Glucose Signaling Is Important For Nutrient Adaptation During Differentiation Of Pleomorphic African Trypanosomes, Yijian Qiu, Jillian E. Milanes, Jessica A. Jones, Rooksana E. Noorai, Vijay Shankar, James C. Morris Oct 2018

Glucose Signaling Is Important For Nutrient Adaptation During Differentiation Of Pleomorphic African Trypanosomes, Yijian Qiu, Jillian E. Milanes, Jessica A. Jones, Rooksana E. Noorai, Vijay Shankar, James C. Morris

Publications

The African trypanosome has evolved mechanisms to adapt to changes in nutrient availability that occur during its life cycle. During transition from mammalian blood to insect vector gut, parasites experience a rapid reduction in environmental glucose. Here we describe how pleomorphic parasites respond to glucose depletion with a focus on parasite changes in energy metabolism and growth. Long slender bloodstream form parasites were rapidly killed as glucose concentrations fell, while short stumpy bloodstream form parasites persisted to differentiate into the insect-stage procyclic form parasite. The rate of differentiation was lower than that triggered by other cues but reached physiological rates …

Bacteria And Competing Herbivores Weaken Top–Down And Bottom–Up Aphid Suppression, Carmen K. Blubaugh, Lynne Carpenter-Boggs, John P. Reganold, Robert N. Schaeffer, William E. Snyder Sep 2018

Bacteria And Competing Herbivores Weaken Top–Down And Bottom–Up Aphid Suppression, Carmen K. Blubaugh, Lynne Carpenter-Boggs, John P. Reganold, Robert N. Schaeffer, William E. Snyder

Publications

Herbivore suppression is mediated by both plant defenses and predators. In turn, plant defenses are impacted by soil fertility and interactions with soil bacteria. Measuring the relative importance of nutritional and microbial drivers of herbivore resistance has proven problematic, in part because it is difficult to manipulate soil-bacterial community composition. Here, we exploit variation in soil fertility and microbial biodiversity across 20 farms to untangle suppression of aphids (Brevicoryne brassicae) through bottom–up and top–down channels. We planted Brassica oleracea plants in soil from each farm, manipulated single and dual infestations of aphids alone or with caterpillars (Pieris …

Examining Predictors Of Implementation Quality In An Emerging International Extension Context, Ryan J. Gagnon, Jonathan Pettigrew Aug 2018

Examining Predictors Of Implementation Quality In An Emerging International Extension Context, Ryan J. Gagnon, Jonathan Pettigrew

Publications

In this article, we explore a component of evidence-based programming—implementation quality—within an emerging international Extension context. Specifically, we examine how the traits, characteristics, and perceptions of 46 program facilitators influenced their support of maintaining implementation quality in a Nicaraguan youth violence and substance abuse prevention program, Dale se REAL. The results indicated that of four potential variables, only facilitator buy-in to the Dale se REAL program was a meaningful predictor of implementation support. The implications of the study findings, relative to evidence-based Extension research in both the United States and an emerging international context, are discussed.

Effects Of Feeding Different Amounts Of Milk Replacer On Growth Performance And Nutrient Digestibility In Holstein Calves To 4 Months Of Age Using Different Weaning Strategies, Rebecca Naomi Klopp

All Theses

A 2x2 factorial design (moderate (MOD) or high (HI) milk replacer (MR) feeding rates and abrupt (AB) or gradual (GR) weaning) was utilized to compare how calf performance and nutrient digestibility is affected pre and post-weaning. The 112 d study consisted of a 56 d nursery and a 56 d grower phase. Calves (n=50) were randomly assigned to one of four pre-weaning treatments. Data were analyzed as a completely randomized design with repeated measures when applicable by PROC MIXED in SAS. Calves assigned to MOD-AB were fed 0.66 kg (DM) MR for first 42 d then 0.33 kg for 7 …

An Evaluation Of Bite Counting As A Form Of Visible Feedback, Jacqueline Elizabeth Mcsorley Aug 2018

An Evaluation Of Bite Counting As A Form Of Visible Feedback, Jacqueline Elizabeth Mcsorley

All Theses

This study aimed to determine if viewing bite count in real time had the same effect as viewing visible food records to decrease intake during a meal. Behavioral intervention is the most common treatment used to lower food consumption. This includes the idea of self-monitoring food intake through a variety of methods like bite counting. Another factor that leads to the reduction of food consumption is the presence of feedback. This feedback can take the form of visible food records such as chicken bones, candy wrappers, or bottle caps. In this study, chicken wings were used as the visible food …

Exploring Interspecific Interactions Of Reniform Nematode (Rotylenchulus Reniformis) And Host Roots, Nathan Wayne Redding Aug 2018

Exploring Interspecific Interactions Of Reniform Nematode (Rotylenchulus Reniformis) And Host Roots, Nathan Wayne Redding

All Dissertations

The semi-endoparastic reniform nematode (Rotylenchulus reniformis) infects over 300 plant species. Females penetrate host roots and induce formation of complex, multinucleate feeding sites called syncytia. While anatomical changes associated with syncytia morphology are well documented, little is known about their molecular basis or the local impact on root development. Using a soybean (Glycine max) split-root system across a twelve-day time course, we identified over 6,000 genes that were differentially expressed between inoculated and control roots (FDR = 0.01, |log2FC| ≥ 1) and 507 gene sets that were significantly enriched or depleted in inoculated roots (FDR = …

Examination Of Carbon Assimilation And Nutrient Movement In Peach Trees (Prunus Persica (L.) Batsch) As A Result Of Variable Fall Climate, Brian Tyler Lawrence Aug 2018

Examination Of Carbon Assimilation And Nutrient Movement In Peach Trees (Prunus Persica (L.) Batsch) As A Result Of Variable Fall Climate, Brian Tyler Lawrence

All Theses

Deciduous fruit tree production worldwide may need to alter management strategies due to an increase of variable climate conditions. These include warmer than average temperatures during the year along with too much or little water during the growing season. Predicting best practices for orchard management will require a thorough understanding of how temperatures and soil moisture alter carbon acquisition and nutrient movement in trees, especially during the end of the growing season when tree reserves are accumulated for the following spring. Over two consecutive years, the effect of warmer fall temperatures, which delayed natural leaf senescence, and variable soil moisture …

The Detrimental Effects Of Cigarette Smoke On Fracture Healing In A Bilateral Femur Fracture Rodent Model, Russell Abbott Reeves Aug 2018

The Detrimental Effects Of Cigarette Smoke On Fracture Healing In A Bilateral Femur Fracture Rodent Model, Russell Abbott Reeves

All Theses

Fractures that fail to heal (nonunions) substantially contribute to patient morbidity and overall healthcare costs. When nonunions occur in the absence of a critical size defect it is the result of impaired healing via one of the two distinct bone healing pathways: primary (intramembranous) and secondary (endochondral) ossification. Furthermore, the recent characterization of circulating osteoprogenitor cells (COPs) presents yet another important mechanism by which fractures may heal. Clinically, patients who smoke cigarettes are at increased risk of suffering from a nonunion; however, the mechanism by which this occurs has still not yet been defined. We sought to determine whether cigarette …

Nickel Tolerance In Cool-Season Grasses, Caleb Randall Patrick

All Theses

Nickel (Ni) toxicity is becoming more prevalent in plants and soils due to increased applications of sewage sludge and waste water. New literature highlighting the Ni-tolerance of cool-season turfgrass species is needed. Herbicide resistant annual bluegrass (Poa annua L.) biotypes are also becoming more prevalent, stemming from application of single mode of action herbicides. Research was conducted to examine Ni tolerance of three cool-season turfgrasses and explore the possibility of Ni use as a suppressant of annual bluegrass. Seaside II creeping bentgrass (Agrostis stolonifera L.), PennLinks II creeping bentgrass (Agrostis stolonifera L.), and a weedy …

Language And Literacy In A Head Start Preschool: Understanding Practice To Inform Theory And Improve Outcomes, M. Deanna Ramey Aug 2018

Language And Literacy In A Head Start Preschool: Understanding Practice To Inform Theory And Improve Outcomes, M. Deanna Ramey

All Dissertations

Head Start is a federally funded program providing comprehensive social and early education services to children from low-income households and their families. Seeking to boost outcomes of Head Start students, in 1998 Congress amended Head Start's statement of purpose, specifying school readiness as the program's top priority. This mission change necessitated that Head Start, long-focused on promoting social competence, shift attention to a new concern—supporting children's development in cognitive domains such as math and literacy. Data suggest that though some progress has been made, Head Start preschoolers still lag behind their peer group in assessments of early math, language, and …

A Decision Support System For Selecting Between Designs For Dynamic Software Product Lines, Ethan Travis Mcgee Aug 2018

A Decision Support System For Selecting Between Designs For Dynamic Software Product Lines, Ethan Travis Mcgee

All Dissertations

When commissioning a system, a myriad of potential designs can successfully fulfill the system's goals. Deciding among the candidate designs requires an understanding of how the design affects the system's quality attributes and how much effort is needed to realize the design. The difficulty of the process compounds if the system to be designed includes dynamic run-time self- adaptivity, the ability for the system to self-modify its architecture at run-time in response to either external or internal stimuli, as the type and location of the dynamic self-adaptivity within the architecture must be co-decided. In this proposal, we introduce a Decision …

The Awareness And Attitudes Of Teachers Towards Educational Restrictions For Immigrant Students, William David Mccorkle Aug 2018

Investigation Of Molecular Mechanisms Involved In Retardation Of Immunosenescence In Caenorhabditis Elegans By Royal Jelly, Ojas Natarajan

All Dissertations

With recent advances in medicine, life expectancy has been steadily increasing. This increasing geriatric populous encounter various age-associated diseases including decline in immunity called immunosenescence, which renders them susceptible to an extended array of pathogens like Staphylococcus aureus. These nosocomial pathogens gain resistance to a variety of antibiotics, hence a strong innate immune system is important for host survival. Caenorhabditis elegans has no complex immune system. Hence, it is an excellent model to study the innate immune response.

Royal jelly, a nutraceutical has been previously shown to prolong the lifespan and UV, oxidative and heat shock resistance in C. …

Development Of A Database-Driven Management System For Retail Food Packaging Eye Tracking Studies, Mengmeng Zhao Aug 2018

Development Of A Database-Driven Management System For Retail Food Packaging Eye Tracking Studies, Mengmeng Zhao

All Dissertations

Within the past three years, approximately two hundred one-off studies have occurred in developing eye tracking methodologies for quantifying consumer attention on packaging design preference. Due to restricted time and resources, most research analyzes consumer attention regarding a relatively small amount of products within specific planogram. However, larger questions concerning category trends and insights are not possible with these smaller, one-off studies. Broad data aggregation and analysis is important for understanding packaging design per product category in order to understand category-wide design trends and insights. Considering the excessive effort in manual data retrieval, analysis, and reporting, a method that can …

The purpose of the following paper is to utilize multinomial regression to study the effect of various demographic and family characteristic variables on both the parental expectations of future postsecondary degree attainment and the parental estimates of college costs. I also explore the effect of college cost estimates as a mediating factor on the parents' expectation of the students' degree attainment. There is significant existing research on college costs, affordability, financial aid, and educational attainment for the various demographic factors found in this study. The missing piece in both the literature and overall understanding of the problem lies in the …

Rapid Screening Of Ellagitannins In Natural Sources Via Targeted Reporter Ion Triggered Tandem Mass Spectrometry, Jeremiah J. Bowers, Harsha P. Gunawardena, Anaëlle Cornu, Ashwini S. Narvekar, Antoine Richieu, Denis Deffieux, Stéphane Quideau, Nishanth Tharayil Jul 2018

Rapid Screening Of Ellagitannins In Natural Sources Via Targeted Reporter Ion Triggered Tandem Mass Spectrometry, Jeremiah J. Bowers, Harsha P. Gunawardena, Anaëlle Cornu, Ashwini S. Narvekar, Antoine Richieu, Denis Deffieux, Stéphane Quideau, Nishanth Tharayil

Publications

Complex biomolecules present in their natural sources have been difficult to analyze using traditional analytical approaches. Ultrahigh-performance liquid chromatography (UHPLC-MS/MS) methods have the potential to enhance the discovery of a less well characterized and challenging class of biomolecules in plants, the ellagitannins. We present an approach that allows for the screening of ellagitannins by employing higher energy collision dissociation (HCD) to generate reporter ions for classification and collision-induced dissociation (CID) to generate unique fragmentation spectra for isomeric variants of previously unreported species. Ellagitannin anions efficiently form three characteristic reporter ions after HCD fragmentation that allows for the classification of unknown …

From The Cradle To The Grave: How The Embryonic Environment Shapes Responses To Climate Throughout The Life Cycle, Michael A. Carlo May 2018

From The Cradle To The Grave: How The Embryonic Environment Shapes Responses To Climate Throughout The Life Cycle, Michael A. Carlo

All Theses

With rapidly warming climates, biologists are challenged to predict organismal responses to environmental variation, changes in biodiversity, and shifts in species distributions. In many cases, organismal responses differ across life stages that exhibit differences in physiological tolerances, habitat requirements, and resource allocations strategies. For instance, mobile life stages can alter behavior in response to changes in the thermal environment, whereas immobile life stages are often vulnerable to warming temperatures due to behavioral constraints and the limited microclimatic conditions experienced over small spatial extents (e.g., an embryo in an egg). Unfavorable developmental conditions can reduce growth, alter developmental rates, and increase …

The Effect Of Hot Fill And Hold Processing On The Performance Of Multilayer Packaging Films, Meredith Leigh Johnson May 2018

The Effect Of Hot Fill And Hold Processing On The Performance Of Multilayer Packaging Films, Meredith Leigh Johnson

All Theses

Heat transfer in thermal processing is crucial to ensure all parts of a product are sufficiently treated to achieve commercial sterility without unacceptable loss of quality. Optimizing pasteurization methods is recommended to preserve quality attributes such as color, texture, and flavor while maintaining food safety integrity. This research evaluated the temperature variability in pouches during a hot fill and hold process and the effect of those identified differences on color quality of a tomato based food simulant. The performance of multilayer films for pasteurized products in accelerated storage conditions were also studied. The research project was separated into two phases. …

The Relationship Between Mentally Unhealthy Days And Access To Healthcare And Healthcare Services Utilization Among The Long-Term Uninsured In South Carolina, Tamara Semaj Dobson-Brown

All Theses

Background: The Patient Protection and Affordable Care Act (ACA) made a tremendous impact on the nation’s nonelderly (ages 18-64) uninsured population. Despite the ACA provisions, they fell short of universal health insurance coverage; especially in states that chose not to expand Medicaid, like South Carolina. Those left uninsured are likely to experience great difficulty with the health care delivery system, especially in areas that have a high concentration of uninsured residents. Purpose: To assess the effect of psychological vulnerability, as measured by mentally unhealthy days, on access to healthcare and healthcare utilization among the long term uninsured in South Carolina. …
