CHANGES in delivery of river-borne nutrients such as dissolved phosphate, nitrate and silicate, owing to land-use changes and anthropogenic emissions, are known to result in eutrophication1— enhanced phytoplankton blooms—and more severe hypoxic events2–1 in many enclosed bays and seas. Although similar ecological effects might be expected on continental shelves, the occurrence of such eutrophication has remained unresolved5. Here we present evidence of eutrophication of the continental shelf near the outflow of the Mississippi river, obtained by quantifying biologically bound silica (BSi) in diatom remnants within dated sediment cores. The human β-globin Locus Control Region (LCR) has two important activities. First, the LCR opens a 200 kb chromosomal domain containing the human ε-, γ- and β-giobin genes and, secondly, these sequences function as a powerful enhancer of ε-, γ- and β-globin gene expression. Erythrold-specific, DNase I hypersensitive sites (HS) mark sequences that are critical for LCR activity. Previous experiments demonstrated that a 1.9 kb fragment containing the 5′ HS 2 site confers position-independent expression in transgenic mice and enhances human β-giobin gene expression 100-fold. A Rationale For Coastal Wetland Restoration Through Spoil Bank Management In Louisiana, Usa, R. Eugene Turner, E. M. Swenson, J. M. Lee

The rationale and outline of an implementation plan for restoring coastal wetlands in Louisiana is presented. The rationale for the plan is based on reversing the consequences of documented cause-and-effect relationships between wetland loss and hydrologic change. The main feature is to modify the extensive interlocking network of dredged spoil deposits, or spoil banks, by reestablishing a more natural water flow at moderate flow velocity (/sec). Guidelines for site selection from thousands of potential sites are proposed. Examples of suitable sites are given for intermediate marshes. These sites exhibit rapid deterioration following partial or complete hydrologic impoundment, implying a strong …

Backfilling Canals To Restore Wetlands: Empirical Results In Coastal Louisiana, R. Eugene Turner, James M. Lee, Christopher Neill

Wetland restoration is largely a developing science and engineering enterprise. Analyses of results are too few and constrained to observations over a few years. We report here on the effectiveness of one restoration technique used sparsely in coastal Louisiana for several decades. Canals have been dredged in coastal Louisiana wetlands since 1938 for oil and gas exploration and extraction. These canals are typically dredged to 2.5 m depth and are 20 to 40 m wide. Canal lengths vary from 100 m to several 1000s m in the case of outer continental shelf pipeline canals that cross the wetlands.

Three tissue culture techniques, somaclone evaluation, in vitro screening of suspension cells, and in vitro screening of protoplasts were evaluated for their potential to produce aluminum (Al) tolerant rice. For somaclone evaluation, a collection of 9,000 rice somaclones was screened. After 11 days of germination in 200 uM Al, the length of the primary root was measured and used as the criterion for selection since a principal effect of Al toxicity is inhibition of root growth. Four superior lines were identified. Root length of the most tolerant line was 50% greater than its control following germination in screening solution. A …

The regulatory enzyme aspartate transcarbamoylase (ATCase), comprising 2 catalytic (C) trimers and 3 regulatory (R) dimers, owes its stability to the manifold interchain interactions among the 12 polypeptide chains. With the availability of a recombinant 70‐amino acid zinc‐containing polypeptide fragment of the regulatory chain of ATCase, it has become possible to analyze directly the interaction between catalytic and regulatory chains in a complex of simpler structure independent of other interactions such as those between the 2 C trimers, which also contribute to the stability of the holoenzyme. The coronavirus strain HECV‐4408 was isolated from diarrhea fluid of a 6‐year‐old child with acute diarrhea and propagated in human rectal tumor (HRT‐18) cells. Electron microscopy revealed coronavirus particles in the diarrhea fluid sample and the infected HRT‐18 cell cultures. This virus possessed hemagglutinating and acetylesterase activities and caused cytopathic effects in HRT‐18 cells but not in MDBK, GBK and FE cells. One of four S‐specific monoclonal antibodies reacted in Western blots with HECV‐4408, BCV‐L9 and BCV‐LY138 but not with HCV‐OC43, and two reacted with BCV‐L9 but not with HECV‐4408, BCV‐LY138 and HCV‐OC43. Three New Species And A New Record Of Neotropical Pocadlus Erichson 1843 (Coleoptera Nitidulidae), R. A.B. Leschen, C. E. Carlton

Three species of nitidulid beetles from Ecuador and Costa Rica are described as new: Pocadius maquipucunensis, P. ephite, and P. ephite and P. jelineki. These species were collected from puffball fungi of the genus Lycoperdon or in flight-intercept traps located in high-altitude cloud forests. Pocadius dimidiatus Jelínek 1977, previously known from Argentina, is reported from Tambopata Province, Peru. Specimens of P. dimidiatus were collected from fungi in the genera Lycoperdon and Geaster or in flight-intercept traps located in lowland tropical rainforest. © 1994 Taylor & Francis Group, LLC.

Results of an in vitro assay revealed that root-associated methane consumption was a common attribute of diverse emergent wetland macrophytes from a variety of habitats. Maximum potential uptake rates (V(maxp)) varied between about 1 and 10 μmol g (dry weight)-1 h-1, with no obvious correlation between rate and gross morphological characteristics of the plants. The V(maxp) corresponded to about 2 x 108 to 2 x 109 methanotrophs g (dry weight)-1, assuming that root-associated methanotrophs have cell- specific activities comparable to those of known isolates. V(maxp) varied seasonally for an aquatic grass, Calamogrostis canadensis, and for the cattail, Typha latifolia, with …

The effects of carbon deprivation on survival of methanotrophic bacteria were compared in cultures incubated in the presence and absence of oxygen in the starvation medium. Survival and recovery of the examined methanotrophs were generally highest for cultures starved under anoxic conditions as indicated by poststarvation measurements of methane oxidation, tetrazolium salt reduction, plate counts, and protein synthesis. Methylosinus trichosporium OB3b survived up to 6 weeks of carbon deprivation under anoxic conditions while maintaining a physiological state that allowed relatively rapid (hours) methane oxidation after substrate addition. The objective was to evaluate bull differences in chromosomal and spermatozoal areas related to the occurrence of the bovine leukocyte adhesion deficiency syndrome. Lymphocyte chromosomes from 30 Holstein bulls and 2 Holstein heifers were measured using image analysis and computer-enhanced video-microscopy. Spermatozoal head areas from 29 of the 30 bulls were measured. Autosomal rank was based on decreasing area. Average total autosomal areas were not the same across bulls. One group of bulls had significantly smaller average chromosomal areas than the others; this group carried bovine leukocyte adhesion deficiency syndrome. ABSTRACT: This study was to verify the usefulness of Nanovid microscopy techniques for evaluating induced modifications in bovine spermatozoal membranes. Frozen thawed bovine sperm were labeled with 20‐nm colloidal gold particles bound to concanavalin A (ConA) or heparin ligands. Sperm membrane changes were induced in vitro by capacitating and acrosome‐reacting procedures. Capacitatton was induced by incubation with 10 μg/ml of heparin for 4 hours at 37°C, 5% CO2, and high humidity. Membrane changes associated with the acrosome reaction were induced by addition of lysophosphatidyicholine (100 μg/ml) and incubation for 15 minutes at 37°C, 5% CO2, and high humidity. This study characterized the oxidative biotransformation capacity of the hepatic cytochrome P-450 system of selected avian species representing the Galliformes (chicken, turkey and Japanese quail) and Anseriformes (goose, Pekin duck, Khaki Campbell duck, and Muscovy duck) orders. Two objectives were addressed; first, important biotransformation pathways were evaluated in economically major and minor food-producing avian species to provide a framework on which to base drug approval decisions in these species. Susceptibility in soybean to red crown rot was investigated and virulence in Calonectria crotalariae was characterized. The following areas were examined: (i) variability and stability in virulence among C. crotalariae isolates, (ii) disease reaction in soybean cultivars in a greenhouse inoculation technique, (iii) disease development on hosts of different ages at time of inoculation, (iv) cultural characteristics and presence of dsRNA in isolates, and (v) the involvement of a toxin(s) in the C. crotalariae/soybean system. Laboratory studies pertaining to effects of soil redox conditions on gas exchange and plant responses were undertaken. A colorimetric method for assaying O$\sb2$ concentration in the root rhizosphere was used to quantify radial O$\sb2$ loss from plant roots. The oxidizing power of seven rice cultivars, grown under aerobic and hypoxic conditions, was measured colorimetrically with titanium-citrate solution. Oxygen release rate was calculated by extrapolation of measured absorbance to standard curves after placing plant roots in the solution for 6 h. Since Ti$\sp{3+}$ ions readily react with O$\sb2$ as it is released from roots, the technique provides a sensitive measurement of …

The appearance of extracellular coats around "eggs" are associated with activation of the oocytes. This coat, termed fertilization envelope (sea urchins), chorion (fish), zona pellucida (mammals), capsule (crab) or hatching envelope (HE) (shrimp) serves as a polyspermy block (Wasserman, 1982) or microenvironment protecting the egg from bacteria or other hostile elements (Kudo and Inoue, 1989; Kudo and Teshima, 1991; Kudo, 1992; Epel, 1993). HE assembly is described for penaeoid shrimp (Lynn, et al., 1991) and is, in contrast to many systems, a fertilization independent event. Race 1 isolates collected from tomato roots were used to develop an orange mutant of Fusarium oxysporum f. sp. lycopersici (FOL) for use in competitive infection tests against nonpathogenic F. oxysporum isolates. Eighty one color mutants were induced by exposure to ultraviolet light. The mutant ORA-22 retained pathogenicity and growth habit similar to the wild-type. Preliminary competitive infection tests indicated differences within the nonpathogenic population in the ability to suppress or enhance infection by FOL orange mutant ORA-22. Four vegetative compatibility groups (VCGs) were tested to determine if competitive infection ability was associated with VCG designation. Results from these tests …

Microbial Rock Plant Filters (MRPFs) are capable of treating municipal wastewaters, particularly for the removal of biochemical oxygen demand (BOD) in an economic manner. There is no consensus on proper design for these treatment systems due to the limited performance data available on these recently developed systems (Reed, 1993). Variability of the BOD removal reaction rate constant is an important observation which has bearing on the design of these systems. A continuous-flow microcosm system was developed to overcome recognized limitations of batch methodology and to facilitate manipulations designed to investigate the ecological role of benthic macrofauna. Experiments comparing traditional batch methodology to continuous-flow methodology indicated that rates of benthic processes were consistently greater when measured using continuous-flow methodology.
