Many older patients often suffer from walking issues such as Drop Foot. Drop Foot is caused by the malfunction of nerves in the foot, resulting in the loss of control of the front foot muscle. Within all of our patients, there has been a common fibular palsy, caused by the entrapment of the peroneal nerve. Due to this, they cannot lift up their foot. Some cases are permanent, while others are temporary. In the temporary cases, we have indicated a Phoenix Sign. Power is diminished more dramatically at higher contraction velocities in older adults. It has been suggested that this may reflect age-related changes in single myofiber morphology or composition. PURPOSE: To examine power, muscle activation, and single myofiber morphology and composition between young (YM) and older (OM) males. METHODS: Power, or torque × velocity, was recorded during isokinetic knee extensions at 60°·s-1 and 180°·s-1 in healthy, untrained YM (n=15; 20.7±2.2 yrs) and OM (n=15; 71.6±3.9 yrs). The purpose of this investigation was to observe the effects of normobaric hypoxia on accumulated oxygen deficit (AOD) with evaluation using both Bayesian and Frequentist analyses. Eighteen recreationally active men performed a graded exercise test (GXT) in normobaric normoxia (N; FiO2~20%) and normobaric hypoxia (H; FiO2~14%) to determine peak power output (PPO). Time to exhaustion trials were later conducted at 110% and 120% PPO under both N, and H. Lower-body power should aid foot pursuit ability; however, there has been minimal investigation of this relationship. This study investigated relationships between absolute and relative lower-body power measured by the vertical jump (VJ) with the 75-yard pursuit run (75PR), which is a change-of-direction (COD) speed test that simulates a foot pursuit in law enforcement recruits. The gut microbiome influences nutrient processing as well as host physiology. Plasma lipid levels have been associated with the microbiome, although the underlying mechanisms are largely unknown, and the effects of dietary lipids on the gut microbiome in humans are not well-studied. We used a compilation of four studies utilizing non-human primates (Chlorocebus aethiops and Macaca fascicularis) with treatments that manipulated plasma lipid levels using dietary and pharmacological techniques, and characterized the microbiome using 16S rDNA. High-fat diets significantly reduced alpha diversity (Shannon) and the Firmicutes/Bacteroidetes ratio compared to chow diets, even when the diets had different compositions …

Atherosclerosis is an inflammatory disease initiated by low and oscillatory shear stress on the endothelium. The inflammatory process recruits leukocytes to the vessel wall by expression of the adhesion molecule VCAM-1. Activation of the NF-κB inflammatory signaling pathway is responsible for the increase in VCM-1 expression. Omega 3 FAs, such as EPA, reduce the risk of atherosclerosis by decreasing this inflammatory response. The pathway by which omega 3 FAs is proposed to inhibit inflammation includes activating FFAR4 to decrease NF-κB activation thereby reducing expression of adhesion molecules. BACKGROUND: Activation of intravesical protease activated receptor 4 (PAR4) leads to release of urothelial macrophage migration inhibitory factor (MIF). MIF then binds to urothelial MIF receptors to release urothelial high mobility group box-1 (HMGB1) and elicit bladder hyperalgesia. Since MIF binds to multiple receptors, we investigated the contribution of individual urothelial MIF receptors to PAR4-induced HMGB1 release in vivo and in vitro and bladder pain in vivo.

Genetically-modified mice lacking the leptin hormone (Ob) or leptin receptor (Db) have reduced testosterone and decreased sperm production leading to infertility. Leptin receptors are also expressed by immune cells, suggesting leptin can regulate immune responses. Recent studies have focused on identifying the immune cells present within the epididymis and found cells expressing markers typically associated with dendritic and macrophage cells. These cells likely function against infection and to induce tolerance to sperm. The effects of leptin on the immune system and the presence of immune cells within the epididymis raises the possibility that alterations in the immune cell profile and/or …

The processes underlying formation and growth of unfolded protein inclusions are relevant to neurodegenerative diseases but poorly characterized in living cells. In S. cerevisiae, inclusions formed by mutant huntingtin (mHtt) have some characteristics of biomolecular condensates but the physical nature and growth mechanisms of inclusion bodies remain unclear. We have probed the relationship between concentration and inclusion growth in vivo and find that growth of mHtt inclusions in living cells is triggered at a cytoplasmic threshold concentration, while reduction in cytoplasmic mHtt causes inclusions to shrink. The growth rate is consistent with incorporation of new material through collision and coalescence. …

It is well-established that reactive oxygen species (ROS), particularly superoxide (O2·-), produced in the vasculature and brain contribute to the pathogenesis of hypertension. We, and others, have previously reported that in vivo scavenging of O2·- via overexpression of superoxide dismutase (SOD) protein or SOD mimics decreases blood pressure in hypertensive animals. While few SOD mimics have transitioned to clinical trials, MnTnBuOE-2-PyP5+ (BuOE), a manganese porphyrin SOD mimic, is currently in clinical trials for normal tissue protection in cancer patients exposed to radiation therapy. However, their potential to improve performance and/or recovery following exercise in non-athletic populations is unknown. Our study evaluated the effects of CS on exercise performance and recovery from a graded maximal treadmill test. Previous research has shown that various modes of exercise may elicit significant increases in resting metabolism for up to 24 hours post-exercise, but typically using untrained or moderately active subjects. The purpose of the present study was to compare excess post-exercise oxygen consumption (EPOC) between circuit-style resistance training (RT) and high-intensity interval training (HIIT) in young, aerobically fit women. Muscle development during embryonic and early post-hatch growth is primarily through hyperplastic growth and accumulation of nuclei through satellite cell contribution. Post-hatch, muscle development transitions from hyperplasia to hypertrophic growth of muscle fibers. Commercial selection for breast yield traditionally occurs at ages targeting hypertrophic rather than hyperplastic growth. This has resulted in the production of giant fibers and concomitant challenges with regard to muscle myopathies. The flexible shell of some oviparous reptiles has led to differences in nutrient mobilization and water relations when compared to their rigid-shelled counterparts. Flexible-shelled eggs gain more water during development, and because of the shell structure, yolk calcium content is higher than that of eggshells. When water availability is altered for reptiles with flexible-shelled eggs, differences in both energy and nutrient utilization are observed. Alzheimer’s Disease (AD) is a progressive brain disorder that destroys memory and cognition thought to be initiated through the build-up of the amyloid-beta (A) peptide. The A peptide aggregates, slowly forming into insoluble plaque substances that destroy the brain and worsen patient’s symptoms over time. Studies have shown that chronic isolation stress (CIS) increases the A peptide soluble and insoluble levels in the brain and that exercise training decreases these levels in transgenic mouse models of AD. The Alligator Snapping Turtle (Macrochelys temminckii) is one of many turtle species facing conservation challenges. Nearly extirpated in the 1980s, it is currently the subject of a head-start initiative, of which, any lasting behavioral or physiological effects were—until now—unknown! To evaluate the ability of captive-reared individuals to excel in natural habitats, and to foresee any future research or conservation challenges regarding this animal, I explored a suite of variables that influence bite performance and behaviors including captive or free-ranging status, and environmental conditions including body temperature and season. Renal medullary carcinoma (RMC) is a lethal cancer that predominantly affects young individuals with sickle cell trait (SCT). It is not currently understood why RMC only affects certain individuals with SCT. We found that patients with RMC more frequently participated in high-intensity exercise than matched controls. Using mouse models of SCT, we demonstrated the significant increase of renal hypoxia in the right kidney following high- but not moderate-intensity exercise. We also demonstrated in cell culture studies that SMARCB1 is ubiquitinated for proteasome-mediated degradation in hypoxia, and the re-expression of SMARCB1 leads to compromised proliferation in renal cells specifically in the …

International Journal of Exercise Science 14(6): 984-993, 2021. Limited research exists on physical performance assessments for women collegiate athletes. The current cross-sectional study compared field-based tests of muscular strength and power and investigated their relationship. Sports included field hockey, volleyball, soccer, and softball. Tests of one repetition maximum (1-RM) back squat, 1-RM bench press, vertical jump, and standing long jump were administered. A one-way analysis of variance (ANOVA) assessed differences across sports. Bivariate Pearson correlation coefficients examined relationships among tests. Strength and conditioning coaches were allegedly involved in pre-COVID-19 nontraumatic injuries/deaths (e.g., exertional heat illness, exertional rhabdomyolysis, cardiorespiratory failure) of NCAA (mainly football) student-athletes during off-season training sessions. During the COVID-19 health crisis, non-pharmaceutical interventions against the SARS-CoV-2 virus included suspension of NCAA seasons, which led to student-athletes exhibiting lower levels of mental health. All NCAA championships have now been reinstated. This summer the off-season is unique, because as several programs had stopped practicing/competing for several months, it may foster additional threats to the wellbeing of the student-athletes. International Journal of Exercise Science 14(3): 1004-1017, 2021. This study evaluated plasma levels of brain-derived neurotropic factor (BDNF), irisin, and lactate in people living with HIV/AIDS who completed a combined physical training program. Nineteen HIV+ participants (age: 39.60 ± 10.96 years; carrier time: 7.75 ± 7.88 years; time of ART: 6.41 ± 5.93 years) performed strength/aerobic training (combined physical training) in the same session for 8 weeks and levels of BDNF, irisin, and lactate were assessed. BDNF (pg/mL) was higher post-CPT (Pre: 1258.73 ± 372.30; Post: 1504.17 ± 322.30; p < 0.001). Irisin (ng/mL) showed no change (Pre: 115.61 ± 72.41; Post: 125.87 ± 81.14; p = 0.973). There is no cure for osteogenesis imperfecta (OI), and current treatments can only partially correct the bone phenotype. Stem cell therapy holds potential to improve bone quality and quantity in OI. Here, we conduct a systematic review and meta-analysis of published studies to investigate the efficacy of stem cell therapy to rescue bone brittleness in mouse models of OI. Identified studies included bone marrow, mesenchymal stem cells, and human fetal stem cells. Effect size of fracture incidence, maximum load, stiffness, cortical thickness, bone volume fraction, and raw engraftment rates were pooled in a random-effects meta-analysis.
