Uterine lumen fluid (ULF) is central to successful pregnancy establishment and maintenance, and impacts offspring wellbeing into adulthood. The current dogma is that ULF composition is primarily governed by endometrial glandular epithelial cell secretions and influenced by progesterone. To investigate the hypothesis that ULF is metabolically semi-autonomous, ULF was obtained from cyclic heifers, and aliquots incubated for various durations prior to analysis by untargeted semi-quantitative metabolomic profiling. Metabolite flux was observed in these ULF isolates, supporting the idea that the biochemical makeup of ULF is semi-autonomously dynamic due to enzyme activities. Stress-inducible promoters are vital for the desirable expression of genes, especially transcription factors, which could otherwise compromise growth and development when constitutively overexpressed in plants. Here, we report on the characterization of the promoter region of a stress-responsive gene SaAsr1 from monocot halophyte cordgrass (Spartina alterniflora). Several cis-acting elements, such as ABRE (ABA-responsive element), DRE-CRT (dehydration responsive-element/C-Repeat), LTRE (low temperature-responsive element), ERE (ethylene-responsive element), LRE (light-responsive element), etc. contributed at varying degrees to salt-, drought- and ABA-enhanced expression of gusA reporter gene in Arabidopsis thaliana under the full-length promoter, pAsr11875 and its deletion derivatives with an assortment of cis-regulatory motifs. …

There is considerable uncertainty regarding which ecosystems are most vulnerable to warming. Current understanding of organismal sensitivity is largely centred on species-level assessments that do not consider variation across populations. Here we used meta-analysis to quantify upper thermal tolerance variation across 305 populations from 61 terrestrial, freshwater, marine and intertidal taxa. We found strong differentiation in heat tolerance across populations in marine and intertidal taxa but not terrestrial or freshwater taxa. This is counter to the expectation that increased connectivity in the ocean should reduce intraspecific variation. Such adaptive differentiation in the ocean suggests there may be standing genetic variation …

Background: Interactions between genetic, metabolic, and environmental factors lead to gestational diabetes mellitus (GDM). We aimed to examine interactive effects of cyclin-dependent kinase 5 regulatory subunit-associated protein1-like 1(CDKAL1) rs7747752 polymorphism with low serum levels of L-carnitine, choline, and betaine for GDM. Methods: A nested case-control study of 207 GDM women and their one-to-one, age-matched controls was organized from a prospective cohort of pregnant women in Tianjin, China. Conditional logistic regressions were used to test associations between CDKAL1 rs7747752 and serum levels of L-carnitine, choline, and betaine, and the risk of GDM. Water is essential for survival, but one in three individuals worldwide (2.2 billion people) lacks access to safe drinking water. Water intake requirements largely reflect water turnover (WT), the water used by the body each day. We investigated the determinants of human WT in 5604 people from the ages of 8 days to 96 years from 23 countries using isotope-tracking (2H) methods. Age, body size, and composition were significantly associated with WT, as were physical activity, athletic status, pregnancy, socioeconomic status, and environmental characteristics (latitude, altitude, air temperature, and humidity). Louisiana, U.S.A., is among the most vulnerable areas globally to coastal natural hazards, with risk vulnerability likely increasing. The risks associated with non-tropical-cyclone hazards in Louisiana’s coastal zone have been understudied. This research enhances present and future (i.e., 2050) Louisiana risk assessment using locally-weighted, model-based hazard frequency/intensity and population projections.
Results suggest that property risks associated with extreme cold temperature and tornado are and will remain costlier than those for hail and lightning. Property risks of extreme cold temperature and hail are projected to decrease with the expected warming temperatures, with those of all four of these hazards peaking in …

This research focused on the Roseau cane (Phragmites australis) dieback assessment in the lower MRD of Louisiana and introduced a comprehensive and systematic multi-source remote sensing method for assessing wetland and Roseau cane dieback and habitat dynamics analysis from three scale levels from large-scale to small scale.
Large-scale historical vegetation/land change analyses were conducted in the lower MRD based on Landsat in the past two decades (2001 - 2021). Landfills are complex geostructures which contains organic and inorganic municipal, and in some cases industrial, wastes and are expected to remain operational for long times. Due to the complex nature of physical, chemical, biological, and thermal reactions that carry on within the depths of a landfill, unexpected incidents such as elevated temperatures could become inevitable. While uncommon to happen, elevated temperatures cause health and environmental issues such as odors, rapid settlements, slope instabilities. Schrenkiella parvula is an extremophyte model related to the most widely studied plant model, Arabidopsis thaliana and Brassica crops in the mustard family (Brassicaceae). It can thrive in highly saline environments where the soil is enriched in Na+, K+, Li+, borates, and chlorides. Understanding how this extremophyte can survive high salinity with genomic adaptations can provide insight into developing stress resilient crops in the future. Carbon dioxide (CO2) outgassing from inland waters is a small but critical component of the global carbon cycle that is currently not well constrained. This thesis is composed of three interrelated studies with an overarching goal to further determine environmental factors impacting carbon dynamics in rivers and lakes. We present dynamical and morphological studies on poly(dimethylsiloxane) (PDMS)-based bottlebrush polymers in solution. A combination of small-angle neutron scattering (SANS), pulsed field gradient nuclear magnetic resonance (PFG NMR), and neutron spin echo (NSE) spectroscopy was used to identify structural changes inherent in changing the side-chain length while keeping the backbone constant. These morphological changes are mirrored in the diffusion coefficient determined by PFG NMR, resulting in two coefficients for the elongated and one for the more spherical sample. Faster motions are tracked by NSE using the advantage of time- and length-scale resolutions with the diffusion coefficient predetermined by PFG NMR. …

To identify novel insecticide target sites that can address the evolution of insecticide resistance, our group has tested the insecticidal potential of two potassium flux pathways: potassium-chloride (K+/Cl−) cotransporter (KCC) and inward rectifier potassium (Kir) channels. GABAergic signaling is the cornerstone of fast synaptic inhibition for neural signaling in arthropods; thus, the γ-aminobutyric acid (GABA)-receptor-chloride-channel (GRCC) complex has been a longstanding target for insecticides. Despite the importance to neuronal homeostasis, the upstream mechanisms enabling GRCC function are poorly defined. Nanotechnology is a multifaceted and a promising strategy in agricultural pest management for the development of insecticides or insecticide carriers. Research has shown that nanoparticles increase stability and efficacy of entrapped chemical pesticides to prevent active ingredient degradation while improving chemical stability and solubility. The following work evaluated nanoparticles synthesized from biopolymers, materials that originate from renewable sources also have an increased biodegradability.

Paraquat is the only herbicide labeled for postemergence control of Italian ryegrass in sugarcane. In 2017, sugarcane farmers in Louisiana reported Italian ryegrass control failures with paraquat applications. Seeds were collected from sugarcane fields in White Castle, St. Gabriel, and Bunkie, Louisiana where control was not achieved (reduced susceptibility). Additionally, Italian ryegrass seeds were collected from a sugarcane field in Welcome, LA with a history of paraquat use and with no reported control issues (susceptible population). Much is known about the development of insecticide resistance associated with targeted application against insect populations. However, off-target selection by applications of insecticides in agricultural and residential sites also impacts development of insecticide resistance and is understudied. Similarly, the impact of selecting one life stage of mosquitoes on the insecticide susceptibility of different life stages is largely unknown. The Gram-negative bacterium, Klebsiella pneumoniae, is a major cause of hospital-acquired pneumonia in the U.S. In particular, the dramatic increase in carbapenem-resistant K. pneumoniae infections poses a serious threat to the public health both in the United States and worldwide. The Formosan subterranean termite, Coptotermes formosanus, is a eusocial insect and an invasive urban pest. Colonies function through a division of labor among castes (i.e., queens and kings, workers, and soldiers). Though soldiers are energetically expensive to maintain, C. formosanus has a relatively large soldier caste (~10% of the colony). Soldiers are physically unable to collect food or build tunnels but are regularly found with foraging workers. In some species, soldiers influence the worker foraging behavior. Mixed species forests may benefit from the reduction in intraspecific competition, although the benefit of species mixing has been shown to vary depending on mixture composition and environmental conditions. This study sought to test the effect of species composition in mixed forest stands on canopy development and growth of loblolly pine (Pinus taeda), cherrybark oak (Quercus pagoda), and sweetgum (Liquidambar styraciflua). Even-aged, grid planted monoculture and mixed species forest plots were established with hardwood monoculture plots at a 741 Trees Per Hectare (TPH), and pine monoculture and mixed species plots established at 1482 TPH. …

Biochar is a porous carbonated substance that is manufactured through pyrolysis of biomass feedstocks in an oxygen-deficiency environment. The production of biochar from agricultural waste materials provides a potential solution for eliminating pollution while improving carbon sequestration. While biochar is shown to provide multiple benefits to soil health and fertility, its use in environmental remediation has been hampered by its low affinity for hazardous anionic species and the difficulty in isolating from water phase. Molecular detection techniques have huge potential in clinical environments. In addition to many other molecular detection techniques, endoscopic Raman spectroscopy has great ability in terms of minimal invasiveness and real-time spectra acquisition. However, Raman Effect is low in sensitivity, limiting the application. Surface-Enhanced Raman Scattering (SERS), addresses this limitation. SERS brings rough nano-metallic surfaces in contact with specimen molecules which enormously enhances Raman signals. Kisspeptins, a family of small peptides derived from the Kiss1 gene, have been described in trophoblast cell dynamics, placentation, and pregnancy outcomes. Furthermore, many other physiological functions have been attributed to kisspeptins, including neuroendocrine regulation of puberty and cyclicity, and peripheral and central regulation of metabolism and adipogenesis. Herein, studies were performed in the Blood Pressure High Subline 5 (BPH/5) mouse and the horse to further investigate the role of kisspeptins in pregnancy. The BPH/5 mouse recapitulates the main features of human preeclampsia (PE), a hypertensive disorder of pregnancy associated with maternal and fetal adverse outcomes.
