Characteristics Of The Recovery Of The Coenzyme A-Synthesizing Protein Complex From Saccharomyces Cerevisiae, Stanley Joseph Tarnowski Jr.

A multienzyme complex contained in Bakers' yeast (Saccharomyces cerevisiae) which synthesizes CoA has been named the coenzyme A-synthesizing protein complex (CoA-SPC). The CoA-SPC has been shown to be insoluble in the crude Bakers' yeast cell lysate formed by exposing the yeast cell to ether and dry ice. Only after solubilization has this multienzyme complex been shown to catalyze the formation of bound dephospho-CoA utilizing the substrates adenosine triphosphate, D-pantothenic acid and L-cysteine. A low molecular weight component or components of the soluble fraction of the yeast cell and chloride ion appears to be responsible for the solubilization of CoA-SPC. This …

Polymorphisms, Linkage And Mapping Of Four Enzyme Loci In The Fish Genus Xiphophorus (Poeciliidae), D C Morizot, M J Siciliano

Electrophoretic variants at four additional enzyme loci--two esterases (Est-2, Est-3), retinal lactate dehydrogenase (LDH-1) and mannose phosphate isomerase (MPI)--among three species and four subspecies of fish of the genus Xiphophorus were observed. Electrophoretic patterns in F1 hybrid heterozygotes confirmed the monomeric structures of MPI and the esterase and the tetrametric structure of LDH in these fishes. Variant alleles of all four loci displayed normal Mendelian segregation in backcross and F2 hybrids. Recombination data from backcross hybrids mapped with Haldane's mapping function indicate the four loci to be linked as Est-2--0.43--Est3--0.26--LDH-1--0.19--MPI. Cell mediated immune (CMI) cytotoxic reactivity of Balb/c mice against H238 cells, a Herpes simplex virus (HSV) Type 2 - induced sarcoma, was measured by the ¹²⁵IUdR release assay. The Balb/c mouse response to the growing HSV-induced tumor, treated with radiation and , was measured by survival rate, tumor growth and immune cytotoxicity of spleen and peritoneal exudate cells (PEC) as determined by ¹²⁵IUdR release assay. Subcutaneous (s.c.) inoculation of 1 x 10⁶ H238 cells (high dose) produced progressive tumor growth while s.c. inoculation of 1 x 10⁴ H238 cells (low dose) produced no tumors. A …

Interplanetary Magnetic Field Sector Polarity And Neuropsychiatric Adaption: A Reanalysis, Hans W. Wendt

Journal of the Minnesota Academy of Science

Recent Russian studies reported associations between adaptation syndromes and (solar activity-related) interplanetary magnetic field (IMF) sector polarity. Certain ambiguities in the original work suggested some reanalyses, based on the graphs and data made available. Syndrome status exhibited a biphasic (semi annual) trend. It also correlated with specific geomagnetic changes during magnetic storms and possibly after sudden impulses. Moreover, lag effects and higher order interactions were noted among all parameters investigated. The restudy tends to support some of the original findings but also suggests new conclusions regarding potential relationships between geophysical/solar and psychophysiological processes.

Staphylococcal protein A and several different immunoglobulins have been radio-labeled to high specific activities (> 106 cpm/pg) by reductive methylation with tritiated (3H) sodium borohydride. The proteins retain excellent functional and antigenic properties. The utility of these reagents in a variety of assays for cell surface antigens is illustrated. The results indicate that this radiolabeling procedure may become the method of choice for many cell surface and solution immunoassays.

Three Linked Enzyme Loci In Fishes: Implications In The Evolution Of Vertebrate Chromosomes, D C Morizot, D A Wright, M J Siciliano

A three-point linkage group comprised of loci coding for adenosine deaminase (ADA), glucose-6-phosphate dehydrogenase (G6PDH), and 6-phospho-gluconate dehydrogenase (6PGD) is described in fish of the genus Xiphophorus (Poeciliidae). The alleles at loci in this group were shown to assort independently from the alleles at three other loci--isocitrate dehydrogenase 1 and 2, and glyceraldehyde-3-phosphate dehydrogenase 1. Alleles at the latter three loci also assort independently from each other. Data were obtained by observing the segregation of electrophoretically variant alleles in reciprocal backcross hybrids derived from crosses between either X. helleri guentheri or X. h. strigatus and X. maculatus. The results of our personal studies of alpha-l-protease inhibitor (APi, formerly termed alpha-l-antitrypsin), the major inhibitor of protease in extracellular fluids, are presented in this dissertation. In the first chapter, we describe techniques used for APi quantitation and determination of Pi phenotypes. After a brief review of the available techniques, we report several improvements on our original technique of Pi phenotyping using thin-layer isoelectric focusing. Particular attention is given to biochemical and immunological methods for verification of the results obtained by isoelectric focusing. Three complementary techniques are reported: protease probe, crossed immunoelectrophoresis, and print immunofixation. Analysis of the results from …

An analog of ATP, 6-mercapto-9+nribofuranosylpurine 5’-triphosphate (SH-TP), functions as a phosphoryl donor for the reaction catalyzed by hexokinase. Incubation of the enzyme with millimolar concentrations of this reagent led to its rapid inactivation. However, the corresponding monophosphate derivative of this reagent was much less effective. A plot of the initial rate of inactivation versus the concentration of SH-TP exhibited saturation, suggesting the formation of a reversible complex between the enzyme and SH-TP prior to the inactivation reaction. Treatment of bovine brain mitochondrial hexokinase with a fivefold molar excess of tetranitromethane (TNM) at pH 8.0 results in a complete loss of activity of the enzyme. Spectral measurements and amino acid analysis showed that only cysteinyl residues were modified by TNM under these conditions. The products of the reaction of TNM with the enzyme were cysteic acid and disulfides as found by amino acid analysis of the carboxymethylated hexokinase. Failure of 2-mercaptoethanol to reverse the effect of TNM suggested that the thiol group that is oxidized to cysteic acid is the one essential for enzyme activity. Substrates glucose and …

A method is described for preparing an enriched fraction of the three major antibodies, immunoglobulins IgG, IgA and IgM, from normal human serum by a batch method. Selective precipitation of these proteins was carried out in successive steps using polyethylene glycol (PEG) of average molecular weight 6,000 at concentrations of 7%, 12% and 14% (16) . Zinc sulfate (10 mM) was used to remove highly immunogenic alpha₂ macroglobulin from the IgM fraction. IgA, IgM and IgG were further purified by adsorption methods using DEAE-cellulose. Quantitation and qualitation of the enriched fractions showed nearly complete separation of each of the …

Food additives are listed by functional category and discussed in terms of the scale of their use and relative risks in terms of long-term human toxicity. The greatest risks appear to lie with the antimicrobial preservative agents, and food colors (which serve only a cosmetic purpose); these and the synthetic sweeteners are discussed in relatively greater detail.

The water permeability was measured for blood cells from normal subjects and for blood cells from patients with leukemia. The method of measurement of the kinetics of water influx employed a stopped flow apparatus. The method of calculation of results was based on irreversible thermodynamics. The measurements and calculations were both performed with the assistance of a computer. The water permeability of the leukemic cells was decreased from normal, and the values were more variable than normal. Leukemia changes the structure of the cell membrane.

Stereospecific Opiate Binding In Human Erythrocyte Membranes And Changes In Heroin Addicts, Leo G. Abood, Holly G. Atkinson, Mary Macneil

Publications and Research

Stereospecific opiate binding has been demonstrated in human erythrocyte membranes, having a Kd of 9-10(-9) M. In most respects the binding characteristics resemble those of synaptic membranes. These included the correlation of binding affinity and pharmacological potency of opiates; competition by naloxone; inhibition by Ca2+ and Na+; and sensitivity to phospholipases and trypsin. A comparison of stereospecific opiate binding in control human subjects and heroin addicts revealed a 43% increase in the addict group.

The Effect Of A Pufa Diet On Human Red Blood Cell Levels Of 2, 3-Dpg, Sandra A. Dassenko

Fasting blood was collected at weekly intervals and the level of 2,3-DPG of the red blood cell was determined in 15 male subjects age 28 to 57 with serum cholesterol levels greater than 250 mg%. A lactovegetarian diet containing no cholesterol, a high level of dietary fiber and a P/S ratio of 4.0 was provided for 28 days.

During the course of the experimental period the 2,3-DPG increased by an average of 0.83> «moles/ml red blood cell which was highly significant (p^.001). This significant increase of 2,3-DPG which relates to an increased oxygen delivery capability of the red blood cells …

The LD₅₀ of Vibrio parahaemolyticus ATCC 17803 for 13- to 18-g male mice, strain ICR, as determined by the 50% endpoint method, was found to be an estimated 7 x 10⁷ CFU, when administered intraperitoneally in 0.5 ml of 24-h broth culture, O. D. 0.5 at 650 nm, diluted to 10^-0.72.

A large degree of variation in fiber tracheid length, vood specific gravity, and time of apical bud formation existed vithin two populations of Liquidambar styraciflua L. from south central Kentucky. The variation exhibited by these populations may be attributed to the self-sterility of Liquidaribar and the variable environment of south central Kentucky. Variation of fiber tracheid length and wood specific gravity within the two populations was significant. Fiber tracheid length was dependent on temperature and photoperiod while wood specific gravity was primarily dependent on temperature. A significant level of variation was also evident between the two populations regarding tracheid length. …

An in vitro study using radioactively-labeled tissue culture cells was conducted over a seven-month period on Grossman's sealer, Rickert's sealer, N₂ Permanent, and Cavit to determine the lasting and relative tissue toxicity of these materials.

From this study it was observed that all materials possess lasting tissue toxicity. When relative toxicity is considered, Grossman's is the most toxic, followed by N₂, Rickert's and Cavit in descending order of toxicity. Both Grossman's and N₂ would be considered highly toxic, with Rickert's displaying moderate toxicity. Cavit would be considered mild to moderate in toxicity.

The Relationship Of Alcohol Consumption In Rats To Morphine And Codeine Intake And The Metabolism Of Dopamine, Hsiu Fen Liu

The effects of morphine and codeine free-choice alcohol consumption in male Sprague-Dawley rats were studied. Intakes of a 10% alcohol solution in milliliters per 100 gm. of body weight were on recorded weekly. No difference was found in the alcohol intake of rats given either a morphine-supplemented diet (0.01% of the diet) or daily injection of morphine sulfate (2 mg./kg.); however, there was a significant lower intake of water (p < .05) in those rats fed the diet with morphine. Total fluid consumption of both groups given the morphine-supplemented diet and daily injection of morphine sulfate was decreased (p < .05). It was found that morphine did not affect the percentage of total fluid consumed as alcohol or the weight gain.

Two levels of codeine phosphate (10 mg./kg. and 20 mg./kg.) were injected subcutaneously into rats twice a day. It was observed that the codeine significantly decreased alcohol intake in rats …

Two series of experiments were conducted in order to determine the effects of diet on neutrophilic phagocytosis. The first series was designed to determine the effects of varying amounts of oral glucose on phagocytosis. Blood samples from 6 to 8 subjects were drawn at fasting, and at various time periods following ingestion of 12.5, 25, 50, 75, and 100 grams of glucose. In the second series of experiments, blood samples were taken from 11 subjects after an overnight fast and 2 hours following ingestion of one of 4 different liquid diets, which included the following: the meal (I), consisting of …
