients. Relationships between GST isoenzyme expression in benign, PIN, and PCA tissue were examined by the Wilcoxon signed-rank test and clinicopathological data were examined by the Spearman correlation rank test. When the benign, PIN, and PCA tissues from these cases were compared according to their staining intensity, GST alpha, pi, mu, and theta expressions in tumor cells were significantly lower than in benign epithelial cells (P < 0.05). The GST alpha class displayed the lowest level of expression in PIN and PCA. Expression of GST pi was lower in PCA tissue than in PIN and benign epithelial tissue (P < 0.05). We hypothesize that carcinogenesis in the prostate results from impaired cellular handling of mutagenic agents owing to reduction or loss of expression of multiple GST isoenzymes and other detoxifying and antimutagenesis agents. This study confirms the down-regulation of GST isoenzymes in PCA of the prostate and shows that the loss of GST isoenzyme expression is a phenotype associated with malignant transformation. There was no statistical relationship between GST isoenzyme expression and the clinicopathological data (age, Gleason score, and total serum prostate-specific antigen levels) (P > 0.05).

Purification And Properties Of An Endoglucanase From Aspergillus Niger Vtcc-F021, Thi Hoa Pham, Dinh Thi Quyen, Ngoc Minh Nghiem

An extracellular endoglucanase (EG) from Aspergillus niger VTCC-F021 was purified 2.09-fold to homogeneity with a yield of 18.4%. The enzyme had a molecular mass of 31 kDa and a specific activity of 14.122 U/mg protein. Optimum temperature was observed at 55 °C and optimum pH at 5. The enzyme was stable up to 50 °C and from pH 5 to 6 with residual activity >80% and 60%, respectively. The kinetic constants K_m and V_{max} determined for EG, with carboxyl methyl cellulose as a substrate, were 8.5815 mg CMC/mL and 20.121 U/mg protein, respectively. A field experiment including 3 sainfoin cultivars (Makedonka, EG Norm, and the Sokobanja population) was set up in a seed production trial with the aim of determining adequate row spacing for seed and forage yield. The chemical fruit composition, phenolic profile, and corresponding total antioxidant activity of bilberry (Vaccinium myrtillus) and 2 commercial blueberry cultivars (Vaccinium corymbosum) were analyzed. Results from this study showed that cultivar Berkeley yielded the highest glucose and fructose contents (70.8 and 88.8 mg/g FW, respectively), and the sweetness index expressed a similar trend, achieving the highest value for cultivar Berkeley (279.2). Citric acid was the major organic acid in the berries tested. Salinity has adverse effects on plant development and ultimately reduces production. Very few reports on the genes induced by salt stress in Gossypium arboreum are available. In this report salt-stress-responsive genes were screened from cotton leaves using the differential display (DD) technique, and gene expression was compared under control and salt stressed conditions. By using a 107 primer combination, 25 gene fragments were found to be up-regulated in response to salt stress. In this study, the water saving and conservation potential of various furrow irrigation management techniques for irrigated cotton were compared. Conventional every-furrow irrigation with open-end furrows (EFO) and blocked-end furrows (EFB), and alternate every-other-furrow management with open-end furrows (AFO) and blocked-end furrows (AFB), were considered. Considerable seasonal water savings were obtained with AFO and AFB flows, on average from 717 mm to 906 mm, respectively, when compared to EFO. Alternate furrows showed the ability to reduce tailwater runoff considerably. When compared with EFO, water use was reduced by 9063 m^3 ha^{-1} (60%) using AFB and 7167 m^3 ha^{-1} (48%) using …

Italian ryegrass (Lolium italicum L. cv. Tetraflorum) seed production is an important industrial activity in Serbia. Cleavers (Galium aparine L.) interferes in Italian ryegrass seed production as well as in many other crops. The effect of stand density of Italian ryegrass on its competitive interaction with cleavers was investigated in western Serbia in the 2005-2006 and 2006-2007 cropping seasons. Safflower (Carthamus tinctorius L.) callus cultures grown in the presence of different concentrations (0-100 µM) of cadmium were assayed for growth, Cd accumulations, and antioxidative responses. Cadmium inhibited the growth of calli by 25% and 46% at 75 and 100 µM Cd concentrations, respectively. Cadmium concentration in the calli increased with an increased Cd supply in the growth medium. Calli accumulated 334 mg Cd kg^{-1} of their dry weight at 100 µM Cd. Cadmium induced oxidative stress, which was indicated by modulating antioxidant (glutathione and \alpha-tocopherol) levels and antioxidative enzymes. The impact of pretreatment with high temperature (45 °C for 45 min) on the UV-B tolerance of 4 barley cultivars (Hordeum vulgare L. 'Bülbül-89', 'Kalaycı-97', 'Tarm-92', and 'Tokak-157/37') was examined. The response of the plants to treatment was evaluated by measuring the pigment content, chlorophyll a fluorescence, oxygen evolution, fraction of oxygen-evolving complex, proline content, UV-B-absorbing compounds (A_{535} and A_{300}), and stress markers (malondialdehyde, H_2O_2, and UV-B marker). Regardless of high temperature pretreatment, UV-B irradiation decreased the photosynthetic pigment content, photosystem II activity, oxygen evolution, and the fraction of oxygen-evolving complex in almost all of the barley cultivars. Ficus carica L. is one of the most important fruit species in Turkey. For this study, 76 fig accessions were collected from Hatay, in the Eastern Mediterranean Region of Turkey, in 2008 and 2009; the morphological diversity of plants and fruits was evaluated. Of the samples studied, 2 accessions were determined to be first crop (breba) and the other 74 accessions were identified as main crop.
