Youth development professionals working in non-formal educational settings face the persistent challenge of translating evidence-based frameworks into effective practice. This explanatory sequential mixed-methods study examines how South Carolina 4-H agent/educators perceive and adopt the 4-H Thriving Model following mandatory statewide training. The study investigates the roles of professional identity, change readiness, and organizational factors in shaping innovation adoption decisions among Extension professionals who work with considerable autonomy within diverse community contexts.

Large Language Models (LLMs) have advanced conversational agents, enabling natural, human-like interactions in domains such as education, programming, and workplace collaboration. Yet, user distrust persists over privacy, accuracy, and bias. As developers work to mitigate these issues and human-AI collaboration expands, reinforcing trust in LLM-driven systems is essential. To address this problem, this dissertation explores the role of anthropomorphic form in LLM-driven conversational agents and its impact on user perception.

The Influence Of Tert-Butylhydroquinone (Tbhq) On Acod1/Irg1-Driven Inflammatory Pathways, Alyssa M. Whisel

The demand for processed foods has led to increased use of synthetic phenolic antioxidants (SPAs), notably tert-butylhydroquinone (TBHQ), to inhibit lipid oxidation and prolong shelf life. While TBHQ is deemed safe at regulated levels, concerns arise from studies indicating potential adverse effects such as oxidative stress, genotoxicity, and impaired immune function. To investigate TBHQ's immunomodulatory effects, RAW 264.7 murine macrophages were pre-treated with TBHQ and subsequently stimulated with lipopolysaccharide (LPS) or polyinosinic-polycytidylic acid (poly I:C, PIC) to simulate bacterial or viral challenges. Results demonstrated that TBHQ at high non-cytotoxic doses reduced nitric oxide and IL-10 production while promoting phagocytosis and …

Ecological interactions such as predation are vital to shaping the distribution of organisms. For interactions to occur, organisms must undertake movement, such as dispersal, that places them in proximity to the organisms with which they may interact. Geographically isolated wetlands (GIW) are productive and biodiverse ecosystems, but little is known on broad geographic scales about the determinants of their fauna. Fishes have difficulty colonizing GIW, whereas amphibians and macroinvertebrates are adept overland dispersers – though they may be sensitive to top-down control by fishes. The characterization of systems encompasses a variety of modeling frameworks designed to capture specific behaviors and components of various system domains. Whatever the framework, the core elements of a system representation are the information of the system and a description of how that information is related. The relations in deterministic systems are functions, which, when composed to form executable processes, can be used to simulate system data. Trypanosoma brucei is a protozoan parasite which causes human African trypanosomiasis and the livestock wasting disease, nagana. These parasites have highly specialized peroxisomes, called glycosomes, which house a variety of essential metabolic pathways, including glycolysis. Glycolysis is the sole means of energy production in the infective life stage of T. brucei making glycosomes and the glycolytic pathway promising targets for therapeutic strategies. In this work, we have studied the glycosome as a therapeutic target from multiple perspectives. The first portion of this work addresses drug trafficking. Understanding musculoskeletal joints from a 3D multiscale perspective, from molecular to anatomical levels, is essential for resolving the confounding relationships between structure and pain, elucidating mechanisms regulating joint health and diseases, and developing new treatment strategies. Here, a musculoskeletal joint immunostaining and clearing (MUSIC) method specifically developed to overcome key challenges of immunostaining and optical clearing of intact joints are introduced. Coupled with large-field light sheet microscopy, this approach achieves 3D high-resolution, microscale neurovascular mapping within the context of whole-joint anatomy without the need for image coregistration across various joints, including temporomandibular joints, knees, and spines, and multiple species, including …

The hydrological urban heat island (HUHI) is a recent facet of the urban heat island (UHI), describing thermal enhancement of bodies of water in response to urbanization. Although the forefront of this work has been studied for large Metropolitan areas, this effect on developing cities is currently unknown. To locally measure and quantify HUHI effects in a developing city, we utilized Multi-Radar Multi Sensor (MRMS) radar-derived rainfall data to estimate rainfall in the Augusta Metropolitan Area (AMA) and the Savannah Metropolitan Area (SMA), two developing communities adjacent to the Savannah River. This discussion activity examines the US Electoral College and its role in animating the anti-Black racism found in the voting systems and practices in the United States of America. Specifically, students are assigned two readings that explore how enslavement and anti-Black racism inform the foundation of the Electoral College. These texts guide students’ subsequent engagement with two videos (e.g., one from NPR and a second from the Harvard Kennedy School) that facilitate an in-class discussion.

Laying hens are often housed in environments that restrict movement, which may contribute to bone fragility and decreased welfare. These confined environments, such as conventional cage systems, limit exercise and result in poor skeletal development. This study investigates the impact of exercise opportunities on bone health in laying hens by comparing the tibiotarsi and humeri bone characteristics in enriched and unenriched housing environments. A total of 200 hens were housed in pens (20 birds/pen) from 0 to 40 weeks of age. Five pens, which served as the experimental group, were equipped with multi-tier perches and ledges, providing exercise opportunities. In …

This study starts by exploring the impact and implementation process of the Express Lane program. This program is a modified schedule initiative developed at Excellent High School in South Carolina to tackle declining graduation rates. The research investigates whether structured, real-time communication protocols and self-pacing learning can help students improve credit completion in order to help students who have the opportunity of graduating high school on time. This investigation is in response to a persistent drop of graduation rates from 90% in 2020 to an alarming 84.3% in 2023-2024. Road networks are a major driver of habitat fragmentation and wildlife mortality. Wildlife crossing structures (WCS) are a common mitigation strategy with the goal of maintaining landscape connectivity, reducing wildlife-vehicle collisions, and facilitating safe passage across highways. Most research on WCS prioritize large ungulates or carnivores, leaving potential gaps in our understanding of how diverse mammalian communities interact with these features. For many years, the United States Army Corps of Engineers (USACE) used the Guidelines for Preparing a Compensatory Mitigation Plan worksheets to guide pre-restoration evaluation and determine post-restoration mitigation credits in South Carolina. Recently, the USACE in South Carolina transitioned to the Stream Quantification Tool (SQT), which provides a more comprehensive assessment framework. Our first objective was to compare these assessment methodologies across different landscape types to determine what the SQT adds to mitigation calculations that were not considered in prior worksheets. The study presents a mixed method and longitudinal assessment of land cover change, estuarine land loss, and policy implemented to address the past and anticipated estuarine losses in the Cooper River and North and South Santee watershed over the past twenty years. This study uses National Land Cover Dataset from 2001 to 2021, NOAA’s SLR projection data and planning documents from 1990 to 2025 to examine policy evolution among ten major themes.
