The purpose of this study was to determine if relationships exist between student demographic or personal data, academic achievement and student perceptions of factors relative to predicting performance on the Registration Dietetic (R.D.) Exam. Perceptions of factors relative to predicting performance on the R.D. exam was also determined from undergraduate dietetic program directors, to explore the extent of agreement between students and educators. This information was used to develop a predictive model explaining a significant proportion of the variance in R.D. exam scores. This exploratory study investigated 11th grade high school chemistry students' understandings of the science concepts of chemical symbol, formula, equation, reaction, reactant, and product. It also investigated students' ideas about the meaning of plus sign, reaction sign, subscript, and coefficient. In addition, it attempted to assess the impact of students' prior mathematical knowledge on their understanding of these fundamental concepts. A combination of quantitative and qualitative methods were employed in a two-stage approach involving a preliminary study and a main study over one academic school year (1990-1991). A panel of eleven monoclonal antibodies of the IgG and IgM class were developed against the HM 175 strain of the Hepatitis A virus. Immunoglobulin specificity for the Hepatitis A virus was determined and the antibodies were applied toward the development of an antigen capture assay linked to a nucleic acid amplification step (RT-PCR), for detection of the Hepatitis A virus. The neutralizing mouse monoclonal antibody and a rabbit polyvalent serum were assessed as capture antibodies for the immunoaffinity stage of the assay. The McrBC (Modified Cytosine Restriction) restriction system has the ability to restrict DNA containing 5-hydroxymethylcytosine, N$\sp4$-methylcytosine, and 5-methylcytosine at specific sequences. The mcrB gene produces two gene products. The complete mcrB open reading frame produces a 53-kDa protein (McrB$\sb{\rm L}$) and a 35-kDa protein (McrB$\sb{\rm S}).$ The smaller McrB polypeptide is produced from an inframe, internal translational start in the mcrB gene. The mcrC gene produces a single 38-kDa protein. Evidence was presented that McrB$\sb{\rm S}$ regulates the activity of McrBC. When McrB$\sb{\rm S}$ was overproduced in a McrBC$\sp+$ background, there was dramatic loss of restriction. Underproduction of this protein …

This study examined the importance of the leaf aerenchyma gas space as a CO$\sb2$ source for photosynthesis in Typha latifolia L. (broadleaf cattail). In the field there was a distinct diurnal pattern of CO$\sb2$ concentration ( (CO$\sb2$)) in the aerenchyma gas space. At dawn the aerenchyma (CO$\sb2$) was 4 to 18 times above atmospheric levels. By midday the aerenchyma (CO$\sb2$) declined to near atmospheric levels and increased again in the late afternoon. It is hypothesized that this diurnal pattern may be controlled by photosynthetic demand for CO$\sb2$. Aerenchyma gas space was estimated as $>$50% of leaf volume, and the …

Although chaperonin-assisted protein folding has been studied in vitro by a number of investigators, the features of an unfolded or partially folded polypeptide that are recognized and bound by chaperonins are not known. I addressed this question using the precursor of the small subunit (pS) of ribulose-1,5-bisphosphate carboxylase as a model substrate for GroEL, the bacterial chaperonin. The precursor protein was expressed in E. coli as a C-terminal fusion to protein A. Protein A-pS (and any associated cellular proteins) was isolated by affinity chromatography. Germline mutation rates measured per generation per locus in three species of Metazoa are used to address questions of the constancy of mutation rates per generation, per year and per germcell division. Null mutation rates per generation are: 3.6 $\times$ 10$\sp{-6}$ for human X-linked disease, 8.9 $\times$ 10$\sp{-6}$ for Specific Locus Test (SLT) in male mice, 3.2 $\times$ 10$\sp{-6}$ for SLT in female mice, 8.0 $\times$ 10$\sp{-6}$ for SLT in male Drosophila melanogaster (D.m.), 1.6 $\times$ 10$\sp{-6}$ for SLRL test in male D.m. and 2.0 $\times$ 10$\sp{-6}$ for SLRL test in female D.m. This dissertation describes extensions of post-structuralism in contemporary curriculum discourses. Post-structuralist thought is mainly associated with the seminal work of Michel Foucault, Jacques Derrida, Gilles Deleuze, Jean-Francois Lyotard, and Michel Serres. Post-structural criticism and analysis challenge prevailing structuralist approaches and question the fundamental assumptions upon which these approaches rest. A key assumption of structural approaches is that all phenomena are constituted by an underlying structure. In curriculum, these structural assumptions (often scientific) remain unacknowledged and thus are immunized against criticism; rather, they are incorporated into the preferred structural analyses, interpretations, and organizations promoted by the promise of order and rationality. …

New methods for assessing solidification/stabilization (S/S) of hazardous waste using solid-state nuclear magnetic resonance are developed for the purpose of diagnosing on a microscopic level the performance of S/S as a hazardous waste disposal technique. An inhibitor activity that prevents the proteolytic processing of Cowpea Mosaic Virus (CPMV) polyprotein has been discovered in crude wheat germ extracts. CPMV is a comovirus that replicates via the synthesis of two polyproteins which are subsequently cleaved by a viral proteinase to yield the functional viral proteins. Since this scheme of replication is common to many other viral groups, a potential target for antivirals is the proteinase responsible for polyprotein processing. Doris Lessing's The Golden Notebook, Alice Walker's The Color Purple, and Margaret Atwood's The Handmaid's Tale are narrated by women whose social identities are partially constructed through activities traditionally associated with mothering. In each text, maternity signifies a biological event as well as the social, sexual, and material relations involved with pregnancy, childbirth, and the nurturing and rearing of children, activities which become grounds for investigating the politics of the body, of sexuality, and consequently, of gender relations. Maternity becomes politicized within the cultural contexts of each novel as maternal experience functions to produce both feminist and postmodernist critiques. In …

Reactions against scale allografts in gulf killifish were studied to examine circadian variation and neuroendocrine regulation of immune function. Although melanophores in autografts remain viable indefinitely, melanophores in scale allografts were broken down predominantly at night (12-h scotophases) regardless of the time of transplantation. Daily treatment with hormones or antagonists of neuroendocrine receptors altered the rhythm of melanophore breakdown when administered at light onset or light offset. Daily rhythms of immune activity may be a direct reflection of growth hormone levels and are entrained by exogenous cortisol treatment. Daily nonphotoperiodic environmental stimuli overrode photoperiodic entrainment of immune activity. Peak melanophore …

Studies were conducted in the field and greenhouse to identify sources of rice water weevil (Lissorhoptrus oryzophilus Kuschel) resistance in rice, Oryza sativa L., and determine the mechanisms of resistance. Rice lines of various sources, including breeding lines, somaclone lines and world collection lines, were evaluated. Tolerance was investigated using replicated insecticide treated and untreated plots in a split-plot design, and antibiosis and/or antixenosis were assessed using caged insect-infested plants in randomized block designs. The objective of this study was to evaluate the antimicrobial properties of glycerol monolaurate (monolaurin) either alone or in combination with organic acids against Listeria monocytogenes in model broth or food. The minimal inhibitory concentration (MIC) of monolaurin was reduced by decreasing the pH value of the medium. The contribution of temperature to monolaurin effectiveness showed that lethal effects of monolaurin increased at higher temperatures and lower pH values, whereas, bacteriostatic effects on growth increased as temperature and pH decreased. In order to study intramolecular enzyme catalysis, chemical models have been designed to quantify the proximity and orientation effects which occur in carboxylate catalyzed hydrolyses. As part of this broader study, various 2,2$\sp\prime$,7,7$\sp\prime$-tetrasubstituted 1,1$\sp\prime$-ethynylenedinaphthalenes are synthesized from 2,7-dihydroxynaphthalene. Two different strategies are used to obtain the title compounds. The first strategy involves palladium-mediated couplings between electron-poor aryl iodides and electron-rich arylethynes and allows synthesis of unsymmetrical binaphthylethynes. The second approach couples two 2,7-disubstituted naphthalenes with tetrachlorocyclopropene in the presence of aluminum chloride, but only symmetrical binaphthylethynes are obtained. Mussels representing two species (Mytilus trossulus and M. galloprovincialis) were collected from 28 littoral locations on the Pacific coast of the USA, and examined for genetic variation at 15 allozyme loci. Eleven samples from a region of suspected hybridization between the two species were also analyzed for variation in seven shell characters. Analysis of allozyme data by principal components analysis revealed three groups based on first principal component scores, and were identified as M. trossulus, M. galloprovincialis and hybrids. Canonical discriminate analysis of seven shell characters was less successful in separating mussels based on morphological criteria alone. To explore the …

The objective of this dissertation was to investigate the small-scale spatial and abundance patterns of meiofauna and their relationship with sediment microtopography and hydrodynamics. Meiofauna are known to be aggregated at small spatial scales. A study was first undertaken to determine, how patchiness differed over seasons and habitats. Significant small-scale aggregations were remarkably consistent across different seasons and habitats for all abundant copepods of an intertidal mudflat. Spatial autocorrelation analysis showed that the abundance of copepods was autocorrelated at short distances (cm scale) on all sample collecting dates, indicating a consistent spatial pattern. The study focused on six at-risk students enrolled in the sixth grade at a large urban Southern middle school. These students, along with their classroom teacher, participated in two interactive reading events using illustrated trade books. The purpose of the study was to examine how these students used the pictures and text to create meaning for the stories. The two illustrated selections differed in story complexity, amount of world knowledge required for interpretation, and relationship of pictures to text (i.e., a close relationship indicated both picture and text communicated similar information, while a distanced relationship indicated one source provided story …

This research focuses on the phenotypic and genotypic characteristics of a temperature sensitive mutant of herpes simplex virus type 1, HSV-1 (tsZ47). This virus was previously reported to carry two different mutations, which caused it to be temperature sensitive for viral replication and transport of glycoproteins to the infected cell surface. However, altered glycoprotein transport to the cell surface was not observed in this study. In addition, cells transformed with unique long gene 28 (UL28), but not glycoprotein B (gB), complemented tsZ47 at the nonpermissive temperature indicating that a single mutation could restore the wild type phenotype to the mutant …

Chemical factors controlling the energy mobilization in three species of insects were identified, partially characterized and their role in the process of energy mobilization were studied. High performance liquid chromatographic fractions were isolated from methanolic extracts of the corpora cardiaca (CC) from the honeybee Apis mellifera, the paper wasp Polistes exclamans and the house cricket Acheta domesticus. Each species had a single active fraction that was able to elevate the lipid titer in Acheta. Pyroglutamate aminopeptidase digestion of isolated active fractions indicated that these molecules are all peptides with a terminal pyroglutamate. Methanolic extracts of CC from Apis, Polistes and …

We proposed that known helix-stabilizing, minor-groove binding agents such as CC-1065 and its analog, U-71,184, might be feasibly targeted to specific mRNA by tethering them to antisense oligonucleotides. The CC-1065-tethered antisense oligonucleotide might be hyperstabilized to the target mRNA and arrest the translation of mRNA. Our hypothesis is that these agents can bind and increase the stability of RNA-DNA duplexes which may occur between mRNA and antisense oligonucleotides. A typical soil under natural forest classifies as fine, oxidic, isomesic, Typic Haplorthods. Cultivated soils classify as fine, oxidic, isothermic Humic Hapludoxs. Acetylation of bacterial alginate by Pseudomonas syringae subsp. phaseolicola ATCC 19304 was independent of alginate biosynthesis. This allowed the development of a process for acetylating seaweed alginate using immobilized P. syringae ATCC 19304 cells. About 50% of the mannuronic acid residues of seaweed alginate were acetylated by carbon immobilized P. syringae cells in a fluidized bed, up-flow reactor system fed continuously with seaweed alginate and gluconic acid. O-Acetylation by this process was found to be specific for the C-2 and/or C-3 position(s) of mannuronate residues. Acetylated seaweed alginate showed altered properties including increased viscosity and changed affinities for some cations.

Mangroves are salt-tolerant trees and shrubs that dominate the intertidal zone in many tropical and subtropical coastal regions. These intertidal forests often exhibit relatively distinct and recurring patterns of species distribution. The movement of agricultural chemical contaminants in alluvial soils with a shallow water table in a warm, humid environment was evaluated in this study. Bromide, a non-adsorbent tracer, was used to assess the pathways of water and solute in the soil profile. The movement and fate of three commonly used herbicides: Trifluralin, Metolachlor, and Metribuzin were determined. The data gathered from the herbicide study were used to compare with the results of simulation to validate the pesticide component of the GLEAMS model. Insect metamorphosis is relatively easy to explain at a superficial level, but a deep understanding of the concept reveals a complex array of interrelationships, most importantly those that address evolution. This is significant because, in biology, evolution is considered to be a central theme. Participants for this study were selected from grades 5, 7, 9, and 11 according to low, middle, or high achievement levels in their science classes, and to scores on a national achievement test. Protective resistance to Strongylus vulgaris infection was examined by immunization of ponies with radiation-attenuated larvae or soluble parasite homogenates. Twelve yearling ponies raised and maintained under parasite-free conditions were divided into four groups which received either radiation-attenuated L$\sb3$; aqueous-soluble adult somatic antigens plus adjuvant; aqueous-soluble larval somatic antigens with excretory/secretory products and adjuvant; or media with adjuvant. Ponies were immunized twice; radiation-attenuated L$\sb3$ were administered orally and soluble antigens or control injections were given intramuscularly.
