Amyloid Accelerator Polyphosphate Fits As The Mystery Density In Α-Synuclein Fibrils, Philipp Huettemann, Pavithra Mahadevan, Justine Lempart, Eric Tse

Open Access archive

Aberrant aggregation of α-Synuclein is the pathological hallmark of a set of neurodegenerative diseases termed synucleinopathies. Recent advances in cryo-electron microscopy have led to the structural determination of the first synucleinopathy-derived α-Synuclein fibrils, which contain a non-proteinaceous, “mystery density” at the core of the protofilaments, hypothesized to be highly negatively charged. Guided by previous studies that demonstrated that polyphosphate (polyP), a universally conserved polyanion, significantly accelerates α-Synuclein fibril formation, we conducted blind docking and molecular dynamics simulation experiments to model the polyP binding site in α-Synuclein fibrils. Here, we demonstrate that our models uniformly place polyP into the lysine-rich pocket, …

Role Of Absolute Versus Relative Voice Rest In Post-Operative Management Of Benign Vocal Fold Lesions, Mihika Sinha, Suresh Pillai, Shama Shetty, Usha Devadas

Open Access archive

Objective Most otolaryngologists advocate absolute voice rest after laryngeal surgery, which proves difficult for patients, so we decided to evaluate the role of absolute voice rest versus relative voice rest in the post-operative management of benign lesions. Methods Forty patients were recruited and divided in two groups: absolute voice rest and relative voice rest. Pre- and post-operative voice analysis (fundamental frequency, jitter, shimmer, voice handicap index, voice-related quality-of-life scale scores and compliance) were noted at one week and one month. Results Voice analysis parameters including jitter (p = 0.035), shimmer (p = 0.020), voice handicap index (p < 0.001) and compliance (p < 0.001) were better in the relative voice-rest group. Frequency, number of voice breaks and voice-related quality of life showed no statistically significant results. Conclusion There was no significant benefit of absolute voice rest on post-operative outcomes as determined by acoustic variables. Compliance and quality-of-life scores were low in the strict voice-rest group. Therefore, we should reconsider post-surgical voice-rest protocol.

Introduction: In India, the burden of diabetes mellitus is rising swiftly, mirroring a global trend that is being observed in both developed and developing countries. Examining the correlation between indices of glycemic status & thyroid and lipid parameters at different glycemic control levels was the goal of the present study. Methods: This case control study included 50 cases (HbA1c > 6.5 %) and 150 controls (HbA1C < 6.5) who were attending an executive health check up at a tertiary level health care hospital attached to a medical college in a coastal city in Southern India. Glycemic indices, thyroid profile and lipid profile were evaluated. Correlation analyses, t-tests, and chi-square tests were carried out. Results: Cases had a mean age of 57.80±10.71 years whereas controls had a mean age of 48.47±13.89 years. The cases and controls did not have discernable differences in the lipid profile characteristics. However, T4 and HbA1C (Glycated Hemoglobin), depicted a weak positive correlation with the value of correlation coefficient r= 0.211 (p= 0.014). With respect to T4, there was a higher mean rank contrast observed among the cases than that of controls (p=0.014). Conclusion: A positive association between thyroid hormone T4 levels and glycemic status was found in this study, even if no correlation was found between glycemic control and lipid profile. This research lays emphasis on closely observing thyroid function in addition to glycemic parameters for the all-encompassing control of diabetes mellitus.

Candidate Ram-Pressure Stripped Galaxies In Six Low-Redshift Clusters Revealed From Ultraviolet Imaging, K. George, B. M. Poggianti, A. Omizzolo, B. Vulcani

Open Access archive

The assembly of galaxy clusters is understood to be a hierarchical process with a continuous accretion of galaxies over time, which increases the cluster size and mass. Late-type galaxies that fall into clusters can undergo ram-pressure stripping, forming extended gas tails within which star formation can happen. The number, location, and tail orientations of such galaxies provide clues about the galaxy infall process, the assembly of the cluster over time, and the consequences of infall for galaxy evolution. Here, we utilise the ∼0.5-degree diameter circular field of the Ultraviolet Imaging Telescope to image six galaxy clusters at z<0.06 that are known to contain 'jellyfish'galaxies. We searched for stripping candidates in the ultraviolet images of these clusters, which revealed 54 candidates showing signs of unilateral extra-planar emission, due to ram-pressure stripping. Seven candidates had already been identified as likely stripping based on optical B-band imaging. We identified 47 new candidates through UV imaging. Spectroscopic redshift information is available for 39 of these candidate galaxies, of which 19 are associated with six clusters. The galaxies with spectroscopic redshifts that are not part of the clusters appear to be within structures at different redshifts identified as additional peaks in the redshift distribution of galaxies, indicating that they might be ram-pressure stripped or disturbed galaxies in other structures along the line of sight. We examine the orbital history of these galaxies based on their location in the position-velocity phase-space diagram and explore a possible connection to the orientation of the tail direction among cluster member candidates. There are limitations due to different integration times and imaging different regions with respect to the cluster centre. The tails of confirmed cluster member galaxies are found to be oriented away from the cluster centre.

"Entwined Derelict" was my first delve into the Twine interactive fiction engine, and the second game that I consider "feature complete". While a significant focus was put on learning the engine as a whole, particular effort for me was put into a sidebar "journal" of sorts, that took narrative flags from the game and recorded the events in a way that allows players to read back what they have experienced in-game at their leisure. While I greatly enjoyed making the game, and I do consider it a complete experience with some unique pieces within, it is a short adventure without …

Lower airway club cells (CCs) serve the dual roles of a secretory cell and a stem cell. Here, we probe how the CC fate is regulated. We find that, in response to acute perturbation of Notch signaling, CCs adopt distinct fates. Although the vast majority transdifferentiate into multiciliated cells, a “variant” subpopulation (v-CCs), juxtaposed to neuroepithelial bodies (NEBs; 5%–10%) and located at bronchioalveolar duct junctions (>80%), does not. Instead, v-CCs transition into lineage-ambiguous states but can revert to a CC fate upon restoration of Notch signaling and repopulate the airways with CCs and multiciliated cells. The v-CC response to …

Fieldnote 19: Intersection Of Mindfulness And Writing, Venus Evans-Winters Dr.

Fieldnotes of a Black Woman Scholar: Research, Culture, and Wellness

Recently, in our culture, there has been an overemphasis on writing for public consumption and profit. Academic writing privileges content that addresses "somebody else's problem" instead of writing for personal growth and imagination. This trend can overshadow the profoundly transformative potential of writing when it's approached as a tool for personal healing and authentic relationship building. Mindfulness awareness allows us to write from a place of authenticity and presence. As we observe our thoughts and emotions, we gain insights into our true selves, much like how staying present in the moment helps us navigate life's complexities. Dr. Venus Evans-Winters shares …
