Precision Delivery Using Nanopipette For Single-Cell Studies, He Zhang, Maksudur Rahman, Yang Tao, Joseph W Sampson, Hang Ren

Nanopipette based scanning probe technique is a versatile tool in non-contact imaging in biology. In addition to the topographic imaging, its capability of localized delivery of bio-active molecules is emerging. In this mini review, we introduce the applications of nanopipette in single-cell researches with a focus on localized delivery. The working principles of three delivery modes including resistive pulse, pressure-driven flow, and electroosmotic flow-driven delivery are summarized and compared. Their applications in single-cell researches are reviewed. The current technical challenges in scanning ion conductance microscopy-based delivery, and their growing influence in medicine and pharmacologic researches are also discussed.

Limb-Girdle Muscular Dystrophy Type 2B/R2 (LGMD2B/R2) is a type of dysferlinopathy, a progressive muscle-wasting disorder caused by mutations in the dysferlin gene. Hallmarks of the disease include chronic inflammation, impaired muscle regeneration, and eventual muscle degeneration. Currently, no FDA-approved therapies are available for dysferlinopathies. Recombinant human Galectin-1 (rHsGal-1) has shown multiple roles in maintaining skeletal muscle homeostasis, promoting muscle repair, enhancing myogenesis, and modulating inflammation by downregulating the NF-κB pathway. OBJECTIVE: Renal cell carcinoma (RCC) is a rare but severe and aggressive pediatric malignancy. While incidence is uncommon, survival is relatively low with respect to acute lymphocytic leukemia (ALL), AML, lymphoma, ependymoma, glioblastoma, and Wilms Tumor. The pediatric renal cell carcinoma (pRCC) incidence, cumulative incidence (period prevalence), and mortality vary by health disparities' indicators, namely sex, race, ethnicity, age at tumor diagnosis, and social determinants of health (SDHs) as well as Epigenomic Determinants of Health (EDHs). The emergence of spatial transcriptomic technologies has opened new avenues for investigating gene activities while preserving the spatial context of tissues. Utilizing data generated by such technologies, the identification of spatially variable (SV) genes is an essential step in exploring tissue landscapes and biological processes. Particularly in typical experimental designs, such as case-control or longitudinal studies, identifying SV genes between groups is crucial for discovering significant biomarkers or developing targeted therapies for diseases. In this article, we present a novel B-Polynomial Approach for approximating solutions to partial differential equations (PDEs), addressing the multiple initial conditions. Our method stands out by utilizing two-dimensional Bernstein polynomials (B-polynomials) in conjunction with their operational matrices to effectively manage the complexity associated with PDEs. This approach not only enhances the accuracy of solutions but also provides a structured framework for tackling various boundary conditions. The PDE is transformed into a system of algebraic equations, which are then solved to approximate the PDE solution. In the Chenier Plain of Louisiana and Texas, coastal marshes are subject to high rates of relative sea level rise (RSLR), restricted sedimentation, and declining elevation attributed to subsidence and decades of intensive management. These marshes were created via mineral sedimentation but now largely accrete via organic matter accumulation. This organic matter accumulation (OMA) – or the balance between plant productivity and decomposition – is essential for maintaining elevation and thus long-term sustainability. This work focuses on the study of bioconvection in a conical region of rotating and stationary cone-disk systems utilizing nanofluids involving gyrotactic micro-organisms. The flow geometry encompasses two different configurations, namely, rotating cone-disk system (RCDS) and stationary cone-disk system (SCDS). For RCDS, four unique configurations are considered: rotating cone static disk (Model-I), static cone rotating disk (Model-II), co-rotating cone-disk (Model-III), and counter-rotating cone-disk (Model-IV), while SCDS includes both swirling and non-swirling flow scenarios. The space environment poses significant dangers and difficulties for human explorers—difficulties so great that it has been argued that space exploration should be exclusively conducted by automated spacecraft. Decomposition is a crucial step in the cycling and recycling of nutrients in the natural world. As things die, detritivores such as bacteria, protists, and fungi break down the material into organic and inorganic matter, releasing nutrients into the soil to be used by living plants. This study aimed to determine the rate of decomposition of a native plant, Baptisia australis, (Blue False Indigo) compared to a non-native plant, Cosmossulphureus (Cosmos). Decomposition rates are important in measuring the health of an ecosystem. They can indicate the rate at which nutrients cycle through the habitat. However, not all leaves will decompose at the same rate, which impacts the rate at which nutrients can be recycled. Plant propagation is a method of growing plants through various techniques that create genetically identical clones of the parent plant. Propagation maintains the preferred genetic material of a plant so that it can be further reproduced on a larger scale. To assess growth variations, sweet potato cuttings were propagated in soil and water, as well as in water with and without rooting hormone. Additionally, Monstera cuttings were propagated in water with and without rocks. Root development was observed across all conditions to determine the most effective propagation methods. This work introduces a new dual-X current conveyor differential input transconductance amplifier (DXCCDITA) based meminductor emulator, alongside its application in chaotic oscillator has also been presented. To realize the designed meminductor emulator, one DXCCDITA, two resistors, and two capacitors are employed. Pinched hysteresis loops are achieved across a wide frequency range spanning from 100 Hz to 1.5 MHz, encompassing both decremental and incremental topologies. Additionally, the proposed circuit offers the flexibility to switch between incremental and decremental configurations using a simple switch. Effective diabetes management relies on precise prediction of blood glucose levels to minimize complications. However, the patterns and fluctuations in blood glucose vary significantly among patients, posing a challenge for existing prediction methods. Many current approaches fail to accommodate these individual differences, leading to less reliable predictions. In response to this challenge, we present LGformer, a novel prediction model based on the Informer architecture, designed to enhance both flexibility and accuracy. Research on the evolution of dinosaur flight has long been dominated by the wing, often overlooking the foot. The foot of Archaeopteryx, the “icon of evolution”, thus offers a unique opportunity to investigate this critical evolutionary stage. Chapter I of this dissertation integrates advanced 3D reconstruction techniques, combining photogrammetry with X-ray data, to create a high-resolution model of the foot of the Thermopolis specimen of Archaeopteryx for visualization and analysis. Consisting of more than 11,000 members distributed over five families, the tautomerase superfamily (TSF) is a large collection of proteins with diverse biological functions. While much attention has been given to individual TSF enzymes, a majority remain structurally and functionally uncharacterized. Given its large size, studying a representative member of each family offers a viable approach for extracting mechanistic insights applicable to the entire superfamily. The advancement and application of non-invasive fisheries survey technologies, such as the scientific echosounder, have significantly enhanced our understanding of fish populations and their habitats, more recently in regions where traditional sampling methods are limited, such as complex reef habitats. This research, conducted as part of the Continental Shelf Characterization, Assessment, and Mapping Project (C-SCAMP) on the West Florida Shelf, leverages acoustic and visual technologies to investigate the distribution of fish densities, fish target strength and length relationships, and the relationship between seafloor topography and fish biomass distribution.

Analyzing lipid assemblies, including synthetically produced liposomes and naturally produced extracellular vesicles (EVs), is challenging due to their size, diverse composition, and tendency to aggregate. EVs contain a complex variety of lipids, proteins, and other metabolites. EVs composition can provide us insight into what is happening inside the body and a subcellular level and have potential for noninvasive biomarkers. Gymnasts are at a high risk of Achilles tendon (AT) injuries due to intense loads during takeoffs and landings. To better understand better the relationship between impact loading and its effects on tendon adaptation and athletic performance, this study uses Inertial Measurement Units (IMUs) to track AT loading across a collegiate gymnastics season. This data were compared against physiological and athletic performance metrics, obtained through force plates and ultrasound imaging, to explore how mechanical load influences reported pain and athletic performance. Cells rely on the secretion of exosomes to communicate with one another and this process is potentially mediated by a protein called tetherin, which anchors exosomes to the cell surface to limit their spread. Tetherin protein has attracted a lot of attention because of its capacity to hinder the propagation of viruses by attaching virions to the surface of host cells. Treatment with interferon-alpha (IFNα) increases the production of tetherin, although it is not known how this affects tetherin localization and exosome-mediated communication. This study investigates the potential of Fe-nontronite, a clay mineral found on Mars, to preserve biological cells and retain biosignatures such as trace metals. The research explores the preservation potential of nontronite by reacting it with cells under the following conditions: 1) reacting cells with nontronite precursors at low temperature and 2) reacting cells with the already synthesized nontronite made at high temperature. More than 40% of the Earth’s surface is covered by drylands. Due to limited availability of water, less surface area is covered by vascular plants. Instead, this space is occupied by microbial communities that exist in close contact with mineral soil, creating a living aggregate on the upper soil layer and are known collectively as biological soil crusts or “biocrusts”. Biocrusts play a vital role in ecosystem functioning in drylands specifically through their influence on hydrological processes (e.g., water retention, infiltration). Ectomycorrhizal fungi (ECM) form symbiotic relationships with their partner plant, creating root systems which allow for the exchange of nutrients among connected plants. The Spring Mountains National Recreation Area (NRA) in Southern Nevada is the study site for examining ECMs due to its diverse ecosystem with numerous inhabiting species. To assess the effects of ECMs on the tree composition, literature review was conducted to outline previous methods. Large-scale wildfires have been historically rare in hot deserts like the Mojave, largely due to the scarcity or uneven distribution of native plant biomass as fuel. Thus, native plants in these regions are ill-adapted to withstand spreading wildfire, resulting in slow post-fire recovery. Here, I test the hypothesis that nonnative grasses respond better to heat cues than native plant species. Seeds from 3 invasive and 13 native plant species were collected from research sites located in the eastern Mojave Desert, near surrounding Clark County Flood Control District weather stations. A crucial part of tissue engineering lies in understanding and characterizing the mechanical and viscoelastic properties of various types of biomaterials. Understanding these properties allows better biomaterials to be produced with characteristics more similar to those of the human body and with better biocompatability. In this study, sodium alginate was chemically crosslinked with calcium chloride and subsequently tested for it's instantaneous Elastic Modulus, instantaneous Shear Modulus, and equivalent viscosity using indentation testing methods consisting of a stress-relaxation test and nonlinear curve-fitting analysis. Timely detection of methane leaks from natural gas infrastructure, like pipelines and valves, is essential for mitigating environmental and safety risks. This research focuses on using mid-wave infrared (MwIR) cameras on unmanned aerial systems (UAS) to detect leaks. By leveraging machine learning, the study aims to develop an efficient, real-time methane inspection system that operates directly on embedded processors on UAS platforms.

Globally, potato (Solanum tuberosum) is a key staple food crop. In Kenya, it is the second most important food crop after maize. Among the various constraints to potato production are plant-parasitic nematodes. In particular, root-knot nematodes (Meloidogyne spp.) are a significant impediment to potato production, suppressing yield and reducing the quality of harvested tubers. Large-scale, high-dimensional data analyses can be computationally prohibitive due to combinatorial explosion of the search space for finding complex patterns; a viable alternative is network modeling for abstraction and quantifying intrinsic data associations. Prominent network analysis methods furnish frameworks for model synthesis and validation but rely on standard correlation measures impaired by semi-supervised biases, latent heterogeneity, and uneven discretization techniques. Here we investigate a holistic measure for encapsulating data heterogeneity for enhanced efficacy of revealing complex patterns through network analysis.
