Understanding the production and differentiation of megakaryocytes from progenitors is crucial for realizing the biology and functions of these vital cells. Previous gene ablation studies demonstrated the essential role of the transcriptional repressor Gfi1b (growth factor independence 1b) in the generation of both erythroid and megakaryocytic cells. However, our recent work has demonstrated the down-regulation of this factor during megakaryocytic differentiation. In this study we identify two new gene targets of Gfi1b, the cytoskeletal proteins Kindlin3 and Talin1, and demonstrate the inverse expression and functions of these cytoskeletal targets relative to Gfi1b, during megakaryocytic differentiation. Stress can significantly impact brain function and increase the risk for developing various psychiatric disorders. Many of the brain regions that are implicated in psychiatric disorders and are vulnerable to the effects of stress are also involved in mediating emotional learning. Emotional learning has been a subject of intense investigation for the past 30 years, with the vast majority of studies focusing on the amygdala and its role in associative fear learning. Photosynthetic coral reef structures extend from the shallow sundrenched waters to the dimly lit, “twilight” mesophotic depths. For their resident endosymbiotic dinoflagellates, primarily from the genus Symbiodinium spp., this represents a photic environment that varies ~15-fold in intensity and also differs in spectral composition. We examined photosynthesis in the scleractinian coral Stylophora pistillata in shallow (3 m) and mesophotic settings (65 m) in the northern Red Sea. Symbiodinium spp. in corals originating from the mesophotic environment consistently performed below their photosynthetic compensation point and also exhibited distinct light harvesting antenna organization. Objective There is a critical need to identify biomarkers and objective outcome measures that can be used to understand underlying neural mechanisms in autism spectrum disorder (ASD). Visual evoked potentials (VEPs) offer a noninvasive technique to evaluate the functional integrity of neural mechanisms, specifically visual pathways, while probing for disease pathophysiology.

Methods Transient VEPs (tVEPs) were obtained from 96 unmedicated children, including 37 children with ASD, 36 typically developing (TD) children, and 23 unaffected siblings (SIBS). Fever is a highly conserved systemic response to infection dating back over 600 million years. Although conferring a survival benefit, fever can negatively impact the function of excitable tissues, such as the heart, producing cardiac arrhythmias. Here we show that mice lacking fibroblast growth factor homologous factor 2 (FHF2) have normal cardiac rhythm at baseline, but increasing core body temperature by as little as 3 C causes coved-type STelevations and progressive conduction failure that is fully reversible upon return to normothermia. FHF2-deficient cardiomyocytes generate action potentials upon current injection at 25 C but are unexcitable at 40 C. The absence …

Metazoan linker histones are essential for development and play crucial roles in organization of chromatin, modification of epigenetic states and regulation of genetic activity. Vertebrates express multiple linker histone H1 isoforms, which may function redundantly. In contrast, H1 isoforms are not present in Dipterans, including D. melanogaster, except for an embryo-specific, distantly related dBigH1. Here we show that Drosophila BEN domain protein Elba2, which is expressed in early embryos and was hypothesized to have insulator-specific functions, can compensate for the loss of H1 in vivo. Bilinguals have to learn two different grammatical systems. Some aspects of these grammars may be similar across the two languages (for example, the active-passive alternation) while others may exist in only one of the two grammars (for example, the distinction between recent and distant past). This dissertation investigates the degree to which grammar information specific to only one language is available when processing the other language. Nanoparticles are often combined with radionuclides for various applications, ranging from waste remediation to imaging and therapy in the medical field. The overarching aim of this body of work is two-fold. The first aim is development of new radiolabeling methods for various nanoparticles that allow stable attachment of a variety of imaging and therapeutic radionuclides. Short-tailed opossums (genus Monodelphis) comprise the most species-rich genus of New World marsupials, with 25 currently recognized species. Monodelphis comprise small, terrestrial species collectively widespread in South America, which makes the group potentially informative about biogeographic processes that have shaped the continental fauna. The genus exhibits striking variation in several phenotypic characters, notably pelage coloration, behavior, and reproductive strategies. This diversity is unique among Neotropical marsupials, and makes the group particularly interesting to investigate the evolution and the adaptive significance of phenotypic trait variation. Poetry is a useful medium for exploring the fluidity and possibilities in language beyond the everyday terms of normative language. For trans- and genderqueer subjects, whose identities cannot be articulated within the linguistic boundaries of binary gender, and whose outward appearance challenges the cultural logic of gendered visibility, poetry becomes a valuable and necessary tool for survival, disruption, activism, and personal and public empowerment. Through syntax, word choice, semantic and non-semantic qualities of language, poetry helps articulate the inexpressible, complex, and unstable gender identities and subject positions, even as they change or multiply. Talk Shows And Language Attitudes: A Sociolinguistic Investigation Of Language Attitudes Towards Taiwan Mandarin Among Chinese Mainlanders, Chun-Yi Peng

This dissertation looks at the effects of media exposure and language ideologies on Mandarin speakers’ acceptability judgments. Although there is a long-standing tradition against citing media exposure as a source of language variation, I show that 1) media exposure to a non-local perceptually salient variant can make people more likely to rate non-local linguistic features as grammatically acceptable, and 2) media exposure shapes people’s language attitudes—a new alignment of attitudes is emerging among the millennials on the mainland.

Data were collected through an online survey consisting of grammaticality judgments, matched-guise tasks, open-ended attitudinal questions, and demographic questions. The data show …

Don DeLillo’s 1997 novel Underworld stands as the framing text for this study of fiction, cultural affect, and resistance in the later part of the 1980’s – the exhausted, waning years of the Cold War – and the 1990’s, the period immediately following its collapse. This paper draws upon the principles of critical discourse analysis in order to examine the production of capitalist and consumerist discourses within contemporary nonhuman animal rights activism. The analysis presents evidence to suggest that the discourses being produced via the websites of People for the Ethical Treatment of Animals (PETA) and The Vegan Society are consistently being constructed through market-centric ideologies that treat activists mainly as middle-class consumers. This paper argues that the consistent presence of neoliberal discourse signals an instructive entanglement with broader sociopolitical issues. Psychodynamic assessment of object relations on projective tests has consistently been shown to contribute to a better understanding of children’s psychological functioning and to guide therapeutic interventions (Tuber, 1992). This research examines the enhanced utility of applying a psychodynamically-derived scale of children’s object relations to a commonly used projective assessment tool, the Thematic Apperception Test (TAT) (Morgan & Murray, 1935; Murray, 1943). This dissertation provides an ethnographic account of the food and parenting practices of a diverse group of middle-class families in the Mount Airy neighborhood of Philadelphia. It starts from the basic premise that the economic pressures on the American middle classes find expression in family life around the socially reproductive work of choosing food and parenting.

This dissertation details the educational histories and linguistic portraits of newcomer immigrant youth in their countries of origin, and provided accounts of their academic trajectories after arrival in the United States. The data provide evidence of poor quality instruction, particularly in literacy, and educational environments in low-income sending countries. The study shows that prior schooling experiences in the country of origin have an impact on school readiness, the degree to which immigrant youth are prepared for education in secondary classrooms in the U.S. The dissertation also provides an analysis of how pre-migration education interacts with educational performance after immigration, leading …

This dissertation suggests that late fourteenth century vernacular poetry in Middle English takes up the idea of a secular Eden by which various non-normative theorizations of time and self are made possible. Using thematic case study methodology, the study found that teachers believed that character education is important and should be taught.
