Transcription factors are important for many aspects of gene regulation in eukaryotes. YY1 (Yin-Yang 1) is a particularly interesting example of a highly conserved zinc-finger transcription factor, involved in transcriptional activation, repression, initiation, and in chromatin modification. YY1 is ubiquitously expressed in mammals, and its binding sites are found in ~10% of human genes as well as in repetitive elements. It is a targeting protein of the Polycomb complex and is involved in mammalian genomic imprinting. Alzheimer’s disease (AD) is a brain disorder named after the German psychiatrist Alois Alzheimer, who first described the disease in 1906. AD is a form of dementia, characterized by impairment of memory and other intellectual abilities, caused by the loss of neurons and synapses (the structures that permit communication between neurons) in certain regions of the brain. Although the mechanism by which neurons are affected is not yet well understood, research points to a small protein, the beta-amyloid peptide (Abeta), as the first suspect. Formation of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) which occurs inexorably from most thermal and combustion processes constitutes a major toxic component of environmental pollutants. Generally, it is well established that transition metal-mediated reactions account for the majority of PCDD/F emissions from combustion sources. Specifically, both copper and iron ions, which occur abundantly in combustion generated particulate matter, are considered probably the most active in promoting PCDD/F formation typically in the low temperature post-combustion zone and flue gas pollution control devices. Whalers from the Caribbean island of St. Vincent and the North Atlantic archipelago of the Faroe Islands hunt pilot whales and a variety of other small cetaceans for food. Vincentian whalers use harpoons, thrown by hand or fired from a modified shotgun mounted on the boat. Faroese whalers, using several dozen boats, work cooperatively to drive an entire pod of whales ashore, where shore-based whalers are waiting to complete the kill with traditional whaling knives. Ludox density gradients were used to enrich for Escherichia coli mutants with conditional growth defects and alterations in membrane composition. A temperature-sensitive mutant named Lud135 was isolated with mutations in two related, nonessential genes: yghB and yqjA. yghB harbors a single missense mutation (G203D) and yqjA contains a nonsense mutation (W92TGA) in Lud135. Both mutations are required for the temperature-sensitive phenotype: targeted deletion of both genes in a wild-type background results in a strain with a similar phenotype and expression of either gene from a plasmid restores growth at elevated temperatures. Neotropical coral reef fish communities are species-poor compared to those of the Indo-West Pacific. An exception to that pattern is the blenny clade Chaenopsidae, one of only three coral reef fish families endemic to the Neotropics. Within the chaenopsids, the genus Acanthemblemaria is the most species-rich. To understand the origin and maintenance of genetic and species diversity in these fishes, I characterized the population genetics for two Acanthemblemaria species, reconstructed the phylogeny of the group, and identified suites of correlated morphological characters responsible for the distinctive skull morphology of these fishes. Biogenic reefs formed by the eastern oyster (Crassostrea virginica) are structurally complex in nature and have been recognized for their potential habitat value in estuarine systems along the Atlantic and Gulf of Mexico coasts. This dissertation outlines the occupation of private investigation and the role that females play within that profession. The difficulties women experience in male-oriented occupations remains noteworthy in sociological research today. Progress has been made, yet many barriers still exist for women. These include structural, social, and cultural factors that influence women and/or the jobs that they hold. With the completion of interviews with twenty-six female private investigators, I was able to analyze the existing structural, interactional, and gender barriers which moderate the number of women that work as private investigators. The addition of serum to embryo culture media may alter gene expression and trigger development of Large Offspring Syndrome. The objectives of this study were to determine gene expression levels in embryos cultured in the absence or presence of 5% calf serum and compare these expression patterns to in vivo derived embryos (IVD), and to determine the effects of serum on the length of day-14 embryos. Women, compared to men, are at higher risk for age-related macular degeneration (AMD), the leading cause of blindness in the elderly. The accumulation of dietary carotenoids, lutein (L) and zeaxanthin (Z) within the macula make up macular pigment and provide important protection to the retina. Macular pigment optical density (MPOD) gives a measure of macula pigment density. MPOD has been shown to be influenced by fruit and vegetable L and Z and docosahexanoic acid (DHA, 22:6n-3), cold water marine fish being the dietary source. A high MPOD may protect against AMD. Differential gene expression in response to internal and external stimuli is studied in detail to understand the intricate mechanisms underlying response to various environmental stressors in microorganisms. MarR family transcriptional regulators have been studied for their involvement in such mechanisms. This work elucidates the mechanism of urate-induced attenuation of DNA binding of HucR, a MarR homolog, and extends this mechanism to describe a novel subfamily of MarR homologs responsive to urate, proposing a physiological relevance of utilizing urate as a signaling molecule.

Old-fashioned, biological, or "Jim Crow" racism is viewed by many in the political science and psychology literature to be largely a relic of the past. In the post-segregation era it has been replaced as a political force by symbolic racism, although its residual effect still operates within symbolic racism as negative racial affect. Symbolic racism is thought of as a coherent belief system that describes whites‘ attitudes not only in the United States, but in some European democracies as well. Tumor initiation and progression are dependent on both aberrant gene expression in tumor cells and the communication between tumor cells and its micro- and systemic microenvironment. Many tumor suppressor genes and oncogenes have been characterized to suppress or promote tumor growth, but fewer genes in tumors are well-characterized as interacting with immune cells in the host to promote or inhibit tumor growth. The interleukin (IL) 27 receptor WSX1 is expressed in immune cells and induces an IL27-dependent immune response. Twelve neonatal Holstein bull calves (38.52 ± 5.87 kg) were fed milk replacer at a fixed or varied meal time to determine the effects on metabolic hormone secretion, average daily intake, growth, and energy metabolism. Body weights were measured every two weeks from birth to 8 weeks. Rumen fluid was collected every two weeks from week 2 through 8. Serial blood collections were conducted every two weeks from week 2 through 8. Blood was collected, beginning at 0530, at 0, 15, 30, 45, 60, 75, 90, 105, 120, 135, and 150 minutes. The Scenic Highway area of the PetroProcessors of Louisiana, Inc. (PPI) Superfund site located near Baton Rouge, LA, has groundwater contaminated with a variety of chlorinated alkanes (e.g., 1,2-dichloroethane, 1,2-dichloropropane) and alkenes (e.g., trichloroethene and vinyl chloride). In March 2009, a field-scale pilot test was conducted to evaluate the efficacy of injecting diluted agricultural feed grade molasses into the groundwater in an effort to stimulate in-situ reductive dechlorination of these chlorinated contaminants. Nicotine is one of the main ingredients of cigarettes and a neurotoxicant which has been purported to affect many aspects of nervous system development causing long-term abnormalities in the offspring. We took advantage of the zebrafish model to investigate the effects of nicotine in nervous system anatomy and function. It was previously reported that embryonic nicotine exposure causes pathfinding errors in secondary motoneuron (SMN) axons through the activation of nicotine acetylcholine receptors (nAChRs). Understanding Zn interaction with mineral surfaces is essential to the understanding of Zn fate and toxicity. In this study, adsorption experiments investigated the impact of ligands and pH on the adsorption of Zn to mineral surfaces. X-Ray Absorption Fine Structure Spectroscopy (XAFS) was used to elucidate the adsorption mechanisms of Zn to mineral surfaces as impacted by ligands. In Louisiana, USA, many municipalities treat wastewater using natural systems, such as lagoons and wetlands, rather than using conventional wastewater treatment technologies and may discharge PhACs into the environment. These treatment systems are not designed to remove PhACs from wastewater, nor is it currently a regulatory requirement. Although birds of the forest canopy are an important component of tropical forest ecosystems, difficulty accessing the forest canopy has limited the advancement of knowledge pertaining to this group of species. Here I test methods for the study of canopy birds in lowland Neotropical rainforests, and identify recurring patterns of community structure in canopy bird assemblages as well as processes potentially responsible for these patterns. Nucleotide excision repair (NER) is a conserved DNA repair mechanism capable of removing a variety of helix-distorting lesions, such as UV-induced cyclobutane pyrimidine dimers (CPDs). NER can be grouped into two pathways: global genomic NER (GGR), which refers to repair throughout the genome, and transcription coupled NER (TCR), which refers to a repair mechanism that is dedicated to the transcribed strand (TS) of actively transcribed genes.
