The Circulome: A Dynamic Reservoir Of Genetic Heterogeneity In Agricultural Pests, Hailey Spier Camposano May 2022

The Circulome: A Dynamic Reservoir Of Genetic Heterogeneity In Agricultural Pests, Hailey Spier Camposano

All Theses

Weed and insect pests cost the agricultural sector billions in losses each year across the world, despite distributing over 1 billion pounds of potentially harmful chemicals into agroecosystems to manage these pests. Two major agricultural pests that are rapidly adapting to multiple xenobiotics are Amaranthus palmeri and Musca domestica. A. palmeri, or Palmer amaranth, is highly productive in terms of vegetative structures and reproductive propagules. Palmer amaranth has evolved multiple resistance mechanisms to several herbicidal modes of action. House fly (M. domestica) presents issues as a vector for pathogenic microbes among livestock, poultry, and humans. This …

Habitat Selection And Breeding Ecology Of Bachman’S Sparrow (Peucaea Aestivalis) In A Wiregrass-Free Ecosystem, Mikayla Thistle

All Theses

Through much of its range, Bachman’s Sparrow (Peucaea aestivalis) uses the wiregrass (Aristida spp.) dominant understory typical of longleaf pine (Pinus palustris) forest. The central South Carolina Coastal Plain, however, lies within the “wiregrass gap” where longleaf pine understories are absent of wiregrass and instead are dominated by bluestem grasses (Schizachyrium spp. and Andropogon spp.), bracken fern (Pteridium aquilinum), and shrubs. Habitat use of Bachman’s Sparrow in this region has yet to be studied and declining Bachman’s Sparrow populations necessitate a better understanding of habitat selection processes and population dynamics …

"A Beauty In The Kitchen": The Introduction Of The Cookstove As A Mechanism Of Change In Charleston's Historic Kitchens, Maria Short

All Theses

While Charleston’s historic houses have long captivated visitors, scholars, and preservationists, the architecture of these properties’ kitchens and the ways people cooked in these historic spaces have long been overlooked, in part because their historic fabric has often been obscured by later alterations or demolition. While interpretation of these historic spaces in certain house museums, such as the Nathaniel Russell House or Heyward-Washington House, now include information on the lives of the enslaved who cooked in these kitchens, the understanding of cooking technology, specifically the transition from hearth cooking to cooking on cookstoves, in Charleston remains largely unstudied. In the …

Evaluating The Accuracy And Quality Of Microgreens Training Materials Available On The Internet: A Content Analysis, Supun Chathuranga Nabadawa Hewage

All Theses

Microgreens are the young, edible seedlings of various vegetables, spices, herbs, and considered as the intermediate stage of sprouts and mature greens, suggesting microgreens may share similar food safety risks with both of these produce. Even though there are no known outbreaks due to contaminated microgreens, multiple product recalls have been reported, indicating food safety risks associated with microgreens should not be underemphasized. A recent national survey of the U.S. microgreens industry reported that almost half of growers (48.3% of 176) learned to grow microgreens by viewing websites and videos on the internet.1 However, it is unknown whether the …

Characterization Of A Potential Glucose Transporter In Trypanosoma Brucei, Matthew Morgan May 2022

Characterization Of A Potential Glucose Transporter In Trypanosoma Brucei, Matthew Morgan

All Theses

Trypanosoma brucei, the African trypanosome, is an organism heavily dependent on glucose for ATP production during the infectious stage of its life cycle. Here, we have explored the role of an uncharacterized protein designated “novel glucose transporter” (NGT) as a potential glucose transporter. Sequence analyses suggests that NGT shares similarities (either at the primary sequence level or structurally) with Trypanosome Hexose Transporters 1 (TbTHT1), and human GLUT3, both of which are membrane sugar transporters. NGT was localized by fluorescence microscopy to subcellular structures consistent with lysosomes. Silencing NGT expression with RNA interference in parasites resulted in a growth defect …

Identification Of Genetic Variants Underlying Maxillary Canine-Tooth Mesioversion And Small Body Size In Shetland Sheepdogs, Sydney R. Abrams

All Theses

The domesticated dog (Canis lupus familiaris) exhibits a unique population structure, with high levels of genetic homogeneity within breeds due to selective breeding. These closed breeding populations can result in breed-specific inherited disorders. Maxillary canine-tooth mesioversion (MCM) is a genetically complex dental anomaly observed nearly exclusively in the small Shetland Sheepdog breed. Here, we utilized a genome-wide approach to discover a locus of major effect on chromosome 9. Using whole-genome resequencing data from a bilaterally affected dog, we identified variants in two genes: FTSJ3, encoding an RNA methyltransferase, and GH1, encoding growth hormone. Independent genome-wide association …

Portable U-Bent Optical Fiber Biosensor For Detecting Microcystin-Lr, Quan Dio Anh Le

All Theses

Cyanobacteria are the only known prokaryotes that can photosynthesize. Due to their unique ability, they are important entities in the global carbon and nitrogen cycles. However, two factors can turn cyanobacteria into a nuisance. In general, eutrophication is the process that supplies external nutrients that are needed for cyanobacteria blooms into bodies of water from the surrounding environment. Climate warming intensifies this process by producing carbon dioxide, methane, and nitrous oxide. With cyanobacteria blooms occurring worldwide, they are becoming a bigger threat to human health, and there has been increased interest in detecting cyanobacteria and their toxic byproducts.

For instance, …

Co-Habitation And Migration: Urban Design For Bird Migratory Pathways, Steve C. Kurtz

All Theses

Urban landscapes are intricate systems interacting throughout the globe. Over the past decades, urban development has widely sprawled and fragmented flyway systems and corridors. This has led to many changes in species composition and has even caused the extinction of some animals (Marzluff 1997). Recent studies have indicated that there exists a protective or restorative design to create stepping-stone connectivity in urban areas. This study looks to understanding potential benefits in urban biodiversity, animal species protection, and urban benefits of green roof designs.

Connecting Process Performance And Microbial Community Dynamics During Anaerobic Wastewater Treatment To Improve Microbial Resource Management, Jessica Anne Deaver May 2022

Connecting Process Performance And Microbial Community Dynamics During Anaerobic Wastewater Treatment To Improve Microbial Resource Management, Jessica Anne Deaver

All Dissertations

Anaerobic wastewater treatment technologies produce renewable energy and recover resources through microbial conversion of organics. Anaerobic digestion is commonly utilized for municipal wastewater sludge treatment, and co-digestion with other organic-rich substrates like fats, oils, and grease (FOG) can improve methane production. Methane can be captured for combined heat and power production. Microbial electrochemical cells (MXCs) convert organic waste to electrical current to produce electricity or other valuable chemicals. Both these technologies would benefit from understanding the microbial conversion processes more thoroughly. FOG co-digestion sometimes inhibits rather than bolsters methane production. MXCs have potential for blackwater (urine and feces) treatment in …

Incorporating Human Effects In Quantifying Mechanisms Of Stream Fish Community Structure Using Metacommunity Theory, Lauren E. Stoczynski

All Dissertations

Metacommunity theory incorporates local and regional factors to understand how biotic communities are structured across the landscape. Despite established knowledge of how humans impact aquatic systems, inclusion of anthropogenic factors in metacommunity studies have been largely ignored. Additionally, alpha, beta, and gamma diversity can all be explored at the metacommunity level to investigate mechanistic drivers of community structure. Beta diversity can be further partitioned into turnover and richness difference components, each with different mechanistic drivers. Streams provide an excellent study system for metacommunity research because of the dendritic structure of watersheds and the natural delineation that watershed boundaries provide. Large-extent …
