Suspension cultures of L-cells were infected with strains representing different biotypes of Chlamydia (C.) psittaci of bovine, ovine, caprine, equine, feline, porcine and guinea pig origin. Biotype-1 chlamydiae multiplied to high titers, and high yields of infectivity were obtained in subsequent passages. Chlamydial strains representative of other biotypes did not infect suspension cultures efficiently. Low rates of infection were obtained on the first passage, and the infectivity was lost while attempting subpassages. L-cells inoculated in suspension with more than one inclusion forming unit/L-cell of the chlamydial strain B577 and allowed to form monolayers, were destroyed within 15 days. However, upon …

Criteria of DSM-III were used as source material for items in a psychopathology inventory series. The inventories provided DSM-III diagnoses by reproducing the DSM-III decision process through diagnostic keys. The personality disorders inventory (ADI) was chosen as initial research instrument. Forty-seven voluntary clients from two sites served as clinical subjects and 29 non-clinical volunteers, given special directions to avoid diagnosis, served as subjects for validity scales. Four clinical psychologists matched DSM-III criteria with ADI items in measurement of content validity. Results showed that the ADI was not a useful psychometric instrument in its present form. Two consecutive summer and winter trials involving low and high-heat increment diets were fed to growing-finishing swine, and three trials evaluating crystalline lysine utilization were conducted during the starting phase of growth. Dehydrated alfalfa meal and vegetable oil in factorial combination were fed during the winter months. Crystalline lysine and vegetable oil, also in factorial combination, were fed during the summer months. In the lysine utilization trials, crystalline or protein-bound lysine were each fed at three levels. Field studies were conducted to document seedling johnsongrass Sorghum halepense (L.) Pers. growth as influenced by date of emergence in sugarcane (Saccharum interspecific hybrids, cultivar 'CP 65-357'). The crop was in the plant cane year of growth in 1982 and 1983, and the first stubble year in 1984. The experiments were randomized complete block designs. Johnsongrass seedling emergence days were approximately three weeks apart, from April or early May through mid-July for a total of five emergence days each year. Part I. We analyze a gravity wave detector consisting of a Weber type resonant antenna, an inductive transducer and a dc SQUID coupling system. We derive the complete set of equations of motion for the system and discuss their implications and solutions in detail. The results obtained are applied to the LSU gravity wave detector, and the optimum system parameters are calculated. We show that it is possible to approach quantum limited sensitivity for the detector if these optimum system parameters and matching conditions are properly satisfied. An experimental dryer with a data acquisition system to analyze single-layer rice drying was built and tested. Data of moisture content and time were collected for long grain Labonnet variety rough rice representing on-farm conditions (35(DEGREES)C to 55(DEGREES)C; 30% to 70% RH; 0.13 to 0.33 m/s drying air velocity) and commercial conditions (55(DEGREES)C to 75(DEGREES)C; 30%; to 70% RH; 0.63 m/s drying air velocity). The initial moisture levels of the rice at these conditions were 28, 33 and 38% (dry basis). Four different drying models were evaluated for the single-layer rice drying data. This dissertation examines the effects of loan and deposit quantity uncertainty and risk aversion on: (1) the spread between the risk averse loan and deposit rates, and (2) the behavior of the intermediary in the Federal funds market. The intermediary's major function is to provide transaction and liquidity services to its customers. Theoretical results indicate that quantity uncertainty from either loans or deposits reduces the spread to a level below that of a profit maximizing intermediary. The pure effects of quantity uncertainty on the intermediary's spread are consistent and mutually reinforcing regardless of whether uncertainty comes from loans or deposits. …

The work presented herein investigates the repair of apurinic/apyrimidinic (AP) sites in Drosophila melanogaster. Two different AP endonucleases were isolated differing in pH optima, reaction requirements, and associated activities. AP endonuclease I flows through phosphocellulose, has a pH optimum of 6.5, and has no obvious cofactor requirements for the incision of AP DNA. AP endonuclease I additionally has associated non-specific endonuclease activity, inseparable throughout the purification. AP endonuclease II is retained by phosphocellulose, eluting between 200-400 mM potassium phosphate. This enzyme has an absolute requirement for magnesium, a pH optimum of 7.0, and no detectable associated enzyme activities. Both enzymes …

Cotton, Gossypium hirsutum L., is a valuable cash crop in Louisiana that is grown primarily for its fiber. The reniform nematode, Rotylenchulus reniformis Linford and Oliveira, is a major pest of cotton in Louisiana, where it was first identified in 1941 and has now been found in 39 of 64 parishes. Nematicides and crop rotation are conventional methods of reniform nematode control, but resistant cultivars offer the most economical and practical method of control. Studies were conducted on apparent phosphorus fixation in 54 soils of Louisiana to determine the effects of different rates of P application on the recovery of P, and to develop appropriate regression model for predicting extractable P and recovery of P by using 4 extractants. Soil samples were equilibrated with aqueous solution of P for 7 days, and were then extracted with the 4 solutions. With the exception of the soils of the Mississippi River area, Bray II solution removed significantly larger quantities of P than did the 3 other extractants. Laboratory, greenhouse, and field investigations were conducted to determine the influence of pendimethalin (N-(1-ethylpropyl)-3,4-dimethyl-2,6-dinitrobenzenamine), trifluralin (2,6-dinitro-N,N-dipropyl-4-(trifluoromethyl)benzenamine), and other selected herbicides on the Bradyrhizobium japonicum (3I1B110)-soybean (Glycine max, (L.) merr.) symbiosis. Bradyrhizobium japonicum, strain 110 growth on yeast extract mannitol (YEM) agar was not inhibited by most herbicides in direct contact with cells when applied at 10 to 2000 mg L('-1). Pendimethalin and trifluralin inhibited B. japonicum growth at levels over 2000 mg L('-1). Bradyrhizobium japonicum growth in YEM broth was not inhibited by pendimethalin or trifluralin at 100 mg L('-1). Metolachlor (2-chloro-N-(2-ethyl-6-methylphehyl)-N-(2-methoxy-1-methylethyl)acetamide at 10 mg L('-1) increased growth in YEM …

Colletotrichum fragariae Brooks, the causal agent of strawberry anthracnose (referred to herein as anthracnose-crown rot), has been reported to infect all above ground parts of the strawberry plant. Colletotrichum acutatum Simmonds is known to cause ripe rot of several fruits including strawberry. Twenty-four Colletotrichum isolates obtained from strawberry plants from several southeastern states and California were compared. Based on conidial morphology and cultural characteristics, 15 of the isolates were identified as C. fragariae and nine as C. acutatum. Two of the C. acutatum isolates were obtained from field grown plants exhibiting crown rot symptoms. This is the first report of …

Part One. Speciation of Mercury by G.C.-A.A.. A new graphite atomic absorption (A.A.) detector for gas chromatography (G.C.) has been developed. It was directly interfaced to a gas chromatograph through a pyrex transfer line. The interfaced G.C.-thermal atomizer A.A. system was used to speciate and determine volatile mercury compounds in perspiration and urine. This interfaced system exhibited the excellent selectivity and high sensitivity to mercury compounds. Because of high sensitivity no sample preparation was necessary, avoiding losses and contamination which often occurs during sample pretreatment. A study combining experimental, empirical modeling, and detailed modeling techniques has been conducted to develop a better understanding of the chemical reactions involved in soot formation during the high-temperature pyrolysis of aromatic and other unsaturated hydrocarbons. The experiments were performed behind reflected shock waves in a conventional shock-tube with soot formation monitored via attenuation of a laser beam at 633nm. Soot-formation measurements were conducted with toluene-argon and benzene-argon mixtures. Detailed kinetic models of soot formation were developed for pyrolyzing acetylene, butadiene, ethylene and benzene. Extensive research has been conducted assessing psychophysiological reactivity to experimental stressors in various populations. However, there is a paucity of empirical investigations concerning the test-retest reliability of these experimental stressors. Establishment of the relative reliability of specific stressor procedures is important so that results can be compared over time and across studies. The purpose of this investigation was to compare test-retest reliability of two mental stressors (i.e. Quiz Electrocardiogram, mental arithmetic) and two physical stressors (i.e. cold pressor, isometric challenge). A study was conducted to evaluate the effectiveness of a food and fitness program implemented by Home Economists of the Cooperative Extension Service. The Fitness Factor program was taught by 49 classroom teachers to 1248 fifth grade students in 20 schools in nine Louisiana parishes. A secondary purpose was to identify and study program management factors that affect success of the program. Three sources of data collection were (a) a youth pretest/posttest instrument measuring food and fitness practices and attitudes, and food consumption, (b) a classroom teacher questionnaire and (c) a Home Economist questionnaire. Aspects of the early life history of 6 species of sciaenids that spawned in west Louisiana shelf waters from December 1981 to April 1982 were studied. This research documented the distribution of larvae and tested a transport hypothesis that detailed advective processes which affected their distribution and, ultimately, recruitment into estuaries in southwest Louisiana, and provided information on larva age and growth. Ranked in order of abundance the six sciaenid species collected were: sand seatrout (Cynoscion arenarius); Atlantic croaker (Micropogonias undulatus); spot (Leiostomus xanthurus); black drum (Pogonias cromis); southern kingfish (Menticirrhus americanus; and banded drum (Larimus fasciatus). Responses of green ash (Fraxinus pennsylvanica Marsh.) and water oak (Quercus nigra L.) to anaerobic root environments were investigated. Initially, analytical methods pursuant to this objective were developed. An extraction solution containing DDT (dithio- threitol) and soluble polyvinylpyrrolidone proved best for oak ADH and protein recovery, while DTT and insoluble polyvinylpolypyroli- done were best for ash. A method for the control of the soil atmosphere surrounding intact roots was developed to study the effects of elevated CO(,2) in the anaerobic soil atmosphere of ash and oak seedlings. Direct and correlated responses from two different sire selection strategies were compared for twelve characteristics of dairy cattle. Daughters of sires selected on genetic merit for milk production (ST) were compared to daughters of sires selected on genetic merit for an index of milk production, fat production and type scores (MT). Data consisted of repeated measures of twelve dairy traits on 275 foundation through third generation Holstein cows in a single herd. A mechanically balanced oscillating inclined digger blade mechanism was designed and constructed to evaluate the effects of vibration on soil break-up and power requirements. Changing the levels of amplitude, frequency, and forward travel speed, twenty-seven combinations of such parameters were tested in two different soil conditions. Soil conditions for this study are based on a silt loam soil with moisture contents of 16 and 15 percent (w.b.), dry densities of 1440 and 1310 Kg/m('3), and cone indices of 0.82 and 0.41 MPa for hard and soft soil conditions, respectively. This study was designed to examine the influence of several potential sources of bias in the rating process. It specifically examined the potential effect of sex of rater and ratee, information about ratee characteristics, information about the job, and two rater personality factors on the way raters use performance information in coming to a final promotion decision. The convergent and discriminant validity of three assessment methods were investigated in relation to the construct of childhood depression for a sample of elementary, intermediate, and secondary students. The three assessment methods of self-report, teacher, and peer rating scales were used to assess the response classes of depression, social withdrawal, social skills, and aggression. The self-report, teacher, and peer rating scales were subjected to a series of analyses to determine their psychometric properties. A new method of approximating a high-order system by a lower-order model in the frequency domain is developed. A reduced-order model of a transfer function obtained by the new method is compared with a reduced-order model obtained by another existing method to illustrate the power of the technique. Furthermore the approximant constructed by this new method is used to design a control system, which is applicable to the plant, to show its usefulness. Purpose of the Study. The purpose of this study was to determine the status of the public secondary school band programs in Louisiana for the 1984-1985 school year. Procedures. Data were gathered by means of questionnaires sent to the directors of all public high school band programs. Information sought was divided into three major sections: (1) Teacher Profile, (2) The Band Program, and (3) Finance and Budget. Responses were obtained from 51.7 percent of the total population, and the data were tabulated and presented with comparison of the findings. Findings. Spatial distribution of fall armyworm, Spodoptera frugiperda (J. E. Smith), (FAW) was studied in cornfields at the St. Gabriel Experiment Station in Louisiana from 1981 to 1983. Corn plants were sampled and FAW larvae counted and divided into categories of small, medium, and large for both single plant and five plant samples. Count data for each category by date were then fitted to Poisson, negative binomial, and Neyman type A distributions. Means and variances for each category by date were also analyzed with the Taylor Power law. Small larvae were more aggregated than the medium larvae which were more aggregated …

This investigation attempted to profile the audience awareness of gifted and non-gifted fifth graders. At three writing sessions, students wrote to a friend, a teacher, and an editor, persuading each to go to the park. Compositions were examined for syntactic complexity, according to t-units, and for the kinds and numbers of persuasive appeals used. A repeated measures design with a 2 x 3 factorial arrangement of between subject treatments (group and sex) and a single repeated factor (audience at three levels) was used. The effects of a stress management program upon the Type A behavior pattern were examined in a sample of patients with coronary heart disease who were enrolled in an on-going cardiac rehabilitation program. Based upon exhibition of Type A behavior (defined by the Structured Inverview), patients were randomly assigned to three experimental conditions: (a) behavioral stress management, (b) supportive (attention placebo control group), and (c) no treatment (waiting list control group). Patients receiving treatment attended 10 sessions of 1-1 1/2 hours duration, as well as pretreatment and posttreatment assessment sessions. The objective of this investigation was the development of quantitative criteria based on the estimation of maximum levels of oxygen demand associated with specific levels of nutrients in algal dominated streams. 1582 intensive survey records were obtained from the Louisiana Department of Environmental Quality (LA DEQ). A sub-set of 225 records was defined by a minimum chlorophyll a of 10.0 ug/l and influential observation analysis to restrict the data to streams where algal processes were likely to be dominant.
