International Journal of Exercise Science 14(4): 971-979, 2021. The purpose of this study was to determine whether the number of warm-up sets and relative intensity impacts the prediction of velocity-based one-repetition maximum (1RM) values. Twenty resistance-trained individuals (males: n = 10, females: n = 10) participated in this study. Warm-up sets consisted of subject’s bench-pressing loads at 50 (five-repetitions), 70 (three-repetitions), and 90% (one-repetition) of estimated 1RM. A maximum of four attempts were performed to determine 1RM, while recording mean concentric velocity (MCV)using a linear position transducer during warm-up and 1RM trials in order to develop load-velocity profiles. International Journal of Exercise Science 14(3): 962-970, 2021. Transcranial direct current stimulation (tDCS) uses a weak electrical current that is sent through the cerebral cortex. The Halo Sport headphones are a user-friendly form of tDCS that is implemented by many athletes purportedly to improve performance. The purpose of this study was to determine the effect of tDCS, using the Halo Sport, on performance variables associated with a high-intensity three-minute cycling test. Eighteen healthy, active individuals (ten men, eight women) volunteered for this study. Notch signaling is required to repress the formation of vertebrate cone photoreceptors and to maintain the proliferative potential of multipotent retinal progenitor cells. However, the mechanism by which Notch signaling controls these processes is unknown. Recently, restricted retinal progenitor cells with limited proliferation capacity and that preferentially generate cone photoreceptors have been identified. Thus, there are several potential steps during cone genesis that Notch signaling could act. Here we use cell type specific cis-regulatory elements to localize the primary role of Notch signaling in cone genesis to the formation of restricted retinal progenitor cells from multipotent retinal progenitor cells. This dissertation focuses on two major topics germane to mechanisms by which animals tolerate extreme water stress. First, the impact of transgenic expression of late embryogenesis abundant proteins (LEA) on water stress tolerance of the fruit fly Drosophila melanogaster was investigated. The overall aim was to extend current understanding of the protective properties of LEA proteins documented with isolated cells to a desiccation-sensitive organism during exposure to drying and hyperosmotic stress. Four lines of D. melanogaster were created that expressed transgenes encoding selected LEA proteins originally identified in embryos of the extremophile Artemia franciscana. An animal's ability to synchronize life-history events or stages with optimal environmental conditions is paramount to successfully reproducing and maximizing fitness. Additional events including migration, foraging, rearing of young, and emergence from hibernation are all examples of processes under environmental control in one species or another. An exciting new mechanism involving neural thyroid hormone metabolism has been elucidated that links environmental control to the neuroendocrine reproductive axis. Freeze-tolerant gray treefrogs survive winter by producing natural cryoprotectants and accommodating ice formation within extracellular spaces. While frozen, gray treefrogs endure hyperglycemia, dehydration, and anoxia due to the halt of all bodily functions. Upon thawing, the frogs’ anoxic cells receive a rapid influx of oxygen, which can cause oxidative damage to vital macromolecules including DNA. Previous studies have suggested freeze-tolerant frogs avoid oxidative damage after freeze-thaw events by elevating antioxidant activity, but recent work has shown upregulated DNA repair encoding genes in post-freeze frogs. Woody breast (WB) myopathy results in poor muscle quality. The increasing incidence of WB over the last several years indicates a need for improved prediction or early diagnosis. We hypothesized that the use of body fluids, including blood, may be more suitable than breast muscle tissue in developing a minimally invasive diagnostic tool for WB detection. Thyroid hormones (THs) in mammalian tissues are crucial for development and maintaining metabolic homeostasis. Iodothyronine deiodinases (Dios) remove iodines from THs by a selenocysteine (Sec) residue, which either activates or inactivates them. Halogen bonding (XB) has been proposed to describe the interaction between the Se and I atoms of the T4-Dio complex. Disruption of TH homeostasis by xenobiotics, such as polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) can cause deleterious effects on the endocrine system. The purpose of this study was to determine the impact of a two-week relaxing music intervention on stress, anxiety, and gastrointestinal (GI) symptoms in exercisers with at least mild anxiety and exercise-related GI symptoms. Block randomization was used to allocate 15 women and two men into control (n = 8) or intervention (n = 9) groups following a one-week baseline. During the baseline period, participants recorded details of their aerobic exercise sessions and discomfort with GI symptoms (nausea, regurgitation/reflux, stomach fullness, bloating, abdominal cramps, gas, the urge to defecate). This study was conducted to distinguish the effects of heat stress (HS) and feed intake (FI) on broiler chicken’s physiological responses. The sodium-hydrogen exchanger isoform 3 (NHE3, SLC9A3) mediates most Na+/H+ exchange in the gastrointestinal tract and the kidney, where it plays crucial roles in Na+ and fluid absorption as well as acid-base homeostasis. Whole body NHE3 knockout (NHE3-/-) mice show overt absorptive defects in both the intestine and kidneys. Genetic mutations of NHE3 gene in humans are associated with congenital sodium diarrhea (CSD). In the kidneys, NHE3 is expressed in the proximal tubule (PT) and the thick ascending limb (TAL). AIM: To provide a detailed gene and protein expression analysis related to mitochondrial biogenesis and assess mitochondrial content in skeletal muscle of children with cerebral palsy (CP).

METHOD: Biceps brachii muscle samples were collected from 19 children with CP (mean [SD] age 15y 4mo [2y 6mo], range 9-18y, 16 males, three females) and 10 typically developing comparison children (mean [SD] age 15y [4y], range 7-21y, eight males, two females). While the desire to uncover the neural correlates of consciousness has taken numerous directions, self-face recognition has been a constant in attempts to isolate aspects of self-awareness. The neuroimaging revolution of the 1990s brought about systematic attempts to isolate the underlying neural basis of self-face recognition. These studies, including some of the first fMRI (functional magnetic resonance imaging) examinations, revealed a right-hemisphere bias for self-face recognition in a diverse set of regions including the insula, the dorsal frontal lobe, the temporal parietal junction, and the medial temporal cortex. Temperature and environmental oxygen availability affect oxygen supply and demand in ectotherms, which are hypothesized to control the geographic limits of many marine species. The oxygen supply capacity (α) is calculated from commonly measured metabolic traits, including the standard metabolic rate (SMR) and critical oxygen partial pressure at SMR (Pcrit). A subset of sensory nerves called nociceptors are stimulated by noxious stimuli and evoke nocifensive responses in different organ systems. Transient Receptor Potential Ankyrin 1 (TRPA1) is a tetrameric, nonselective, cation channel that initiates the generator potential that evokes afferent signaling in nociceptive nerves. TRPA1 is activated many harmful irritants, such as food chemicals, environmental pollutants, reactive oxygen species and other endogenous mediators. Circadian rhythms are generated by cell autonomous circadian clocks that perform a ubiquitous cellular time-keeping function and cell type-specific functions important for normal physiology. Studies show inducing the deletion of the core circadian clock transcription factor Bmal1 in adult mouse cardiomyocytes disrupts cardiac circadian clock function, cardiac ion channel expression, slows heart rate, and prolongs the QT-interval at slow heart rates. This study determined how inducing the deletion of Bmal1 in adult cardiomyocytes impacted the in vivo electrophysiological phenotype of a knock-in mouse model for the arrhythmogenic long QT syndrome (Scn5a+/ΔKPQ). Osteosarcoma (OS) is an aggressive bone cancer originating in the mesenchymal lineage. Prognosis for metastatic disease is poor, with a mortality rate of approximately 40%; OS is an aggressive disease for which new treatments are needed. All bone cells are sensitive to their mechanical/ physical surroundings and changes in these surroundings can affect their behavior. Introduction: Burnout in emergency medicine and in residency training has been well-described. The impact of demographic, individual, and programmatic factors on burnout have not previously been determined in a national survey of emergency medicine residents. This study aimed to identify personal and environmental factors impacting resident burnout in a national sample of emergency medicine residents.

Methods: A prospective Emergency Medicine Resident Wellness Survey was administered in 2017. Law enforcement agencies often conduct academy training classes that include recruits hired by both the larger hosting agency (HA), and by smaller participating agencies (PA). In alignment with efforts to mitigate the negative health consequences of Parkinson’s Disease (PD), the purpose of this investigation was to examine if participation in a community-based boxing program (CBP) was associated with improvements in balance and fall risk reduction among individuals with PD. In this retrospective cross-sectional study, de-identified data from 12 individuals with PD participating in a CBP was examined. The promotion of ketone body (KB) metabolism via ketosis has been suggested as a strategy to increase exercise performance. However, studies in humans and animals have yielded inconsistent results. The purpose of the current study was to examine the effects of ketosis, achieved via fasting or a short-term ketogenic diet (KD), on endurance exercise performance in female mice. After 8 h of fasting, serum KB significantly increased and serum glucose significantly decreased in fasted compared to fed mice. Heart failure (HF) is a common cause of morbidity and mortality in the United States that may disproportionately affect certain racial/ethnic groups. Compared with White individuals, HF may affect Black individuals at a younger age with less favorable prognosis, and this excess risk may be partially explained by differences in HF risk factor burden. This study examined the physiological changes resulting from training on a lower body positive pressure treadmill (LBPPT) at three different levels of body weight support (BWS). Thirty-three healthy college aged students (22.3 ± 3.1 years) completed the study. Participants performed a graded exercise test (GXT) to exhaustion and were placed into one of three experimental groups corresponding to 100%, 75%, and 50% of their normal BW.
