Functional Characterization Of A Zinc Finger Protein Aebp2, Hana Kim

AEBP2 is a zinc finger protein that has been shown to interact with the mammalian Polycomb Repression Complex 2 (PRC2). I characterized this unknown protein and tested its potential targeting roles for the PRC2. AEBP2 is an evolutionarily well-conserved gene that is found in animals ranging from flying insects to mammals. The transcription of mammalian AEBP2 is driven by two alternative promoters and produces multiple transcripts that give rise to at least two isoforms of the protein. These isoforms show developmental stage-specific expression patterns: the larger adult-specific form (52 kDa) and the smaller embryo-specific form (31 kDa). The AEBP2 protein …

In this dissertation I explored the processes that generated high species diversity in a continental adaptive radiation of passerine birds: the family Furnariidae. Although there is a vast amount of information on the microevolutionary mechanisms that generate speciation and adaptation, much less is known about the processes underlying large-scale patterns of diversity. I used new techniques that exploit information contained in phylogenetic trees to investigate the diversification process in Furnariidae at a macroevolutionary scale. Nicotine, a drug of abuse, has been shown to have deleterious outcomes on nervous system development in vertebrates. Using zebrafish behavior as a diagnostic tool, we aimed to understand the effects of embryonic nicotine exposure on spinal neuron development. We initially quantified nicotine incorporation into zebrafish embryos. We found that only a fraction of the waterborne concentration of nicotine was incorporated into embryos. Also, when embryos were exposed to epibatidine and ABT 418, potent alpha 4/beta 2 nAChRs agonists, the embryos exhibited a robust swim-like behavioral response similar to that of nicotine. In riverine wetlands, the rate and magnitude of water exchanged between river channels and adjacent wetlands have a major influence on hydrologic function and associated aquatic habitat and water quality. River channels are generally separated from backswamps by natural levees; however, preferential flow may allow potentially substantial water and solute exchange between them. Determining the influence of preferential flow on water and solute transport in natural levees requires an innovative approach that allows measurement of transport rates at a scale large enough to capture field-scale variability. In wetland sediments, organic matter provides a substrate for microbial activity. During metabolism, microbes release extracellular polymeric substances, which accumulate to bind soil particles. A similar concept can be implemented on a large scale to reduce wetland loss in Louisiana. Hypothetically, hydraulically dredged sediment can be amended with polymer and deposited on subsiding marshes as a restoration method where the polymer increases sediment stabilization until plants become established. To control gene expression in vivo with spatial and temporal precision remains a significant hurdle in laboratory studies of development as well as clinical genetic therapies. Here we demonstrate such control over gene expression by use of photochemistry to reversibly inactivate the hybridization of a nucleic acid analog used for specific protein knockdown. A morpholino oligonucleotide, commonly used for knockdown of protein expression in developmental studies, was “caged” using carbodiimide conjugate chemistry which yielded photocleavable adducts that can be removed with light exposure. This research assessed the current conditions of water quality in a shallow subtropical lake, influenced by a highly developed urban environment. Specifically, the research focused on the dynamics of phosphorus, nitrogen, and dissolved oxygen, as well as the effects of stormwater runoff on lake water quality. Furthermore, the research quantified gross primary production, net primary production, and respiration in order to discern seasonal variation in lake metabolism. Microfluidic interfaces were developed for off-line matrix-assisted laser desorption/ionization mass spectrometry (MALDI). Microfluidic interfaces allow samples to be manipulated on-chip and deposited onto a MALDI target plate for analysis. For this research, microfluidic culturing devices and automated digestion and deposition microfluidic chip platforms were developed for the identification of proteins. The microfluidic chip components were fabricated on a poly(methyl methacrylate), PMMA, wafer using the hot embossing method and a molding tool with structures prepared via micromilling. High stress environments can reduce species diversity. How such stress-induced reduction in predator diversity impacts prey survival is less well studied. Brackish waters in estuaries are stressful, species depauperate areas, but also prime oyster habitat in Louisiana. Surveys revealed reduced bivalve predator diversity at the low salinity (high stress) site. Exclosure experiments indicated highest prey mortality at the high salinity (low stress) site. Predator enclosures corroborated the field study results, with reduced consumption rates at the low salinity site for both stone crabs, Menippe adina and oyster drills Stramonita haemastoma, but not blue crabs, Callinectes sapidus. This dissertation research aimed to quantify current soil organic carbon (SOC) stocks across Louisiana’s landscape, examine the spatial relationships between SOC and terrain factors at the watershed and river basin scales, and predict SOC changes in surface soils during future climate change. Using Louisiana as an example, a spatially-explicit modeling framework was developed that is conducive to watershed-scale prediction of soil carbon stock and change. SOC densities at the watershed scale were estimated using the USDA NRCS Soil Geographic Database (STATSGO). Multitudes of pathogenic and infectious microbes are known to spread via contaminated aerosols. Dental personnel have an increased incidence of respiratory infections. Ultrasonic scaling procedures are reported to produce the largest amounts of contaminated aerosols of any dental procedure. Traditionally, women have been relegated to the margins of society, history, and culture in male-dominated environments. Patriarchal systems have long denied women to play an appropriate role in nation building and to enter the public sphere, as is the case in Mexico. The female participation during one of the country’s most critical periods, the Mexican Revolution, has largely been ignored. Advances in cancer therapies continue to be improved, yet cancer continues to be one of the deadliest diseases in the world. Harnessing the power of the body’s immune system to attack cancer is a promising strategy that can further improve therapies for neoplastic diseases. As part of this strategy, cytokines such as interleukin (IL) 2 and interferon á are currently accepted cancer treatments, and other cytokines such as IL12 and GM-CSF also show potential as new treatments. Salt dome dissolution in the Gulf of Mexico sedimentary basin is a primary cause for elevated pore water salinities in the subsurface. Temperature, pressure, salinity, lithology and fluid density are parameters often used to identify preferential conduits and driving forces for fluid migration. These parameters were calculated using 20 wireline logs covering 40 km² on the south flank of a salt dome on the continental shelf, offshore Louisiana. 3-D seismic has been utilized to determine location of faults, to aid in sand correlation, and to provide a structural overview of the dome. Integrin conformational changes mediate integrin activation and signaling triggered by intracellular molecules or extracellular ligands. Even though it has been shown that TM and/or cytoplasmic α and β domains associate in the resting state and separation of these domains is required for integrin signaling, it is still not clear how this signal is transmitted from the transmembrane domain through two long extracellular legs to the ligand-binding headpiece. Lyme disease is a multisystem disorder caused by the tick vector Borrelia burgdorferi. During its life cycle between the tick vector and the mammalian host, Borrelia up- and down-regulates the expression of its surface lipoproteins. Of its many surface lipoproteins, the Outer surface protein C (OspC) is crucial for initial mammalian infection. OspC has a common role shared with other lipoproteins of protection against host innate defences and a unique function in facilitating the dissemination of B. burgdorferi in the murine host. The need to characterize offshore resources as borrow areas for beach restoration has initiated interest of the impact of storms to the inner continental shelf. While numerous studies have investigated the response of coastal systems to major storms, very little is known about the geological response of inner shelves to frequent and intense storms. While algae are often used in forensic science for the determination and confirmation of death by drowning, their utility for the estimation of postmortem submersion interval (PMSI) has been underutilized. Algae are present in all water systems and will grow upon decomposing matter; yet, very little published literature exists on their use in PMSI estimation. The spatial and temporal variation of soil properties was evaluated on three sites in close proximity to Bayou Wikoff, St. Landry Parish, Louisiana. A total of 1,068 surface (0-5 cm) samples were collected, geo-located, and transported to Louisiana State University for physical and chemical analyses. Physical and chemical properties were spatially and temporally displayed using ArcGIS. Interpolation techniques such as spline, kriging, and inverse distance weighting were evaluated to determine the best fit model for the project. Spline and inverse distance weighting were found to be the least accurate interpolation models. Kriging provided the most accurate model of spatial and …

Neutron production is of principal concern when designing proton therapy vault shielding. Conventionally, neutron calculations are based on analytical methods, which do not accurately consider beam shaping components and nozzle shielding. The goal of this study was to calculate, using Monte Carlo modeling, the neutron spectral fluence and neutron dose equivalent generated by a realistic proton therapy nozzle and evaluate how these data could be used in shielding calculations. We modeled a contemporary passive scattering proton therapy nozzle in detail with the MCNPX simulation code.
