Breeding for grain quality traits and resistance to sheath blight (SB), a disease caused by Rhizoctonia solani Kuhn, are important objectives for the rice (Oryza sativa L.) industry. Grain quality traits and SB resistance play an important role in the economic prosperity of commercial rice markets. The objectives of our research were to: (1) Explore performance and stability for SB resistance among doubled-haploid (DH) lines of the SB2 mapping population using GGE biplots (2) Exploit whole genome sequences of 13 inbred lines to identify non-synonymous SNPs (nsSNPs) and candidate genes for SB resistance. Sweetpotato potyviruses [Sweet potato feathery mottle virus (SPFMV), Sweet potato virus G (SPVG) and Sweet potato virus 2 (SPV2)] commonly infect sweetpotato and weedy morning glories in the USA. These viruses are transmitted in a non-persistent manner by various aphid species and cause up to 15% yield loss. Sweetpotato is vegetatively propagated, and in the USA growers are supplied with virus tested propagation material to minimize impact of viruses. However the rapid re-infection of these materials with viruses warranted further studies to determine factors that influence the epidemiology of these viruses. Insulin resistance in type 2 diabetes is associated with impaired glucose and protein metabolism in skeletal muscle. The impaired insulin signaling in skeletal muscle affects muscle mass by tilting the balance between skeletal muscle protein synthesis and degradation toward degradation, a process that is primarily regulated by the ubiquitin-proteasome system. Studies have shown that an extensively characterized ethanol extract of Artemisia dracunculus L (Russian Tarragon), termed PMI 5011, enhances insulin signaling in human primary skeletal muscle cells and in a rodent model of insulin resistance. The inception of No Child Left Behind (NCLB) has focused national attention on improving the academic achievement of all students. In response to this federal legislation, educators, policymakers and others have sought remedies to turnaround chronically low-performing schools. The academic achievement outcomes of implementing such strategies have been mixed. Some schools have experienced clear, unambiguous growth. Others have remained stagnant. Others have regressed. For these three areas, crystallography has played and continues to play a huge role by providing researchers with the means to determine the structure of a compound and relate these to its properties. This dissertation is highly interdisciplinary, focusing on structure determination of complex systems and materials with tailored applications. This dissertation explores the effects of stress and disturbance on fungal endophyte diversity and composition in obligate wetland plants in southeast Louisiana. I explored two types of endophytes: root and foliar. Each of these groups is known to live within apparently healthy host plants without causing symptoms of disease; therefore, plants are thought to benefit from this association. Few studies, however, have shown that plants in wetlands associate with fungi. In this dissertation, I show that both root and foliar endophytes not only exist in wetland plants living in flooded and saline environments, but also are abundant within these plants. …

Salvaging of epididymal sperm from injured or deceased animals allows for the propagation of favorable traits from endangered or genetically superior males. Techniques developed in domestic species can also serve as the foundation for the collection, cryopreservation and utilization of epididymal sperm in exotic breeds. The need to preserve and utilize epididymal sperm, in the most efficient manner, is of the utmost importance. In a series of experiments, ejaculated and epididymal sperm from the same four mature, fertile, Holstein bulls were collected, cryopreserved, cultured and used for in vitro fertilization. The gut of insects and plant tissues are productive sources for the isolation of undescribed species of yeasts. In particular, the gut of lignicolous insects is colonized by yeasts that can carry out the fermentation of several sugars. The consistent association between xylose-fermenting (X-F) yeasts and the gut of lignicolous insects has been used as evidence of a symbiotic relationship between them. In general passalid beetles (Passalidae) and wood-roaches (Cryptocercidae) feed on rotted wood where they spend most of their lives. Children diagnosed with Autistic Disorder (AD), Asperger’s Disorder (AS), and Pervasive Developmental Disorder Not Otherwise Specified (PDD-NOS) share overlapping diagnostic criteria. As a result, there has been an enduring debate regarding the appropriateness of the current categorical classification system used to diagnose this group of disorders, commonly referred to as Autism Spectrum Disorders (ASD). The goal of this dissertation was to integrate multiple disciplines for application of high-throughput sperm cryopreservation to aquatic species. The blue catfish Ictalurus furcatus was selected for research due to its role in production of hybrid catfish with eggs from channel catfish Ictalurus punctatus. A high-throughput cryopreservation pathway was developed by systematically evaluating each process factor and using automated processing systems. Large quantities of cryopreserved sperm produced from this protocol were evaluated in commercial-scale hatcheries in cooperation with aquaculture producers. Storage root development is the most important physiological process in sweetpotato. Understanding the underlying genetics is the overall objective of this research. A secondary objective is examining the impact of abiotic stress on gene expression and storage root development. A comprehensive analysis of a high-throughput RNA sequencing data (sweetpotato root, stem, and leaf) and public Expressed Sequences Tags was done to generate a genome-wide transcriptome assembly. About 33 million sequences were assembled into 77,663 unigenes; 52,322 (69.55%) of these unigenes matched to a protein sequence from UniprotKB. RNA interference (RNAi) is a phylogenetically conserved gene regulation mechanism that modulates a wide variety of biological functions through suppressing gene expression at transcriptional or posttranscriptional levels (Bass 2000; Sharp 2001). One of the major natural functions of RNAi is antiviral defense in cytosol. Research indicates that the average age of the American farmer is 57 years old. There is a need for new programs to be developed to encourage youth to pursue careers in agriculture. This study developed and implemented a horticultural curriculum at a summer camp targeting participants aged 9-12. A treatment group consisted of campers that signed up for the Victory Garden camp track and a control group consisted of randomly selected campers from a different camp track. RNA Polymerase III (Pol III) is best characterized for its transcription of tRNA molecules. Interestingly, Pol III and its associated complexes have been found to be involved in the regulation of transcription of genes transcribed by RNA Polymerase II (Pol II) (Kleinschmidt, LeBlanc et al. 2011). Extra TFIIIC sites (ETC sites) are chromosomally located sites discovered in S. cerevisiae that bind the Pol III transcription factor TFIIIC only through B-box interactions. Herpes simplex virus type 1 (HSV-1) is a DNA virus that is a ubiquitous pathogen of humans. The hallmark of the HSV-lifecycle is infection of a mucosal surface with spread to sensory neurons where the virus establishes a latent infection with periodic recurrences for the life of the host. The most common course of symptomatic disease with HSV-1 is the typical mucocutaneous lesion that is self-limiting. HSV-1 can also cause acute encephalitis and ocular pathology on reactivation. Under Louisiana climatic conditions, production of sugarcane (Saccharum spp.) is limited to a maximum growth period of nine months. To increase sucrose concentration in the crop, ripener is applied prior to harvest. The chemical ripeners, glyphosate and trinexapac-ethyl were applied to the sugarcane cultivars HoCP 96-540, L 99-226, L 99-233, HoCP 00-950, and L 01-283 eight weeks prior to harvest. When glyphosate was applied at 210 g ae/ha, TRS for the cultivars was increased 10 to 28% compared with the nontreated. The goal of this dissertation is to investigate electroosmotic flow (EOF) and electric field dynamics during capillary electrophoresis (CE) experiments using methods based on periodic photobleaching of fluorophores added to the separation buffer at nanomolar concentrations. The methods provide time resolved EOF and local electric field information during an experiment, which can be applied to fundamental studies to provide better understanding of CE techniques. Crayfish harvested from the Atchafalaya River Basin (ARB) represent the majority of Louisiana wild crayfish landings. However, excluding water level influences, it is difficult to elucidate inter-annual harvest differences and intra-annual population variability among habitats. This research investigated ecological influences on population characteristics of red swamp crayfish Procambarus clarkii in the southeastern ARB as well as physiological biomarkers of hypoxic stress in P. clarkii. Biotic and abiotic effects on P. clarkii populations were examined throughout the 2008 and 2009 commercial crayfish seasons. In Louisiana, current N rate recommendations for sugarcane production are based on multiple year N response trials and refined based on soil and crop variables. Without accounting for current growing conditions, recommendations can potentially lead to over- or under-application of N. Ecological speciation is a process involving adaptation to different environments leading to reproductive isolation between populations. Hybridization between Clapper and King rails (Rallus longirostris and R. elegans) is frequent but restricted to narrow bands of brackish marsh along the Gulf of Mexico and Atlantic coastlines of North America. In the second chapter I used mitochondrial and nuclear sequences to infer phylogenetic relationships in this species complex. The complex falls out into three distinct groups: South America; western North America; and eastern North America and the Caribbean. Unionoids (freshwater mussels) are important in aquatic ecosystems, yet despite their continued decline in diversity, little is known about their distribution and ecology. To study Louisiana mussel species richness, abundance, and distribution I sampled 65 sites within six major watersheds in the Pine Hill region, Louisiana. Second through sixth order streams were surveyed for local instream variables, land use, hydrology, and co-occurring fish assemblage. The research goals presented here were to evaluate the solvent concentrations over which Dehalogenimonas lykanthroporepellens strain BL-DC-9T and Dehalogenimonas sp. IP3-3 are able to reductively dechlorinate 1,2-dichloroethane (1,2-DCA), 1,2-dichloropropane (1,2-DCP), and 1,1,2-trichloroethane (1,1,2-TCA). Both of these novel bacteria were isolated from contaminated groundwater at the PetroProcessors of Louisiana, Inc. Superfund site located near Baton Rouge. Both cluster in the phylum Chloroflexi, related to, but distinct from, the widely studied, reductively dechlorinating “Dehalococcoides” strains .

Chapter 1 is a description of the basic fundamental concepts of porphyrins and BODIPYs, (including synthetic methodologies), a general mechanism of the processes of EGFR, and computational concepts that are elaborated upon in future chapters. Chapter 2 is a discussion solely on the porphyrins that were synthesized to be used as fluorophores for EGFR-targeted conjugates, both peptidic and non-peptidic. The porphyrins were primarily synthesized via the Lindsey method as well as by O-alkylation of tetrahydroxy porphyrins. Synthesis of PEG linkages are discussed as well through three methodologies. Advice In Troubles Talk Conversations Between Strangers: The Role Of Problem Seriousness And The Impact Of Advice On Helper Supportiveness And The Desire For Future Interactions, Andrea Jean Vickery

In troubles talk conversations, problems are disclosed and discussed. When responding to problem disclosures, advice is one common response where the respondent recommends how to think, feel or act in response to a problem. This thesis focuses on extending our understanding of advice messages, with a main research question focused on determining if advice occurs in initial interactions between strangers. Through an analysis of 125 transcribed conversations, advice was present in 38.4% (n = 44) of the conversations. Advice was offered in response to less serious problems, supporting the first hypothesis. There was no support found for the positive association …

This dissertation examines gender policies in former Soviet republics. Gender policies are depicted as traditional policies (including such policies as child support, spousal support, and inheritance rights), violence against women policies (domestic violence, marital rape, sexual assault, and sexual harassment), and human trafficking policies. It builds upon previous works with a multi-methodological approach (content analysis, logistical regression, and qualitative survey analysis). The focus here is on the factors that influence adoption of formal policies in former Soviet republics from 1993-2008.
