Measurement Of Acetate Concentrations In Marine Pore Waters By Using An Enzymatic Approach, G. M. King

Acetate concentrations in marine and freshwater matrices were measured by an enzymatic technique which coupled the synthesis of acetyl coenzyme A to AMP production. The resulting AMP was assayed by a sensitive and relatively rapid high-pressure liquid chromatography method, using an aqueous, isocratic mobile phase for elution. The method was insensitive to the presence of seawater salts and required no sample prepurification or distillation. Propionate caused a minor, but statistically insignificant, interference when equimolar with acetate; butyrate caused no interference, even at relatively high concentrations. Detection limits for acetate were approximately 100 nM with a precision of about 5%. Pore …

Specific and sensitive amplification of major outer membrane protein (MOMP) gene DNA sequences of Chlamydia psittaci was achieved in a two-step polymerase chain reaction. First, oligonucleotide primers specific for 5' and 3' nontranslated regulatory regions of the MOMP gene were used in a polymerase chain reaction to amplify a DNA fragment of approximately 1,400 bp. A portion of this DNA fragment was amplified in a second reaction using a degenerate oligonucleotide primer specific for a DNA sequence contained within the 1,400-bp DNA fragment and one of the first-step primers. The entire nucleotide sequences of the spike glycoprotein (S) genes of the highly virulent bovine coronavirus (BCV) strain BCV-LY138, the avirulent BCV-L9 and related Norden Vaccine (BCV-Vaccine) strains were determined using the polymerase chain reaction (PCR) to amplify cDNAs obtained by reverse transcription of viral RNA, and to produce single strand cDNAs for DNA sequencing. The S gene sequences of these viral strains were compared with those of recently published strains BCV-Mebus, BCV-Quebec, and BCV-F15. An open reading frame of 4092 nucleotides, encoding a protein of 1363 amino acid residues, was found in all six strains. Frameshifts and insertions or …

Monoclonal antibodies (MAbs) against two major structural proteins of the cell-adapted Mebus strain of bovine coronavirus (BCV-L9) were produced and characterized. Seven MAbs reacted with the peplomeric glycoprotein, gp100/S, while three MAbs reacted with the nucleoprotein p53/N in Western blot analysis of BCV polypeptides. MAbs to gp100/S reacted with discontinuous epitopes of gp100/S in Westerns under mild but not under standard denaturing conditions. In contrast, MAbs to p53/N reacted in both types of Westerns, and those epitopes were thus continuous. MAbs to p53/N failed to neutralize BCV infectivity, while 4 MAbs to gp100/S neutralized BCV effectively. Cross reactivity of MAbs …

Sodium zeolite A (SZA), a synthetic sodium aluminosilicate having high ion-exchange capacity, has been shown to increase eggshell specific gravity in laying hens and to improve Ca utilization in chickens. A 4 x 2 factorial arrangement of treatments was used to investigate the effect of dietary Ca (.6, .8, 1.0, and 1.2%) and SZA (0 and .75%) on growth, plasma, and tibia characteristics of chicks from 5 to 15 days of age. Increasing dietary Ca linearly increased (P less than .05) Ca and alkaline phosphatase (AP) in plasma and increased tibia shearing force and percentage ash, Ca, and P in …

A simple procedure that yields highly purified intact chloroplasts from Chlamydomonas reinhardtii is described. This procedure involves breakage of cell wall-deficient cells by passing them through a narrow bore syringe needle. The intact chloroplasts are then purified from the crude homogenate by differential centrifugation and Percoll gradient centrifugation. This procedure generates relatively high yields of chloroplasts capable of CO2 fixation. These chloroplasts were characterized by electron microscopy, marker enzyme analysis, and ferricyanide exclusion. Transmission electron microscopy indicates that these chloroplasts retain their pyrenoids and eyespots. The Soviet-American Gallium Experiment is the first experiment able to measure the dominant flux of low energy p-p solar neutrinos. Four extractions made during January to May 1990 from 30 tons of gallium have been counted and indicate that the flux is consistent with 0 SNU and is less than 72 SNU (68% CL) and less than 138 SNU (95% CL). This is to be compared with the flux of 132 SNU predicted by the Standard Solar Model. © 1991.

The six individual catalytic polypeptide chains within the two catalytic trimers of Escherichia coli aspartate transcarbamoylase (ATCase; EC 2.1.3.2) are folded into two discrete structural domains interconnected in part by helix 12, which comprises residues 285-305 and is located near the carboxyl terminus of the chain. The essential role of this helix in folding of the chains and their assembly into ATCase was demonstrated by introducing a stop codon at the position corresponding to amino acid 284, 291, or 299. Cells containing these mutations are pyrimidine auxotrophs lacking ATCase-like protein in cell extracts. In contrast, stable active enzyme is formed …

This text is intended for either an applied algebra course or a modern algebra course that includes more applications than has been traditional. It is at an advanced undergraduate (junior-senior) level and is suitable for a one-semester or two-quarter course. We assume that students have already had a course in linear algebra (although we briefly review concepts from linear algebra when they are needed in the sections on fields and linear codes).

My subject is the relationship between rhetoric and the range of possible reaction to Marlowe's protagonists in his five major plays--Tamburlaine I and II, Doctor Faustus, The Jew of Malta, and Edward II. In a very broad sense, I conceive of the rhetoric of each play in terms of opposing rhetorical forces, amplification and irony, and attempt to account for the relative intensity of amplification and irony in each play as I study the rhetoric of the protagonist. Since increasing more complex RIM molds are being used in industry, the goal of this research is to develop a model which can be used to predict the dynamic filling front, hot spots, and air entrapment in molds. In this research a method to simulate the RIM process by considering both the filling and curing stages in variously shaped molds for unsteady state three dimensional state cases was developed. Let ℱ be a collection of 3-connected matroids, none a proper minor of another, such that if M is a 3-connected matroid having a proper ℱ-minor and e is an element of M, then M has an ℱ-minor avoiding e. This paper proves that there are precisely two collections ℱ with this property: {U2,4} and {U2,4, M(K4)}. Let n be an integer exceeding one and M be a matroid having at least n + 2 elements. In this paper, we prove that every n-element subset X of E(M) is in an (n + 1)-element circuit if and only if (i) for every such subset, M X is disconnected, and (ii) for every subset Y with at most n elements, M Y is connected. Various extensions and consequences of this result are also derived including characterizations in terms of connectivity of the 4-point line and of Murty's Sylvester matroids. We present the theory and technique for obtaining the distributional solutions for the Wiener-Hopf integral and integro-differential equations. This is achieved by identifying a class of kernels for which these equations are well defined and are of the Fredholm type. Consequently, the associated operators and their images are of finite dimensions. Furthermore, we define the operators in such a way that the corresponding equations hold at the end points; otherwise, the equations are usually ill-behaved. The effects of elevated levels of ozone on growth and frost resistance were studied using broadleaf‐evergreen citrus and avocado trees. ‘Ruby Red’ grapefruit (Citrus parodist L.) trees on either Volkamer lemon (Citrus volkanieriana Ten. & Pasq.) or sour orange (Citrus aurantium L.) rootstocks and ‘Simmonds’ or ‘Pancho’ avocado trees (Persea americana Mill.) on the rootstock ‘Waldin’ were exposed to ozone in open‐top chambers for 4 months in 1988 and for 8 months in a second experiment in 1989. Various mechanisms have been suggested for the quenching of Cd ion activity in plant vacuoles. These include solution complexation with organic acids and sulfhydryl-containing peptides and precipitation as sulfides. Because direct experimental support for these mechanisms is lacking and difficult to obtain, we have used a computer model to evaluate the quenching role of possible organic and inorganic ligands of tobacco cultured cells exposed to Cd. Results of this thermodynamic evaluation, which assumes that a chemical equilibrium state is met in the vacuole, support the conclusion that sulfhydryl-containing peptides and certain organic acids may form soluble Cd complexes. Acrosomal integrity of bovine spermatozoa was evaluated using naphthol yellow S-erythrosine B stain with bright field microscopy, .2% glutaraldehyde fixation with Nomarski optics, and live wet smears with Nomarski optics to compare the evaluation techniques. Four ejaculates per two Holstein bulls were enriched through a Percoll gradient, capacitated with heparin, and acrosome-reacted with lysophosphatidylcholine to prepare spermatozoa for in vitro fertilization. Spermatozoa samples were taken after each preparatory step, and percentage of intact acrosomes was evaluated with the stain, .2% glutaraldehyde, and live wet smear. The electron affinities of atomic hydrogen and deuterium have been determined by tunable-laser threshold-photodetachment spectroscopy: the electron affinities of H (F=0) and D (F=1/2) are 6082.990.15 and 6086.20.6 cm-1, respectively. The value for H is a 20-fold improvement over previous experimental determinations. © 1991 The American Physical Society.

One solution to overcome the shortage of hospital nurses is to establish and implement clinical career ladder programs. The purpose of this study was to examine hospital clinical nurses' perceptions of ladder programs as a job enrichment strategy and to determine individual and work-related variables contributing most to nurses' participation or nonparticipation in available clinical ladder programs. A random sample of 600 clinical nurses employed full time in five regional medical center hospitals located in Louisiana and Mississippi were the study subjects. Respondents were 106 (88.3%) of the 120 ladder program participants and 385 (80.2%) of the 480 nonparticipant nurses. …

Infection cushions were formed in a similar manner on soybean leaves by Rhizoctonia solani AG-1 IA (aerial blight) and AG-1 IB (web blight) isolates, however, AG-1 IB produced more mucilagenous material. Leaf topography did not induce infection cushion formation. More infection cushions were formed by isolates of AG-1 IA and IB on collodion membranes placed over leaves of susceptible compared to resistant cultivars. The potential of tropical Africanized honey bees to survive through winter was investigated to predict their possible geographical range and density in the United States. Experiments were conducted in cold rooms, at high elevations in tropical mountains, and during a winter season in Germany. The performance and survival of isolated groups of workers and of intact Africanized colonies were compared with those of European origin, and in some experiments, with those of hybrid crosses. Calyptospora funduli is a pathogenic coccidium that infects the liver and pancreas of estuarine killifishes belonging to the genus Fundulus. Experimental infection of the atypical fish host Rivulus marmoratus with this intracellular protozoan parasite caused the development of hepatic macrophage aggregates (MA's) and granulomas. The Rivulus/Calyptospora host-parasite system thus represents a new laboratory model for investigations of MA biology. In the present study, this model was used to: (1) examine the morphology and sequential development of the parasite-elicited hepatic MA's in R. marmoratus, (2) define the role that these structures play in the pathogenesis of an infectious disease process, and …
