Glutamate is well established as an excitatory neurotransmitter in the vertebrate retina. Its role as a modulator of retinal function, however, is poorly understood. We used immunocytochemistry and calcium imaging techniques to investigate whether metabotropic glutamate receptors are expressed in the chicken retina and by identified GABAergic amacrine cells in culture. Antibody labeling for both metabotropic glutamate receptors 1 and 5 in the retina was consistent with their expression by amacrine cells as well as by other retinal cell types. In double-labeling experiments, most metabotropic glutamate receptor 1-positive cell bodies in the inner nuclear layer also label with anti-GABA antibodies. …

Important parameters of estuarine variability include morphology, flushing times, nutrient loading rates, and wetland: water ratios. This variability both reflects and disguises underlying relationships between the physics and biology of estuaries, which this comparative analysis seeks to reveal, using the Gulf of Mexico (GOM) estuaries as a starting point. A question used to focus this analysis is: are the GOM estuaries unique? The GOM receives the Mississippi River, a uniquely large, world-class river, which dominates the freshwater and nutrient inflows to the GOM continental shelf, whose margins include 35 major estuarine systems. Erythroid Kruppel-like Factor (EKLF) is an erythroid-specific transcription factor that plays a critical role in γ- to β-globin gene switching during development. To identify essential domains required for EKLF transactivation function, we cotransfected a human erythroleukemia cell line (K562) with a locus control region γ/Luc-β/Cat reporter and an EKLF expression vector. In this assay EKLF mediates a 500-fold induction of β/CAT expression compared with controls. To map essential transactivation domains, progressive NH2-terminal and internal deletion mutants of EKLF were constructed. Interferon-γ (IFN-γ) is known primarily for its roles in immunological responses but also has been shown to affect fat metabolism and adipocyte gene expression. To further investigate the effects of IFN-γ on fat cells, we examined the effects of this cytokine on the expression of adipocyte transcription factors in 3T3-L1 adipocytes. Although IFN-γ regulated the expression of several adipocyte transcription factors, IFN-γ treatment resulted in a rapid reduction of both peroxisome proliferator-activated receptor (PPAR) protein and mRNA. A 48-h exposure to IFN-γ also resulted in a decrease of both CCAAT/enhancer-binding α and sterol regulatory element binding protein (SREBP-1) expression. The …

Intermycelial transfer of Streptomyces plasmid pIJ101 occurs prior to cellular differentiation and is mediated by plasmid functions that are also required for production of zones of growth-inhibited recipient cells (i.e., pocks) that develop around individual donors during mating on agar medium. Several other pIJ101 functions, including that of the kilB gene, whose unregulated expression on pIJ101 is lethal, are required for normal pock size and so have been postulated to mediate intramycelial spread of the plasmid throughout recipient cells. Field trials were conducted in cabbage (Brassica oleracea var capitata), cauliflower (B oleracea var botrytis) and knol khol (B oleracea gongylodes) crops at two different locations in Karnataka State (India) to optimize the timing of insecticide applications to control the diamondback moth, Plutella xylostella, using sex pheromone traps. Our results indicate that applications of cartap hydrochloride as insecticide during a 12-24h period after the pheromone traps had caught on average 8, 12 and 16 males per trap per night in cabbage, cauliflower and knol khol, respectively, were significantly more effective than regular insecticide sprays at 7, 9, 12 or 15 …

Bone marrow is the primary site of metastasis in patients with advanced stage prostate cancer. Prostate carcinoma cells metastasizing to bone must initially adhere to endothelial cells in the bone marrow sinusoids. In this report, we have modeled that interaction in vitro using two bone marrow endothelial cell (BMEC) lines and four prostate adenocarcinoma cell lines to investigate the adhesion mechanism. Highly metastatic PC3 and PC3M-LN4 cells were found to adhere rapidly and specifically (70-90%) to BMEC-1 and trHBMEC bone marrow endothelial cells, but not to human umbilical vein endothelial cells (15-25%). The present investigation examined the psychosocial correlates of eating disorder symptoms among a young sample of Black female adolescents. Recent epidemiological investigations have documented higher rates of disordered eating in Black females than had previously been reported. Because of the high prevalence of these symptoms both in majority and in minority populations, many eating disorder researchers are promoting preventive strategies to stem further increases in symptom levels. To this end, several longitudinal investigations have been conducted to delineate the risk factors of disordered eating among White adolescent populations. However, Black adolescents have been minimally represented in these samples. Thus, there …

The Effect Of Domestic Violence On Welfare Use, Employment And Mental Health: A Quantitative And Qualitative Analysis., Taryn Patricia Lindhorst

This dissertation is an empirical policy analysis which investigates the effect of domestic violence on a woman's welfare status, employment, and mental health within the context of new policies instituted by welfare reform. It uses quantitative data from a panel survey, and qualitative narratives from respondents who reported current domestic abuse. Multivariate analysis of welfare outcomes finds that domestic violence affects welfare outcomes indirectly through its effect on depression. Depression is associated with unemployment, continuing on TANF, and being sanctioned off welfare. Domestic violence is one of the strongest predictors of the level of reported depression. Abuse is not directly …

Streptococcus iniae was first isolated in 1972 from an Amazon River dolphin. It reappeared as a fish pathogen in Japan several years later. Since then it has steadily increased in incidence and host range worldwide, and is a known zoonotic agent. In the United States it is the most significant pathogen affecting growth of tilapia aquaculture. Histopathologic descriptions are incomplete and little is known concerning pathogenesis of the disease or virulence mechanisms of the organism. A composite of lesions was compiled from diseased tilapia evaluated by the Aquatic Animal Disease Diagnostic Laboratory, Louisiana State University from 1995--2001. In common with …

The goal of this research is to identify compounds which act as inhibitors for the synthesis of guanosine 5'-diphospho-L -fucose (GDP-fucose). The enzymes involved in this pathway are guanosine 5'-diphospho-D-mannose 4,6-dehydratase (GDP-D-mannose 4,6-dehydratase) and guanosine 5' -diphospho-4-keto-6-D-deoxymannose epimerase/guanosine 5'-diphospho-4-keto-6-L-galactose reductase (also known as FX protein and GDP-fucose synthetase). In this research project, GDP-mannose 4,6-dehydratse, the first enzyme in this pathway was studied. The plasmid DNA that contained the gene for GDP-mannose 4,6-dehydratase was transformed into E. coli bacteria; the enzyme was expressed and purified for use in this research. Introduction. Approximately 600 industrial workers reported exposure to sub-lethal levels of sulfur mustard gas and nitrogen mustard gas. A sample of 203 of these workers were administered standardized measures of perceptual, motor, intellectual, academic, attentional, memory, abstract reasoning, and psychological functioning, as well as level of effort during testing. Mean time from exposure to testing was 1.18(SD = .35) years. Males comprised 97.5% of the participants and 79.3% were Caucasian. Mean education was 11.67(SD = 1.73) years and mean age at exposure was 36.74( SD = 10.31) years. Method. Mustard gas exposure severity was defined by the presence of obvious …

This study examines the portrayal of Southern Appalachian people and their culture in American drama, discussing works from time periods that range from the 1880s to the 1990s. The plays are grouped into categories that are reflective of mainstream America's perceptions of Appalachian culture: (1) the importance of family and gender roles, including the insider/outsider romance plot, (2) issues of violence and conflict between both internal and external forces within the region in the context of wars, feuds, and environmental and labor abuses, (3) the importance of folk practice and belief including tales of the supernatural, superstitious and astrological traditions, …

A new methodology is introduced to produce nanometer-sized protein patterns. The approach includes two main steps, nanopatterning of self-assembled monolayers using atomic force microscopy (AFM)-based nanolithography and subsequent selective immobilization of proteins on the patterned monolayers. The resulting templates and protein patterns are characterized in situ using AFM. Compared with conventional protein fabrication methods, this approach is able to produce smaller patterns with higher spatial precision. In addition, fabrication and characterization are completed in near physiological conditions. The adsorption configuration and bioreactivity of the proteins within the nanopatterns are also studied in situ.

The complete genome sequences are reported here of two field isolates of bovine coronavirus (BCoV), which were isolated from respiratory and intestinal samples of the same animal experiencing fatal pneumonia during a bovine shipping fever epizootic. Both genomes contained 31028 nucleotides and included 13 open reading frames (ORFs) flanked by 5′- and 3′-untranslated regions (UTRs). ORF1a and ORF1b encode replicative polyproteins pp1a and pp1ab, respectively, that contain all of the putative functional domains documented previously for the closest relative, mouse hepatitis virus. The genomes of the BCoV isolates differed in 107 positions, scattered throughout the genome except the 5′-UTR. Differences …

A Phylogenetic Analysis Of Variation In Reproductive Mode Within An Australian Lizard (Saiphos Equalis, Scincidae), Sarah A. Smith, Christopher C. Austin, Richard Shine

Saiphos equalis, a semi-fossorial scincid lizard from south-eastern Australia, is one of only three reptile species world-wide that are known to display geographic variation in reproductive mode. Uniquely, Saiphos equalis includes populations with three reproductive modes: oviparous with long (15-day) incubation periods; oviparous with short (5-day) incubation periods; and viviparous (0-day incubation periods). No Saipho populations show 'normal' scincid oviparity (>30-day incubation period). We used mitochondrial nucleotide sequences (ND2 and cytochrome b) to reconstruct relationships among populations from throughout the species' distribution in New South Wales, Australia. We have analyzed 35 widely distributed, polymorphic Alu loci in 715 individuals from 31 world populations. The average frequency of Alu insertions (the derived state) is lowest in Africa (.42) but is higher and similar in India (.55), Europe (.56), and Asia (.57). A comparison with 30 restriction-site polymorphisms (RSPs) for which the ancestral state has been determined shows that the frequency of derived RSP alleles is also lower in Africa (.35) than it is in Asia (.45) and in Europe (.46). Neighbor-joining networks based on Alu insertions or RSPs are rooted in Africa and show African populations as separate …

Phylogenetic relationships within Celastraceae were inferred using a simultaneous analysis of 61 morphological characters and 1123 base pairs of phytochrome B exon 1 from the nuclear genome. No gaps were interred, and the gene tree topology suggests that the primers were specific to a single locus that did not duplicate among the lineages sampled. This region of phytochrome B was most useful for examining relationships among closely related genera. Fifty-one species from 38 genera of Celastraceae were sampled. The Celastraceae sensu lato (including Hippocrateaceae) were resolved as a monophyletic group. Loesener's subfamilies and tribes of Celastraceae were not supported. The …

Hybrid male sterility, hybrid inviability, sexual isolation, and a hybrid male courtship dysfunction reproductively isolate Drosophila pseudoobscura and D. persimilis. Previous studies of the genetic bases of these isolating mechanisms have yielded only limited information about how much and what areas of the genome are susceptible to interspecies introgression. We have examined the genetic basis of these barriers to gene exchange in several thousand backcross hybrid male progeny of these species using 14 codominant molecular genetic markers spanning the five chromosomes of these species, focusing particularly on the autosomes. We consider a method of approximating Weir and Cockerham's θ, an unbiased estimator of genetic population structure, using values readily available from published studies using biased estimators (Wright's FST or Nei's GST). The estimation algorithm is shown to be useful for both model populations and real-world avian populations. However, the correlation between Wright's FST and Weir and Cockerham's θ is strong when compared among 39 empirical avian datasets. Thus, the advantage of approximating an unbiased estimator is unclear considering the small actual effect of θ's bias-removing power on empirical datasets.
