Microwell Based Solid Phase Competitive Enzyme Immunoassay For Estradiol And Estriol, Jeng-Ting Yang

A sensitive and simple microwell based competition enzyme immunoassay for the quantitative determination of estradiol (E2) and estriol (E3) was developed. Nicrowells coated with antibody were incubated with antigen followed by adding horseradish peroxidase (HRPO) conjugate. The assay, which can be performed within two hours at room temperature, involved simultaneous incubation of E2 - or E3-HRPO conjugate and serum sample in polystyrene microwells coated with anti- E2 or anti-E3 gamma globulin fraction.
Gamma globulin was isolated from whole anti-serum by DEAE-cellulose chromatography. A carbodiimide coupling method was utilized to prepare the …

An enzyme-labeled immunoassay of sufficient range and sensitivity was developed for serum testosterone determination. In this method, the testosterone in the serum sample and testosterone-HRPO conjugate compete with each other for the limited antibody binding sites on the anti-testosterone gamma globulin coated microwell. The assay can be performed in only 2.5 hours.
The gamma globulin fraction was isolated from the anti-testosterone whole serum by DEAE-cellulose column chromatography. Testosterone-peroxidase conjugate was prepared by a carbodiimide coupling method. The interference which arises from the binding of testosterone to steroid hormone binding protein was eliminated by the addition of 17 beta-estradiol. All assay …

Triphenylethylene antiestrogen compounds are used to stimulate ovulation in anovulary women and spermatogenesis in oligozoospermatic males. Clomiphene Citrate (CC) is given therapeuticaly as a mixture of the trans (E) and cis (Z) isomers. Each of these isomers has its own characteristic agonist-antagonist profiles. Only the trans form of Tamoxifen (T) is administered therapeutically.
HPLC methodologies were developed for the determination of CC and Tin human blood and seminal plasma. The methodologies involved extraction of the triphenylethylene compounds using hexane, followed by evaporation of the hexane under N2. The residue was then taken up in a volume of HPLC …

Replication and amplification of RNA genomes of defective interfering (DI) particles of vesicular stomatitis virus (VSV) depend on the expression of viral proteins and have untD now been attained only in ceUs coinfected with helper VSV. In the work described in this report, we used a recombinant vaccinia virus-T7 RNA polymerase expression system to synthesize individual VSV proteins in cells transfected with plasmid DNAs that contain cDNA copies of the VSV genes downstream of the T7 RNA polymerase promoter. Advocacy/Empowerment: An Approach To Clinical Practice For Social Work, Stephen M. Rose
The Journal of Sociology & Social Welfare
Social Work has been embedded in a structural and ideological contradiction throughout its history. The profession, its employing institutions, and the problems confronted by its clients are all produced by the same political economy that pays its workers and supports its schools. Ideologically, the profession has avoided the confrontation implied by its dependency upon individual defect explanatory or causal analysis frameworks that constitute a betrayal of its real constituencies. An advocacy! empowerment paradigm is offered as an alternative.

Remarkable success has been achieved in the transplantation of allogeneic cardiac grafts into newborn infants at Loma Linda University Medical Center. The superior graft survival rate documented in these patients has not correlated with the degree of immunosuppression rendered, or the selection of genetically matched donors. However, the clinical success has correlated with the age of the recipient at the time of receiving a transplant. Patients receiving an allograft within the first few weeks of life are unique in that they seem to accept the alloantigens of their cardiac graft while responding aggressively to antigens in their environment. The purpose of this study was to determine the relative safety of headgear safety straps. Samples from five different safety headgear straps simulated orthopedic and orthodontic use and data were collected from a series of pulls on an Instron machine. The straps were compared by the distance the face bow traveled before the safety mechanism released, the force built into the face bow at safety release and the force required to remove the inner bow out of the molar tube. The study of the regulation of gene expression in complex genes can be facilitated by the use of operon fusions which place a well characterized lacZ gene under the control of a promoter of interest. An hsd::MudX128 operon fusion mutant of E. coli K-12 isolated by Prakash (1986) was observed to have a high β-galactosidase activity (1000 units), ten to twenty times higher than other hsd::MudX mutants. This β-galactosidase activity was suppressed to a very low level (25 units) by the introduction of an F’ plasmid, F'101, which carries the 98 min to 2 min region of the …

This clinical study evaluated a modified Nance appliance designed by the author to distalize molars unilaterally for Class II correction. This fixed appliance did not rely upon extraoral traction or intraoral elastics. Twelve unilateral Class II patients aged 13 to 17 were treated nonextraction with the appliance. Pretreatment and posttreatment photographs were taken of the palatal soft tissue and did not reveal any untoward changes as a result of the Nance acrylic button. All molars were corrected to an Angle's Class I relationship. Molar correction was evaluated visually and molar distalization was measured using an acrylic jig fitted to the …

Modulation of surface transferrin receptor (TfR) activity has been associated with leukemia cell differentiation and proliferation. To examine the mechanisms involved in regulating this event, surface receptor protein and mRNA levels were measured in HL-60 promyelocytic leukemia cells induced to differentiate along the myelocytic and monocytic pathways. Surface TfR down-regulation which occurs during granulocytic differentiation by dimethylsulfoxide, retinoic acid, or aclacinomycin A appears to be kinetically compatible with reduced biosynthesis resulting from reductions in the level of steady-state mRNA. In contrast, biosynthetic modulation does not appear to mediate the intial surface receptor down-regulation seen during 12-O-tetradecanoylphorbol-13-acetate-induced macrophagic differentiation. The purpose of this research was to conceptualize, construct and establish psychometric properties of an instrument to examine unmeasured dimensions of autonomy in practice through a process of retroduction and triangulation. In the context of ongoing rapid change and increasing competition in the health care delivery arena there is valid reason for nursing and other health professions to be concerned with their autonomous practice status. However, research and instrumentation activities have been hindered by the abstract nature of autonomy with only limited theoretical dimensions empirically measured. The purpose of the following study was to determine whether contraceptive use could be predicted from health locus of control, contraceptive self-efficacy, and sexual self-concept scores in a selected group of single college students ages 18-24. In order to better understand contraceptive behaviors, it is desirable to determine which personality variables may influence contraceptive behavior in young adults. A sample of 122 single undergraduate college students, both male and female, ages 18-24, completed a self-reported questionnaire including tools to measure health locus of control, contraceptive self-efficacy, and sexual self-concept, as well as demographic data. This study identifies the occupational interests and predominant vocational personality types of freshmen students (undeclared majors, nursing majors, and non-nursing majors), explores perceptions about selected professional nursing functions and characteristics among the students, and examines perceptual differences between groups (majors).
A non-experimental ex post facto design is used to analyze data from 312 students in a large urban university. Descriptive and inferential statistics are applied in data analysis. The following findings support the hypotheses: (1) Nursing continues to attract the traditional student who has Holland's Social vocational personality type as measured by the Self-Directed Search, although many Social types are …

Multiple sclerosis causes demyelination of central nerve fibers, and it is this pathology which results in most of the symptoms of the disease. The repair of the fibers by remyelination has been advanced as a potential symptomatic therapy, but at present the conduction properties of remyelinated central fibers are not well understood. In this study the conduction properties of Schwannian or oligodendrocyte remyelinated central nerve fibers have been determined. In addition, the status of the blood-brain barrier has been examined in lesions characterized by Schwann cell remyelination. These lesions chronically lack astrocytes, a cell believed to be involved in the …

The purpose of the present study was to conduct an empirical investigation of parallel process. The study used a cross-sectional design in which 30 therapy relationships and the corresponding supervision relationships were studied. The therapist assessed the behavior manifested by the patient during a targeted therapy session. Following the subsequent supervision session, the supervisor assessed the behavior manifested by the supervisee during the supervision session. In addition, each of the triad participants (patient, therapist, supervisor) rated the level of anxiety they experienced during the targeted therapy and supervision sessions. Most methods for isolation of RNA from yeast require tedious vortexing with glass beads, and give low yields when scaled down to 10 ml cultures (1). In addition, it is frequently desirable to prepare RNA from several different yeast strains grown under a variety of growth conditions, and preparations using glass beads are impractical when dealing with multiple samples.

Actinomycotic infections, once common in humans and cattle, are now rare causes of disease in man. This general group of organisms belongs taxonomically between the true bacteria and the fungi; however, the organisms behave clinically like true anaerobes. The organism, although phagocytized by the host cells, is not killed. Therefore, d is defined as a facultative intracellular parasite of the host. The protean manifestations of actinomycotic infections often mimic infectious processes, such as osteomyelitis or granulomatous disease, as well as neoplasia. The conventional estimation of motor, sensory, and mixed nerve conduction velocities reflects activity in the fastest conducting, heavily myelinated nerve fibers that are only a small proportion of the total. Unmyelinated and thinly myelinated fibers are not evaluated by this technique and numerically represent the largest group of fibers in human cutaneous nerves. The availability of new quantitative techniques to study this aspect of sensory function is an important addition to standard electrodiagnosis. Patient understanding and cooperation is essential because subjective responses are evaluated. We evaluated a reference range for 20 healthy subjects as well as variability on repeated testing. …

Evaluation Of The Predictive Power Of Progesterone Receptor Levels In Primary Breast Cancer: A Comparison With Other Criteria In 559 Cases With A Mean Follow-Up Of 74.8 Months, Robert A. Huseby, Helen E. Ownby, Sam Brooks, Jose Russo

A total of 559 women with primary breast cancer treated by modified radical mastectomy were followed for a mean of 74.8 months to evaluate the relationship of sex hormone receptor content in the tumor with time to first recurrence and to death due to breast cancer. The prognostic significance of progesterone receptor (PgR) status was evaluated in terms of estrogen receptor (ER) status, age (< 49 years, > 50 years), extent of lymph node involvement, tumor size, and morphologic characteristics. Overall, patients with PgR positive (> 9 femtomoles/10 mg wet weight tissue) tumors experienced a significantly longer period to both first recurrence and …
