This research sought to define how typical eye tracking studies are executed and improve the process with qualitative and quantitative methods. Eye tracking is a tool to collect and analyze the behavioral biometrics of consumers. Temporomandibular joint (TMJ) disorders affect over 10 million people in the US with an annual economic cost of more than $4 billion. Due to intriguing etiological factors of TMJ disorders, objective and accurate diagnosis is difficult and so is with the treatment. Consequently, there is the high recurrence rate. To make the situation even worse, many other types of diseases, including cancer, share symptoms and signs with TMJ disorders. Early non-invasive, objective, subject-specific and accurate diagnosis is vital because incorrect or delayed diagnosis can cause patients to miss optimal treatment times, increase unnecessary therapy expenses and even endanger patients’ lives. …

Late gestation and post-weaning are two critical periods a calf needs to receive proper nutrition to insure adequate growth for the rest of the animal’s lifespan. Supplementation at these critical time points may be necessary in order to meet nutritional requirements. The number of Clostridium difficile infection (CDI) cases associated with healthcare and community is increasing. Increased isolation of this pathogen from environmental samples of soil, water, livestock animals and their feces, and produce suggests its dissemination in the environment. Identification, molecular characterization, and revealing the relationship of those isolates with human pathogenicity would be very helpful in understanding the epidemiology of CDI. Livestock manure is a rich source of nutrients for the growth of many crops, and composted manure is widely used as a fertilizer in organic farming. This exploratory study examined an existing data set containing outcome scores of adolescents and adults on the autism spectrum who were enrolled in 18 different outdoor behavioral health programs in the United States, all of whom are members or cooperating programs of the National Association of Therapeutic Schools and Programs (NATSAP) Practice Research Network. The dataset contained demographic characteristics and self-reported psychosocial functioning of the participants at intake and again at discharge. Data suggest that the self-reported outcome scores for both adolescents and adults demonstrated statistically significant change after the intervention, but when examined for clinical significance, adolescent paired scores …

Förster Resonance Energy Transfer (FRET) is a promising methodology which is used by researchers in determining structural information of biomolecules. Other methodologies such as X-ray diffraction, nuclear magnetic resonance (NMR) and electron microscopy (EM), and X-ray scattering (SAXS) and circular dichroism (CD) can only give snapshots of the functional conformers. In the applications of most cases, FRET can only be used to give a qualitative result such as “yes” or “no” pattern. In our lab, we are able to expand this ability by applying FRET to quantitatively determine the structure and dynamic information in the biological system with high accuracy. …

Subventricular zone (SVZ) neural stem cells (NSCs) are the cornerstone of the perinatal neurogenic niche. Microglia are immune cells of the nervous system that are enriched in the neonatal SVZ. Although microglia regulate NSCs, the extent to which this interaction is bi-directional is unclear. Extracellular vesicles (EVs) are cell-derived particles that encase miRNA and proteins. Here, we demonstrate that SVZ NSCs generate and release EVs. Neonatal electroporated fluorescent EV fusion proteins were released by NSCs and subsequently cleared from the SVZ. EVs were preferentially targeted to microglia. Small RNA sequencing identified miRNAs within the EVs that regulate microglia physiology and …

The Privilege Of Healthy Eating: A Qualitative Study Exploring The Local Food Choices Of Low-Income Families From Appalachia, Iryna Sharaievska, Stephanie West, Melissa Weddell

Using qualitative semi-structured interviews, 15 low-income women of different ages were asked to discuss their perceptions of healthy eating, local farmers’ markets, as well as their visitation of farmers’ markets. The participants were also asked to share what features of farmers’ markets they found appealing. The results showed that most of the participants had either a deep or moderate understanding of what it means to eat healthy. However, many of them also believed they could improve their own patterns of eating. The data also showed that the low-income women who took part in the study had positive attitudes overall toward …

Soil inorganic carbon (SIC) is currently not included in the list of key soil properties related to ecosystem services (e.g., provisioning, regulating, cultural, and supporting services). Placental efficiency (PE) describes the relationship between placental and fetal weights and is defined as fetal weight divided by placental weight. Within pig litters, PE can vary drastically, resulting in similarly sized pigs associated with very different placenta, up to a 50% weight difference. However, the means enabling the smaller placenta to grow a similarly sized littermate is unknown. This study utilized an online survey administered to Kentucky and South Carolina in conjunction with discrete choice modeling to examine: i) the impact of several factors including demographic characteristics, purchasing behaviors, and frequency of seafood consumption on consumer's preferences for specific seafood attributes (e.g. fresh over frozen, wild-caught over farm-raised), ii) the impact of the aforementioned consumer characteristics, on the probability that an individual will be interested in joining a CSF arrangement in the future. The results indicate that demographic characteristics, and lifestyle preferences have a statistically significant effect on consumer's preferences for both tangible and intangible seafood characteristics. The objective of this study was to evaluate the growth and reproductive development of young growing beef bulls on a complete forage ration (F) to development of similar young growing beef bulls on a forage ration supplemented with concentrate (CT). Data was collected from the 665 beef bulls, grouped into breed codes (BC) of continental (CN; n = 103) or British (B; n = 562) that participated in the Edisto Forage Bull Test (EFBT) in Blackville, SC from 2006 to 2016 with the exception of 2012. Mass flow sensor algorithms were developed for a hay yield monitoring system using ultrasonic sensors to measure windrow height. The algorithms indicated accuracies of 17.34% to 9.51%, with a wide tolerance to mounting positions. The developed mass flow algorithms were used in a nitrogen rate strip test on a 25 ac irrigated Tifton 85 bermudagrass field. Results suggested that yield, crude protein content and profit was maximized with the highest nitrogen rate only during the first harvest of 2017. All other harvest suggested a lower rate of nitrogen should be applied in order to maximize profit. A second method of …

Acute gastroenteritis (AGE), a major cause of illness, results in 179 million AGE cases every year in the United States. AGE bacteria that have low-infectious dose include E. coli O157:H7 (<10-100 cells) and L. monocytogenes (<1000 cells). Because of their low-infectious dose and high environmental resistance, contaminated surfaces, such as the exterior surface of food packages, could be a source for disease transmission. Our aim was to determine the persistence of E. coli O157:H7 and L. monocytogenes on three packaging materials - oriented polyethylene terephthalate (OPET), oriented polypropylene (OPP), and nylon-6. Coupons (25 cm2) from each material were sterilized under ultraviolet light for 5 minutes. Spot and spread inoculation was done on treatment coupons with ca. 7 log CFU of a 3-strain-mixture of green fluorescent protein (GFP)-labeled E. coli O157:H7. Chapter 1 describes new hydrothermal synthesis of carbon nanofibers from cellulose nanocrystals. Diet-induced milk fat depression is a multifactorial disorder characterized by sustained reduction in milk fat. Feeding high amounts of starch and unsaturated fatty acids have been shown to reduce milk fat yield and concentration, and alter ruminal biohydrogenation. In addition, starch with high rates of degradability may lead to a decrease in ruminal pH and an increase in the outflow of ruminal biohydrogenation intermediates. It is possible that starch sources of high degradability could still be utilized in diets for the recovery of milk fat depression, provided the unsaturated fat level is reduced. The value of a carcass is largely dependent on lipid deposition. The subcutaneous and visceral depots are often reported to develop and fill first. These depots also have little to no value, and often result in carcass price discounts. The intramuscular depot is well correlated with consumer palatability and carcass value. The ability to increase deposition of intramuscular lipid while minimizing the filling of low value fat depots provides ample opportunity to improve carcass value. The perceptions that parents, teachers, and early childhood education leaders have about school readiness, and how these individuals promote readiness in children, influences how prepared children are to enter the school setting. In this study, I drew upon an ecological systems model to examine how parents, teachers, and early childhood school leaders perceive and promote school readiness. In this study, I also examined the strategies that early childhood education leaders' used to build relationships with families and community members, and to foster parent involvement.
