Electrogenic Uptake Of Gamma-Aminobutyric Acid By A Cloned Transporter Expressed In Xenopus Oocytes, Michael Kavanaugh, Jeffrey L. Arriza, R. Alan North, Susan G. Amara

GAT-1, a gamma-aminobutyric acid (GABA) transporter cloned from rat brain, was expressed in Xenopus oocytes. Voltage-clamp measurements showed concentration-dependent, inward currents in response to GABA (K0.5 4.7 microM). The transport current required extracellular sodium and chloride ions; the Hill coefficient for chloride was 0.7, and that for sodium was 1.7. Correlation of current and [3H]GABA uptake measurements indicate that flux of one positive charge occurs per molecule of GABA transported. Membrane hyperpolarization from -40 to -100 mV increased the transport current approximately 3-fold. Four copies of the coding sequence for a voltage-dependent potassium channel (RBK1, rat Kv1.1) were ligated contiguously and transcribed in vitro. The resulting RNA encodes four covalently linked subunit domains ([4]RBK1). Injection of this RNA into Xenopus oocytes resulted in the expression of voltage-dependent potassium currents. A single amino acid substitution, Tyr-->Val, located within the outer mouth of the pore, introduced into the equivalent position of any of the four domains, reduced affinity for external tetraethylammonium by approximately the same amount. The widely expressed Na(+)-independent transporter for basic amino acids (system y+) is the cell surface receptor (ecoR) for ecotropic host-range mouse retroviruses (murine leukemia viruses (MuLVs)), a class of retroviruses that naturally infects only mice or rats. Accordingly, expression of mouse ecoR cDNA in mink CCL64 fibroblasts yields cells (CEN cells) that have y+ transporter activity above the endogenous background and that bind and are infected by ecotropic MuLVs. The effect of ecotropic MuLV infection on expression of y+ transporter was analyzed in mouse and in mink CEN fibroblasts. Transgenic animals will be created to study human genetic disease as soon as the technological capability exists to do so. Extant laws permit such animals to be created. The mindset of the research community makes it inevitable. It is also clear that such diseases can cause enormous amounts of pain and suffering. Responsible researchers need to explore all possible avenues for controlling such pain and suffering. Thus far the research community has not engaged this issue vis a vis animals. Small mammals were trapped for 2 years at 16 localities on the central-Argentine pampa. Six species (Akodon azarae, Calomys musculinus, C. laucha, Bolomys obscurus, Oligory-zomys flavescens and Mus musculus) accounted for >95% of captures. The major breeding season, as assessed by pregnancies, was September or October through April or May. Mild weather in late autumn and winter of the second season resulted in a relatively longer breeding season during the 2nd year of the study. Females of all six species comprised significantly 50% of captures during the height of the breeding season. For most species, there was …

Mushrooms from the genus Marasmius have the ability to revive -- viz., dried specimens regain their original shape when moistened. The purpose of this study was to determine if the basidiocarps are metabolically active upon revival or whether they simply absorb moisture like a sponge. The reduction of tetrazolium chloride to a red, water-insoluble formazan pigment was used as a measure of respiratory activity. Basidiocarp samples were incubated in a phosphate buffer containing 1% tetrasolium chloride. Murder, She Wrote Or Was It Merely Selective Nontreatment?, George P. Smith Ii
Scholarly Articles
This article will both explore and thereby establish the medical, ethical, and legal validity of selective nontreatment of severely handicapped newborns. A construct for principled decision-making, tied to a basic recognition of the right of self-determination, as shaped by compassion and validated principles of triage and cost-benefit analysis, will be seen as the most effective means for the states-and not the federal government-to evaluate the intensely complex issues associated with allocating scarce medical resources to defective infants. Governmental intrusions into the familial decision- making forum in these circumstances must be kept to a minimum and allowed only in grave cases.

Rex protein, the posttranscriptional regulator of human T-cell leukemia virus type I (HTLV-I), is required for the control of viral structural protein expression and virus replication. Rex is a phosphoprotein found predominantly in the cell nucleolus, whose function is thought to be regulated by its nucleolar localization and phosphorylation. Therefore, we investigated the in vivo phosphorylation of Rex protein in more detail. Phosphorylation of Rex occurred in all HTLV-I-infected cell lines examined in vivo, primarily at serine residues and to a very small extent at threonine residues. Previous studies suggested that teniposide is a strong clastogen, and that the DNA breakage effect of this drug is mediated by the nuclear enzyme topoisomerase II. Ripley et al found evidence for a correspondence between sites of acridine-induced frameshift mutations in bacteriophage T4 and sites of in Vitro DNA cleavage by T4 topoisomerase II. To identify the sequence specificity of teniposide-induced deletion and insertion mutations in mammalian cells, the CHO-D422 cell line, which is hemizygous at the aprt locus, was employed in this study. This study utilized a quasi-experimental pretest-posttest design that compared the effect of body position changes on arterial oxygen saturation in obesity as measured noninvasively by pulse oximetry. The null hypothesis, that there would be no difference between oxygen saturation values measured by pulse oximetry with position change from sitting to supine in obese subjects as compared to nonobese subjects, was tested. Fourteen obese, American Society of Anesthesiologist (ASA) physical class status II subjects and 14 nonobese ASA physical class status I subjects were included in the sample population. The purpose of this research was to identify the values influencing the nurse's perception and choice of behavior in a hypothetical clinical situation. The theoretical framework was Rokeach's (1973) Theory on the Nature of Human Values and Value Systems. A descriptive study using a mailed survey was conducted on a random sample of 331 members of the National Association of Neonatal Nurses. Data on individual nurses' values, perception of information and behavioral choices were collected with an investigator developed questionnaire, consisting of a values scale (alpha =.82) and an information scale and choice alternatives related to three hypothetical vignettes: a …

Over the past years, the use of epidural anesthesia as an intervention for pain relief has increased in the labor and delivery setting. Exploration of the literature has not identified what effects the addition of this technology has had on the normal childbirth process and those participating in childbirth, specifically the new father. The purpose of this study was to explore first-time fathers' perceptions of their feelings and concerns during the various phases of the labor and delivery experience in which epidural anesthesia is used.
