Efficient Vasoactive Intestinal Polypeptide Hydrolyzing Autoantibody Light Chains Selected By Phage Display, Sonia Tyutyulkova, Qing-Sheng Gao, Austin Thompson, Stephen I. Rennard, Sudhir Paul

An immunoglobulin light chain (L chain) library derived from the peripheral blood lymphocytes of a patient with asthma was cloned into a phagemid vector. Phage particles displaying L chains capable of binding vasoactive intestinal polypeptide (VIP) were isolated by affinity chromatography. Two VIP binding L chains were expressed in Escherichia coli in soluble form and purified to electrophoretic homogeneity by metal chelating and protein L affinity chromatography. Both L chains catalyzed the hydrolysis of [tyr10-125I]VIP substrate. The catalytic activity eluted at the molecular mass of the monomer form of the L chain (28 kDa) from a gel filtration column. Adolescents are becoming sexually active at increasingly younger ages despite the well-known health risks. The adolescent pregnancy rate in the United States has been considered the highest of any country in the Western world. The purpose of this study was to identify, examine, and describe family relationships, race and ethnic origin, and sexuality education in pregnant adolescents. Pender's Health Promotion Model was used as the theoretical framework to guide this descriptive study. Trypanosoma cruzi. a protozoan parasite, causes American trypanosomiasis or Chagas' disease. The infective stage of this parasite resides in the hindgut of the reduviid bug (family Reduviidae), which is both host and vector, and is transmitted to the mammalian host through fecal material released during a blood meal. Chagas' disease, an acute to chronic infection resulting in fever, malaise, and heart and liver enlargement, is becoming a concern in the United States due to the large increase in Latin American immigrants, and the lack of safe and effective therapeutic treatment and vaccination. Trypanosoma cruzi is an obligate intracellular parasite infecting 18-20 million people in Latin America. It is the causative agent of Chagas' disease, commonly referred to as American Trypanosomiasis. The C3H murine model is widely used to study experimental Chagas' disease. It is well documented that when these highly susceptible mice are infected with the Brazil strain of T. cruzi and maintained at room temperature (RT), mice die between day 35-45 of infection. However, when held at an elevated temperature of 36°C, C3H mice survive an otherwise lethal infection with T. cruzi. When held at 36°C, the body temperature of the …

An Investigation Of Hexatylis Contracta Blomquist (Southern Heartleaf): Individuals, Hybrids, Populations And Species, Patrick Carroll

Hexastylis contracta Blomquist is a rare herbaceous perennial plant endemic to southeastern North America. The plant exhibits a disjunct distribution, being found on the Cumberland Plateau of Tennessee and Kentucky and also 300 km east in the Blue Ridge Mountains of North Carolina. This plant was the subject of a population inventory supported through the Kentucky State Nature Preserves Commission (KNPC) which expanded the number of known populations of the species (28) by 68%, with 47 populations now known to exist. A morphological analysis of this species was done using principal components analysis. This analysis was used to answer questions …

Glass ionomers chemically and mechanically bond to enamel. Repeated bonding may deplete calcium in enamel and reduce the bond strength. Seventy-five non-carious molars were randomly divided into three equal groups and metal brackets were bonded with either Ketac-Cem, Photac-Bond, or Fuji Ortho LC. The samples were stored in 100% humidity at 37° C for 7 days then shear/peel tested with an Instron machine. New brackets were repeatedly cemented with the same cement to the same tooth and shear/peel tested a total of five times. Relationships Of Job And Family Involvement, Family Social Support, And Work–Family Conflict With Job And Life Satisfaction, Gary A. Adams, Lynda A. King, Daniel W. King

A model of the relationship between work and family that incorporates variables from both the work-family conflict and social support literatures was developed and empirically tested. This model related bidirectional work-family conflict, family instrumental and emotional social support, and job and family involvement to job and life satisfaction. Data came from 163 workers who were living with at least 1 family member. Results suggested that relationships between work and family can have an important effect on job and life satisfaction and that the level of involvement the worker assigns to work and family roles is associated with this relationship. Interleukin-10 (IL-10) is a recently described pleiotropic cytokine secreted mainly by type 2 helper T cells. Previous studies have shown that IL-10 suppresses cytokine expression by natural killer (NK) and type 1 T cells, thus down-regulating cell-mediated immunity and stimulating humoral responses. We here report that injected IL-10 protein is an efficient inhibitor of tumor metastasis in experimental (B16-F10) and spontaneous (M27 and Lox human melanoma) metastasis models in vivo at doses that do not have toxic effects on normal or cancer cells. Johnson's Behavioral Systems Model (1968) provided the conceptual framework. The relationship between chronic illness and sexuality has long been recognized, and diabetes mellitus is a chronic illness affecting 16 million individuals in the United States. The purpose of this descriptive comparative study was to explore sexuality in the insulin-dependent diabetic female. The research hypothesis that guided the study was there is no difference in perceived sexuality for women with insulin-dependent diabetes and women without chronic illness. Cationic liposomes may be valuable for the delivery of anti-sense oligonucleotides, ribozymes, and therapeutic genes into human immunodeficiency virus type 1 (HIV-1)-infected and uninfected cells. We evaluated the toxicity of three cationic liposomal preparations, Lipofectamine, Lipofectin, and 1,2-dimyristyloxypropyl-3-dimethyl-hydroxyethyl ammonium bromide (DMRIE) reagent, to HIV-infected and uninfected cells. Monocyte/macrophages were infected with HTV-1(BaL) and treated with liposomes in medium containing 20% fetal bovine serum (FBS) for 4 h or 24 h at 37°C. Uninfected monocytic THP-1 cells and chronically infected THP-1/HIV-1(IIIB) cells were treated with phorbol 12-myristate 13-acetate (PMA) and exposed to liposomes in the presence of 10% FBS. The deiodination of thyroid hormones in extrathyroidal tissues plays an important role in modulating thyroid hormone action. The type II deiodinase (DII) converts thyroxine to the active hormone 3,5,3'-triiodothyronine, and in the rat is expressed in the brain, pituitary gland, and brown adipose tissue (BAT). Complementary DNAs (cDNAs) for the types I and III deiodinases (DI and DIII, respectively) have been isolated and shown to code for selenoproteins. However, information concerning the structure of the mammalian DII remains limited, and the pattern of its expression in human tissues is undefined. Program admissions committees seek students who will perform the best, didactically and clinically, completing the program and demonstrating the potential to pass the professional exams. One Way To Be Born? Legislative Inaction And The Posthumous Child, Karin M. Mika

This article argues that the posthumous child and the rights and responsibilities relating to such a child are directly related to the fundamental right to procreate. It argues that legislation must necessarily incorporate that right in sorting out issues related to the posthumous child and deviate from the standard principles of contract laws which have been applied in the past. This article examines the history, case law, federal decisions, and current legislation pertaining to artificial insemination. It argues that such legislation is inadequate and that legislatures must act promptly to address the realities of the posthumous child.

Dental anxiety, a pervasive problem in the general population, is an important client management concern for dental hygienists. This study sought to determine the type and amount of instruction in United States dental hygiene programs in terms of behavioral strategies for the management of anxious clients, the difference in instruction between associate and baccalaureate degree dental hygiene programs, and the opinions of dental hygiene educators regarding these strategies. Second-Order Conditioning Detects Unexpressed Morphine-Induced Salt Aversion, Rick A. Bevins, Timothy A. Delzer, Michael T. Bardo

Morphine failed to condition a salt taste aversion at a dose (15 mg/kg) sufficient to produce a robust aversion to a saccharin taste. Indeed, three different concentrations of salt (1%, 1.5%, and 2%) paired with the same morphine dose yielded no direct evidence for conditioned aversion. Yet, when a novel saccharin taste was paired in compound with the previously conditioned salt conditioned stimulus, we found evidence for a conditioning to the saccharin cue alone in three separate experiments. Control groups eliminated alternative accounts such as neophobia and differential exposure to morphine. Combined, these findings indicate that morphine conditioned a salt …
