The Phylogeny Of The Prasinophyceae And Pleurastrophyceae (Chlorophyta) Inferred From Ribosomal Rna Genes And Morphology., Thomas Sinclair Kantz

The phylogeny of the problematic green algal classes Prasinophyceae and Pleurastrophyceae (Chlorophyta) sensu Mattox and Stewart (1984) was inferred from ultrastructural and biochemical data, and from ribosomal RNA gene sequence data. Analysis of a data set of ultrastructural and biochemical characters (with a predominance of flagellar and cell covering features) revealed a monophyletic Pleurastrophyceae and a monophyletic Prasinophyceae. Rerooting experiments indicated that the ancestral flagellate was likely Pedinomonas-like or Mamiellales-like. The cladograms generated from the ribosomal RNA gene sequence data show that the Pleurastrophyceae is not monophyletic; however, pleurastrophycean taxa are more closely related to the Chlorophyceae than to any …

In a Gorenstein local ring R, two ideals A and B are said to be linked by an ideal I if the two relations A = (I: B) and B = (I: A) hold. In the case that I is a complete intersection, or a Gorenstein ideal, it is known that linkage preserves the Cohen-Macaulay property. That is, if A is a Cohen-Macaulay ideal, then so is B. However, if I is allowed to be a generically Gorenstein, Cohen-Macaulay ideal, easy examples show that this type of linkage does not preserve the Cohen-Macaulay property. The purpose of the study was to investigate the effects of classroom practices on the distribution of achievement within the classroom as well as on mean levels of achievement through the use of the Hierarchical Linear Model (Raudenbush & Bryk, 1986). The investigation focused on sixty classrooms--thirty from schools labeled as effective and thirty from schools labeled as ineffective. Data on teacher behaviors were gathered through classroom observations during which six dimensions of effective teaching were evaluated. A novel post-translationally modified asparagine, $\gamma$-N-methylasparagine (NMA), has been characterized at the $\beta$-72 site in many phycobiliproteins. The behavior of a NMA-containing peptide under Edman degradation conditions employed for sequence analysis was examined; NMA can be reproducibly identified in protein sequences by careful attention to repetitive yields and the presence of minor peaks. The effects of asparagine methylation on photosynthetic rates in the cyanobacterium Synechococcus PCC 7942 and two methylase-minus mutants was measured by steady-state oxygen evolution in whole cells. This dissertation investigates an emerging stock market: Istanbul Stock Exchange. First the organizational properties and the history of the exchange are presented. The dissertation then provides an extensive statistical study of the time series properties of the Turkish stock prices. It shows that the Turkish stocks are nonnormal and highly leptokurtic. Absolute value of the stocks show significant dependence. The dissertation examines the effects of Decree No. 32 of August 11, 1989 on the stock distributions. After this date the stock market removed the restrictions on foreign investors. It is shown that the mean of stock price changes has stayed …

A semigroup has the congruence extension property (CEP) provided that each congruence on each subsemigroup can be extended to a congruence on the semi-group. This property, the ideal extension property (IEP), and other related concepts are studied from both an algebraic and a topological perspective in this work. A characterization of semigroups with CEP is given in terms of the lattice of congruences. A similar result is obtained for IEP. Semigroups in which the relation "is an ideal of" is transitive (t-semigroups) are explored. It is shown that each of CEP and IEP implies this condition and that these are …

The localization of the 36-kD polypeptide of Chlamydomonas reinhardtii induced by photoautotrophic growth on low CO2 concentrations (0.03% in air [v/v], low CO2-grown cells) has been investigated. This polypeptide was specifically localized to the chloroplast envelope membranes isolated from low CO2-grown cells and was not present in the chloroplast envelopes isolated from high (5% CO2 in air [v/v]) CO2-grown cells. The 36-kD protein does not show carbonic anhydrase activity and was not present on the plasma membranes isolated from low CO2-grown cells. Synaptic Transmission Between Pairs Of Retinal Amacrine Cells In Culture, E. Gleason, S. Borges, M. Wilson

We have examined synaptic transmission between isolated pairs of chick GABAergic amacrine cells, maintained in sparse culture and identified by their binding of an amacrine cell-selective antibody. Using the perforated- patch method to whole-cell clamp both cells of a pair, postsynaptic currents were examined for step depolarizations of the 'presynaptic' cell. Synaptic transmission, frequently reciprocal, was calcium dependent and reversibly blocked by bicuculline. Postsynaptic currents, excluding those due to ohmic electrical coupling, were elicited only for presynaptic voltage steps positive to about -40 mV and were always very noisy, suggesting that they were summed from relatively small numbers of quanta. …

To identify sequence‐specific motifs associated with the formation of an ionic pore, we systematically evaluated the channel‐forming activity of synthetic peptides with sequence of predicted transmembrane segments of the voltage‐gated calcium channel. The amino acid sequence of voltage‐gated, dihydropyridine (DHP)‐sensitive calcium channels suggests the presence in each of four homologous repeats (I–IV) of six segments (S1–S6) predicted to form membrane‐spanning, α‐helical structures. Only peptides representing amphipathic segments S2 or S3 form channels in lipid bilayers. To generate a functional calcium channel based on a four‐helix bundle motif, four‐helix bundle proteins representing IVS2 (T4CaIVS2) or IVS3 (T4CaIVS3) were synthesized. Both proteins …

This study explored the relationship between the developmental appropriateness of kindergarten classroom instruction and first-grade report card grade overall averages and averages in reading, language, spelling, math, science, and social studies. The interactive roles that gender, SES, and kindergarten classroom type play in children’s later achievement were also examined. The sample consisted of166first-grade children who hadattended kindergarten classrooms with teaching practices identified as predominately developmentally appropriate or developmentally inappropriate. Findings indicated that firstgraders from more appropriate kindergarten classrooms had higher reading averages than children from less appropriate kindergarten classrooms. Females had higher overall and subject area averages than males. Significant …

Glucose uptake was monitored on a seasonal basis, using [6-3H]glucose and undisturbed cores collected from an intertidal mud flat. The fate of glucose carbon, including the formation of CO2 and biomass, was assayed by using undisturbed cores and [U-14C]glucose; the production of short-chain fatty acids was monitored with [U-14C]glucose and sediment slurries. Rate constants for glucose uptake varied temporally, with temperature accounting for much of the variability; turnover times ranged from about 2 to 10 min. Rate constants decreased with increasing sediment depth and in the following order for several common monosaccharides: glucose>galactose>mannose~fucose. Time course analyses of 14CO2 …

Methane Consumption In Temperate And Subarctic Forest Soils: Rates, Vertical Zonation, And Responses To Water And Nitrogen, A. P.S. Adamsen, G. M. King

Rates of methane consumption were measured in subarctic coniferous and temperate mixed-hardwood forest soils, using static chambers and intact soil cores. Rates at both sites were generally between 1 and 3 mg of CH4 m-2 day-1 and decreased with increasing soil water contents above 20%. Addition of ammonium (1 μmol g of soil-1) strongly inhibited methane oxidation in the subarctic soils; a lesser inhibition was observed for temperate forest samples. The response to nitrogen additions occurred within a few hours and was probably due to physiological changes in the active methane-consuming populations. The tetrazolium salt sodium 3'-{1-[(phenylamino)-carbonyl]-3,4- tetrazolium}-bis (4-methoxy-6-nitro)benzene-sulfonic acid hydrate (XTT) was examined for use as a colorimetric indicator of viability in respiring bacteria. XTT was reduced to an orange, water-soluble formazan product by Methylosinus trichosporium OB3b, Pseudomonas putida, Escherichia coli, and Bacillus subtilis. Formazan production was proportional to live cell biomass, and XTT was reduced by all cultures in the absence of added electron-coupling agents. XTT reduction by M. trichosporium OB3b was linear over several hours and was stimulated by the presence of an exogenous substrate (methanol). We have developed a new NMR-based procedure for studying waste/cement interactions. This is the first use of deuterium NMR spectroscopy to study waste solidification/stabilization (S/S). The main feature of deuterium NMR spectroscopy is the ability to monitor molecular reorientations over a wide range of reorientation rates. This technique allows one to determine if a particular deuterated organic waste is effectively solidified/stabilized and to determine the lower limit of the bond strength between the waste and the cement matrix. Conceptualizations of pre-Hispanic staple transport remain underdeveloped. Conventional wisdom has long maintained that while "prestige goods" could demand long-distance transport, staple transport was short distance. A quantitative model reveals the fallacy of that argument and establishes the possibility of long-distance, overland staple transport in Mesoamerica by using maize tribute transport between Zempoala and Tenochtitlan as an example. This conclusion has implications for understanding Mesoamerican interregional exchange, ecology, and society.

DNA sequences coding for 81% of the ompA gene from 24 chlamydial strains, representing all chlamydial species, were determined from DNA amplified by polymerase chain reactions. Chlamydial strains of serovars and strains with similar chromosomal restriction fragment length polymorphism had identical ompA DNA sequences. The ompA sequences were segregated into 23 different ompA alleles and aligned with each other, and phylogenetic relationships among them were inferred by neighbor-joining and maximum parsimony analyses. The neighbor-joining method produced a single phylogram which was rooted at the branch between two major clusters. One cluster included all Chlamydia trachomatis ompA alleles (trachoma group). Three amber mutations were introduced proximal to the syn3 locus of the herpes simplex virus type 1 glycoprotein B (gB) gene specifying gB derivatives lacking the carboxy-terminal 28, 49, or 64 amino acids. A complementation system that utilized gBs expressed in COS cells to complement gB-null virus KΔT was established. The 49- or 64-amino-acid-truncated gBs failed to complement gB-null virus KΔT, while the 28-amino-acid-truncated gB complemented KΔT efficiently. Mutant herpes simplex virus type 1 KOS (amb1511-7) specifying the 28-amino-acid-truncated gB fused Vero cells extensively.

Glycine And Alanine Supplementation Of Culture Medium Enhances Development Of In Vitro Matured And Fertilized Cattle Embryos, K. Moore, K. R. Bondioli

One-cell cattle embryos were prepared by in vitro oocyte maturation and fertilization (IVM/IVF) and cultured with or without oviductal cells. Embryos were evaluated after 7 days in culture to determine the percentage developing from the 1-cell stage to the morula or blastocyst stage. The combination of glycine (2 mM) and alanine (1 mM) with oviductal cells (experiment 1) improved embryo development over that in control culture (29 vs. 13%; p < 0.05). An optimum response was obtained with 10 mM glycine and 1 mM alanine in coculture (experiments 2 and 3). In experiment 4, the effects of glycine (0 or 10 mM), alanine (0 or 1 mM), and the presence or absence of oviductal cells were tested. In the absence of oviductal cells, the addition of glycine, alanine, or glycine and alanine combined improved embryonic development over that in control medium (45, 33, 42 vs. 24%, p < 0.001, p < 0.05, p < 0.001, respectively). However, the effect of the combination of glycine and alanine was not different from that of glycine alone when oviductal cells were absent (42 vs. 45%, p > 0.10). In the presence of oviductal cells, glycine or the combination of glycine and alanine improved embryonic development over that in the control medium with cells (47, 55 vs. 37%, …

In vitro matured (IVM) bovine oocytes were examined to determine their potential viability in embryo cloning. Activation competence, as monitored by pronuclear formation, increased with oocyte age. Oocytes readily formed a pronucleus when challenged with an electrical pulse 30 h after the onset of maturation. Developmental competence of IVM oocytes tended to increase with oocyte age (P= 0.079). Selection of IVM oocytes on the basis of the presence of a polar body 24 h after the onset of maturation and the size of the follicle from which the oocyte was derived improved development of nuclear transfer embryos (polar body positive …

Two experiments were conducted to assess the interactive effects of dietary sodium zeolite A (SZA) and experimental Eimeria acervulina infection on growth and tissue mineral concentrations in chicks. The average initial weight of the chicks was 71.8 g, and the experimental periods were from 5 to 15 days posthatching. In both experiments, the corn-soybean meal basal diet was supplemented with 0 or .75% SZA and fed to uninfected chicks or to chicks infected with 4 x 10(5) sporulated E. acervulina oocysts on Days 0, 3, and 6 of the experiments. Both coccidial infection and SZA reduced (P < .05) gain and feed intake; however, feed efficiency was reduced (P < .01) only in the coccidiosis-infected chicks. Two experiments were conducted with chicks from 5 to 15 days posthatching to study the effect of sodium zeolite A (SZA) on Zn utilization. The corn-soybean meal basal diet was supplemented with ZnCO3 to provide three levels of dietary Zn (35, 40, and 85 ppm) in Experiment 1, and two levels of dietary Zn (85 and 4,000 ppm) in Experiment 2. Twenty-four high producing (36.7 kg/d, initially) Holstein cows in early lactation were used in a 63-d lactation trial to study the effects of low (28%), medium (31%). and high (37%) nonstructural carbohydrate concentrations in diets with added fat. Diets with isonitrogenous (17% CP) and fed as TMR with 41% sorghum silage. Nonstructural carbohydrate concentrations were altered by substitution of corn gluten feed and soybean hulls (50:50) for wheat and corn in the concentrate portion of the diet. Twenty-four high producing (36.7 kg/d, initially) Holstein cows in early lactation were used in a 63-d lactation trial to study the effects of low (28%), medium (31%). and high (37%) nonstructural carbohydrate concentrations in diets with added fat. Diets with isonitrogenous (17% CP) and fed as TMR with 41% sorghum silage. Nonstructural carbohydrate concentrations were altered by substitution of corn gluten feed and soybean hulls (50:50) for wheat and corn in the concentrate portion of the diet. A 20-ton neutrino detector located near the Los Alamos Meson Physics Facility beam stop is used search for ν̄e generated via neutrino oscillations from any of the three neutrino types, νμ, ν̄μ, and νe, which radiate from the beam stop. The analysis of three years of data provides limits on the oscillation modes ν̄μ→ν̄e, νe→ν̄e, and νμ→ν̄e, and the lepton-number-violating decay process μ+→e++ν̄e+νμ. The 90%-confidence-level limits for ν̄μ→ν̄e oscillations are δm2≤0.14 eV2 for maximal mixing, and sin22θ≤0.024 for large δm2. © 1993 The American Physical Society.

Pulsar measurements performed by the experiment COMPTEL, aboard the Compton Gamma Ray Observatory, are described. The main results refer to the Crab and Vela pulsars whose pulse shape characteristics are given in some detail and light curves are compared with those above 50 MeV, as observed by the COS-B satellite. No other gamma-ray pulsars have been detected to date by COMPTEL, the upper limit on the pulsed signal from Geminga being compatible with indications by other experiments. © 1993.

The energy spectrum of galactic cosmic ray Helium was measured in two different balloon experiments launched four days apart from Canada: SMILI‐I on 1‐Sept‐1989 and MASS on 5‐Sept‐1989. A slow Forbush decrease began on 4‐Sept‐1989 and had not reached its maximum at the time of the MASS flight. Comparison of the balloon measurements shows a fractional decrease of 0.37 to 0.15 in the Helium flux between 200 and 450 MeV/nucleon (1.2–2.0 GV). The rigidity dependence is analyzed in two models and found to be steeper than previous observations. Interplanetary particle data and ground‐based Neutron Monitor results are consistent with the …
